Abstract:Mosquito-borne diseases constitute a significant global impact on public and animal health. Climatic variables are recognized as major drivers in the mosquitoes' life history, principally rainfall and temperature, which directly influence mosquito abundance. Likewise, urbanization changes environmental conditions, and understanding how environmental variables and urbanization influence mosquito dynamics is crucial for the integrated management of mosquito-borne diseases, especially in the context of climate change. In this study, our aim was to observe the effect of temperature, rainfall, and the percentage of impervious surface on the abundance of mosquito species over a temporal scale of one complete year of fortnightly samplings, spanning from June 2021 to June 2022 in Yucatan, Mexico. We selected nine localities along an urbanization gradient (three natural, three rural, and three urban) from Mérida City to Reserva de la Biosfera Ría Celestún. Using BG-traps, mosquitoes were collected biweekly at each locality. Additionally, we estimated the percentage of impervious surface. Daily data of the maximum, mean and minimum temperatures, diurnal temperature range and rainfall were accumulated weekly. We calculated the accumulated quantities of temperatures and rainfall and lagged from one to four weeks before sampling for each locality. Generalized linear mixed models were then performed to study the influence of environmental variables and percentage of impervious surfaces on each of the 15 most abundant species. A total of 131,525 mosquitoes belonging to 11 genera and 49 species were sampled with BG-Sentinel traps baited with BG-lure and dry ice. The most frequently significative variable is the accumulated precipitation four weeks before the sampling. We observed a positive relationship between Cx. quinquefasciatus and Cx. thriambus with the diurnal temperature range. For Ae. aegypti , we observed a positive relationship with minimum temperature. Conversely, the percentage of impervious surface serves as a proxy of anthropogenic influence and helped us to distinguishing species exhibiting habitat preference for urban and rural environments, versus those preferring natural habitats. Our results characterize the species-specific effects of environmental variables (temperature, rainfall and impervious surface) on mosquito abundance.

Modeling the Impact of Seasonality on Mosquito Population Dynamics: Insights for Vector Control Strategies.

Abstract:Mosquitoes are important vectors for the transmission of some major infectious diseases of humans, i.e., malaria, dengue, West Nile Virus and Zika virus. The burden of these diseases is different for different regions, being highest in tropical and subtropical areas, which have high annual rainfall, warm temperatures, and less pronounced seasonality. The life cycle of mosquitoes consists of four distinct stages: eggs, larvae, pupae, and adults. These life stages have different mortality rates and only adults can reproduce. Seasonal weather may affect the population dynamics of mosquitoes, and the relative abundance of different mosquito stages. We developed a stage-structured model that considers laboratory experiments describing how temperature and rainfall affects the reproduction, maturation and survival of different Anopheles mosquito stages, the species that transmits the parasite that causes malaria. We consider seasonal temperature and rainfall patterns and describe the stage-structured population dynamics of the Anopheles mosquito in Ain Mahbel, Algeria, Cape Town, South Africa, Nairobi, Kenya and Kumasi, Ghana. We find that neglecting seasonality leads to significant overestimation or underestimation of mosquito abundance. We find that depending on the region, mosquito abundance: peaks one, two or four times a year, periods of low abundance are predicted to occur for durations ranging from six months (Ain Mahbel) to not at all (Nairobi); and seasonal patterns of relative abundance of stages are substantially different. The region with warmer temperatures and higher rainfall across the year, Kumasi, Ghana, is predicted to have higher mosquito abundance, which is broadly consistent with reported malaria deaths relative to the other countries considered by our study. Our analysis reveals distinct patterns in mosquito abundance across different months and regions. Control strategies often target one specific life stage, for example, applying larvicides to kill mosquito larvae, or spraying insecticides to kill adult mosquitoes. Our findings suggest that differences in seasonal weather affect mosquito stage structure, and that the best approaches to vector control may differ between regions in timing, duration, and efficacy.

