Abstract:Florida faces the challenge of repeated introduction and autochthonous transmission of arboviruses transmitted by Aedes aegypti and Aedes albopictus. Empirically-based predictive models of the spatial distribution of these species would aid surveillance and vector control efforts. To predict the occurrence and abundance of these species, we fit a mixed-effects zero-inflated negative binomial regression to a mosquito surveillance dataset with records from more than 200,000 trap days, representative of 53% of the land area and ranging from 2004 to 2018 in Florida. We found an asymmetrical competitive interaction between adult populations of Aedes aegypti and Aedes albopictus for the sampled sites. Wind speed was negatively associated with the occurrence and abundance of both vectors. Our model predictions show high accuracy (72.9% to 94.5%) in validation tests leaving out a random 10% subset of sites and data since 2017, suggesting a potential for predicting the distribution of the two Aedes vectors.

Abstract:In this paper, a periodic Chikungunya model with temperature and rainfall effects is proposed and studied, which incorporates time-dependent extrinsic incubation period, time-dependent maturation delay, asymptomatic and symptomatic infectious humans. Two threshold parameters for the extinction and persistence of mosquitos and the virus are derived, respectively: the mosquito reproduction number <math>Rm</math> and the basic reproduction number <math>R0</math>. Then the analytic results are verified by numerical simulation with the temperature and rainfall data of the state of Ceará, where the largest outbreak in Brazil's history occurred in 2017. And the effects of rainfall, seasonality, and asymptomatic infection in humans on mosquito population and the Chikungunya transmission are explored. Our simulations show that if these factors are not taken into account, the number of mosquito population and people infected may be overestimated. Finally, the relationships between <math>Rm</math> and <math>R0</math> and some parameters are established.

Abstract:Chikungunya virus (CHIKV) is a vector-borne alphavirus transmitted by the bites of mosquitoes, specifically infected, female mosquitoes of the invasive Aedes species. In nature, CHIKV can be maintained by vertical transmission, a phenomenon that relates to the transfer of CHIKV from the infected parent to their offspring within the ovary or during oviposition. In the present study, we conducted laboratory experiments to determine vertical transmission with Ae. albopictus populations from Brazil and Florida. Parental Ae. albopictus females were orally infected with the emergent Asian genotype of CHIKV in the first gonotrophic cycle (infectious blood meal) and tested for vertical transmission following the second (non-infectious blood meal) gonotrophic cycle. CHIKV infection and CHIKV viral titer in parental females were significantly related to population origin, with Brazilian Ae. albopictus showing higher viral dissemination and viral titer than the Florida population. Experimental vertical transmission of CHIKV was documented in one pool of female and four pools of male Ae. albopictus from Brazil (minimum infection rate, MIR, of 0.76% and 2.86%, respectively, for females and males). For the Florida population of Ae. albopictus, only one pool of males was positive for CHIKV infection, with an MIR of 1.06%. Our results demonstrate that Ae. albopictus populations from Brazil and Florida show heterogeneous CHIKV dissemination and vertical transmission, which may contribute to the epidemiology of CHIKV and may be particularly relevant to virus survival during inter-epidemic periods.

Abstract:Dengue is an infectious disease that is transmitted by the Aedes aegypti mosquito. Each stage of the life cycle is influenced by climate variation. The transmission of the dengue virus can be related to increased mosquito survival due to rain and temperature conditions that are optimal for the mosquito’s maturation. The aim of this paper is to propose a mathematical model to study how temperature influences each stage of the mosquito’s life cycle dynamics by representing transitions and death rates as an explicit function of temperature. The model is thus able to show the influence of temperature on dengue transmission. It can also be used as an operational tool due to its simplicity regarding data requirements and computational effort. The model demonstrates that an expected increase in global temperature will influence the mosquito’s life cycle and, consequently, increase the incidence of dengue cases in areas that were previously free from the disease.

