Abstract:One of the most dramatic changes occurring on our planet is the ever-increasing extensive use of artificial light at night, which drastically altered the environment to which nocturnal animals are adapted. Such light pollution has been identified as a driver in the dramatic insect decline of the past years. One nocturnal species group experiencing marked declines are moths, which play a key role in food webs and ecosystem services such as plant pollination. Moths can be easily monitored within the illuminated area of a streetlight, where they typically exhibit disoriented behavior. Yet, little is known about their behavior beyond the illuminated area. Harmonic radar tracking enabled us to close this knowledge gap. We found a significant change in flight behavior beyond the illuminated area of a streetlight. A detailed analysis of the recorded trajectories revealed a barrier effect of streetlights on lappet moths whenever the moon was not available as a natural celestial cue. Furthermore, streetlights increased the tortuosity of flights for both hawk moths and lappet moths. Surprisingly, we had to reject our fundamental hypothesis that most individuals would fly toward a streetlight. Instead, this was true for only 4% of the tested individuals, indicating that the impact of light pollution might be more severe than assumed to date. Our results provide experimental evidence for the fragmentation of landscapes by streetlights and demonstrate that light pollution affects movement patterns of moths beyond what was previously assumed, potentially affecting their reproductive success and hampering a vital ecosystem service.

Abstract:Crickets serve as a well-established model organism in biological research spanning various fields, such as behavior, physiology, neurobiology, and ecology. Cricket circadian behavior was first reported over a century ago and prompted a wealth of studies delving into their chronobiology. Circadian rhythms have been described in relation to fundamental cricket behaviors, encompassing stridulation and locomotion, but also in hormonal secretion and gene expression. Here we review how changes in illumination patterns and light intensity differentially impact the different cricket behaviors as well as circadian gene expression. We further describe the cricket's circadian pacemaker. Ample anatomical manipulations support the location of a major circadian pacemaker in the cricket optic lobes and another in the central brain, possibly interconnected via signaling of the neuropeptide PDF. The cricket circadian machinery comprises a molecular cascade based on two major transcriptional/translational negative feedback loops, deviating somewhat from the canonical model of Drosophila and emphasizing the significance of exploring alternative models. Finally, the nocturnal nature of crickets has provided a unique avenue for investigating the repercussions of artificial light at night on cricket behavior and ecology, underscoring the critical role played by natural light cycles in synchronizing cricket behaviors and populations, further supporting the use of the cricket model in the study of the effects of light on insects. Some gaps in our knowledge and challenges for future studies are discussed.

Abstract:Abstract To counteract insect decline, it is essential to understand the underlying causes, especially for key pollinators such as nocturnal moths whose ability to orientate can easily be influenced by ambient light conditions. These comprise natural light sources as well as artificial light, but their specific relevance for moth orientation is still unknown. We investigated the influence of moonlight on the reproductive behavior of privet hawkmoths ( Sphinx ligustri ) at a relatively dark site where the Milky Way was visible while the horizon was illuminated by distant light sources and skyglow. We show that male moths use the moon for orientation and reach females significantly faster with increasing moon elevation. Furthermore, the choice of flight direction depended on the cardinal position of the moon but not on the illumination of the horizon caused by artificial light, indicating that the moon plays a key role in the orientation of male moths.

Abstract:The relationship between light and the circadian system has long been a matter of discussion. Many studies have focused on entrainment of light with the internal biological clock. Light also functions as an environmental stimulus that affects the physiology and behaviour of animals directly. In this study, we used light as an unexpected stimulus for flies at different circadian times. We found that wildtype flies showed circadian changes in light-induced locomotion responses. Elevation of locomotor activity by light occurred during the subjective night, and performance in response to light pulses declined to trough during the subjective day. Moreover, arrhythmic mutants lost the rhythm of locomotion responses to light, with promotion of activity by light in timeless(01) mutants and inhibition of activity by light in Clock(ar) mutants. However, neither ablation of central oscillators nor disturbance of the functional clock inside compound eyes was sufficient to disrupt the rhythm of light responses. We show that, compound eyes, which have been identified as the control point for normal masking (promotion of activity by light), are sufficient but not necessary for paradoxical masking (suppression of activity by light) under high light intensity. This, taken together with the clear difference of light responses in wildtype flies, suggests that two different masking mechanisms may underlie the circadian modulation of light-induced locomotion responses.