Abstract:Understanding how variation in key abiotic and biotic factors interact at spatial scales relevant for mosquito fitness and population dynamics is crucial for predicting current and future mosquito distributions and abundances, and the transmission potential for human pathogens. However, studies investigating the effects of environmental variation on mosquito traits have investigated environmental factors in isolation or in laboratory experiments that examine constant environmental conditions that often do not occur in the field. To address these limitations, we conducted a semi-field experiment in Athens, Georgia using the invasive Asian tiger mosquito ( ). We selected nine sites that spanned natural variation in impervious surface and vegetation cover to explore effects of the microclimate (temperature and humidity) on mosquitoes. On these sites, we manipulated conspecific larval density at each site. We repeated the experiment in the summer and fall. We then evaluated the effects of land cover, larval density, and time of season, as well as interactive effects, on the mean proportion of females emerging, juvenile development time, size upon emergence, and predicted per capita population growth (i.e., fitness). We found significant effects of larval density, land cover, and season on all response variables. Of most note, we saw strong interactive effects of season and intra-specific density on each response variable, including a non-intuitive decrease in development time with increasing intra-specific competition in the fall. Our study demonstrates that ignoring the interaction between variation in biotic and abiotic variables could reduce the accuracy and precision of models used to predict mosquito population and pathogen transmission dynamics, especially those inferring dynamics at finer-spatial scales across which transmission and control occur.

Abstract:Over 50% of the world population is at risk of mosquito-borne diseases. Female Ae. aegypti mosquito species transmit Zika, Dengue, and Chikungunya. The spread of these diseases correlate positively with the vector population, and this population depends on biotic and abiotic environmental factors including temperature, vegetation condition, humidity and precipitation. To combat virus outbreaks, information about vector population is required. To this aim, Earth observation (EO) data provide fast, efficient and economically viable means to estimate environmental features of interest. In this work, we present a temporal distribution model for adult female Ae. aegypti mosquitoes based on the joint use of the Normalized Difference Vegetation Index, the Normalized Difference Water Index, the Land Surface Temperature (both at day and night time), along with the precipitation information, extracted from EO data. The model was applied separately to data obtained during three different vector control and field data collection condition regimes, and used to explain the differences in environmental variable contributions across these regimes. To this aim, a random forest (RF) regression technique and its nonlinear features importance ranking based on mean decrease impurity (MDI) were implemented. To prove the robustness of the proposed model, we compared it to other machine learning techniques and statistical models. Our results show that machine learning techniques perform better than linear statistical models for the task at hand, and RF performs best. By ranking the importance of all features based on MDI in RF and selecting the subset comprising the most informative ones, a more parsimonious but equally effective and explainable model can be obtained...

Focal segmental glomerulosclerosis: a morphologic diagnosis in evolution.

Abstract:CONTEXT The diagnosis of focal segmental glomerulosclerosis (FSGS) is a descriptive pathologic diagnosis that in certain clinical situations (ie, primary or idiopathic) becomes its own disease. The clinical diversity, varied histology, and nonspecific morphologic features of FSGS all contribute to the complexity and problematic nature in making a pathologic diagnosis of FSGS. The definitions of the disease and of the morphologic features have evolved during the last century. OBJECTIVE To review historic and morphologic features of FSGS in order to demonstrate a practical approach in achieving a pathologic diagnosis of FSGS on kidney tissue. DATA SOURCES In 2004 a working proposal on the pathologic (morphologic) classification of FSGS was published in an attempt to unify the complexity of diagnosing FSGS, and it has shown to be both reproducible and with unique clinical implications for each defined FSGS variant. CONCLUSIONS An accurate diagnosis of FSGS can be challenging. During the last few decades, numerous new scientific discoveries have enriched our knowledge of pathogenetic mechanisms of nephrotic syndrome. Thus, it is expected there will be a need for a further modification to a morphologic classification and that the pathologist's role in diagnosing FSGS will remain in evolution. This review recapitulates the history of the pathologic diagnosis of FSGS and a current morphologic classification, hopefully opening up a discussion for further modifications that reflect the status of knowledge evolving in the 21st century.

