Abstract:Even as the incidence of mosquito-borne diseases like West Nile Virus (WNV) in North America has risen over the past decade, effectively modelling mosquito population density or, the abundance has proven to be a persistent challenge. It is critical to capture the fluctuations in mosquito abundance across seasons in order to forecast the varying risk of disease transmission from one year to the next. We develop a process-based mechanistic weather-driven Ordinary Differential Equation (ODE) model to study the population biology of both aqueous and terrestrial stages of mosquito population. The progression of mosquito lifecycle through these stages is influenced by different factors, including temperature, daylight hours, intra-species competition and the availability of aquatic habitats. Weather-driven parameters are utilised in our work, are a combination of laboratory research and literature data. In our model, we include precipitation data as a substitute for evaluating additional mortality in the mosquito population. We compute the \textit{Basic offspring number} of the associated model and perform sensitivity analysis. Finally, we employ our model to assess the effectiveness of various adulticides strategies to predict the reduction in mosquito population. This enhancement in modelling of mosquito abundance can be instrumental in guiding interventions aimed at reducing mosquito populations and mitigating mosquito-borne diseases such as the WNV.

Abstract:The invasion of vector-borne diseases depends on the type of specific features of the vector and hosts at play. Within the Culex pipiens complex, differences in ecology, biology, and vector competence can influence the risk of West Nile virus (WNV) outbreaks. To determine which life-history traits affect WNV invasion into susceptible communities the most, we constructed an epidemiological Susceptible-Exposed-Infectious-Recovered model with three vector (eco)types, Culex pipiens pipiens, Cx. pip. molestus, and their hybrids, and two vertebrate hosts, birds (as amplifying hosts) and humans (as dead-end hosts). We investigated how differences in feeding preferences and transmission rates influenced WNV transmission across different habitats and two seasons (Spring versus Summer), to investigate the impact of increasing mosquitoes on the WNV transmission risk. Our results showed that vector feeding preferences and the transmission rate between mosquitoes and birds were the parameters that most influenced WNV invasion risk. Even though our model did not predict WNV invasion across any of the studied environments, we found that natural habitats displayed the highest susceptibility to WNV invasion. Pipiens (eco)type acted as the primary vector in all habitats. Hybrids, contrary to common opinion, showed minimal involvement in WNV transmission. However, it is important to interpret our study results with caution due to the possibility of idealized spring and summer seasons being reflected in the field-collected data. Our study could be a tool to enhance current vector surveillance and control programs by targeting specific vector types in specific environments, especially in natural habitat, which are most responsive to environmental shifts. The joint approach based on epidemiological modelling based on field collected data can help to reduce wasted time and economic costs while maximizing the efficiency of local public health authorities.

Abstract:West Nile virus (WNv) is a mosquito-borne infection, causing health challenges to bird and human populations. We propose a general nonlocal reaction-diffusion model to explore the combined effect of alternative hosts, vector-host movement and spatial–temporal environmental structure on WNv transmission dynamics. The model is partially degenerate and its solution maps lack compactness due to the absence of diffusion terms in larval equations. For this tightly coupled disease model, we introduce the mosquito reproduction number R0V$${\mathcal {R}}_0^V$$ and WNv reproduction number R0$${\mathcal {R}}_0$$, respectively, and further show that these two aforementioned reproduction numbers completely determine the threshold behavior of the disease spread. Numerical simulations support our analytic results and suggest that environmental heterogeneity plays an important role in shaping WNv dynamics. Moreover, bird migration may increase the disease risk.

Abstract:In Canada, the periodic circulation of West Nile Virus (WNV) is difficult to predict and, beyond climatic factors, appears to be related to the migratory movements of infected birds from the southern United States. This hypothesis has not yet been explored in a spatially distributed model. The main objective of this work was to develop a spatially explicit dynamic model for the transmission of WNV in Canada, that allows us to explore non-climate related hypotheses associated with WNV transmission. A Cellular Automata (CA) approach for multiple hosts (birds and humans) is used for a test region in eastern Ontario, Canada. The tool is designed to explore the role of host and vector spatial heterogeneity, host migration, and vector feeding preferences. We developed a spatialized compartmental SEIRDS-SEI model for WNV transmission with a study region divided into 4 km2 rectangular cells. We used 2010–2021 bird data from the eBird project and 2010–2019 mosquito data collected by Ontario Public Health to mimic bird and mosquito seasonal variation. We considered heterogeneous bird densities (high and low suitability areas) and homogeneous mosquito and human densities. In high suitability areas for birds, we identified 5 entry points for WNV-infected birds. We compared our simulations with pools of WNV-infected field collected mosquitoes. Simulations and sensitivity analyses were performed using MATLAB software. The results showed good correspondence between simulated and observed epidemics, supporting the validity of our model assumptions and calibration. Sensitivity analysis showed that a 5% increase or decrease in each parameter of our model except for the biting rate of bird by mosquito ( c(B,M)) and mosquito natural mortality rate ( dM ), had a very limited effect on the total number of cases (newly infected birds and humans), prevalence peak, or date of occurrence. We demonstrate the utility of the CA approach for studying WNV transmission in a heterogeneous landscape with multiple hosts.