Abstract:As a goal-directed behavior, foraging for nectar functions on the basis of a sequence of innate stereotyped movements mainly regulated by sensory input. The operation of this inherited program is shaped by selective pressures acting on its efficiency, which is largely dependent upon the way the system handles sensory information. Flowers offer a wealth of signals, from odors acting as distant attractants, to colors eliciting approximation and feeding responses, to textures guiding feeding responses toward a reservoir of nectar. Thus, animals use different signals in the regulation of particular motor outputs. Nevertheless, the use of these sensory signals can be user-specific (e.g. species, motivation, experience, learning) as well as context-dependent (e.g. spatiotemporal patterns of stimulation, availability of signals, multimodal integration). The crepuscular/nocturnal hawkmoths Manduca sexta experience a wide range of illuminations during their foraging activity, which raises the question of how these environmental changes might affect the use of two important floral signals, odor and visual display. In a flight cage, we explored the use of these signals under different illuminances. Under conditions of starlight and crescent moonlight, moths showed very low levels of responsiveness to unscented feeders (artificial flowers). However, responsiveness was recovered either by increasing illumination, or by offering olfactory signals. Additionally, we recorded a bias toward white over blue feeders under dim conditions, which disappeared with increasing illumination. We discuss how this kind of experimental manipulation may provide insights to the study of how innate behavioral programs, and their underlying neural substrates, overcome selective forces imposed by the uncertainty of natural, ever-changing environments.

Abstract:Artificial light at night (ALAN) is predicted to have far-reaching consequences for natural ecosystems given its influence on organismal physiology and behaviour, species interactions and community composition. Movement and predation are fundamental ecological processes that are of critical importance to ecosystem functioning. The natural movements and foraging behaviours of nocturnal invertebrates may be particularly sensitive to the presence of ALAN. However, we still lack evidence of how these processes respond to ALAN within a community context. We assembled insect communities to quantify their movement activity and predation rates during simulated Moon cycles across a gradient of diffuse night-time illuminance including the full range of observed skyglow intensities. Using radio frequency identification, we tracked the movements of insects within a fragmented grassland Ecotron experiment. We additionally quantified predation rates using prey dummies. Our results reveal that even low-intensity skyglow causes a temporal shift in movement activity from day to night, and a spatial shift towards open habitats at night. Changes in movement activity are associated with indirect shifts in predation rates. Spatio-temporal shifts in movement and predation have important implications for ecological networks and ecosystem functioning, highlighting the disruptive potential of ALAN for global biodiversity and the provision of ecosystem services. Abstract:Vehicle headlights create pulsed artificial light at night (pALAN) that is unpredictable, intense and extends into previously dark areas. Nocturnal insects often have remarkable low-light vision, but their slow pupillary light responses may leave them vulnerable to pALAN, which has important ecological consequences. To test this, we exposed nocturnal moths—important pollinators and prey—to four pALAN treatments. These comprised ‘cool’ and ‘warm’ lights, either emitted from phosphor-coated light-emitting diodes (LEDs) or RGB (red-green-blue) LEDs, matched in colour (CCT) and intensity to human vision. We assessed the initial behavioural response, likely crucial to the survival of an organism, of 428 wild-caught moths comprising 64 species. We found that exposure to a cool phosphor-coated LED light pulse increased instances of erratic flight and flight-to-light that are likely detrimental as they increase the risks of impact with a vehicle, predation or excess energy expenditure. Our findings suggest that pALAN can cause a wide range of behavioural responses in nocturnal moths, but that the most harmful effects could be minimized by reversing the current shift towards high CCT (cool) phosphor-coated LED car headlights. Lower CCT or RGB alternatives are likely to provide benefits for road safety while reducing ecological harm.