Abstract:Phenology of adult host-seeking female mosquitoes is a critical component for understanding potential for vector-borne pathogen maintenance and amplification in the natural environment. Despite this importance, long-term multi-species investigations of mosquito phenologies across environments and differing species' life history traits are rare. Here we leverage long-term mosquito control district monitoring data to characterize annual phenologies of 7 host-seeking female mosquito species over a 20-year time period in suburban Illinois, USA. We also assembled data on landscape context, categorized into low and medium development, climate variables including precipitation, temperature and humidity, and key life history traits, i.e. overwintering stage and Spring–Summer versus Summer–mid-Fallseason fliers. We then fit linear mixed models separately for adult onset, peak abundances, and flight termination with landscape, climate and trait variables as predictors with species as a random effect. Model results supported some expectations, including warmer spring temperatures leading to earlier onset, warmer temperatures and lower humidity leading to earlier peak abundances, and warmer and wetter fall conditions leading to later termination. However, we also found sometimes complex interactions and responses contrary to our predictions. For example, temperature had generally weak support on its own, impacting onset and peak abundance timing; rather temperature has interacting effects with humidity or precipitation. We also found higher spring precipitation, especially in low development contexts, generally delayed adult onset, counter to expectations. These results emphasize the need to consider how traits, landscape and climatic factors all interact to determine mosquito phenology, when planning management strategies for vector control and public health protection.

Abstract:Understanding drivers of disease vectors' population dynamics is a pressing challenge. For short-lived organisms like mosquitoes, landscape-scale models must account for their highly local and rapid life cycles. Aedes aegypti , a vector of multiple emerging diseases, has become abundant in desert population centers where water from precipitation could be a limiting factor. To explain this apparent paradox, we examined Ae. aegypti abundances at > 660 trapping locations per year for 3 years in the urbanized Maricopa County (metropolitan Phoenix), Arizona, USA. We created daily precipitation layers from weather station data using a kriging algorithm, and connected localized daily precipitation to numbers of mosquitoes trapped at each location on subsequent days. Precipitation events occurring in either of two critical developmental periods for mosquitoes were correlated to suppressed subsequent adult female presence and abundance. LASSO models supported these analyses for female presence but not abundance. Precipitation may explain 72% of Ae. aegypti presence and 90% of abundance, with anthropogenic water sources supporting mosquitoes during long, precipitation-free periods. The method of using kriging and weather station data may be generally applicable to the study of various ecological processes and patterns, and lead to insights into microclimates associated with a variety of organisms' life cycles.

Abstract:The spread of arboviruses like yellow fever, dengue, chikungunya, and Zika, transmitted by the invasive mosquito Aedes aegypti has led to the development of many strategies to suppress mosquito populations. Given the rapid development of resistance to common chemical larvicides and adulticides in some Ae . aegypti populations, as well as the ever-shrinking chemical options for mosquito control, there is a pressing need for new tools and deployment of those innovative tools as a component of integrative mosquito management programs. Prior to the adoption of any mosquito population intervention, be it conventional or innovative, understanding the baseline population is essential to evaluate the efficacy of the control measure. The Lee County Mosquito Control District in Florida has collected a three-year-long period of baseline entomological surveillance data collection for Ae . aegypti on Captiva and Sanibel Islands as foundational information prior to implementation of a new integrative mosquito management approach. We identified 18 mosquito species and described their population dynamics during the rainy and dry seasons. The two islands had no significant differences in species richness, diversity, dominance, or evenness overall. Yet, there were clear differences between the high rain season and low rain season in the Shannon diversity index, Simpson dominance index, and Pielou species evenness index within each site. Our data suggest that any innovative intervention should begin before mid to late April when the mosquito population is at its lowest and certainly before populations build up to their summer peak between June and September. These data also show the spatial distribution of Ae . aegypti is dynamic in space and time, identifying hotspots of mosquito abundance to focus on for future interventions. Overall, our study emphasizes the importance of entomological data collection to understand the population dynamics of Ae . aegypti mosquitoes, including the impact of environmental factors such as temperature and precipitation.