Abstract:West Nile Virus (WNV) is a mosquito-borne pathogen that primarily infects birds. Infections can spillover to humans and cause a spectrum of clinical symptoms, including WNV neuroinvasive disease. The extrinsic incubation period (EIP) is the time taken for a mosquito to become infectious following the ingestion of an infected blood meal. Characterising how the EIP varies with temperature is an essential part of predicting the impact and transmission dynamics of WNV. We re-analyse existing experimental data using Bayesian time delay models, allowing us to account for variation in how quickly individual mosquitoes become infectious with WNV. In these experiments, cohorts of Culex pipiens mosquitoes were infected with WNV and kept under different temperature conditions, being checked for disseminated infection at defined timepoints. We find that EIPs are best described with a Weibull distribution and become shorter log-linearly with temperature. Under 18oC, less than 1% of infected Cx. pipiens had a disseminated infection after 5 days, compared to 9.73% (95% CrI: 7.97 to 11.54) at 25oC and 42.20% (95% CrI: 38.32 to 46.60) at 30oC. In the hottest experimental temperature treatment (32oC), the EIP 50 was estimated at 3.78 days (CrI: 3.42 to 4.15) compared to over 100 days in the coolest treatment (15oC). The variance of EIPs was found to be much larger at lower temperatures than higher temperatures, highlighting the importance of characterising the time delay distribution associated with the EIP. We additionally demonstrate a competitive advantage of WNV strain WN02 over NY99, where the former infects mosquitoes more quickly at colder temperature than the latter. This research contributes crucial parameters to the WNV literature, providing essential insights for modellers and those planning interventions.

Abstract:Temperature influences the transmission of mosquito-borne pathogens with significant implications for disease risk in the context of climate change. Mathematical models of mosquito-borne infections rely on functions that capture mosquito-pathogen interactions in response to temperature to better estimate transmission dynamics. To derive these functions, experimental studies provide valuable data on the temperature sensitivity of mosquito life-history traits and pathogen transmission. However, the scarcity of experimental data and inconsistencies in methodologies for analysing and comparing temperature responses across mosquito species and pathogens present challenges to accurately modelling mosquito-borne infections. In this study, we investigated the thermal biology of West Nile virus (WNV), a major mosquito-borne infection. We critically reviewed existing experimental studies, obtaining temperature responses for eight mosquito-pathogen traits across 15 mosquito species. Using this data, we employed Bayesian hierarchical models to estimate temperature response functions for each trait and estimate their variation between species and experiments. We incorporate the resulting functions into mathematical models to estimate the temperature sensitivity of WNV transmission, focusing on six competent mosquito species of the genus Culex. Our study finds similarities in the temperature response among Culex species, with a general optimal transmission temperature around 24°C. We demonstrate that differing mechanistic assumptions in published models can result in a temperature optimum variation exceeding 3°C, underscoring the need for model scrutiny. Additionally, we identify substantial variability between trait temperature responses across experiments on the same mosquito species, indicating significant intra-species variation in trait performance. We provide recommendations for future experimental studies, emphasizing the critical need for additional data on all mosquito-pathogen traits, except for mosquito larva-to-adult development rate and survival, which are relatively well-documented. Incorporating additional data into our multi-species approach enhances the accuracy of temperature trait response estimates. These improvements are critical for forecasting future shifts in mosquito-borne disease risk due to climate change.

A hierarchical network approach for modeling Rift Valley fever epidemics with applications in North America