Abstract:Temporal ecological niche partitioning is an underappreciated driver of speciation. While insects have long been models for circadian biology, the genes and circuits that allow adaptive changes in diel-niches remain poorly understood. We compared gene expression in closely related day- and night-active non-model wild silk moths, with otherwise similar ecologies. Using an ortholog-based pipeline to compare RNA-Seq patterns across two moth species, we find over 25 pairs of gene orthologs showing differential expression. Notably, the gene disco, involved in circadian control, optic lobe and clock neuron development in Drosophila , shows robust adult circadian mRNA cycling in moth heads. Disco is highly conserved in moths and has additional zinc-finger domains with specific nocturnal and diurnal mutations. We propose disco as a candidate gene for the diversification of temporal diel-niche in moths.

Abstract:Night-flying pollinators, such as hawkmoths, are particularly vulnerable to the global spread of urban artificial lighting which is changing nighttime environments worldwide, impacting organisms and their interactions. Incident light quality can alter flower and leaf color perception by insects, depending on the emission spectra of light sources and the spectral sensitivity of insects. We asked, using Manduca sexta visual models, whether color contrast against natural backgrounds is altered by artificial lights for flowers and leaves of 16 plant species with an estimated long history of coevolution with hawkmoth pollinators. Specifically, we compared the perception of flowers and leaves by hawkmoths under artificial lights, including light-emitting diodes (5000 K LED), mercury vapor (MV), and high-pressure sodium (HPS) artificial lights, with the perception under natural illuminations. The models we implemented estimate that LED and HPS lighting change hawkmoth perception of flowers and leaves, with color loci appearing nearer to each other in hawkmoths perceptual space than they would be under natural nighttime conditions. Receptor Noise Limited models show that under the different lighting conditions hawkmoths would still discriminate flowers from their leaves in most but not all species. Consequently, artificial lights likely alter perception by hawkmoths of floral and leaf signals possibly affecting interactions and fitness of plants and pollinators. Our results emphasize the intricate and insidious ways in which human-made environments impact species interactions. Further studies should confirm whether light pollution represents a novel selective force to nocturnal interacting partners as emerging evidence suggests. Addressing the effects of artificial lighting is crucial for designing infrastructure development strategies that minimize these far-reaching effects on ecosystem functioning.

Neural Summation in the Hawkmoth Visual System Extends the Limits of Vision in Dim Light

Abstract:Most of the world's animals are active in dim light and depend on good vision for the tasks of daily life. Many have evolved visual adaptations that permit a performance superior to that of manmade imaging devices [1]. In insects, a major model visual system, nocturnal species show impressive visual abilities ranging from flight control [2, 3], to color discrimination [4, 5], to navigation using visual landmarks [6-8] or dim celestial compass cues [9, 10]. In addition to optical adaptations that improve their sensitivity in dim light [11], neural summation of light in space and time-which enhances the coarser and slower features of the scene at the expense of noisier finer and faster features-has been suggested to improve sensitivity in theoretical [12-14], anatomical [15-17], and behavioral [18-20] studies. How these summation strategies function neurally is, however, presently unknown. Here, we quantified spatial and temporal summation in the motion vision pathway of a nocturnal hawkmoth. We show that spatial and temporal summation combine supralinearly to substantially increase contrast sensitivity and visual information rate over four decades of light intensity, enabling hawkmoths to see at light levels 100 times dimmer than without summation. Our results reveal how visual motion is calculated neurally in dim light and how spatial and temporal summation improve sensitivity while simultaneously maximizing spatial and temporal resolution, thus extending models of insect motion vision derived predominantly from diurnal flies. Moreover, the summation strategies we have revealed may benefit manmade vision systems optimized for variable light levels [21].
Nocturnal city lighting elicits a macroscale response from an insect outbreak population