Abstract:The invasive Asian tiger mosquito Aedes albopictus is nowadays broadly distributed with established populations in all continents except Antarctica. In the invaded areas, this species represents an important nuisance for humans and, more relevant, it is involved in the local transmission of pathogens relevant under a public health perspective. Aedes albopictus is a competent vector of parasites such as Dirofilaria and viruses including dengue virus, Zika virus, and chikungunya virus, among others. The mosquito microbiota has been identified as one of the major drivers of vector competence, acting upon relevant vector functions as development or immunity. Here, we review the available literature on the interaction between Ae. albopictus microbiota and pathogen transmission and identify the knowledge gaps on the topic. Most studies are strictly focused on the interplay between pathogens and Wolbachia endosymbiont while studies screening whole microbiota are still scarce but increasing in recent years, supported on Next-generation sequencing tools. Most experimental trials use lab-reared mosquitoes or cell lines, exploring the molecular mechanisms of the microbiota-pathogen interaction. Yet, correlational studies on wild populations are underrepresented. Consequently, we still lack sufficient evidence to reveal whether the microbiota of introduced populations of Ae. albopictus differ from those of native populations, or how microbiota is shaped by different environmental and anthropic factors, but especially, how these changes affect the ability of Ae. albopictus to transmit pathogens and favor the occurrence of outbreaks in the colonized areas. Finally, we propose future research directions on this research topic.
Present and future suitability of invasive and urban vectors through an environmentally-driven Mosquito Reproduction Number

Abstract:Temperature and water availability significantly influence mosquito population dynamics. We've devised a method, integrating experimental data with insights from mosquito and thermal biology, to calculate the basic reproduction number (RM) for urban mosquito species, Ae. albopictus and Ae. aegypti. RM represents the number of female mosquitoes produced by one female during her lifespan, indicating suitability for growth. Environmental conditions, including temperature, rainfall and human density influence RM> by altering key mosquito life cycle traits. Validation using data from Spain and Europe confirms the approach's reliability. Our analysis suggests that temperature increases may not uniformly benefit Ae. albopictus proliferation but could boost Ae. aegypti expansion. We suggest using vector RM maps, leveraging climate and environmental data, to predict areas susceptible to invasive mosquito population growth. These maps aid in resource allocation for intervention strategies, supporting effective vector surveillance and management efforts.

Abstract:Recent outbreaks of the West Nile virus have been reported in southern Spain, a region with important wetland habitats for migratory birds. Here, we analyzed the role of species association and abiotic parameters on the abundance of seven mosquito species in the Doñana National Park, Spain. We applied the Joint Species Distribution Models (JSDM), using the Hierarchical Modeling of Species Communities approach to simultaneously model the effect of habitat type, normalized difference vegetation index, hydroperiod, distance to rivers, land surface temperature, and the abundance of seven mosquito species. We created three models with varied parameters and evaluated the effects of abiotic parameters and species-to-species associations, which served as proxies for species interactions. Our models estimated the species-specific responses of the mosquito community to abiotic parameters. A positive association was identified within the Culex community with potential biotic interactions. However, Ochlerotatus caspius, Ochlerotatus detritus, and Anopheles atroparvus had no statistically significant association with each other and with the Culex species after accounting for the effect of abiotic parameters. We also found that the potential species associations estimated at particular sites and abiotic parameters influenced the model projection for the average abundance of mosquito species. The JSDM will allow spatial projection of the abundance of each mosquito species, which is an important parameter for epidemiological models. The JSDM inference of species association is important because predation, competition, and facilitation affect the distribution and abundance of different species.

Abstract:The vectorial capacity of mosquitoes, which influences the dynamics of vector-borne disease transmission, is intricately linked to mosquito abundance and the composition and diversity of their associated microbiomes. However, the influence of environmental factors on mosquito populations and microbiome diversity remains underexplored. Here we examined the effects of seasonality and developed land cover on Culex mosquito abundance and bacterial diversity. Biweekly field sampling of female Culex mosquitoes was conducted using gravid and CDC light traps, spanning summer to autumn across varying developed land cover levels in two urban areas in Central Illinois. Mosquito abundance was assessed by the number of mosquitoes captured per trap night and compared across seasons and developed levels. The mean mosquito abundance for gravid and light traps was 12.96 ± 2.15 and 7.67 ± 1.44, respectively. Notably, higher levels of developed land cover exhibited higher Culex abundance than the low level for light traps, but no significant difference was found between summer and early autumn. In gravid traps, no significant differences were detected across seasons or developed levels. Microbial analysis of the mosquito microbiome revealed that Proteobacteria and Wolbachia , with a mean relative abundance of 80.77 and 52.66% respectively, were identified as the most dominant bacterial phylum and genus. Their relative abundance remained consistent across seasons and developed land cover levels, with negligible variations. Alpha diversity, as measured by observed species, Chao1, Shannon, and Simpson, showed slightly higher values in early-autumn compared to late-summer. A notable pattern of bacterial diversity, as indicated by all four diversity indexes, is evident across varying levels of land development. Significantly, high or intermediate developed levels consistently showed reduced alpha diversity when compared to the lower level. This underscores the pronounced impact of anthropogenic ecological disturbances in shaping mosquito microbiomes. Beta diversity analysis revealed no significant dissimilarities in bacterial community composition across seasons and developed levels, although some separation was noted among different levels of developed land cover. These findings highlight the significant role of environmental factors in shaping mosquito abundance and their associated microbiomes, with potential implications for the vectorial capacity in the transmission of vector-borne diseases.

