Abstract:We have described a turn-on near-infrared fluorescent probe Cy-NO(2) based on nitro group reduction for intracellular H(2)S detection. The probe employs cyanine dye as a fluorophore, and is equipped with a nitro group as a fluorescent modulator. It is readily employed for assessing intracellular H(2)S level changes, and confocal imaging is achieved successfully.

Abstract:A novel selective fluorescent chemosensor 1 has been synthesized with a phenanthrene-fused dipyrromethene structure. Selective binding of Cu2+ by 1 results in a complex that displays high selectivity and sensitivity for H2S. The signal transduction occurs via reversible formation-separation of the Cu-1 complex and CuS. Its potential utility for biological applications was confirmed by fluorescence imaging of H2S in live cells.

Abstract:Since Hydrogen Sulfide (H2S) was recognized as a gas transmitter, its detection and quantification have become a hot research topic among chemists and biologists. In this area, fluorescent probes have shown great advantages: fast and strong response, low detection limit and easy manipulation. Here we developed a new fluorescent probe that detected H2S selectively among various bioactive and inorganic salts. This probe was based on the core structure of fluorescein and reacted with H2S through azide-reduction. Great linearity was achieved correlating fluorescence intensity and H2S concentrations in solution. The detection of H2S in cancer cells was also achieved.

Abstract:An theoretical study on the luminescent and reaction mechanism of dansyl‐based fluorescence probe for detecting hydrogen sulfide is conducted and the results can explain the "turn‐on" effect of fluorescent probe in the presence of H2S. The photophysical and photochemical properties of the sulfonyl azide‐based fluorescent probe DNS–Az and its reduction product DNS by hydrogen sulfide (H2S) have been investigated theoretically. The calculated results indicated the first excited states of DNS–Az was dark state (oscillator strength less than 0.03) and DNS was bright state (oscillator strength more than 0.1), which determined the predicted radiative rate kr of DNS–Az was much smaller than that of DNS, meanwhile, due to more larger reorganization energy of DNS–Az, its predicted internal conversion rate kic was four times larger than that of DNS; moreover, owing to the effect of heavy atom from sulfur atom in DNS–Az, its predicted intersystem crossing rate kisc was seven times larger than that of DNS, thus the calculated fluorescence quantum yield of DNS–Az was only 2.16% and that of DNS was more than 77.2%, the above factors is the basis for DNS–Az molecule to function as a fluorescent probe. Regarding both DNS‐Az and DNS molecules, their maximum Huang‐Rhys factors, which are less than unity, signify the reliability of 0–0 transitions between their S0 and S1 electronic states. In addition, for DNS, our simulated emission peak of the 0–0 transition is 515 nm, a value that exhibits enhanced accuracy and coherence when compared to the experimental datum of 528 nm. The reaction mechanism of DNS‐Az generating DNS by H2S has been investigated too, according to the potential energy profile, we found that the fluorescent probe firstly protonated, then this organic ion broke down into DNS with the aid of a proton.

Abstract:As a signal transduction molecule in organism, H2S plays an important role in many physiological processes. In this paper, a novel H2S fluorescent probe SBD was designed based on 1,8-naphthalimide and dinitrobenzenesulfonyl group. In DMSO-PBS solution, probe SBD could recognize H2S and change the color of solution from yellow to pink, realizing the naked eye recognition for H2S. In addition, probe SBD displayed high fluorescence enhancement rate (45-fold), good selectivity, strong anti-interference ability and the detection limit was as low as 152 nM. Most importantly, probe SBD showed very low cytotoxicity and had been successfully applied to image exogenous H2S in living cells.

Abstract:Novel cyanine-based fluorescent probes for the detection of H2S were developed. The probes developed are stable under physiological conditions. The water soluble fluorescent probe 2 displayed ultrafast and specific response to H2S displaying NIR fluorescence of 115-fold turn-on with the detection limit of 11 nM without assistance of organic solvent or surfactant. Cell imaging experiments indicated that probe 2 was cell-permeable and was able to detect H2S sensitively in lysosomes. Moreover, our probe was able to detect H2S intrinsically produced H2S through enzymatic/non-enzymatic biosynthetic pathway from Cys/GSH. Moreover, we applied probe 2 to detect H2S in living mice and demonstrated the fast metabolism of H2S. Thus, probe 2 shows great promise as a reporter for H2S.