Abstract:Anthropogenic environmental change affects organisms by exposing them to enhanced sensory stimuli that can elicit novel behavioural responses. A pervasive feature of the built environment is artificial nocturnal lighting, and brightly lit urban areas can influence organism abundance, distribution and community structure within proximate landscapes. In some cases, the attractive or disorienting effect of artificial light at night can draw animals into highly unfavourable habitats, acting as a macroscale attractive ecological sink. Despite their significance for animal ecology, identifying cases of these phenomena and determining their effective scales and the number of organisms impacted remains challenging. Using an integrated set of remote-sensing observations, we quantify the effect of a large-scale attractive sink on nocturnal flights of an outbreak insect population in Las Vegas, USA. At the peak of the outbreak, over 45 million grasshoppers took flight across the region, with the greatest numbers concentrating over high-intensity city lighting. Patterns of dusk ascent from vegetated habitat toward urban areas suggest a daily pull toward a time-varying nocturnal attractive sink. The strength of this attractor varies with grasshopper density. These observations provide the first macroscale characterization of the effects of nocturnal urban lighting on the behaviour of regional insect populations and demonstrate the link between insect perception of the built environment and resulting changes in spatial and movement ecology. As human-induced environmental change continues to affect insect populations, understanding the impacts of nocturnal light on insect behaviour and fitness will be vital to developing robust large-scale management and conservation strategies.

Abstract:Moths and other insects are attracted by artificial light sources. This flight-to-light behaviour disrupts their general activity focused on finding resources, such as mating partners, and increases predation risk. It thus has substantial fitness costs. In illuminated urban areas, spindle ermine moths Yponomeuta cagnagella were reported to have evolved a reduced flight-to-light response. Yet, the specific mechanism remained unknown, and was hypothesized to involve either changes in visual perception or general flight ability or overall mobility traits. Here, we test whether spindle ermine moths from urban and rural populations—with known differences in flight-to-light responses—differ in flight-related morphological traits. Urban individuals were found to have on average smaller wings than rural moths, which in turn correlated with a lower probability of being attracted to an artificial light source. Our finding supports the reduced mobility hypothesis, which states that reduced mobility in urban areas is associated with specific morphological changes in the flight apparatus.

Abstract:Seasonal change in daylength (photoperiod) is widely used by insects to regulate temporal patterns of development and behaviour, including the timing of diapause (dormancy) and migration. Flexibility of the photoperiodic response is critical for rapid shifts to new hosts, survival in the face of global climate change and to reproductive isolation. At the same time, the daily circadian clock is also essential for development, diapause and multiple behaviours, including correct flight orientation during long-distance migration. Although studied for decades, how these two critical biological timing mechanisms are integrated is poorly understood, in part because the core circadian clock genes are all transcription factors or regulators that are able to exert multiple effects throughout the genome. In this chapter, we discuss clocks in the wild from the perspective of diverse insect groups across eco-geographic contexts from the Antarctic to the tropical regions of Earth. Application of the expanding tool box of molecular techniques will lead us to distinguish universal from unique mechanisms underlying the evolution of circadian and photoperiodic timing, and their interaction across taxonomic and ecological contexts represented by insects. Abstract:Ever-present in our environments, light entrains circadian rhythms over long timescales, influencing daily activity patterns, health and performance. Increasing evidence indicates that light also acts independently of the circadian system to directly impact physiology and behaviour, including cognition. Exposure to light stimulates brain areas involved in cognition and appears to improve a broad range of cognitive functions. However, the extent of these effects and their mechanisms are unknown. Intrinsically photosensitive retinal ganglion cells (ipRGCs) have emerged as the primary conduit through which light impacts non-image-forming behaviours and are a prime candidate for mediating the direct effects of light on cognition. Here, we review the current state of understanding of these effects in humans and mice, and the tools available to uncover circuit-level and photoreceptor-specific mechanisms. We also address current barriers to progress in this area. Current and future efforts to unravel the circuits through which light influences cognitive functions may inform the tailoring of lighting landscapes to optimize health and cognitive function.

