Abstract:Klotho (KL) was originally characterized as an aging suppressor gene, and has been identified as a tumor suppressor gene in a variety of cancers including colon cancer. However, the potential role and molecular events for KL in colon cancer remain unclear. The present study aimed to investigate the expression of KL in human colon cancer by immunohistochemistry, and to analyze the correlation between KL expression and clinicopathological characteristics of patients with colon cancer. Functional analysis after lentivirus-mediated gain of KL expression was used to assess the tumor growth and invasion in colon cancer cells in vitro and in vivo. The rate of KL expression was significantly decreased in cancer tissues compared with that in adjacent non-cancer tissues (ANCT) (60.3 vs.77.9%, P=0.022), and KL expression was negatively associated with Dukes staging (P=0.034) and depth of tumor invasion (P=0.008). Overexpression of KL in vitro inhibited cell proliferative activities and invasive potential in colon cancer cells, companied with decreased expression of p-IGF1R, p-PI3K, p-AKT, PCNA and MMP-2. In addition, the tumor volumes in the HT-29 subcutaneous tumor model treated with lentivirus‑mediated KL vector (Lv-KL) was significantly smaller than those of the negative control (NC) group (P<0.01). Taken together, our findings indicate that the expression of KL is downregulated in human colon caner and correlates with tumor invasion and Dukes staging, while overexpression of KL suppresses growth and invasion through inhibition of IGF1R-mediated PI3K/AKT pathway in colon cancer cells, suggesting that KL may serve as a potential therapeutic target for the treatment of colon cancer.

Abstract:To overcome Yb lasing, a kilowatt-level 1535 nm fiber laser is utilized to in-band pump an Er:Yb co-doped fiber (EYDF) amplifier. The output power of a 301 W narrow-linewidth EYDF amplifier operating at 1585 nm, with 3 dB bandwidth of 150 pm and M-2 < 1.4, is experimentally demonstrated. To the best of our knowledge, it is the highest output power achieved in L-band narrow-linewidth fiber amplifiers with good beam quality. Theoretically, a new ion transition behavior among energy levels for in-band pumping EYDF is uncovered, and a spatial-mode-resolved nonlinearity-assisted theoretical model is developed to understand its internal dynamics. Numerical simulations reveal that the reduction in slope efficiency is significantly related to excited-state absorption (ESA). ESA has a nonlinear hindering effect on power scaling. It can drastically lower the pump absorption and slope efficiency with increasing pump power for in-band pumped EYDF amplifiers. Meanwhile, optimized approaches are proposed to improve its power to the kilowatt level via in-band pumping.

Abstract:In this paper, Er/Yb co-doped fiber amplifiers (EYDFAs) with an Yb-band fiber Bragg grating (FBG) at the pump end to improve the performance of the amplifier is systematically studied. The influence of the reflectivity and center wavelength of the FBG along with the gain-fiber length on the performance of an EYDFA are numerically analyzed. The results show that the wavelength of the FBG has critical influence on the efficiency of the EYDFA, whereas the requirement to its reflectivity is relaxed. It is an effective and promising way to improve the efficiency of a high-power pumped EYDFA by introducing a suitable Yb-band FBG at the pump end. Based on the analysis of the underlying principles, suggestions for the practical design and possible further improvement strategies are also proposed.

Abstract:To improve the radiation resistance of Yb-doped fiber lasers, we investigate the influence of pre-radiation and photo-bleaching on the gamma-radiated laser's performance. When the gamma radiation dose is within 10 krad(Si) with a radiation dose rate less than 0.4 rad(Si)/s, compared to the output power of a non-pre-radiated Yb-doped fiber laser, the pre-radiation technique could enhance the radiation resistance against gamma-ray. However, the mode instability threshold was decreased, which was caused by the cumulated radiation-induced attenuation of pre-radiation and radiation. Based on an electronic probe micro-analyzer, the Yb-doped active fiber was Yb-doped aluminophosphosilicate ternary fiber; therefore, the radiated defects were mainly hole-related defects. A laser diode centered at 532 nm was chosen as the photo-bleaching laser source, which could recover 45.2% of the radiated-induced attenuation and increase the mode instability threshold. This work demonstrates the influence of pre-radiation and photo-bleaching on the radiation resistance against the gamma-ray of Yb-doped fiber lasers, which are of significance in the design and fabrication of related fiber lasers.
High-efficiency cladding-pumped Er/Yb co-doped alumino-phosphosilicate fiber for an extended L-band amplification

