Abstract:Abnormal levels of endogenous sulfur dioxide in organisms will cause cardiovascular, respiratory and neurological diseases. Therefore, the development of an effective detecting method for real-time, quantitative detection of sulfur dioxide derivatives in organisms is of great importance in physiological and pathological aspects. In this work, a red-emitting SO2 fluorescent probe based on benzopyrylium derivatives were designed and synthesized to specifically detect SO2 derivatives by nucleophilic addition of unsaturated carbon-carbon double bonds. The probe is characterized with rapid response, “naked eye” visualization, broad detection range and high sensitivity (8.1 μM) for SO2 derivatives. In addition, it has been applied to track the dynamic, real-time, visual detection of exogenous and endogenous SO2 in cells, and imaging of exogenous SO2 derivatives in zebrafish.

Abstract:On the basis of a unique nucleophilic addition reaction, a novel water-soluble broadly emitting (500-700 nm) fluorescent Probe 1 was developed for the rapid detection of SO2 derivatives in aqueous media. The positively-charged benzopyrylium moiety in Probe 1 provides both excellent water solubility, making this probe applicable in 100% aqueous environments, and the ability to function as a fluorescence quencher of the coumarin moiety. Probe 1 generates a nearly instantaneous strong fluorescence signal in response to SO2 derivatives having an 8.3 nM detection limit for bisufite. The resulting Probe 1-sulfite adduct emits in the green/red spectral region (lambda(max) = 585 nm) with a large Stokes shift (139 nm). The probe exhibits excellent selectivity toward SO2 derivatives over other potential interfering agents including reactive sulfur-containing species. Importantly, we demonstrate that Probe 1 can be used for the real-time sensing and bioimaging of SO2 derivatives in living cells.

Abstract:Hydrogen sulfide (H2S) is a signaling gasotransmitter that plays important roles in modulating the functions of different systems. We have designed and synthesized a 3-hydroxychromone-based fluorescent probe A for H2S detection. Probe A is based on a H2S-induced thiolysis reaction and an excited-state intramolecular proton transfer (ESIPT) process. The probe shows high selectivity and sensitivity for the detection of H2S over glutathione, cysteine and homocysteine. Moreover, this probe was successfully applied for imaging exogenous and endogenous H2S in living cells.

Abstract:A novel chromenylium-based fluorescent probe was exploited for sulphur dioxide (SO₂) detecting. The probe displayed a remarkable fluorescence turn-on response towards SO₂ based on the nucleophilic addition reaction to the carbon-carbon double bond with 105 nm Stock shift. The probe was successfully applied for the quantification of SO₂.The linear detection range was from 0-160 μM with the detection limit as low as 99.27 nM. It also exhibited high selectivity for SO₂ than other reactive species and amino acids. Furthermore, cell staining experiments indicated that the probe was cell membrane permeable and could be used for high-performance imaging of SO₂ in living cells. The superior properties of the probe made it highly promising for use in chemical and biological applications.

Abstract:: Sulfur dioxide and its derivatives (HSO 3 - /SO 3 2- ) play a crucial role in maintaining the redox balance of biological systems. Therefore, the development of effective detection methods for sulfur dioxide and its derivatives is of paramount importance. Herein, a novel fluorescent probe, QLN , was designed and synthesized using small molecule quinoline, demonstrating the capacity to recognize HSO 3 - through Michael addition reaction. The probe QLN displayed good selectivity and anti-interference capabilities for HSO 3 - recognition, effectively operating within a wide pH range (5∼9) and exhibiting a rapid response time (< 5 s). Practical applications revealed the potential of probe QLN for detecting HSO 3 - in living cells and food samples.

Abstract:A novel highly active fluorescence chemical sensor (TBQN) for HSO3- was synthesized by the Knoevenagel reaction based on triphenylamine-benzothiazole as a new fluorophore. The probe possessed good selectivity toward HSO3- and anti-interference ability with common ions. The fluorescence and UV-vis spectra of the TBQN probe were significantly changed after the addition of HSO3-. At the same time, the probe solution released obvious green fluorescence. Moreover, the limit of detection for HSO3- was calculated to be 3.19 x 10(-8) M. The TBQN probe displayed a rapid response to HSO3- and it took about 3 min to complete the recognition. The detection mechanism is the nucleophilic addition reaction between HSO3- and -C = C- in the probe molecule. The pi-conjugation and ICT (intramolecular charge transfer) transition in the TBQN molecule were destroyed by this addition, which resulted in the change of the fluorescence before and after the addition of HSO3-. Then, the mechanism was verified by theoretical calculations, H-1 NMR measurements and mass spectroscopy. In addition, the probe showed low cytotoxicity and could be used for biological imaging in RAW264.7 cells.

