Abstract:Sulfur dioxide (SO2) is an important marker in the human body, and maintaining its balance is very significant for human health. The high concentration of SO2 will cause harm to human health because of the transient nature of SO2. To measure the level of SO2, it is urgent to find a suitable detection method for the real-time monitoring of sulfite. Herein, we designed and exploited a Near-Infrared (NIR) turn-on fluorescent probe L derived from rhodamine for the detection of SO32- in living cells and zebrafish. The addition of sulfite would attack the 4-position of the carbonyl group of levulinic acid, resulting in the cleavage of the ester bond and the formation of the fluorophore. The intramolecular charge transfer (ICT) was restored thus the monitoring system emitted the corresponding long NIR fluorescence at 646 nm. Probe L had high selectivity, a short response period within 10 min, and the detection limit of 0.6 mu M. Besides, Probe L had a long excitation wavelength (>580 nm), which was in favor of more deeply biological imaging properties. Moreover, Probe L had low cell cytotoxicity through MTT assay, thus it was confirmed with good biocompatibility. In addition, probe L could successfully real-time monitor exogenous and endogenous sulfite in living cells, and be applied in zebrafish. The confocal results suggested that probe L had strong potential application prospect of outstanding capabilities for detecting sulfite in clinical diagnostics.

Abstract:In daily life, sulfite and bisulfite have been extensively used as antimicrobial agent and antioxidant for food products, beverages and pharmaceutical products, but, SO3 2−/HSO3 − are fantastically harmful to human health, so it is meaningful to develop an effective probe for these two anions. In this work, a new fluorescent material probe 1 was designed and constructed, which was able to sense sulfite and bisulfite through fluorescence “turn-on” manner. Compared with correlative fourteen other anions used, probe 1 showed great selectivity toward sulfite or bisulfite. Furthermore, the probe can detect SO3 2−/HSO3 − with fast response (within 4min) as well as low detection limit (8.8nM) compared with the previous work. We characterized the adduct through NMR and X-ray diffraction analysis and further confirmed the detection mechanism of probe 1. Finally, fluorescence imaging of MCF-7 cells indicated that 1 could be used for monitoring SO2 derivatives in living cells with low cytotoxicity.

Abstract:Sulfite (SO32-) is considered as a monitor of a wide range of physiological processes. However, cells and tissues are adversely affected when the body ingests high level of sulfite. Here, we designed and synthesized a "turn on" fluorescent probe ImiSft-1 with 2-cyano-N-methylacetamide as the specific recognition site of SO32-. This probe predominantly achieved high response intensity to SO32- and desirable properties such as large Stokes shift (similar to 180 nm), fast response time (within 15 s), and high sensitivity (LOD = 0.12 mu M). Importantly, the probe was highly selective for sulfite from other bio-species including biological thiols. Other functional properties included broad pH adaptability (5.0-10.0) and low cytotoxicity. Given these advantages and the fluorescence imaging in living MCF-7 cells, it was demonstrated that probe ImiSft-1 could monitor the changes of sulfite concentration in living cells.

Abstract:SO2 and its derivatives (SO32-/HSO3-) are used widely in food, beverages, and pharmaceutical production. However, they could induce multiple diseases in respiratory, nervous, and cardiovascular systems. Although several fluorescent probes have been developed for detecting SO32-/HSO3-, reports on rapid fluorescent probes for the on-site detection of SO2 derivatives are scarce. Herein, a colorimetric and ratiometric fluorescent probe 1 based on the intramolecular charge transfer (ICT) was reported. Probe 1 resulted in a 122 nm blue-shift in fluorescent emission and decrement of absorbance at 500 nm upon the addition of sulfite. Therefore, probe 1 could quantify SO32-/HSO3- using both UV-Vis and fluorescent methods (LOD: UV-Vis method 34 nM; fluorescent method 51 nM). Importantly, probe 1 was used for a rapid (60 s) and convenient (1 step, on-site) measurement of the SO2 derivatives in real samples (LOD: 0.47 µM) using smartphone based on the colorimetric method. The SO32-/HSO3--sensing mechanism was confirmed as the Michael addition reaction. Furthermore, the probe was used for the real-time monitoring of SO32-/HSO3- in A549 cells and zebrafish. In summary, an all-in-one fluorescent probe was successfully developed for the accurate quantification, on-site detection, and bioimaging of SO32-/HSO3-.

Abstract:Sulfur dioxide (SO2) and its derivatives (SO32- and HSO3-), are important active sulfur species that play significant roles in physiological processes. Fluorescence probe imaging technology, due to its high temporal and spatial resolution, real-time non-invasive and non-destructive detection, has emerged as a valuable tool for studying SO2 in biological systems. In this study, we presented a colorimetric fluorescent probe for the detection of HSO3-. The structure of probe TPN-BP consists of a triphenylamine group and a benzopyrylium group that are connected by a vinyl double bond. The benzopyrylium group in probe TPN-BP, which carries a positive charge, serves two important functions: enhancing water solubility, allowing for its effective use in fully aqueous environments, and acting as a fluorescence quencher for the triphenylamine group. Upon interaction with HSO3-, probe TPN-BP exhibited significantly increase in fluorescence at 480 nm, causing the solution to change from blue to colorless. Spectral experiments showed that probe TPN-BP showed quick response time (10 s), high sensitivity (12.7 nM), and excellent selectivity towards HSO3-. It is worth noting that probe TPN-BP has been successfully used for fluorescence imaging and detection of HSO3- in plants and zebrafish. The results of this study indicated that probe TPN-BP can be used as a promising tool for the research and monitoring of SO2 in living organisms.