Abstract:Extending the gain bandwidth of L-band optical fiber amplifier has provoked a widespread interest. To date, achieving a high-efficiency extended L-band amplification remains a challenge. Here, we report a cladding-pumped Er/Yb co-doped alumino-phosphosilicate fiber, prepared by the modified chemical vapor deposition process. We demonstrate the efficiency of alumino-phosphosilicate glass for cladding-pumped Er/Yb co-doped fiber, with a gain per unit fiber length of 0.45 dB/m at 1625 nm and a gain ripple of ∼9.4 dB. For 0.8 W pump power, the fiber exhibits a 20 dB gain bandwidth covering 1575-1625 nm and 6.9 dB noise figure at 1625 nm. Additionally, the utilization of multi-mode laser diode enables further significant power savings and cost reduction. To the best of our knowledge, Er/Yb co-doped fiber in alumino-phosphosilicate glass is first proposed, with a cladding-pumped scheme for enhancing an extended L-band performance.

Abstract:In this Letter, a high-power Er/Yb-codoped fiber amplifier (EYDFA) with a high-reflection Yb-band fiber Bragg grating (FBG) at the pump end is experimentally investigated. The FBG was inscribed on a piece of double-clad fiber with a center wavelength of 1032 nm. Due to the selective reflection of the backward Yb-band amplified spontaneous emission (Yb ASE) by the FBG, a co-pump-propagating Yb-band auxiliary signal was generated. Because of the stimulated amplification and reabsorption of the auxiliary signal, the Yb ASE was dramatically suppressed and the pump conversion efficiency (PCE) of the EYDFA was notably improved. An output power of 6.48 W was achieved at a pump power of 16.5 W, which is equivalent to a PCE of ∼39%. The slope efficiency relative to applied pump power was ∼40%. The maximum output power was improved ∼20% because of the introduction of the FBG.

Abstract:In this work, Na+ ions doping into the Yb-doped fiber is proposed to improve the photo-darkening resistance. The results show that the photo-darkening induced excess loss at equilibrium state at 633nm, 702 nm, 810 nm, and 1041 nm is 115.54dB/m, 86.87dB/m, 25.51 dB/m, 2.92 dB/m, respectively, when co-doping with Na+ ions. More than 30% excess loss is reduced comparing to the Yb-doped fiber without Na+ ions. The mechanism of Na+ ions doping to mitigate the PD excess loss is discussed. Besides, we measured the laser efficiency of Yb/Al/Na co-doped fiber to be 76.1%. This result remains almost the same with Yb-doped fiber and proves that the addition of Na+ ions do not deteriorate the fiber slope efficiency. The background loss of the two fibers also stays close. The results indicate this method is promising in high power fiber laser development.

Abstract:The experimental results of a high power 1080-nm ytterbium-doped fiber amplifier (YDFA) tandem-pumped by a 1018-nm fiber laser for different temperatures have been reported. For a 1.64 m of active fiber, the power conversion efficiency (PCE) of our YDFA has reached 91.1% at room temperature. And for a 0.82 m of active fiber, i.e., half-length of 1.64-m active fiber, the PCE has been improved from 58.7% to 81.3% by increasing the heating temperature from room temperature to 302 °C. In other words, we have traded off $sim 10$ % PCE for a 50% reduction of active fiber length. The temperature-dependent signal amplification model simulations agree well with the experiments. Our experimental and theoretical studies confirm that the temperature-dependent absorption and emission cross sections and the long-wavelength pump source are the physical causes of the improved PCE.

Abstract:A comprehensive model of erbium-doped fiber amplifiers (EDFA) is proposed in this paper, taking exited-state absorption (ESA), amplified spontaneous emission (ASE), background absorption, mode field, and Erbium ion radial distribution into account. Computer programs capable of handling forward, backward, and different wavelength bidirectional pumping are developed. Among other things, it is found that under typical conditions, Gaussian field approximations may cause considerable errors. The optimum V-value for small signal gain is much smaller than that for minimum Gaussian or Desurvire spot size; ASE propagating opposite to the pump beam can be several times higher than at propagating along the pump beam, resulting in a faster-than-linear decrease of the pump power at its beginning portion. Experimental results of domestic-made erbium-doped fibers pumped by 980 nm and 1480 nm laser diodes are also given.