Abstract:Arboviruses can emerge rapidly and cause explosive epidemics of severe disease. Some of the most epidemiologically important arboviruses, including dengue virus (DENV), Zika virus (ZIKV), Chikungunya (CHIKV) and yellow fever virus (YFV), are transmitted by Aedes mosquitoes, most notably Aedes aegypti and Aedes albopictus . After a mosquito blood feeds on an infected host, virus enters the midgut and infects the midgut epithelium. The virus must then overcome a series of barriers before reaching the mosquito saliva and being transmitted to a new host. The virus must escape from the midgut (known as the midgut escape barrier; MEB), which is thought to be mediated by transient changes in the permeability of the midgut-surrounding basal lamina layer (BL) following blood feeding. Here, we present a mathematical model of the within-mosquito population dynamics of DENV (as a model system for mosquito-borne viruses more generally) that includes the interaction of the midgut and BL which can account for the MEB. Our results indicate a dose-dependency of midgut establishment of infection as well as rate of escape from the midgut: collectively, these suggest that the extrinsic incubation period (EIP)—the time taken for DENV virus to be transmissible after infection—is shortened when mosquitoes imbibe more virus. Additionally, our experimental data indicate that multiple blood feeding events, which more closely mimic mosquito-feeding behavior in the wild, can hasten the course of infections, and our model predicts that this effect is sensitive to the amount of virus imbibed. Our model indicates that mutations to the virus which impact its replication rate in the midgut could lead to even shorter EIPs when double-feeding occurs. Mechanistic models of within-vector viral infection dynamics provide a quantitative understanding of infection dynamics and could be used to evaluate novel interventions that target the mosquito stages of the infection. Aedes mosquitoes are the main vectors of dengue virus (DENV), Zika virus (ZIKV) and yellow fever virus (YFV), all of which can cause severe disease in humans with dengue alone infecting an estimated 100–400 million people each year. Understanding the processes that affect whether, and at which rate, mosquitoes may transmit such viruses is, hence, paramount. Here, we present a mathematical model of virus dynamics within infected mosquitoes. By combining the model with novel experimental data, we show that the course of infection is sensitive to the initial dose of virus ingested by the mosquito. The data also indicate that mosquitoes which blood feed subsequent to becoming infected may be able to transmit infection earlier, which is reproduced in the model. This is important as many mosquito species feed multiple times during their lifespan, and any reduction in time to dissemination will increase the number of days that a mosquito is infectious and so enhance the risk of transmission. Our study highlights the key and complementary roles played by mathematical models and experimental data for understanding within-mosquito virus dynamics.

Abstract:Aedes Aegypti is the vector of several deadly diseases, including Zika. Effective and sustainable vector control measures must be deployed to keep A. aegypti numbers under control. The distribution of A. Aegypti is subject to spatial and climatic constraints. Using agent-based modeling, we model the population dynamics of A. aegypti subjected to the spatial and climatic constraints of a neighborhood in the Key West. Satellite imagery was used to identify vegetation, houses (CO2 zones) both critical to the mosquito lifecycle. The model replicates the seasonal fluctuation of adult population sampled through field studies and approximates the population at a high of 986 (95% CI: [979, 993]) females and 1031 (95% CI: [1024, 1039]) males in the fall and a low of 316 (95% CI: [313, 319]) females and 333 (95% CI: [330, 336]) males during the winter. We then simulate two biological vector control strategies: 1) Wolbachia infection and 2) Release of Insects carrying a Dominant Lethal gene (RIDL). Our results support the probability of sustained Wolbachia infection within the population for two years after the year of release. egies, our approach provides a realistic simulation environment consisting of male and female Aedes aegypti, breeding spots, vegetation and CO2 sources.

Abstract:BACKGROUND: Estimates of the spatiotemporal distribution of different mosquito vector species and the associated risk of transmission of arboviruses are key to design adequate policies for preventing local outbreaks and reducing the number of human infections in endemic areas. In this study, we quantified the abundance of Aedes albopictus and Aedes aegypti and the local transmission potential for three arboviral infections at an unprecedented spatiotemporal resolution in areas where no entomological surveillance is available.METHODS: We developed a computational model to quantify the daily abundance of Aedes mosquitoes, leveraging temperature and precipitation records. The model was calibrated on mosquito surveillance data collected in 115 locations in Europe and the Americas between 2007 and 2018. Model estimates were used to quantify the reproduction number of dengue virus, Zika virus, and chikungunya in Europe and the Americas, at a high spatial resolution.FINDINGS: In areas colonised by both Aedes species, A aegypti was estimated to be the main vector for the transmission of dengue virus, Zika virus, and chikungunya, being associated with a higher estimate of R0 when compared with A albopictus. Our estimates highlighted that these arboviruses were endemic in tropical and subtropical countries, with the highest risks of transmission found in central America, Venezuela, Colombia, and central-east Brazil. A non-negligible potential risk of transmission was also estimated for Florida, Texas, and Arizona (USA). Modeling the Impact of Seasonality on Mosquito Population Dynamics: Insights for Vector Control Strategies.