Abstract:A simple 4-hydroxynaphthalimide-derived colorimetric and ratiometric fluorescent probe ( 1 ) containing a receptor of levulinate moiety was designed and synthesized to monitor sulfite. Probe 1 could quantificationally detect sulfite by a ratiometric fluorescence spectroscopy method with high selectivity and sensitivity. Specially, probe 1 exhibited a 100 nm red-shifted absorption spectrum along with the color changes from colorless to yellow, and 103 nm red-shifted emission spectra upon the addition of sulfite. Thus, 1 can serve as a “naked-eye” probe for sulfite. Further, the recognition mechanism of probe 1 for sulfite was confirmed using nuclear magnetic resonance and electrospray ionization mass spectrometry. Also, the preliminary practical application demonstrated that our proposed probe provided a promising method for the determination of sulfite.

Abstract:: Sulfur dioxide and its derivatives (HSO 3 - /SO 3 2- ) play a crucial role in maintaining the redox balance of biological systems. Therefore, the development of effective detection methods for sulfur dioxide and its derivatives is of paramount importance. Herein, a novel fluorescent probe, QLN , was designed and synthesized using small molecule quinoline, demonstrating the capacity to recognize HSO 3 - through Michael addition reaction. The probe QLN displayed good selectivity and anti-interference capabilities for HSO 3 - recognition, effectively operating within a wide pH range (5∼9) and exhibiting a rapid response time (< 5 s). Practical applications revealed the potential of probe QLN for detecting HSO 3 - in living cells and food samples.

Abstract:The emission of venenous sulfur dioxide (SO2) and its derivatives from industrial applications such as coking, transportation and food processing has caused great concern about public health and environmental quality. Probes that enable sensitivity and specificity to detect SO2 derivatives play a crucial role in its regulations and finally mitigating its environmental and health impacts, but fluorescent probes that can accurately, rapidly and on-site detect SO2 derivatives in foodstuffs and environmental systems rarely reported. Herein, a near-infrared (NIR) fluorescent probe (ZTX) for the ratiometric response of bisulfite (HSO3-) was designed and synthesized by regulating the structure of high-performance HSO3- fluorescent probe SL previously reported by us based on structural analyses, theoretical calculations and related literature reports. The Michael addition reaction between the electronic-deficient C=C bond and HSO3- destroys ZTX's π-conjugation system and blocks its intramolecular charge transfer (ICT) process, resulting in a significant fading of the fuchsia solution and the bluish-purple fluorescence turned light blue fluorescence. Fluorescent imaging of HSO3- in live animals utilizing ZTX has been demonstrated. The quantitative analysis of HSO3- in food samples using ZTXvia a smartphone has been also successfully implemented. Simultaneously, the ZTX-based test strips were utilized to quantificationally determine HSO3- in environmental water samples by a smartphone. Consequently, probe ZTX could provide a new method to understand the physiopathological roles of HSO3-, evaluate food safety and monitor environment, and is promising for broad applications.

Abstract:An A-pi-A' structural ratiometric fluorescent probe 1 bearing a benzo[e]indolium moiety was developed for the detection of HSO3- in PBS buffer solution. HSO3- was expected to react with probe 1 via a Michael addition reaction, causing dramatic changes in its emission spectrum (from 578 nm to 466 nm) and an obvious color change from orange red to blue which could be observed by the naked eye. The reaction could be complete in 30 seconds in the sensing system, and displayed a high selectivity and sensitivity for HSO3- with a detection limit of 15 nM. The practical value of the probe was confirmed for application to detect the level of HSO3- in sugar samples with good recovery and in living cells.

Abstract:In view of the vital roles in biological systems, it is significant and challenging to construct a fluorescent probe that can simultaneously detect and discriminate the most important four reactive sulfur species (H 2 S, Cys, Hcy/GSH). Herein, a smart fluorescent probe Indole-NI-NBD based on indole-fused 1,8-naphthalimide derivative has been rationally designed and synthesized. This probe displayed fast, selective and sensitive responses for these four reactive sulfur species with distinct fluorescent turn-on signal changes at 595 nm. Moreover, the unique absorbance enhancement at 575 nm and the distinct color change achieved "naked eye" detection of H 2 S. The distinctive fluorescence enhancement at 560 nm only for Cys achieved its discrimination from H 2 S, and Hcy/GSH. More importantly, using Indole-NI-NBD -loaded test strips and RGB experiment, a smartphone sensing platform was constructed and realized the convenient, reliable, and on-site detection of H 2 S and Cys. Finally, Indole-NI-NBD was applied to image H 2 S, Cys, Hcy/GSH and distinguish Cys from them in living cells and zebrafish via different fluorescence signals at a single excitation wavelength.