Abstract:We demonstrate a high repetition rate, high average power narrow-linewidth fiber amplifier at 1.55 μm eye-safe waveband by using a very-large-mode-area active fiber doping Er and Yb, having a 60-μm-diameter core and a 600-μm-diameter inner cladding. A maximum average power reaches 16 W at a 10 kHz repetition rate for 600 ns pulses in the stimulated Brillouin scattering (SBS) free operation, which calculates to a 1.6 mJ pulse energy and 2.7 kW peak power. In addition, the peak power rises to 4.9 kW with 148 ns pulses at the onset of SBS. The signal peak is located at 1,551.15 nm with a 36 dB signal-to-noise ratio (SNR). In order to eliminate the pulse steepening, the pulse shaping technology using a triangular-like wave with a slower rising edge is applied on the input pulses. The half-width of the frequency-intensity spectrum from high-average-power fiber amplifier is about 1.02 MHz, approaching the Fourier transform limit.

Abstract:Based on a two-stage fiber, erbium doped fiber amplifiers (EDFAs) pumped by 980nm and 1480nm laser diodes (LD) are investigated. Experimental comparisons of gain, noise figure and output power between the intermediate isolator configuration and the loop-coupling configuration are discussed. A small signal gain of 42.8 dB of EDFA has been obtained for the loop-coupling configuration. The minimum noise figure and maximum output power at wavelength of 1553.5nm are 4.4 dB and 15.2 dBm, respectively.

Abstract:We present a numerical analysis of noise performance improvement of the erbium-doped fiber amplifier by flatting the mode field intensity distribution within the interaction region of erbium ions. Three different refractive index types of erbium-doped fiber were used to study the effect of the pump mode intensity distribution on noise performance. The results show that taking advantage of the depressed refractive index erbium-doped fiber which has a flat mode distribution in the fiber core, the maximum reduction of nearly 0.53 dB for noise figure was achieved across the full C-band. Based on the center depressed refractive index profile, the noise figure was further decreased by about 0.1 dB by reducing the doping radius to 0.71 m. Finally, maintaining the same gain level of 38.3 dB, a NF reduction of 0.63 dB was realized by introducing the center depressed refractive index profile and narrowing the erbium doping region. It is suggested that our investigation shows great potential for improving the noise performance of the erbium-doped fiber amplifier.

Abstract:High-energy irradiation significantly increases the optical losses and noise coefficients of laser materials, leading to a substantial decrease in the slope efficiency or gain performance of laser output. To address this issue, we propose a strategy to enhance the radiation resistance of glasses/fibers by introducing phase interfaces. Based on the sol–gel method, through phase-separation techniques and high-temperature annealing treatments, silica-rich and phosphorus-rich phases were formed in erbium-ytterbium co-doped high-phosphorus silica glass, and nanoscale phase interfaces with specific densities, stability levels, and homogeneous distributions of doped elements were constructed between the phases. Using high-resolution transmission electron microscopy, nuclear magnetic resonance, and spectroscopic analyses, we tracked the evolution of the internal microstructure of the glasses at the atomic level. The findings confirmed that annealing effectively controlled the density of the phase interfaces formed. Under 1 kGy X-ray irradiation, glasses with effective phase interfaces exhibited significant reduction in radiation-induced attenuation (RIA) and improvement in photoluminescence intensity compared to pristine glasses. This indicated that effective phase interfaces could act as complex centers for irradiation-induced point defects, absorbing radiant energy and trapping these defects, thus mitigating high-energy radiation-induced damages. Furthermore, online irradiation tests on the Er 3+ /Yb 3+ co-doped silica fibers supported this result. Compared to pristine fiber, fibers annealed for 3 h and annealed for 20 h with different phase interfacial densities showed 45% and 73% lower RIA at 1080 nm, respectively.

Abstract:The fiber gain and noise figure in erbium (Er)-ytterbium (Yb) codoped fiber amplifiers are experimentally studied and theoretically modeled. The different fiber parameters for the model are found in separate characterization measurements. The comparison between the measured and calculated data show excellent agreement. In addition, we demonstrate the design process for a specific cladding-pumped fiber to find the optimum fiber length.