Abstract:Mosquitoes are important vectors for the transmission of some major infectious diseases of humans, i.e., malaria, dengue, West Nile Virus and Zika virus. The burden of these diseases is different for different regions, being highest in tropical and subtropical areas, which have high annual rainfall, warm temperatures, and less pronounced seasonality. The life cycle of mosquitoes consists of four distinct stages: eggs, larvae, pupae, and adults. These life stages have different mortality rates and only adults can reproduce. Seasonal weather may affect the population dynamics of mosquitoes, and the relative abundance of different mosquito stages. We developed a stage-structured model that considers laboratory experiments describing how temperature and rainfall affects the reproduction, maturation and survival of different Anopheles mosquito stages, the species that transmits the parasite that causes malaria. We consider seasonal temperature and rainfall patterns and describe the stage-structured population dynamics of the Anopheles mosquito in Ain Mahbel, Algeria, Cape Town, South Africa, Nairobi, Kenya and Kumasi, Ghana. We find that neglecting seasonality leads to significant overestimation or underestimation of mosquito abundance. We find that depending on the region, mosquito abundance: peaks one, two or four times a year, periods of low abundance are predicted to occur for durations ranging from six months (Ain Mahbel) to not at all (Nairobi); and seasonal patterns of relative abundance of stages are substantially different. The region with warmer temperatures and higher rainfall across the year, Kumasi, Ghana, is predicted to have higher mosquito abundance, which is broadly consistent with reported malaria deaths relative to the other countries considered by our study. Our analysis reveals distinct patterns in mosquito abundance across different months and regions. Control strategies often target one specific life stage, for example, applying larvicides to kill mosquito larvae, or spraying insecticides to kill adult mosquitoes. Our findings suggest that differences in seasonal weather affect mosquito stage structure, and that the best approaches to vector control may differ between regions in timing, duration, and efficacy.

Autochthonous Chikungunya Transmission and Extreme Climate Events in Southern France

Abstract:Background: Extreme precipitation events are increasing as a result of ongoing global warming, but controversy surrounds the relationship between flooding and mosquito-borne diseases. A common view among the scientific community and public health officers is that heavy rainfalls have a flushing effect on breeding sites, which negatively affects vector populations, thereby diminishing disease transmission. During 2014 in Montpellier, France, there were at least 11 autochthonous cases of chikungunya caused by the invasive tiger mosquito Aedes albopictus in the vicinity of an imported case. We show that an extreme rainfall event increased and extended the abundance of the disease vector Ae. albopictus, hence the period of autochthonous transmission of chikungunya. Methodology/principal findings: We report results from close monitoring of the adult and egg population of the chikungunya vector Ae. albopictus through weekly sampling over the entire mosquito breeding season, which revealed an unexpected pattern. Statistical analysis of the seasonal dynamics of female abundance in relation to climatic factors showed that these relationships changed after the heavy rainfall event. Before the inundations, accumulated temperatures are the most important variable predicting Ae. albopictus seasonal dynamics. However, after the inundations, accumulated rainfall over the 4 weeks prior to capture predicts the seasonal dynamics of this species and extension of the transmission period. Conclusions/significance: Our empirical data suggests that heavy rainfall events did increase the risk of arbovirus transmission in Southern France in 2014 by favouring a rapid rise in abundance of vector mosquitoes. Further studies should now confirm these results in different ecological contexts, so that the impact of global change and extreme climatic events on mosquito population dynamics and the risk of disease transmission can be adequately understood.
Disclosing temperature sensitivity of West Nile virus transmission: novel computational approaches to mosquito-pathogen trait responses