Abstract:Hydrogen sulfide (H 2 S) is traditionally considered as an ecologically toxic gas, has been proven to be the third endogenous signaling gasotransmitter and plays important roles in physiological and pathophysiological conditions. Herein, we designed and synthesized a novel "turn off" fluorescent probe for the detection of hydrogen sulfide(H 2 S) in vitro. Probe A could detect H 2 S in CH 3 CN/PBS system with significant fluorescence quenching. Probe A also showed obviously aggregation- induced emission (AIE) effects. Fluorescence titration experiments revealed probe A combine with H 2 S in a 1: 1 ratio with a detection limit of 6.54 μM. Cell imaging results demonstrated the potential applicability of probe A for H 2 S detection in living cells. The supramolecular assembly of probe A and β-cyclodextrin further revealed the interaction with H 2 S.

Abstract:Hydrogen sulfide or H2S is known as an important gasotransmitter involved in regulation of many physiological and pathological processes. Since a small amount of H2S is produced by endogenous cells in the body and microorganisms in the gastrointestinal tract (GI tract), the current detection probes are limited for their use in measurement of unpretentious change of H2S level both in vitro and in vivo. Therefore, development of a highly sensitive and responsive fluorescent probe for measurement of H2S in vivo is of great significance for investigation of H2S function and toxicity. Here, we designed a novel fluorescent probe AC-N-3 through introducing azide in 6-position of chroman dye based on the hypothesis that 6-position (para-position) of chromone may be a potential trigger of fluorescent properties. The probe AC-N-3 exhibits a better selectivity without interference from analytes, high sensitivity and little cytotoxicity. In addition, we confirmed that the conversion of electron donating group in 6-position of chromone can trigger the change of fluorescence, which can be applied to design of other fluorescent probes.

Abstract:A new Near-infrared fluorescent probe for hydrogen sulfide detection was synthesized by employing dicyanoisophorone based fluorescence dye as a fluorophore and methyl 3-(2-(carbonyl)phenyl)-2-cyanoacrylate group as the response unit. The Probe DCI-H 2 S showed a long emission wavelength (λem = 674 nm). Based on the H 2 S-induced addition–cyclization of deprotecting methyl 3-(2-(carbonyl)phenyl)-2-cyanoacrylate group, the probe DCI-H 2 S showed high selectivity, sensitivity and response speed toward hydrogen sulfide under room temperature. These numerous advantages of the probe DCI-H 2 S make it to potentially detect endogenous hydrogen sulfide in living organisms.

Abstract:As a common gaseous signaling molecule, hydrogen sulfide (H2S) plays a vital role in physiology and pathology. The development of fluorescent probes for detecting H2S has attracted widespread attention. However, most of the reported fluorescent probes with nitrobenzoxadiazole (NBD) as the recognition group have been widely used to simultaneously detect biothiols and H2S, instead of specifically detecting H2S. Herein, a novel NBD-based near-infrared (NIR) fluorescent probe named CX-N for the detection of H2S is synthesized. The selectivity of CX-N for H2S is significantly higher than that for biothiols and other potential interferences. After reacting with H2S, CX-N shows a significant increase in NIR fluorescence (75-fold), large Stokes shift (155 nm) and fast response (4 min). And the possible response mechanism of CX-N to H2S is given and confirmed by HPLC and HRMS. Based on the low cytotoxicity of CX-N, it has been used for H2S imaging in live cells and zebrafish. More importantly, CX-N has also been successfully applied for the real-time imaging of H2S in inflammatory and tumor mice based on its NIR emission, which provides a reliable platform for the specific recognition of H2S in complex biological systems.