Abstract:Mosquito vectors of pathogens (e.g., Aedes, Anopheles, and Culex spp. which transmit dengue, Zika, chikungunya, West Nile, malaria, and others) are of increasing concern for global public health. These vectors are geographically shifting under climate and other anthropogenic changes. As small-bodied ectotherms, mosquitoes are strongly affected by temperature, which causes unimodal responses in mosquito life history traits (e.g., biting rate, adult mortality rate, mosquito development rate, and probability of egg-to-adult survival) that exhibit upper and lower thermal limits and intermediate thermal optima in laboratory studies. However, it remains unknown how mosquito thermal responses measured in laboratory experiments relate to the realized thermal responses of mosquitoes in the field. To address this gap, we leverage thousands of global mosquito occurrences and geospatial satellite data at high spatial resolution to construct machine-learning based species distribution models, from which vector thermal responses are estimated. We apply methods to restrict models to the relevant mosquito activity season and to conduct ecologically plausible spatial background sampling centered around ecoregions for comparison to mosquito occurrence records. We found that thermal minima estimated from laboratory studies were highly correlated with those from the species distributions (r = 0.87). The thermal optima were less strongly correlated (r = 0.69). For most species, we did not detect thermal maxima from their observed distributions so were unable to compare to laboratory-based estimates. The results suggest that laboratory studies have the potential to be highly transportable to predicting lower thermal limits and thermal optima of mosquitoes in the field. At the same time, lab-based models likely capture physiological limits on mosquito persistence at high temperatures that are not apparent from field-based observational studies but may critically determine mosquito responses to climate warming. Our results indicate that lab-based and field-based studies are highly complementary; performing the analyses in concert can help to more comprehensively understand vector response to climate change.

Abstract:Aedes aegypti, the vector for dengue, chikungunya, yellow fever, and Zika, poses a growing global epidemiological risk. Despite extensive research on Ae. aegypti's life history traits and behavior, critical knowledge gaps persist, particularly in integrating these findings across varied experimental contexts. The plasticity of Ae. aegypti's traits throughout its life cycle allows dynamic responses to environmental changes, yet understanding these variations within heterogeneous study designs remains challenging. A critical aspect often overlooked is the impact of using lab-adapted lines of Ae. aegypti, which may have evolved under laboratory conditions, potentially altering their life history traits and behavioral responses compared to wild populations. Therefore, incorporating field-derived populations in experimental designs is essential to capture the natural variability and adaptability of Ae. aegypti. The relationship between larval growing conditions and adult traits and behavior is significantly influenced by the specific context in which mosquitoes are studied. Laboratory conditions may not replicate the ecological complexities faced by wild populations, leading to discrepancies in observed traits and behavior. These discrepancies highlight the need for ecologically relevant experimental conditions, allowing mosquito traits and behavior to reflect field distributions. One effective approach is semi-field studies involving field-collected mosquitoes housed for fewer generations in the lab under ecologically relevant conditions. This growing trend provides researchers with the desired control over experimental conditions while maintaining the genetic diversity of field populations. By focusing on variations in life history traits and behavioral plasticity within these varied contexts, this review highlights the intricate relationship between larval growing conditions and adult traits and behavior. It underscores the significance of transstadial effects and the necessity of adopting study designs and reporting practices that acknowledge plasticity in adult traits and behavior, considering variations due to larval rearing conditions. Embracing such approaches paves the way for a comprehensive understanding of contextual variations in mosquito life history traits and behavior. This integrated perspective enables the synthesis of research findings across laboratory, semi-field, and field-based investigations, which is crucial for devising targeted intervention strategies tailored to specific ecological contexts to combat the health threat posed by this formidable disease vector effectively.