Abstract:Temperature influences the transmission of mosquito-borne pathogens with significant implications for disease risk in the context of climate change. Mathematical models of mosquito-borne infections rely on functions that capture mosquito-pathogen interactions in response to temperature to better estimate transmission dynamics. To derive these functions, experimental studies provide valuable data on the temperature sensitivity of mosquito life-history traits and pathogen transmission. However, the scarcity of experimental data and inconsistencies in methodologies for analysing and comparing temperature responses across mosquito species and pathogens present challenges to accurately modelling mosquito-borne infections. In this study, we investigated the thermal biology of West Nile virus (WNV), a major mosquito-borne infection. We critically reviewed existing experimental studies, obtaining temperature responses for eight mosquito-pathogen traits across 15 mosquito species. Using this data, we employed Bayesian hierarchical models to estimate temperature response functions for each trait and estimate their variation between species and experiments. We incorporate the resulting functions into mathematical models to estimate the temperature sensitivity of WNV transmission, focusing on six competent mosquito species of the genus Culex. Our study finds similarities in the temperature response among Culex species, with a general optimal transmission temperature around 24°C. We demonstrate that differing mechanistic assumptions in published models can result in a temperature optimum variation exceeding 3°C, underscoring the need for model scrutiny. Additionally, we identify substantial variability between trait temperature responses across experiments on the same mosquito species, indicating significant intra-species variation in trait performance. We provide recommendations for future experimental studies, emphasizing the critical need for additional data on all mosquito-pathogen traits, except for mosquito larva-to-adult development rate and survival, which are relatively well-documented. Incorporating additional data into our multi-species approach enhances the accuracy of temperature trait response estimates. These improvements are critical for forecasting future shifts in mosquito-borne disease risk due to climate change.

Abstract:Many infectious diseases are deadly to humans. The Aedes aegypi mosquito is the principal vector of infectious diseases that include chikungunya, dengue, yellow fever, and zika. Some factors such as survival time and aging are vital in its development and capacity to transmit the pathogens, which in turn are affected by environmental factors such as temperature. In this paper, we consider aging as the biological wear and tear presented in some mosquito populations over time, whereas survival is considered as the maximum time that a mosquito lives. We propose statistical methods that are commonly used in engineering for reliability analysis to compare transmission riskiness among different mosquitoes. We conducted a case study in three Colombian cities: Bello, Riohacha, and Villavicencio. In this study, we detected that the Aedes aegypi female mosquitoes in Bello live longer than in Riohacha and Villavicencio, and the females in Riohacha live longer than those in Villavicencio. Regarding aging, the females from Riohacha age slower than in Villavicencio and the latter age slower than in Bello. Mosquito populations that age slower are considered young and the other ones are old. In addition, we detected that the females from Bello in the temperature range of 27 ∘C–28 ∘C age slower than those in Bello at higher temperatures. In general, a young female has a higher risk of transmitting a disease to humans than an old female, regardless of its survival time. These findings have not been previously reported in studies of this type of infectious diseases and contributed to new knowledge in biomedicine.

Abstract:Use has been made of the mechanism of DNA deoxyribose damage by the ene-diyne-containing chromophore of the antitumor antibiotic neocarzinostatin to provide chemical evidence for the structure of the activated drug-DNA complex. Radical centers at C-2 and C-6 of the diradical form of the glutathione-activated chromophore abstract hydrogen atoms from C-1' of the C residue and C-5' of the T residue in AGC.GCT to generate a bistranded lesion consisting of an abasic site at C and a strand break at T. This laboratory has proposed a molecular model for the drug-DNA interaction in which the naphthoate moiety of the chromophore intercalates between A.T and G.C, placing the diradical core in the minor groove, so that the radical centers at C-6 and C-2 are close to C-5' of T and C-1' of C, respectively. To determine which radical center abstracts one of the hydrogen atoms from C-5', the self-complementary oligodeoxynucleotide GCAGCGCTGC was synthesized with 2H at both 5' positions of the T residue and treated with glutathione-activated chromophore. Sequencing-gel electrophoresis showed that drug attack was limited to the T and C residues and that abstraction of 2H from C-5' exhibited a small isotope selection effect of 1.25. 1H NMR spectroscopic examination of the reacted chromophore, isolated by HPLC, indicated that 2H was selectively abstracted by C-6, providing experimental corroboration of the model and further elucidating the chemical mechanism. Since direct strand breakage at the T residue exceeds (44% more) abasic site formation at the C residue, other models of drug-DNA interaction leading to only single-strand breaks are also considered.
Impact of venereal transmission on the dynamics of vertically transmitted viral diseases among mosquitoes

