Abstract:A thermal desorption low temperature plasma (TD-LTP) ionization source was developed for the direct, rapid and sensitive detection of pesticides by mass spectrometry without complex sample pretreatment. A thermal desorption sampler was added in front of the plasma generator, and the analyte was collected on a polytetrafluoroethylene (PTFE) swab and desorbed into gas molecules, and then gas molecules were transported to the plasma generator by carrier gas to be ionized. The utilization of thermal desorption sampler helped to transform the interaction between gas phase plasma and sample from gas-solid or gas-liquid to gas-gas, which greatly increased the sensitivity and stability especially for non-volatile sample (e.g. pesticides) compared with conventional LTP ionization source. The TD-LTP ionization source was coupled to a homemade rectilinear ion trap mass spectrometry. The operation parameters of the TD-LTP ionization source were optimized and the characteristic ions of 12 different pesticides were successfully obtained and investigated. Then the TD-LTP ionization source was connected with commercial ACQUITY TQD mass spectrometer, and the pesticide residue level in broomcorn was evaluated.
High Performance Liquid Chromatography Method for Residues Analysis of Thidiazuron in Apple and Soil

Abstract:A rapid, sensitive and reliable analytical method for thidiazuron residues in apple and soil was established. The residual levels of the pesticide in apple and soil were determined by high performance liquid chromatography (HPLC) with UV detector. Samples of apple and soil were extracted with acetonitrile-water solutions, and then cleaned up by Florisil or C-18 cartridges. The results showed good linearity (r(2) = 1.000) over the concentration range of 0.01-5.0 mg/L. Limits of quantification (LOQ) of the method were 0.01 mg/kg for both soil and apple. Recovery from the apple and soil samples were 83.36%-84.08% and 85.27%-89.83%, respectively, and the corresponding relative standard deviations (RSDs) of the recovery data were 0.155%-0.524% and 0.475%-4.79% for the three fortified levels (0.01, 0.1, 0.5 mg/kg). The analyte in the samples were further confirmed by electrospray ionization tandem mass spectrometry (ESI-MS/MS). It was demonstrated that the proposed method was simple and efficient, and particularly suitable for detecting thidiazuron residues in apple and soil.
Establishment of a Method for the Detection of Indaziflam, Spirotetramat, Cyantraniliprole, and Their Metabolites and Application for Fruit and Vegetable Risk Assessment

Abstract:The presence of pesticides in foodstuffs has received increasing amount of attention worldwide. In this study, an efficient and sensitive QuEChERS/HPLC-MS/MS-based method was established for the simultaneous detection of indaziflam, cyantraniliprole, spirotetramat, and their metabolites on fruits and vegetables. The purification procedure and detection condition parameters were optimized. Good precision and reproducibility were found for the method, and the average recoveries of the target analytes ranged from 71 to 118%, with the relative standard deviation ranging from 2 to 12%. The limits of quantification and the limits of detection were 1-5 and 0.3-1.5 μg kg-1, respectively. The proposed method was used to detect three pesticides and their metabolites in fruit and vegetable samples collected from China's major producing regions. Furthermore, the dietary risk posed by the pesticide residues on fruits and vegetables was evaluated by risk quotients (RQs) and risk probabilities (RPs). The RQ and RP values were less than 100% for Chinese consumers. This study not only provides a reliable analytical method for monitoring purposes but also serves as a significant guide for food safety and export.
A Novel Integrated APCI and MPT Ionization Technique As Online Sensor for Trace Pesticides Detection