Abstract:The power scaling of Er-Yb co-doped fiber lasers and amplifiers has been limited by the bottleneck effect of energy-transfer saturation between Yb ions and Er ions. The emerging method of Er-Yb co-doped fiber amplifiers pumped by Yb-doped fiber lasers is considered as an approach to enhance the threshold of the bottleneck effect. In this paper, we quantitatively characterize the threshold of the bottleneck effect via the method of extreme value analysis of the second-order derivative. The method facilitates the optimization of the amplifier configuration. Afterward, we numerically investigate the bottleneck effect of various Er-Yb co-doped fiber amplifiers off-peak cladding-pumped by 10××nm Yb-doped fiber lasers for what we believe, to the best of our knowledge, is the first time. The result shows that the most optimal configuration is long gain fiber over 20 m pumped by a 1020-1025 nm fiber laser, with more than two times the output limit of a conventional laser diode pumping scheme. The essential factors of an amplifier are discussed afterward, including the pump-launching direction, the optimization of large-mode-area fiber, the core-cladding ratio, the concentration of doping ions, the nonlinearity limit, and the distribution of the heat load.

Abstract:By focusing on a typical emitting wavelength of 1120 nm as an example, we present the first demonstration of a high-efficiency, narrow-linewidth kilowatt-level all-fiber amplifier based on hybrid ytterbium-Raman (Yb-Raman) gains. Notably, two temporally stable, phase-modulated single-frequency lasers operating at 1064 nm and 1120nm, respectively, were applied in the fiber amplifier, to alleviate the spectral broadening of the 1120 signal laser and suppress the stimulated Brillouin scattering (SBS) effect simultaneously. Over 1 kW narrow-linewidth 1120 nm signal laser was obtained with a slope efficiency of ~ 77% and a beam quality of M2~1.21. The amplified spontaneous emission (ASE) noise in the fiber amplifier was effectively suppressed by incorporating an ASE-filtering system between the seed laser and the main amplifier. Further examination of the influence of power ratios between the two seed lasers on the conversion efficiency had proved that the presented amplifier could work efficiently when the power ratio of 1120 nm seed laser ranged from 31% to 61%. Overall, this setup could provide a well reference for obtaining high power narrow-linewidth fiber lasers operating within 1100-1200 nm.

Abstract:Fiber amplifiers operating in the L-band can benefit from further extending their gain bandwidth and improving overall operation costs. This work addresses both aspects through an optimization of the host matrix using phosphorus/aluminum (P/Al) as glass modifier and secondly through the use of yttrium as an alternative co-dopant to ytterbium. Fiber samples were fabricated using the modified chemical vapor deposition (MCVD) and solution doping technique. Using a maximum amount of phosphorus consistent with a flat refractive index profile, we find the optimum alumino-phospho-silicate glass matrix. The achieved amplifier exhibits a large gain amplification ranging from 15 to 25 dB, with its peak gain occurring at 1610 nm when pumped with a total power of 1.4 W at 978 nm. The maximum noise figure was 6.3 dB, while the gain ripple within the wavelength range of 1575 to 1626 nm was 76%. Moreover, we have obtained a low level of pair-induced quenching (<3%) using Y rather than Yb as a de-clustering agent for Er3+ concentration of 1.2 ×1025 m-3. The result provides a viable solution to the 980 nm pumping scheme in a quest to achieve cost-effective and low-noise L-band EDFAs.

Abstract:A finite-difference time-domain (FDTD) simulation of Yb-doped cladding-pumped, mJ-level, excitation-balanced fiber pulse amplifiers (EBFAs) is presented. In EBFAs, two pumps, one above (anti-Stokes pump, or ASP) and one below (Stokes pump, or SP) the signal wavelength, are utilized to reduce the net thermal energy generated due to the quantum defect. From the results of the FDTD simulation, detailed analyses on the fiber length optimization, excited Yb3+ population evolution, pump and signal power evolution, optical-to-optical (o-o) conversion efficiency, wall plug efficiency, as well as thermal energy generation are performed. For example, with an ASP at 990 nm and a SP at 975 nm, only 2.3 µJ of thermal energy is produced when generating a 2 mJ output pulse at 985 nm, whereas a pulse amplifier with only SP pumping rendering the same 2 mJ output gives more than 10 times the thermal energy. In the meantime, the system maintains an o-o efficiency of 8.43% and wall plug efficiency of 6.6%. The results here indicate the feasibility of the power-scaling of excitation-balanced laser systems, and the FDTD model will be beneficial for the design and optimization of such systems. The first half of this paper presents the FDTD model and provides an example calculation outlining the modeling procedure. The remaining half details the impact of varying laser parameters on system performance. These include pumping and input signal energies, repetition rates, and selection of the ASP, SP, and signal wavelengths. The results presented herein can also be extended to excitation balancing in other solid-state laser systems, such as Yb:YAG and Tm:YAG lasers.