Abstract:Despite centuries of enormous control efforts, mosquito-borne diseases continue to show upward trend of morbidity. According to WHO reports, malaria caused 438000 deaths in the year 2015 and dengue cases have been increased 30-fold over the last five decades. To control these diseases, it is necessary to understand the transmission dynamics of them among mosquitoes. There are some vertically transmitted mosquito-borne diseases which can also be spread among mosquitoes through sexual contact (e.g., dengue, zika, chikungunya). Recent experimental observations indicate that for virus persistence in mosquito population, the role of venereal transmission cannot be ignored. It is therefore important to investigate which transmission route is more responsible for the persistence of the virus when there is no host. To this aim, we propose and analyze a novel compartmental model considering mosquito population only. To the best of authors knowledge, this is the first attempt to take into account both vertical and sexual transmission of the virus in a mathematical model. Expression representing the basic reproduction number is derived using Jacobian approach. Local stability conditions for disease-free equilibrium and complete infection equilibrium are obtained. Global sensitivity analysis of the system is performed with respect to an epidemiologically important response. While investigating the impact of sexual transmission in presence of vertical transmission, we observed that sexual transmission route has the potential to drive the equilibrium from disease free to endemic states. Further numerical experiments reveal that the virus will have higher half life in fertilized infected female mosquitoes for vertical transmission only than for venereal transmission alone. Furthermore, when both transmission pathways are active, a variety of parameters indicate threshold like behavior of the infection.
Seasonal insights for integrative mosquito management from multi-year baseline entomological data on Aedes aegypti in Lee County, Florida

Abstract:The spread of arboviruses like yellow fever, dengue, chikungunya, and Zika, transmitted by the invasive mosquito Aedes aegypti has led to the development of many strategies to suppress mosquito populations. Given the rapid development of resistance to common chemical larvicides and adulticides in some Ae . aegypti populations, as well as the ever-shrinking chemical options for mosquito control, there is a pressing need for new tools and deployment of those innovative tools as a component of integrative mosquito management programs. Prior to the adoption of any mosquito population intervention, be it conventional or innovative, understanding the baseline population is essential to evaluate the efficacy of the control measure. The Lee County Mosquito Control District in Florida has collected a three-year-long period of baseline entomological surveillance data collection for Ae . aegypti on Captiva and Sanibel Islands as foundational information prior to implementation of a new integrative mosquito management approach. We identified 18 mosquito species and described their population dynamics during the rainy and dry seasons. The two islands had no significant differences in species richness, diversity, dominance, or evenness overall. Yet, there were clear differences between the high rain season and low rain season in the Shannon diversity index, Simpson dominance index, and Pielou species evenness index within each site. Our data suggest that any innovative intervention should begin before mid to late April when the mosquito population is at its lowest and certainly before populations build up to their summer peak between June and September. These data also show the spatial distribution of Ae . aegypti is dynamic in space and time, identifying hotspots of mosquito abundance to focus on for future interventions. Overall, our study emphasizes the importance of entomological data collection to understand the population dynamics of Ae . aegypti mosquitoes, including the impact of environmental factors such as temperature and precipitation.

Abstract:Vectors of Dengue, Chikungunya, Zika, and Yellow Fever are emerging in new areas, posing increasing public health risks. Aedes albopictus, a key vector for these diseases, is expanding its range beyond its tropical and subtropical origins, driven by suitable climate, population mobility, trade, and urbanization. Since its introduction to Europe, Ae. albopictus has rapidly spread and triggered recurrent outbreaks. Past model attempts have handled vector suitability and vector introduction as independent drivers. Here we develop a novel, highly predictive spatio-temporal vector diffusion model based on minimum temperature, median temperature, relative humidity and human population as predictors. The model predicts areas of presence or absence with an accuracy of 99% and 79% for new established vector populations. The model explains how short- and long-range spread of Ae. albopictus interacts with vector suitability. These results show that the expansion of Ae. albopictus in Europe is predictable and closely linked to climate suitability, and human population. The new model integrates in one simultaneous model framework the climate and mobility drivers, providing a better basis for anticipating future outbreaks in situations of dependent interacting co-drivers.