Thus, it is exceedingly promising and significant to develop a highly sensitive detection technique for tracing H2S. Inspired by this, we designed and synthesized a new fluorescent probe 2-[3-[2-[3-bromo-4-(2,4- dinitrobenzenesulfonate)] ethenyl]-5,5-dimethyl2-cyclohexen-1-ylidene]propanedinitrile (SP-Br) for hydrosulfide ion detection by introducing bromine atom. Compared with reported H2S probes based on the same fluorescent parent, SP-Br has longer fluorescence emission (gem = 670 nm), shorter response time (3 min), lower detection limit (149 nM), and wider detection range (0-30 nM). SP-Br can emit weak yellow fluorescence, and the emission intensity at 670 nm is considerably enhanced in the presence of hydrosulfide ions. The identification mechanism of hydrosulfide ion by SP-Br was verified by high-resolution mass spectrometry, fluorescence, and UV-vis absorption spectroscopy. In addition, SP-Br has been successfully applied to the monitoring of actual water samples and beer samples and has certain development prospects and value in the fields of environmental pollution and food quality analysis.

Abstract:As an important gas signal molecule, sulfur dioxide(SO2) has attracted increasing attention in recent decades.It has been revealed that SO2 and its derivatives are closely associated with various symptoms and diseases including cancer, cardiovascular diseases, neurological disorders and respiratory problems.Hence, developing analytical methods that could realize concentration determination of SO2 and its derivatives are highly needed.In this work, we reported a novel turn-on fluorescent probe(3) for selectivie sensing of SO2 derivatives based on the conjugate of benzo[e]indolium and pyrrole.The intermediate 2 was prepared by using 1,1,2-trimethyl-1H-benz[e]indole and iodomethane as raw materials.3 was synthesized via condensation of 2 with 5-formyl-2,4-dimethyl-1H-pyrrole-3-carboxylic acid.The structure was characterized by 1H NMR, 13C NMR and HR-MS.3 exhibited excellent selectivity for SO2 over other anions and reductive sulfurs.With addition of sulfur dioxide, the emission intensity at 450 nm increased accordingly, and the emission intensity was linear function with the sulfur dioxide concentration over the range from 0 to 100 μmol·L-1(R2=0.997).

Abstract:Sulfur dioxide is one of the reactive sulfur species, which has significant physiological functions in cells. Some physiological processes are closely related to SO2 in organisms, and the high concentration of SO2 in living cells can cause many diseases. In order to investigate the unique function of SO2 at the subcellular level, developing a molecular tool which could detect of SO2 within organelles is imperative. Hence, we developed a cationic dye named HQ-SO2 as a new fluorescent probe to specifically monitor SO2 , which was easy to obtain through one-step reaction. It took Michael addition reaction as the mechanism of reaction for detection of SO2 . In addition, this probe showed a series of highly favorable properties such as rapid response rate, low cytotoxicity, high selectivity, low detection limit, and good photostability, which enabled the probe to track SO2 in living cells.

Abstract:As one of the members of reactive sulfur species, hydrogen polysulfide (H 2 S n , n > 1) plays an important role in enzyme activity and nervous system regulations, and the sensing mechanism study is of great significance for the design of novel efficient probes. Herein, we investigated the sensing mechanism of an efficient triphenylamine-benzofuran-based probe (TBF-SS) towards H 2 S n using DFT method. The inherent fluorescence quenching of the probe is dominated by the twisted intramolecular charge transfer (TICT) as revealed by the torsional potential curve calculations. When the nitro fluorophenyl group is replaced by a hydroxyl group in the reaction with H 2 S n , the TICT is eliminated and the excited state can return to the ground state in a radiative way, leading to strong fluorescence emission.