Abstract:In this work, through the orthogonal design of two fluorophores and two recognition groups, a series of fluorescent probes were developed from the flavone derivatives for hydrogen sulfide (H 2 S). The probe FlaN-DN stood out from the primarily screening on the selectivity and response intensities. It could respond to H 2 S with both the chromogenic and fluorescent signals. Among the recent reported probes for the H 2 S detection, FlaN-DN indicated the most highlighted advantages including the rapid response (within 200 s) and the high response multiplication (over 100 folds). FlaN-DN was sensitive to the pH condition, thus could be applied to distinguish the cancer micro-environment. Moreover, FlaN-DN suggested practical capabilities including a wide linear range (0-400 μM), a relatively high sensitivity (limit of detection 0.13 μM), and high selectivity towards H 2 S. As a low cytotoxic probe, FlaN-DN achieved the imaging in living HeLa cells. FlaN-DN could detect the endogenous generation H 2 S and visualize the dose-dependent responses to the exogenous H 2 S level. This work provided a typical case of natural-sourced derivatives as functional implements, which might inspire the future investigations.

Abstract:Hydrogen sulfide (H 2 S) plays a key role in the physiology and pathology of organisms, and H 2 S in the environment is easily absorbed and harmful to health. It is of great significance to develop a probe with good selectivity, high sensitivity and good stability that can detect hydrogen sulfide inside and outside organisms. In this work, we designed a novel "turn-on" fluorescent probe CIM-SDB for the detection of H 2 S. The probe CIM-SDB used indene-carbazole as the fluorophore and 2,4-dinitrobenzenesulfonyl as the recognition site. The probe CIM-SDB exhibited high selectivity and sensitivity to H 2 S (detection limit as low as 123 nM). Moreover, the probe CIM-SDB was successfully applied to the detection of intracellular exogenous and endogenous H 2 S, and the test strips prepared by the probe CIM-SDB could realize the convenient and rapid detection of H 2 S.

Abstract:A fluorescence turn-on probe, 2-butyl-1,3-dioxo-2,3-dihydro-1H-benzo[de]isoquinolin-6-yl 2,4-dinitrobenzenesulfonate (NT-SH), has been constructed for sensing of hydrogen sulfide (H2S). NT-SH exhibited excellent detection performance including favorable water solubility, low fluorescence back-ground, high enhancement (45-fold), large linear response range (0-50 mu M) and low detection limit (80.01 nM) for H2S in aqueous. In addition, the response mechanism of NT-SH for H2S was confirmed by the theoretical calculation and mass spectral analysis. More importantly, the imaging experiments of H2S in vitro and in vivo confirmed that NT-SH had low cytotoxicity, and favorable biocompatibility. In addition, it illustrated that NT-SH was able to detected exogenous H2S in living cells and zebrafish. These results suggested that NT-SH can be act as a potential molecular tool for detecting of H2S in aqueous solution, in vitro and in vivo. (C) 2021 Published by Elsevier B.V.

Abstract:We present a new two-photon fluorescent probe (T-HS) for the detection of H2S. With the addition of hydrogen sulfide, the absorption and fluorescence spectra of the probe show regular changes. The probe exhibited favorable properties, such as large turn-on fluorescence signal, good selectivity and low cytotoxicity. Moreover, the probe T-HS was successfully used for the fluorescence imaging of H2S in live cells and zebrafish.

Abstract:A novel near-infrared fluorescent probe (L) based on a 1,4-diethyl-1,2,3,4-tetrahydro-7H-pyrano[2,3-g]quinoxalin-7-one scaffold has been synthesized and characterized. Probe L displays highly selective and sensitive recognition to H2S over various anions and biological thiols with a large Stokes shift (125 nm) in THF/H2O (6/4, v/v, Tris-HCl 10 mM, pH = 7.4). This probe exhibits turn-on fluorescence for H2S through HS- induced thiolysis of dinitrophenyl ether. Confocal laser scanning micrographs of MCF-7 cells incubated with L confirm that L is cell-permeable and can successfully detect H2S in living cells.
