Abstract:Self-assembled catalyst: A new kind of chiral supramolecular organocatalyst, self-assembled from chiral amines and poly(alkene glycol)s, was developed. The resulting self-assemblies were found to be highly efficient in the asymmetric catalysis of the unusual Diels–Alder reaction between cyclohexenones and nitrodienes, nitroenynes or nitroolefins, giving excellent chemo-, regio- and enantioselectivities (see scheme). Abstract:A cascade vinylogous 1,6-Michael addition/1,4-proton shift/aza-Michael addition/hemiaminal formation sequence of 1,2-diaza-1,3-dienes and β-substituted 2-butenals has been developed under the influence of dienamine activation of a chiral secondary amine. This method exhibits high regio- and chemoselectivity and provides an efficient and straightforward approach to bicyclic l,8-diazabicyclo[3.3.0]octane skeletons with a tetrasubstituted stereogenic center with fair to excellent enantioselectivity.

Abstract:A bimetallic relay catalysis protocol for tandem allylic C–H amination and asymmetric [2,3]-sigmatropic rearrangement has been developed with the use of an achiral Pd 0 catalyst and a chiral N , N ′-dioxide–Mg II complex in a one-pot operation. A series of anti-α-amino derivatives containing two stereogenic centers were prepared from readily available allylbenzenes and glycine pyrazolamide with good yields and high stereoselectivities. Moreover, the synthetic potential of this protocol was further demonstrated by the product transformations, and a catalytic cycle was proposed to illustrate the reaction process.

Abstract:Using terminal alkyne as a nontraditional one-carbon (C1) unit and silylborane as an external silicon pronucleophile, a relay palladium/copper-catalyzed silylative [5 + 1] benzannulation of 3-acetoxy-1,4-enynes for producing polysubstituted arylsilanes, especially including bioactive motif-based analogues, in a single reaction step through benzene ring skeleton assembly and silyl intermolecular incorporation cascades is developed. Abstract:The asymmetric rearrangement of allylic sulfilimines is an effective route to synthetic attractive targets such as allylic sulfenamides and others. The current methods are limited to chirality transfer from chiral allylic sulfilimine precursors. Herein, we report a general and fundamentally new rearrangement route accessing optically enriched allylic sulfenamides and their derivatives. The process involves a S-alkylation and an unusual S-to-N rearrangement step. The chiral nickel complex enables the transformation of a broad scope of sulfenamides and vinyl α-diazo pyrazoleamides under mild conditions. Various allylic sulfenamides have been synthesized with excellent γ-regioselectivity and enantioselectivity, which can be efficiently converted to sulfinamide and 4-aminobutenoic acid derivatives. In addition, DFT calculations demonstrate the connection between the spin state and conformation of nickel vinyl carbenoid, as well as an unknown rearrangement process.

Abstract:An asymmetric three‐component reaction of α‐H diazoacetate with sulfonamide and imine has been developed under the co‐catalyzation of Rh2(OAc)4 and a chiral phosphoric acid (CPA), which proceed through Mannich‐type interception of transient ammonium ylide. The reaction offers a convenient access to α, β‐diamine acid derivatives with two adjacent chiral centers in 59%‐74% yields with 91%‐99% ee and 92:8 to > 95:5 dr. Moreover, the generated products could be easily converted to free chiral diamines and other useful chiral motifs.

Abstract:The asymmetric Csp3-Csp3 bond formation has been a grand pursuit in synthetic chemistry. The regio- and enantionselective hydroalkylation of 1,3-diene has emerged as an appealing approach for constructing chiral allylic Csp3-Csp3 bonds. However, this method is presently confined to the use of stabilized Csp3 nucleophilic substrates. Herein, we present a nickel-catalyzed asymmetric hydroalkylation of 1,3-dienes with simple unstablized alkyl carbanion enabled by hydrazones umpolung under mild reaction conditions. A range of simple alkylated chiral allylic compounds were generated in good to excellent yields, with enantiomeric ratio (er) of up to 2:98 in forming the Csp3-Csp3 bond. The protocol is applicable to heterocycles, polyenes and unsaturated hydrazones, as well as amenable for late-stage functionalizations of various complex pharmaceuticals. Density functional theory calculations elucidate the mechanism and enantioselectivity of the reaction. An enantiocontrol model is also proposed, emphasizing the crucial role of a chiral NHC ligand in facilitating this asymmetric reaction, as revealed by the two-layer two-dimensional (2D) contour maps.

Abstract:AbstractA unique Pd‐catalyzed approach for asymmetric (4+1) annulations via cascade allylation and transient σ‐alkyl‐Pd(II) initiated methylene Csp3−H activation is reported. The enolate fragment derived from the decarboxylation of vinyl methylene carbonate is crucial to stabilize the key intermediate. These reactions enable the synthesis of various useful dihydrobenzofurans with excellent enantioselectivity, typically >95 : 5 er, and exclusive (Z)‐stereoselectivity. Compared with the well‐established annulations via Heck‐type C−H activations, this protocol showcases a conceptually new way to generate σ‐alkyl‐Pd(II) species that could initiate challenging asymmetric Csp3−H activations.

Abstract:We herein describe an asymmetric α-allylic allenylation of β-ketocarbonyls and aldehydes with 1,3-enynes. A synergistic chiral primary amine/Pd catalyst was identified to facilitate the utilization of 1,3-enynes as atom-economic and achiral allene precursors. The synergistic catalysis enables the construction of all-carbon quaternary centers-tethered allenes bearing non-adjacent 1,3-axial central stereogenic centers in high level of diastereo- and enantio-selectivity. By switching the configurations of ligands and aminocatalysts, diastereodivergence can be achieved and any of the four diastereoisomers can be accessed in high diastereo- and enantio- selectivity.