Abstract:Rift Valley fever is a vector-borne zoonotic disease which causes high morbidity and mortality in livestock. In the event Rift Valley fever virus is introduced to the United States or other non-endemic areas, understanding the potential patterns of spread and the areas at risk based on disease vectors and hosts will be vital for developing mitigation strategies. Presented here is a general network-based mathematical model of Rift Valley fever. Given a lack of empirical data on disease vector species and their vector competence, this discrete time epidemic model uses stochastic parameters following several PERT distributions to model the dynamic interactions between hosts and likely North American mosquito vectors in dispersed geographic areas. Spatial effects and climate factors are also addressed in the model. The model is applied to a large directed asymmetric network of 3,621 nodes based on actual farms to examine a hypothetical introduction to some counties of Texas, an important ranching area in the United States of America (U.S.A.). The nodes of the networks represent livestock farms, livestock markets, and feedlots, and the links represent cattle movements and mosquito diffusion between different nodes. Cattle and mosquito (Aedes and Culex) populations are treated with different contact networks to assess virus propagation. Rift Valley fever virus spread is assessed under various initial infection conditions (infected mosquito eggs, adults or cattle). A surprising trend is fewer initial infectious organisms result in a longer delay before a larger and more prolonged outbreak. The delay is likely caused by a lack of herd immunity while the infections expands geographically before becoming an epidemic involving many dispersed farms and animals almost simultaneously.

Abstract:Mathematical models within the Ross–Macdonald framework increasingly play a role in our understanding of vector-borne disease dynamics and as tools for assessing scenarios to respond to emerging threats. These threats are typically characterized by a high degree of heterogeneity, introducing a range of possible complexities in models and challenges to maintain the link with empirical evidence. We systematically identified and analysed a total of 77 published papers presenting compartmental West Nile virus (WNV) models that use parameter values derived from empirical studies. Using a set of 15 criteria, we measured the dissimilarity compared with the Ross–Macdonald framework. We also retrieved the purpose and type of models and traced the empirical sources of their parameters. Our review highlights the increasing refinements in WNV models. Models for prediction included the highest number of refinements. We found uneven distributions of refinements and of evidence for parameter values. We identified several challenges in parametrizing such increasingly complex models. For parameters common to most models, we also synthesize the empirical evidence for their values and ranges. The study highlights the potential to improve the quality of WNV models and their applicability for policy by establishing closer collaboration between mathematical modelling and empirical work.

Abstract:West Nile virus (WNV) is the most significant arbovirus in the United States in terms of both morbidity and mortality. West Nile exists in a complex transmission cycle between avian hosts and the arthropod vector, Culex spp. mosquitoes. Human spillover events occur when humans are bitten by an infected mosquito and predicting these rates of infection and therefore the risk to humans may be associated with fluctuations in environmental conditions. In this study, we evaluate the hydrological and meteorological drivers associated with mosquito biology and viral development to determine if these associations can be used to forecast seasonal mosquito infection rates with WNV in the Coachella Valley of California. We developed and tested a spatially resolved ensemble forecast model of the WNV mosquito infection rate in the Coachella Valley using 17 years of mosquito surveillance data and North American Land Data Assimilation System-2 environmental data. Our multi-model inference system indicated that the combination of a cooler and dryer winter, followed by a wetter and warmer spring, and a cooler than normal summer was most predictive of the prevalence of West Nile positive mosquitoes in the Coachella Valley. The ability to make accurate early season predictions of West Nile risk has the potential to allow local abatement districts and public health entities to implement early season interventions such as targeted adulticiding and public health messaging before human transmission occurs. Such early and targeted interventions could better mitigate the risk of WNV to humans.

Abstract:We study the disease-spreading dynamics of the West Nile virus (WNv) epidemic model under shifting climatic conditions. A WNv epidemic model is developed incorporating a shifting net growth term to depict the evolving mosquito habitat. First, we comprehensively characterize the spreading dynamics of mosquitoes for any given climate change speed compared with the intrinsic spreading speed of mosquitoes. Utilizing the results from mosquito dynamics, we determine the spreading dynamics of infected birds and mosquitoes, taking into account relationships among the shifting speed and the spreading speeds of mosquito and WNv. Ultimately, we find that infected mosquitoes and birds propagate, and their population densities converge to a stable positive endemic state. This paper provides crucial insights into the impact of climate change on the spread of vector-borne diseases such as WNv.

Abstract:West Nile virus is the most common mosquito borne disease in North America and the leading cause of viral encephalitis. West Nile virus is primarily transmitted between birds and mosquitoes while humans are incidental, dead-end hosts. Climate change may increase the risk of human infections as climatic variables have been shown to affect the mosquito life cycle, biting rate, incubation period of the disease in mosquitoes, and bird migration patterns. We develop a zero-inflated Poisson model to investigate how human West Nile virus case counts vary with respect to mosquito abundance and infection rates, bird abundance, and other environmental covariates. We use a Bayesian paradigm to fit our model to data from 2010–2019 in Ontario, Canada. Our results show mosquito infection rate, temperature, precipitation, and crow abundance are positively correlated with human cases while NDVI and robin abundance are negatively correlated with human cases. We find the inclusion of spatial random effects allows for more accurate predictions, particularly in years where cases are higher. Our model is able to accurately predict the magnitude and timing of yearly West Nile virus outbreaks and could be a valuable tool for public health officials to implement prevention strategies to mitigate these outbreaks.