Abstract:Abstract Although diurnality is widespread across Lepidoptera and has evolved many times independently, its causes and ecological implications are yet poorly understood. The ‘Salient Aroma Hypothesis’ (SAH) postulates that diurnal insect herbivores are overall more specialised in dietary breadth than species active at night. It is furthermore assumed that diurnality evolved more frequently in species that live in cooler environments. Using European geometrid moths as a model group, we tested whether diurnal activity in adults is associated with an increased larval dietary breadth as predicted by the ‘Salient Aroma Hypothesis.’ We further investigated whether species that exclusively occur in colder regions or whose flight period is restricted to cool seasons are more likely to exhibit a diurnal flight activity. Contrary to expectation, we found no consistent differences in larval dietary breadth between diurnal and nocturnal species, and thus no support for the ‘Salient Aroma Hypothesis.’ Diurnal activity occurred more frequently in species restricted to cold regions, but not in species restricted to cool seasons. We conclude that diurnality could serve as an advantageous adaptation in cold environments, depending on further factors such as resource availability or predation pressure, but has no immediate consequences for larval dietary breadth.

Abstract:Microglia are increasingly recognized to contribute to brain health and disease. Preclinical studies using laboratory rodents are essential to advance our understanding of the physiological and pathophysiological roles of these cells in the central nervous system. Rodents are nocturnal animals, and they are mostly maintained in a defined light-dark cycle within animal facilities, with many laboratories investigating the molecular and functional profiles of microglia exclusively during the animals' light (sleep) phase. However, only a few studies have considered possible differences in microglial functions between the active and sleep phases. Based on initial evidence suggesting that microglial intrinsic clock genes can affect their phenotypes, we sought to investigate differences in transcriptional, proteotype and functional profiles of microglia between light (sleep) and dark (active) phases, and how these changes are affected in pathological models. We found marked transcriptional and proteotype differences between microglia harvested from male mice during the light or dark phase. Amongst others, these differences related to genes and proteins associated with immune responses, motility, and phagocytosis, which were reflected by functional alterations in microglial synaptic pruning and response to bacterial stimuli. Possibly accounting for such changes, we found RNA and protein regulation in SWI/SNF and NuRD chromatin remodeling complexes between light and dark phases. Importantly, we also show that the time of microglial sample collection influences the nature of microglial transcriptomic changes in a model of immune-mediated neurodevelopmental disorders. Our findings emphasize the importance of considering diurnal factors in studying microglial cells and indicate that implementing a circadian perspective is pivotal for advancing our understanding of their physiological and pathophysiological roles in brain health and disease.
The impact of artificial light at night on nocturnal insects: A review and synthesis

Abstract:In recent decades, advances in lighting technology have precipitated exponential increases in night sky brightness worldwide, raising concerns in the scientific community about the impact of artificial light at night (ALAN) on crepuscular and nocturnal biodiversity. Long-term records show that insect abundance has declined significantly over this time, with worrying implications for terrestrial ecosystems. The majority of investigations into the vulnerability of nocturnal insects to artificial light have focused on the flight-to-light behavior exhibited by select insect families. However, ALAN can affect insects in other ways as well. This review proposes five categories of ALAN impact on nocturnal insects, highlighting past research and identifying key knowledge gaps. We conclude with a summary of relevant literature on bioluminescent fireflies, which emphasizes the unique vulnerability of terrestrial light-based communication systems to artificial illumination. Comprehensive understanding of the ecological impacts of ALAN on diverse nocturnal insect taxa will enable researchers to seek out methods whereby fireflies, moths, and other essential members of the nocturnal ecosystem can coexist with humans on an increasingly urbanized planet.