Abstract:The misuse of pesticides poses a tremendous threat to human health. Excessive pesticide residues have been shown to cause many diseases. Many sensor detection methods have been developed, but most of them suffer from problems such as slow detection speed or narrow detection range. So, the development of rapid, direct and sensitive means of detecting trace amounts of pesticide residues is always necessary. A novel online sensor technique was developed for direct analysis of pesticides in complex matrices with no sample pretreatment. The portable sensor ion source consists of an MPT (microwave plasma torch) with desolventizing capability and an APCI (atmosphere pressure chemical ionization), which provides abundant precursor ions and a strong electric field. The performance which improves the ionization efficiency and suppresses the background signal was verified by using pesticide standard solution and pesticide pear juice solution measurements with an Orbitrap mass spectrometer. The limit of detection (LOD) and the limit of quantization (LOQ) of the method were measured by pear juice solutions that were obtained in the ranges of 0.034–0.79 μg/L and 0.14–1 μg/L. Quantitative curves were obtained ranging from 0.5 to 100 μg/L that showed excellent semi-quantitative ability with correlation coefficients of 0.985–0.997. The recoveries (%) of atrazine, imidacloprid, dimethoate, profenofos, chlorpyrifos, and dichlorvos were 96.6%, 112.7%, 88.1%, 85.5%, 89.2%, and 101.9% with the RSDs ranging from 5.89–14.87%, respectively. The results show that the method has excellent sensitivity and quantification capability for rapid and direct detection of trace pesticide.

Abstract:n-Octadecylamine was adopted as a cleanup agent to develop a novel solidification of a switchable solvent-based QuEChERS method. At higher temperatures (such as 55 degrees C), n-octadecylamine can melt into a liquid, allowing effective extraction of matrix interferences in acetonitrile solution (i.e., in dispersive liquid-liquid microextraction). At lower temperatures, n-octadecylamine carrying matrix interferences can rapidly solidify and easily separate from the acetonitrile solution. The results demonstrated that n-octadecylamine possessed a better ability to remove matrix interferences and reduce matrix effects than those of traditional solid-phase dispersive extraction cleanup agents of primary secondary amine and octadecyl bonded silica gel. By coupling it with gas chromatography-mass spectrometry, the proposed method was applied to the detection of 16 pesticides in cucumber. The recoveries were from 80.9 to 112.6% with relative standard deviations less than 12.9%. Satisfactory results were also obtained for the detection of 16 pesticides in pear, orange, apple, pepper, lettuce, and tomato.
Low-background interference detection of glyphosate, glufosinate, and AMPA in foods using UPLC-MS/MS without derivatization

Abstract:The abuse of herbicides has emerged as a great threat to food security. Herein, a low-background interference detection method based on UPLC-MS was developed for the simultaneous determination of glufosinate, glyphosate, and its metabolite aminomethylphosphonic acid (AMPA) in foods. Initially, this study proposed a simple and rapid pretreatment method, utilizing water extraction and PRiME HLB purification to isolate glyphosate, glufosinate, and AMPA from food samples. After the optimization of pretreatment conditions, the processed samples are then analyzed directly by ultra-performance liquid chromatography–tandem mass spectrometry (UPLC-MS) without pre-column derivatization. The method can effectively reduce interference from by-products of pre-column derivatization and background substrates of food sample, showing low matrix effects (ME) ranging from − 24.83 to 32.10%. Subsequently, the method has been validated by 13 kinds of food samples. The recoveries of the three herbicides in the food samples range from 84.2 to 115.6%. The limit of detection (LOD) is lower to 0.073 mg/kg, 0.017 mg/kg, and 0.037 mg/kg, respectively. Moreover, the method shows an excellent reproducibility with relative standard deviations (RSD) within 16.9%. Thus, the method can provide high trueness, reproducibility, sensitivity, and interference-free detection to ensure human health safety.

Simultaneous determination of triazine herbicides and their metabolites in shellfish by HPLC-MS/MS combined with Q/E-Orbitrap HRMS

Abstract:Triazine herbicides are used extensively in agriculture and aquaculture worldwide because of their broad effectiveness in weed control. However, after they are discharged into the sea, they seriously contaminate aquatic ecosystems and threaten aquatic organisms, especially shellfish. Currently, there are no established methods for the detection and confirmation of triazine herbicides and their metabolites in biological matrixes. Hence, the food safety of aquatic products cannot be accurately evaluated, which creates a technical barrier against international aquatic product trade. In this study, for the first time, a method was developed for the analysis and confirmation of seven triazine herbicides and 13 metabolites in shellfish, based on alkaline acetonitrile extraction and neutral Al2O3 cartridge purification coupled with internal standard calibration. Specifically, quantitative and qualitative analysis was conducted using high-performance liquid chromatography-triple quadrupole mass spectrometry (HPLC-MS/MS), and accurate identification was carried out by quadrupole orbitrap high-resolution mass spectrometry (Q/E Orbitrap HRMS). The results showed that target analytes demonstrated good linearity within the corresponding range (R2 > 0.995). The limit of detection and limit of quantitation of the proposed method were 0.1 and 0.3 μg/kg, respectively. The average recoveries of analytes were between 70.0% and 120% when spiked at three levels with blank oyster (Crassostrea gigas) as the matrix, and the relative standard deviations (RSDs) were all less than 12% (n=6). The proposed method was successfully applied for the detection of triazine herbicide residues in oyster samples during actual breeding, and the presence of DIP, HP, DEHA, and other metabolites in positive samples was confirmed by Q/E Orbitrap HRMS. This method exhibits high accuracy, high sensitivity, and good reproducibility. It has promising application prospects in the field of hazard analysis and the positive identification of aquatic products.