Abstract:Sulfur dioxide is a common air pollutant and extremely harmful to human health. After inhalation, SO2 is easily hydrated to two derivatives, sulfite and bisulfite. So it is valuable to develop an effective probe for these anions. In this work, a green-emissive fluorescent probe 1 is designed and synthesized, which employ 4-hydrazinyl-1, 8-naphthalimide as intramolecular charge transfer (ICT) fluorophore. Probe 1 is able to sense sulfite and bisulfite through fluorescence “turn-on” manner, and the detection limit for sufite is 0.56μM. The green emission induced by sulfite and bisulfate can be observed by naked eyes under UV irradiation. Compared with other anions used, probe 1 shows great selectivity toward sulfite or bisulfite. We illustrate the possible recognition mechanism of probe 1 and sulfite by NMR analysis and molecular modeling experiments. The real-time monitoring of SO2 release and cell imaging of sulfite is also investigated.

Abstract:Sulfur dioxide derivatives (HSO3− and SO32−) play an important role in food preservative, antibacterial, antioxidant and other aspects, so it is urgent for us to develop more efficient detection methods to broaden their application in biochemical research and related disease diagnosis. Fluorescent probes are of particular interest because of their simplicity and high temporal and spatial resolution. Herein, we constructed a new near-infrared (NIR) fluorescence probe, CQC, composed of coumarin fluorophore and quinoline fluorophore, for detecting SO2 derivatives. The near-infrared emission probe CQC with a large Stokes shift (260 nm) not only kept the distance between the two emission peaks large enough (165 nm), but also had a particularly high energy transfer efficiency (99.5%), and was particularly sensitive to the detection of HSO3−/SO32− (LOD: 0.1 μM). The powerful probe CQC succeeded in real-time visualizing endogenous HSO3−/SO32− in living cells.

Abstract:Monitoring sulfur dioxide (SO2) derivatives in environment is of great significance due to their harmful effects to the environment and human health. In this study, a fluorescent probe (CZBT) for SO2 derivatives was prepared from 9-ethyl-9H-carbazole-3,6-dicarboxaldehyde and 2-methyl-benzothiazolium, which displayed a noticeable color change from yellow to colorless along with a remarkable fluorescence change from yellow to blue in response to HSO3-. The probe could quantitatively determine the concentration of HSO3- with excellent selectivity, high sensitivity and low limit of detection. 1H NMR and HR-MS spectra demonstrated that a selective 1, 4-nucleophilic addition occurred on the bridge double bond in CZBT. The probe was successfully used to determine the SO2 derivatives in several real water samples with good recovery. Furthermore, the probe was employed for monitoring the level of intracellular HSO3- in HeLa (human cervical cancer) cells by fluorescence imaging. These results indicated that CZBT has a good capability for monitoring SO2 derivatives in environment and living cells.

Abstract:As a signal molecule, SO2 participates in many complex physiological processes. Therefore, in view of the addition properties of SO2 derivatives to unsaturated olefins, we reasonably designed a set of ratiometric fluorescent probes (NB1, NB2, NB3), and evaluated the influence of the coumarin moiety on the optical properties of the probes. Amongst the probes when the 7-position is substituted by a fused quinolizine (julolidine) unit, the probe NB2 shows a change from red to green fluorescence upon reaction with SO2, and has excellent optical properties: 136 nm emission band gap, 230 s response time, high selectivity, and excellent sensitivity. More valuable is the fact that NB2 can be used in cell imaging experiments, real sample detection, and employed to prepare fluorescent test paper for the qualitative test of SO2 gas, indicating that NB2 can be used as a powerful tool for the detection of SO2 derivatives.

Abstract:Two fluorescence-enhanced probes, 4-(2,4-dinitrophenoxy)- N -(2-hydroxyethyl)-1,8-naphthalimide(NTE-1) and 4-(2,4-dinitrophenoxy)- N -(4-(2,4-dinitrophenoxy)phenyl)-1,8-naphthalimide(NTE-2), have been designed and synthesized for detection of H 2 S. 4-Hydroxy-1,8-naphthalimide as fluorophore in combination with 2,4-dinitrophenyl ether as H 2 S response site constructed the fluorescence probes. The consequences showed that both NTE-1 and NTE-2 displayed large red-shift(excess 100 nm) in absorption spectra and more than 30-fold fluorescence enhancement in response to H 2 S. Moreover, the dual site probe, NTE-2, displayed wider linear range between fluorescence intensity and concentration of H 2 S(0—40 μmol/L) compared with single site probe, which can be applied to quantitative detection of high concentration of H 2 S. The photoinduced electron transfer(PET) response mechanism of probe was further studied by analyzing the distributions of molecular orbital. Importantly, the probes have potential practical applications in detection of H 2 S.
