Abstract:1,2-Diaminocyclohexane-hexanedioic acid has been demonstrated to catalyze the asymmetric aldol reactions of cycloketones and various isatin derivatives efficiently in MeOH-H2O. The corresponding products were obtained in good yields (70-90%) with high diastereoselectivity (up to 99:1 anti/syn) and enantioselectivity (up to 99% ee).

Abstract:Catalytic asymmetric 1,3-dipolar cycloaddition reactions of activated isocyanides with various 2 pi dipolarophiles have been intensively studied, affording a wide range of enantioenriched five-membered N-heterocycles. In sharp contrast, the catalytic enantioselective higher-order cycloaddition of activated isocyanides has not been achieved yet. We present here our recent work on the development of an unprecedented silver-catalyzed highly diastereo- and enantioselective [3+3] cycloaddition of activated isocyanides with azomethine imines. This method features high efficiency, good to excellent stereocontrol, wide substrate scope, as well as operational simplicity. It is also noteworthy that the same catalytic system was proved to be suitable for not only the late-stage functionalization of complex bioactive molecules but also the kinetic resolution of racemic azomethine imines.

Abstract:The search for new catalysts with superior performance and broad applicability remains a central theme in asymmetric organocatalysis. Abstract:We report a cobalt-catalyzed desymmetrizing isomerization of exo-cyclic alkenes to generate chiral 1-methylcyclohexene derivatives with good yields and enantioselectivities. A novel chiral thiazolinyl iminoquinoline ligand and its cobalt complex were designed and synthesized to control the establishment of tertiary or quaternary carbon centers at a remote position. This protocol is operationally simple, and a model for the stereochemical outcome has been proposed.

Abstract:Compared with the well-explored enamine catalysis with secondary amines, the development of efficient enamine-based primary amine catalysts has remained as an elusive goal until recently. We present herein that a simple chiral primary-tertiary diamine 1d in combination with TfOH acts as an efficient enamine-based primary amine catalyst. The 1d-TfOH catalytic system could effectively catalyze the asymmetric direct aldol reaction with broad substrate scope as well as high regio- and diastereoselectivity and enantioselectivity under ambient temperature. Significantly, the reactions accommodate the synthetically important but challenging substrates, such as linear aliphatic ketones, with high regioselectivity and unprecedented syn diastereoselectivity. These results are in sharp contrast with the secondary amine mediated similar reactions wherein anti diastereoselectivity was normally observed.

Abstract:The authors present an asymmetric aldol reaction promoted by β-cyclodextrin-supported chiral primary amine 1. Supramolecular recognition of the cyclodextrin moiety generates a synergistic effect on catalytic activity and enantioselectivity. A slightly acidic environment was found to be optimal for the catalysis. Substrate binding mode and conformational change of the catalyst during the reaction course was confirmed using a sophisticated spectroscopic analysis.

Abstract:In prime condition: The hydrolysis of a chiral diimine precursor can be carried out by a Brønsted acid to form an in situ primary amine-imine intermediate as a bifunctional primary aminocatalyst, which promoted direct asymmetric aldol reactions between α-keto esters and ketones in high yields (up to 88 %) and with excellent enantioselectivities (see scheme; R 1, R 2, R 4=alkyl; R 3=aryl).