Abstract:Pesticide residues on plant surfaces pose a severe threat to food security, yet most research has focused on monitoring the liquid matrix, with few reports conducting in-situ analysis of the residues. This study was the first to attempt to utilize portable attenuated total reflectance Fourier transform infrared spectroscopy (ATR-FTIR) for in-situ characterization of broad-spectrum fungicide boscalid residues on plant surfaces. ATR-FTIR scanning of tomato fruits with pre-determined concentrations of boscalid residues was conducted without any pre-treatment, and the obtained spectra were then processed using chemometrics methods. The results demonstrated a negative correlation between the residual concentrations and their corresponding absorption intensities of several well-resolved peaks from the spectra, resulting in a high accuracy of 93.33% for the classification model created by probabilistic neural network (PNN) coupled with principal component analysis (PCA). By employing correlation analysis and the interval partial least squares method (iPLS), quantitative analysis was conducted on the wavenumber ranges of 1000–1800 cm −1 and 2700–2900 cm −1 from the spectra. The regression model, established through partial least squares regression (PLSR), demonstrated exceptional performance with an R 2 value of 0.80, RMSE of 1.02 μg/cm 2 , RPD of 2.0, and RPIQ of 2.1 for validation. Meanwhile, the detection limit (LOD) of the model was calculated as 3.06 μg/cm 2 . This report highlights the potential of using portable ATR-FTIR for conducting qualitative and quantitative monitoring of pesticide residues both in-situ and on-site. It also provides references for other measuring techniques.

Abstract:In this work, an analytical method for GC using direct solid sample introduction was developed to tackle the problem regarding quick detection of pesticide residue in crops and large-scale screening of samples. 10mg of the crop solid sample without sample pre-treatment was directly introduced into a modified split/splitless injector for GC analysis. A split/splitless injector was modified to quickly remove oxygen and low boiling-point matrices of the sample. The whole sampling procedure was simple and it required less than 5min. The experimental parameters including injector-port temperature, removal of oxygen and low boiling point matrices, size and the amount of the solid sample, oven temperature program were studied. Satisfactory recoveries of 6 pesticides (methyl parathion, fenitrothion, aldrin, dieldrin, endosulfan, o,p′-DDT) were obtained in maize and rice sample. Relative standard deviation was less than 15%. Experimental results showed that the proposed method was quick and reliable for pesticide residues analysis in crops.

Abstract:An automated online gel permeation chromatography–gas chromatograph mass spectrometer (GPC–GC/MS) was developed for the rapid determination of residual pesticides in agricultural products. Pesticides were extracted from homogenized food samples with acetonitrile and decontaminated via the matrix solid-phase dispersion (MSPD) technique, using a primary secondary amine as sorbent prior to GPC–GC/MS analysis. A slightly modified preparation method and automated GPC step proved useful in minimizing matrix interference. To evaluate the performance of the system, 97 target pesticides were spiked at a concentration of 0.1mg/kg into a range of food types, including potato, cabbage, carrot, apple, orange, cucumber, and rice. A low flow rate of 0.1mL/min in GPC resulted in a 40-fold reduction in solvent consumption compared with conventional GPC column applications. The combination of MSPD technique and GPC–GC/MS for the analysis of the 97 pesticides can be accomplished within 90min. Most pesticides were recovered in the range of 70–120%, with relative standard deviation generally less than 10%. The results demonstrate that the method can be successfully applied with acceptable recoveries to a broad range of target pesticides within a diverse range of food types.
Deep learning enabled miniature mass spectrometer for rapid qualitative and quantitative analysis of pesticides on vegetable surfaces