Abstract:Fragmented landscapes in Mexico, characterized by a mix of agricultural, urban, and native vegetation cover, presents unique ecological characteristics that shape the mosquito community composition and mosquito-borne diseases. The extent to which landscape influences mosquito populations and mosquito-borne diseases is still poorly understood. This work assessed the effect of landscape metrics -agriculture, urban, and native vegetation cover- on mosquito diversity and arbovirus presence in fragmented tropical deciduous forests in Central Mexico during 2021. Among the 21 mosquito species across six genera we identified, Culex quinquefasciatus was the most prevalent species, followed by Aedes aegypti, Ae. albopictus, and Ae. epactius. Notably, areas with denser native vegetation cover displayed higher mosquito species richness, which could have an impact on phenomena such as the dilution effect. Zika and dengue virus were detected in 85% of captured species, with first reports of DENV in several Aedes species and ZIKV in multiple Aedes and Culex species. These findings underscore the necessity of expanding arbovirus surveillance beyond Ae. aegypti and advocate for a deeper understanding of vector ecology in fragmented landscapes to adequately address public health strategies.
Aedes albopictus in a recently invaded area in Spain: effects of trap type, locality, and season on mosquito captures

Abstract:Mosquitoes are primary vectors of pathogens impacting humans, wildlife, and livestock. Among them, the Asian tiger mosquito, Aedes albopictus , stands out as an invasive species with a global distribution, having established populations on every continent except Antarctica. Recent findings incriminate Ae. albopictus in the local transmission of several pathogens causing human diseases, including dengue, chikungunya, and Zika viruses and worm parasites as Dirofilaria . In Spain, the establishment of Ae. albopictus occurred in 2004 and it rapidly expanded, currently reaching southern provinces and creating novel epidemiological scenarios in recently invaded areas. In this study, we conducted captures of Ae. albopictus from May to November 2022 in two provinces, Granada and Malaga, situated near the current edge of the species' expanding range in Spain. The objective was to identify the primary factors influencing their captures in these regions. Mosquitoes were captured using BG-Sentinel traps baited with CO 2 and BG-Lure, and miniature CDC-UV traps in five different localities. Our findings underscore the influence of both extrinsic factors, such as locality, and intrinsic factors, including mosquito sex, on the abundance of captured Ae. albopictus . A higher abundance of Ae. albopictus was observed in the Malaga province compared to localities in the Granada province. Furthermore, similar numbers of Ae. albopictus mosquitoes were captured in more urbanized areas of Granada, while the lowest counts were recorded in the less urbanized area. These results were compared to captures of another common species in the area, specifically Culex pipiens . Overall, these results represent the first monitoring of invasive Ae. albopictus in the area and are discussed in the light of the potential importance of the species as a nuisance for humans and vectors of pathogens of public health relevance.

Abstract:In recent centuries, the mosquito Aedes aegypti has spread into most urban areas throughout the tropics. This species is considered the main vector of the chikungunya, dengue, yellow fever and Zika viruses and causes major public health issues. The aim of this study is to investigate the relative influence of biotic and abiotic parameters on immature populations of Ae. aegypti. During a one-year-long field experiment, we monitored 108 macroinvertebrate aquatic communities inhabiting four types of water containers across three different urbanized sites in a Neotropical city. A multimodel inference approach revealed that, in addition to abiotic parameters, biotic interactions with aquatic organisms had an important influence on the abundance of Ae. aegypti and that the urbanized site considered influences the outcomes of the interactions. Controphic species other than mosquitoes aided Ae. aegypti development, suggesting a mechanism of facilitation through a chain of processes. However, the abundance of Ae. aegypti was lowered by competition with native mosquito species in the slightly urbanized area and by predation in more urbanized areas. Competitive displacement and reduction, as well as predation by native aquatic organisms, can be considered a form of ecosystem service. The conservation and/or augmentation of natural enemies should improve the short- and long-term success of incompatible and/or sterile insect techniques, thus opening up perspectives for the future of mosquito management.