Spatiotemporally Explicit Epidemic Model for West Nile Virus Outbreak in Germany: An Inversely Calibrated Approach

Abstract:Since the first autochthonous transmission of West Nile Virus was detected in Germany (WNV) in 2018, it has become endemic in several parts of the country and is continuing to spread due to the attainment of a suitable environment for vector occurrence and pathogen transmission. Increasing temperature associated with a changing climate has been identified as a potential driver of mosquito-borne disease in temperate regions. This scenario justifies the need for the development of a spatially and temporarily explicit model that describes the dynamics of WNV transmission in Germany. In this study, we developed a process-based mechanistic epidemic model driven by environmental and epidemiological data. Functional traits of mosquitoes and birds of interest were used to parameterize our compartmental model appropriately. Air temperature, precipitation, and relative humidity were the key climatic forcings used to replicate the fundamental niche responsible for supporting mosquito population and infection transmission risks in the study area. An inverse calibration method was used to optimize our parameter selection. Our model was able to generate spatially and temporally explicit basic reproductive number (R0) maps showing dynamics of the WNV occurrences across Germany, which was strongly associated with the deviation from daily means of climatic forcings, signaling the impact of a changing climate in vector-borne disease dynamics. Epidemiological data for human infections sourced from Robert Koch Institute and animal cases collected from the Animal Diseases Information System (TSIS) of the Friedrich-Loeffler-Institute were used to validate model-simulated transmission rates. From our results, it was evident that West Nile Virus is likely to spread towards the western parts of Germany with the rapid attainment of environmental suitability for vector mosquitoes and amplifying host birds, especially short-distance migratory birds. Locations with high risk of WNV outbreak (Baden-Württemberg, Bavaria, Berlin, Brandenburg, Hamburg, North Rhine-Westphalia, Rhineland-Palatinate, Saarland, Saxony-Anhalt and Saxony) were shown on R0 maps. This study presents a path for developing an early warning system for vector-borne diseases driven by climate change.

Abstract:In this paper, a reaction-diffusion system is proposed to model the spatial spreading of West Nile virus in vector mosquitoes and host birds in North America. Infection dynamics are based on a simplified model for cross infection between mosquitoes and birds, and the free boundary is introduced to model and explore the expanding front of the infective region. The spatial-temporal risk index $R_0^F(t)$, which involves time and characters of the region, is defined for the simplified model with the free boundary to compare with other related threshold values, including the usual basic reproduction number $R_0$. Sufficient conditions for the virus to vanish or spread are given. Our results suggest that the virus will be in a scenario of vanishing if $R_0\leq 1$, and the virus will spread to the whole region if $R_{0}^F(t_0)\geq 1$ for some $t_0\geq 0$, while if $R^F_0(0)<1<R_0$, the spreading or vanishing of the virus depends on the initial numbers of infected mosquitoes and birds, the area of the infected region and diffusion rates. Moreover, some remarks on the basic reproduction numbers and the spreading speed are presented and compared.

Abstract:Wolbachia is possibly the most studied reproductive parasite of arthropod species. It appears to be a promising candidate for biocontrol of some mosquito borne diseases. We begin by developing a sex-structured model for a Wolbachia infected mosquito population. Our model incorporates the key effects of Wolbachia infection including cytoplasmic incompatibility and male killing. We also allow the possibility of reduced reproductive output, incomplete maternal transmission, and different mortality rates for uninfected/infected male/female individuals. We study the existence and local stability of equilibria, including the biologically relevant and interesting boundary equilibria. For some biologically relevant parameter regimes there may be multiple coexistence steady states including, very importantly, a coexistence steady state in which Wolbachia infected individuals dominate. We also extend the model to incorporate West Nile virus (WNv) dynamics, using an SEI modelling approach. Recent evidence suggests that a particular strain of Wolbachia infection significantly reduces WNv replication in Aedes aegypti. We model this via increased time spent in the WNv-exposed compartment for Wolbachia infected female mosquitoes. A basic reproduction number $R_0$ is computed for the WNv infection. Our results suggest that, if the mosquito population consists mainly of Wolbachia infected individuals, WNv eradication is likely if WNv replication in Wolbachia infected individuals is sufficiently reduced.