Abstract:Excessive pesticide use poses a significant threat to food safety. Rapid on-site detection of multi-target pesticide residues in vegetables is crucial due to their widespread distribution and limited shelf life. In this study, a rapid on-site screening method for pesticide residues on vegetable surfaces was developed by employing a miniature mass spectrometer. A direct pretreatment method involves placing vegetables and elution solution into a customized flexible ziplock bag, allowing thorough mixing, washing, and filtration. This process effectively removes pesticide residues from vegetable surfaces with minimal organic solvent usage and can be completed within 2 min. Moreover, this study introduced a deep learning algorithm based on a one-dimensional convolutional neural network, coupled with a feature database, to autonomously discriminate detection outcomes. By combining full scan MS and tandem MS analysis methods, the proposed method achieved a qualitative recognition accuracy of 99.62%. Following the qualitative discrimination stage, the target pesticide residue and internal standard can be simultaneously isolated and fragmented in the ion trap, thus enabling onsite quantitative analysis and warning. This method achieved a quantitative detection limit of 10 mu g/kg for carbendazim in cowpea. These results demonstrate the feasibility of the proposed analytical system and strategy in food safety applications.

Abstract:An analytical method for the determination of 34 pesticide residues in plant-derived foods was established using an automated QuEChERS sample preparation system combined with high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). The sample extraction and dispersive solid-phase extraction in the manual QuEChERS method were combined using the vortex vibration and centrifugation functions of the automated QuEChERS sample preparation system. The operating parameters and pretreatment steps were optimized, and the analytes were detected in the multiple reaction monitoring (MRM) mode. The quantification analysis was performed by the matrix-matched external standard method. The automated and the manual QuEChERS methods were compared from the methodological verification standpoint. The calibration curves showed good linearity in a certain range, and the correlation coefficients (R2) were greater than 0.99. The limits of detection and limits of quantification were 0.76-3.60 μg/kg and 2.28-10.80 μg/kg, respectively. Moreover, the recoveries ranged from 53.0% to 125.2%, and the relative standard deviations (RSDs) were less than 1.59% (n=5). The results obtained by the manual QuEChERS method were not significantly different from those obtained by the automated QuEChERS method. This method can effectively reduce the labor intensity and probability of error in the determination of pesticide residues.

Abstract:Chlorpyrifos (CPF) and 2,4-dichlorophenoxyacetic acid (2,4-D) are insecticides and herbicides which has been widely used on farms. However, CPF and 2,4-D residues on corps can bring high risks to human health. Accurate detection of pesticide residues is important for controlling health risks caused by CPF and 2,4-D. Therefore, we developed a fast, sensitive, economical, and lossless surface-enhanced Raman spectroscopy (SERS)-based method for pesticide detection. It can rapidly and simultaneously determine the CPF and 2,4-D mixed pesticide residues on an apple surface at a minimum of 0.001 mg L−1 concentration, which is far below the pesticide residue standard in China and the EU. The limits of detection reach down to 1.28 × 10−9 mol L−1 for CPF and 2.47 × 10−10 mol L−1 for 2,4-D. The limits of quantification are 4.27 × 10−9 mol L−1 and 8.23 × 10−10 mol L−1 for CPF and 2,4-D. This method has a great potential for the accurate detection of pesticide residues, and may be applied to other fields of agricultural products and food industry.