Abstract:Sulfur dioxide derivatives (HSO3− and SO32−) play an important role in food preservative, antibacterial, antioxidant and other aspects, so it is urgent for us to develop more efficient detection methods to broaden their application in biochemical research and related disease diagnosis. Fluorescent probes are of particular interest because of their simplicity and high temporal and spatial resolution. Herein, we constructed a new near-infrared (NIR) fluorescence probe, CQC, composed of coumarin fluorophore and quinoline fluorophore, for detecting SO2 derivatives. The near-infrared emission probe CQC with a large Stokes shift (260 nm) not only kept the distance between the two emission peaks large enough (165 nm), but also had a particularly high energy transfer efficiency (99.5%), and was particularly sensitive to the detection of HSO3−/SO32− (LOD: 0.1 μM). The powerful probe CQC succeeded in real-time visualizing endogenous HSO3−/SO32− in living cells.

Abstract:The fluorescent probe is constructed by incorporating an α, β-unsaturated pyridinium to a pyrene fluorophore. The chemodosimeter has shown a selective and sensitive response to sulfite anion over other various anions and biological thiol through a Michael addition of the sulfite to the alkene of the probe. Meanwhile, it can be easily observed that the color of the probe for sulfite changes from yellow to colorless by the naked eye, and from yellow to blue under UV lamp immediately after the sulfite is added.

Abstract:A novel chromenylium-based fluorescent probe was exploited for sulphur dioxide (SO₂) detecting. The probe displayed a remarkable fluorescence turn-on response towards SO₂ based on the nucleophilic addition reaction to the carbon-carbon double bond with 105 nm Stock shift. The probe was successfully applied for the quantification of SO₂.The linear detection range was from 0-160 μM with the detection limit as low as 99.27 nM. It also exhibited high selectivity for SO₂ than other reactive species and amino acids. Furthermore, cell staining experiments indicated that the probe was cell membrane permeable and could be used for high-performance imaging of SO₂ in living cells. The superior properties of the probe made it highly promising for use in chemical and biological applications.

Abstract:Six coumarin-based fluorescent probes were designed and synthesized for the detection of sulfite. The probes were constructed based on "fluorophore-receptor/linker-steric group" mode with N,N-diethyl-7-aminocoumarin (DEA) or 2,3,5,6-1H,4H-tetrahydro-quinolazino-(9,9a,1-gh)coumarin (THQ) as the fluorophore, 2-naphthyl ketone (N) or phenyl ketone (B) as the steric group and vinyl (1) or 1,3-butadienyl (2) as the receptor/linker. The effects of the fluorophore, the linker and the steric group on the photophysical properties and sensing performance were systematically studied. Among all the probes, DEA-N2 exhibited the most excellent spectral responses toward sulfite: upon addition of sulfite, the absorption and emission maxima shifted from 488 nm and 684 nm to 405 nm and 484 nm, respectively. I 484 /I 688 displayed good linear relationship with sulfite concentration in the range of 3-50 μM, with a detection limit of 0.31 μM. The experimental results demonstrate that DEA-N2 has potential in ratiometric fluorescent imaging of mitochondrial sulfite in living cells.

Abstract:A novel 1-hydroxy-2,4-diformylnaphthalene-based fluorescent probe L was synthesized by a Knoevenagel reaction and exhibited excellent sensitivity and selectivity towards sulfite ions (SO32−) and bisulfite ions (HSO3−). The detection limits of the probe L were 0.24 μM using UV-Vis spectroscopy and 9.93 nM using fluorescence spectroscopy, respectively. Furthermore, the fluorescent probe L could be utilized for detection in real water samples with satisfactory recoveries in the range 99.20%~104.30% in lake water and 100.00%~104.80% in tap water by UV-Vis absorption spectrometry, and in the range 100.50%~108.60% in lake water and 102.70%~103.80% in tap water by fluorescence spectrophotometry.

Abstract:Sulfite is one of the main existing forms of sulfur dioxide (SO<sub>2</sub>) in living system, which has been recognized as an endogenous mediator in inflammation. Evidence has accumulated to show that abnormal level of sulfite is associated with many inflammatory diseases, including neurological diseases and cancers. Herein, a novel fluorescent probe named QX-OA was designed and synthesized to detect sulfite. QX-OA was constructed by choosing quinolinium-xanthene as the fluorophore and levulinate as the specific and relatively steady recognition reaction. The probe showed remarkable green turn-on signal at 550 nm, together with high sensitivity (90-fold) and excellent selectivity to sulfite over other possible interfering species. In the meantime, QX-OA was successfully applied to visualize endogenous and exogenous sulfite in Hela cells. In the LPS-induced inflammation model, QX-OA could visualize the dose-dependent increase of sulfite level (0-2 mg/mL). Consequently, QX-OA was determined to be a potential method for detecting sulfite in pre-clinical diagnosis.