Abstract:The endocytosis and degradation of 125I-labeled anti-mu monoclonal antibody DA4-4 by a Burkitt's lymphoma cell line was investigated using biochemical, chromatographic, electrophoretic, radioautographic, and electron microscopic techniques. 125I-DA4-4 was rapidly internalized by Ramos cells and routed from endosomes to lysosomes. Proteolysis of radiolabeled antibodies began in a late endosomal compartment, but lysosomes were primarily responsible for the terminal degradation of 125I-DA4-4. Catabolism of 125I-DA4-4 could be inhibited by 74-95% by blocking its delivery to late endosomes and lysosomes by incubation at 18 degrees C, by neutralizing the pH in intracellular organelles with monensin or ammonium chloride, or by inhibiting lysosomal enzymes with leupeptin. Radiolabeled antibodies synthesized using the chloramine T or Iodo-Gen techniques were degraded three times faster than conjugates made using a nonmetabolizable 125I-tyramine cellobiose adduct. Five major intermediate metabolites (Mr 48,000, 42,000, 25,000, 15,000, and 10,000) were generated during the intracellular catabolism of 125I-DA4-4, but 125I-tyrosine was responsible for 95% of the small-molecular-weight metabolites released by cells into the culture medium. We anticipate that a full comprehension of the catabolism of radiolabeled antibodies by tumor cells will make possible the development of clinical interventions which will enhance the retention of radioimmunoconjugates by hematologic malignancies and improve the efficacy of radioimmunotherapy.
Remote sensing of temperature‐dependent mosquito and viral traits predicts field surveillance‐based disease risk

Abstract:Mosquito‐borne diseases contribute substantially to the global burden of disease, and are strongly influenced by environmental conditions. Ongoing and rapid environmental change necessitates improved understanding of the response of mosquito‐borne diseases to environmental factors like temperature, and novel approaches to mapping and monitoring risk. Recent development of trait‐based mechanistic models has improved understanding of the temperature dependence of transmission, but model predictions remain challenging to validate in the field. Using West Nile virus (WNV) as a case study, we illustrate the use of a novel remote sensing‐based approach to mapping temperature‐dependent mosquito and viral traits at high spatial resolution and across the diurnal cycle. We validate the approach using mosquito and WNV surveillance data controlling for other key factors in the ecology of WNV, finding strong agreement between temperature‐dependent traits and field‐based metrics of risk. Moreover, we find that WNV infection rate in mosquitos exhibits a unimodal relationship with temperature, peaking at ~24.6–25.2°C, in the middle of the 95% credible interval of optimal temperature for transmission of WNV predicted by trait‐based mechanistic models. This study represents one of the highest resolution validations of trait‐based model predictions, and illustrates the utility of a novel remote sensing approach to predicting mosquito‐borne disease risk.

Abstract:We propose and analyze an epidemiological model for vector borne diseases that integrates a multi-stage vector population and several host sub-populations which may be characterized by a variety of compartmental model types: subpopulations all include Susceptible and Infected compartments, but may or may not include Exposed and/or Recovered compartments. The model was originally designed to evaluate the effectiveness of various prophylactic measures in malaria-endemic areas, but can be applied as well to other vector-borne diseases. This model is expressed as a system of several differential equations, where the number of equations depends on the particular assumptions of the model. We compute the basic reproduction number $\mathcal R_0$, and show that if $\mathcal R_0\leqslant 1$, the disease free equilibrium (DFE) is globally asymptotically stable (GAS) on the nonnegative orthant. If $\mathcal R_0>1$, the system admits a unique endemic equilibrium (EE) that is GAS. We analyze the sensitivity of $R_0$ and the EE to different system parameters, and based on this analysis we discuss the relative effectiveness of different control measures.

Abstract:Mathematical models play a crucial role in assisting public health authorities in timely disease control decision-making. For vector-borne diseases, integrating host and vector dynamics into models can be highly complex, particularly due to limited data availability, making system validation challenging. In this study, two compartmental models akin to the SIR type were developed to characterize vector-borne infectious disease dynamics. Motivated by dengue fever epidemiology, the models varied in their treatment of vector dynamics, one with implicit vector dynamics and the other explicitly modeling mosquito-host contact. Both considered temporary immunity after primary infection and disease enhancement in secondary infection, analogous to the temporary cross-immunity and the Antibody-dependent enhancement biological processes observed in dengue epidemiology. Qualitative analysis using bifurcation theory and numerical experiments revealed that the immunity period and disease enhancement outweighed the impact of explicit vector dynamics. Both models demonstrated similar bifurcation structures, indicating that explicit vector dynamics are only justified when assessing the effects of vector control methods. Otherwise, the extra equations are irrelevant, as both systems display similar dynamics scenarios. The study underscores the importance of using simple models for mathematical analysis, initiating crucial discussions among the modeling community in vector-borne diseases.