Abstract:A rapid and accurate integrated QuEChERS method was established for the determination of multi-pesticide residues in fruits. Poly-dopamine-modified magnetic nanomaterial (Fe 3 O 4 -pDA) was homemade and characterized. The prepared Fe 3 O 4 -pDA has the functional group of absorbing the saccharides, and can be used as co-adsorbent with 3-(N, n-diethyl amino) propyl trimethoxy-silane (PSA) in the developed integrated QuEChERS method to purify the fruit matrix, thus achieve the accurate determination of multi-pesticides residue. Grape was used as the representative sample to explore the influence of the salting out agent and each purification adsorbent on the pesticide recoveries. Under the optimized conditions, the proposed method showed good linearity for 92.6 % of pesticides in the concentration range of 1-150 μg L −1 with method limit of quantitative (mLOQs) ranged from 10 to 18 μg kg −1 . Spiked recoveries experiments were performed on four kinds of grapes and other fruits (apple, watermelon, pear, jujube and peach), in which satisfactory recoveries and precision were obtained for most of the pesticides. Meanwhile, comparison experiments also verified this method was superior to the traditional QuEChERS method in terms of convenient operation, high efficiency and low reagent consumption. The further real sample analysis was performed using this method, and the overall detection rate was 52 %, while 2 % of samples were exceeding the maximum residue limits. All results confirmed that the proposed method could be used for the rapid, simple, low-costing and effective analyses of trace multi-pesticides residue in fruit samples.

Homer conferencing - A multimedia test bed for various experiments and measurements

Abstract:Transferring multimedia data from and to customers is one of the biggest challenges of current network engineering. A key challenge, researchers are confronted with, is to understand what kind of real-time support is needed, what the network could provide to the application and how the application could utilize codecs, quality settings and transmission protocols in order to assure the service. In this paper we introduce the open source software Homer-Conferencing (HC) as multimedia test bed for research purposes. Our approach was developed during the last four years. The main goal of our work is to allow researchers implementing deep analysis of multiple kinds of multimedia traffic. For this purpose, exchangeable codecs and adjustable transmission qualities can be used in a video conferencing application. In this paper, we describe the architecture of HC and highlight based on selected experiments the benefits the HC system provides for experiments and measurements in currently used networks as well as future Internet platforms.

Abstract:Residues of plant growth regulators (PGRs) in homologous materials of medicine and food threaten public health. This study aimed to develop a rapid, sensitive, and high-throughput method for simultaneously determining 16 PGR residues in homologous materials of medicine and food. Furthermore, the established method was applied to actual samples to assess the potential exposure risk of multi-PGR residues. A modified high-throughput quick, easy, cheap, effective, rugged, and safe (QuEChERS) method coupled with ultra-high-performance liquid chromatography-tandem mass spectrometry (UPLC–MS/MS) was developed and validated. The extraction solvent, type of extraction method, and subsequent purification techniques were investigated to achieve a better analysis of the target. Risk assessment was based on chronic dietary risk assessment. Ultrasonic extraction with 1% formic acid–acetonitrile was employed, and MgSO 4 + NaAc was selected as the clean-up sorbent. The 16 PGRs showed a good linear relationship in the range of 1 ~ 200 μg/L ( r ≥ 0.9960), with detection limits ranging from 0.3 to approximately 3 μg/kg. The recovery rate ranged from 65 to 109%, with RSD from 0.01 to 10% ( n = 6). The total detection rate of 16 PGRs in the samples was 87%. The risk assessment indicated that the multi-residues of PGRs in homologous materials of medicine and food would not pose a potential risk to human health. This work provides a valuable reference for the monitoring of multiple PGRs. It has also improved our understanding of the possible exposure risk of PGR residues in homologous materials of medicine and food.

Abstract:Rationale: Chlorothalonil (CHT), a broad-spectrum fungicide, has been employed widely to control foliar diseases, whereas with a major metabolite of polar 4-hydroxychlorothalonil (CHT-4-OH), only an acceptable nonpolar CHT residue is allowed by most countries. This study involves the method development for CHT residue in vegetables/fruits using liquid chromatography-tandem mass spectrometry (LC-MS/MS) with a novel modified discharge-adaptor (DA) interface. Methods: CHT residue was analyzed using LC-MS/MS with DA interface (LC-DA-MS/MS), developed in our previous works. A DA was placed on the electrospray tip to switch the ionization modes. A modified quick, easy, cheap, effective, rugged, and safe (QuEChERS) method was applied to extract CHT residue of vegetables/fruits efficiently with less sample preparation time and analysis cost. Results: CHT and CHT-4-OH spiked in four different vegetables/fruits were extracted using the modified QuEChERS method. After LC with isocratic elution, CHT and CHT-4-OH were separated within 3 min. Using LC-DA-MS/MS, the ion signals of CHT were improved two to three times, and the limit of quantification of 5 ng/g and linearity (r2 > 0.99) in the range of 5-200 ng/g were achieved using 10 g of vegetables/fruits. The precision and accuracy were within 15% each. The modified QuEChERS and LC-DA-MS/MS were applied to examine eight field-grown vegetables/fruits; 9.5 and 2588.9 ng/g of CHT were detected in two vegetables/fruits. Conclusion: LC-DA-MS/MS combined with modified QuEChERS was successfully applied to determine CHT residue <10 ng/g in vegetables/fruits and with satisfied validation results. The developed method could reduce both analysis cost and time, attributing to simplifications in modified QuEChERS, isocratic elution, and DA interface in LC-DA-MS/MS.

Abstract:Pesticides residues in foodstuffs are longstanding of great concern to consumers and governments, thus reliable evaluation techniques for these residues are necessary to ensure food safety. Emerging ambient ionization mass spectrometry (AIMS), a transformative technology in the field of analytical chemistry, is becoming a promising and solid evaluation technology due to its advantages of direct, real-time and in-situ ionization on samples without complex pretreatments. To provide useful guidance on the evaluation techniques in the field of food safety, we offered a comprehensive review on the AIMS technology and introduced their novel applications for the analysis of residual pesticides in foodstuffs under different testing scenarios (i.e., quantitative, screening, imaging, high-throughput detection and rapid on-site analysis). Meanwhile, the creative combination of AIMS with high-resolution mass analyzer (e.g., orbitrap and time-of-flight) was fundamentally mentioned based on recent studies about the detection and evaluation of multi-residual pesticides between 2015 and 2021. Finally, the technical challenges and prospects associated with AIMS operation in food industry were discussed.

Multiresidue Analysis of Pesticides in Straw Roughage by Liquid Chromatography-Tandem Mass Spectrometry

Abstract:A multiresidue analytical method using a modification of the "quick, easy, cheap, effective, rugged, and safe" (QuEChERS) sample preparation approach combined with liquid chromatography-tandem mass spectrometry (LC-MS/MS) analysis was established and validated for the rapid determination of 69 pesticides at different levels (1-100 ng/g) in wheat and rice straws. In the quantitative analysis, the recoveries ranged from 70 to 120%, and consistent RSDs ≤ 20% were achieved for most of the target analytes (53 pesticides in wheat straw and 58 in rice straw). Almost all of the analytes achieved good linearity with R(2) > 0.98, and the limit of validation levels (LVLs) for diverse pesticides ranged from 1 to 10 ng/g. Different extraction and cleanup conditions were evaluated in both types of straw, leading to different options. The use of 0.1% formic acid or not in extraction with acetonitrile yielded similar final outcomes, but led to the use of a different sorbent in dispersive solid-phase extraction. Both options are efficient and useful for the multiresidue analysis of targeted pesticides in wheat and rice straw samples.

Abstract:A new analytical method for gas chromatography (GC) or GC-mass spectrometry (MS) using the direct sampling technique is described. This direct sampling technique, which bypasses the conventional complicated sample pretreatment process, is applicable to cases of fast detection of pesticide residues in foods and large-scale screening of samples by portable GC in field detection. By a direct sampling technique, the vegetable sample is ground into paste, and 30 mg is placed directly into the evaporating chamber for GC-MS identification and quantitation (by full-scan mode). The GC column used is an HP-5 (30.0-m x 250-microm x 0.25-microm, 5% phenyl methyl siloxane). Chlorpyrifos, bromophos, fenpropathrin, gamma-666, and pp'-DDT are chosen to represent organophosphorus, pyrethrins, and organochlorine pesticides because they are chief objects of the detection of pesticide residues in vegetables. Rape, a common and mass-consumed vegetable in China, is chosen as the sample in this study. The detection limits for these pesticides by the full-scan mode are all below the maximum pesticide residue limit of vegetables set by the Ministry of Agriculture of China, and the reproducibility of this method is acceptable. This analysis method is proven to be simple, quick, and reliable and is suitable for multipesticide residues analysis of vegetables. It can also be used in the analysis of vegetable components and signal chemicals.
