Abstract:Male moths detect and find their mates using species-specific sex pheromones emitted by conspecific females. Olfaction plays a vital role in this behavior. Since the first discovery of an insect sex pheromone from the silkmoth Bombyx mori, great efforts have been spent on understanding the sensing of the pheromones in vivo. Much progress has been made in elucidating the molecular mechanisms that mediate chemoreception in insects in the past few decades. In this review, we focus on pheromone reception and detection in moths, from the molecular to the behavioral level. We trace the information pathway from the capture of pheromone by male antennae, binding and transportation to olfactory receptor neurons, receptor activation, signal transduction, molecule inactivation, through brain processing and behavioral response. We highlight the impact of recent studies and also provide our insights into pheromone processing.

Abstract:Many insects are capable of developing into either long-winged or short-winged (or wingless) morphs, which enables them to rapidly match heterogeneous environments. Thus, the wing polymorphism is an adaptation at the root of their ecological success. Wing polymorphism is orchestrated at various levels, starting with the insect's perception of environmental cues, then signal transduction and signal execution, and ultimately the transmitting of signals into physiological adaption in accordance with the particular morph produced. Juvenile hormone and ecdysteroid pathways have long been proposed to regulate wing polymorphism in insects, but rigorous experimental evidence is lacking. The breakthrough findings of ecdysone receptor regulation on transgenerational wing dimorphism in the aphid Acyrthosiphon pisum and of insulin signaling in the planthopper Nilaparvata lugens greatly broaden our understanding of wing polymorphism at the molecular level. Recently, the advent of high-throughput sequencing coupled with functional genomics provides powerful genetic tools for future insights into the molecular bases underlying wing polymorphism in insects.
A Dynamic Optical Signal in a Nocturnal Moth

Abstract:The wings of butterflies and moths generate some of the most spectacular visual displays observed in nature [1-3]. Particularly striking effects are seen when light interferes with nanostructure materials in the wing scales, generating bright, directional colors that often serve as dynamic visual signals [4]. Structural coloration is not known in night-flying Lepidoptera, yet here we show a highly unusual form of wing coloration in a nocturnal, sexually dimorphic moth, Eudocima materna (Noctuidae). Males feature three dark wing patches on the dorsal forewings, and the apparent size of these patches strongly varies depending on the angle of the wing to the viewer. These optical special effects are generated using specialized wing scales that are tilted on the wing and behave like mirrors. At near-normal incidence of light, these "mirror scales" act as thin-film reflectors to produce a sparkly effect, but when light is incident at ∼20°-30° from normal, the reflectance spectrum is dominated by the diffuse scattering of the underlying, black melanin-containing scales, causing a shape-shifting effect. The strong sexual dimorphism in the arrangement and architecture of the scale nanostructures suggests that these patterns might function for sexual signaling. Flickering of the male's wings would yield a flashing, supernormal visual stimulus [5] to a viewer located 20°-30° away from the vertical, while being invisible to a viewer directly above the animal. Our findings reveal a novel use of structural coloration in nature that yields a dynamic, time-dependent achromatic optical signal that may be optimized for visual signaling in dim light.
Identification of a Female-Produced Sex Attractant Pheromone of the Winter Firefly, Photinus corruscus Linnaeus (Coleoptera: Lampyridae)

Abstract:Firefly flashes are well-known visual signals used by these insects to find, identify, and choose mates. However, many firefly species have lost the ability to produce light as adults. These "unlighted" species generally lack developed adult light organs, are diurnal rather than nocturnal, and are believed to use volatile pheromones acting over a distance to locate mates. While cuticular hydrocarbons, which may function in mate recognition at close range, have been examined for a handful of the over 2000 extant firefly species, no volatile pheromone has ever been identified. In this study, using coupled gas chromatography - electroantennographic detection, we detected a single female-emitted compound that elicited antennal responses from wild-caught male winter fireflies, Photinus corruscus . The compound was identified as (1 S )- exo -3-hydroxycamphor (hydroxycamphor). In field trials at two sites across the species' eastern North American range, large numbers of male P. corruscus were attracted to synthesized hydroxycamphor, verifying its function as a volatile sex attractant pheromone. Males spent more time in contact with lures treated with synthesized hydroxycamphor than those treated with solvent only in laboratory two-choice assays. Further, using single sensillum recordings, we characterized a pheromone-sensitive odorant receptor neuron in a specific olfactory sensillum on male P. corruscus antennae and demonstrated its sensitivity to hydroxycamphor. Thus, this study has identified the first volatile pheromone and its corresponding sensory neuron for any firefly species , and provides a tool for monitoring P. corruscus populations for conservation and further inquiry into the chemical and cellular bases for sexual communication among fireflies.

Abstract:Mating swarms of flying male insects are a form of collective behaviour, albeit one different from flocks and schools as they do not display ordered collective movements. In recent years, much progress has been made in uncovering the emergent mechanical-like and thermodynamic-like of such swarms. Nonetheless, two basic properties of this swarming behaviour remain unexplained. Namely, why do individual insects have erratic rather than regular flight patterns? And why are the swarms elliptical rather than circular? Here I account for this seemingly undue complexity. I show that regular flight patterns weaken an individual's attraction to the swarm centre, making swarms less resilient to the presence of environmental disturbances. I then show that the elliptical shape of swarms of the non-biting midge Chironomus riparius optimizes the trade-off between maximizing swarm size (target size for females) and maximizing swarm stability. Finally, I show that the observed excess velocity kurtosis of swarming C. riparius maximizes swarm cohesiveness. Taken together the new results provide the first tentative evidence for fine-tuning in insect mating swarms driven by selection pressure for advantageous behaviours.

Abstract:Moths and other insects are attracted by artificial light sources. This flight-to-light behaviour disrupts their general activity focused on finding resources, such as mating partners, and increases predation risk. It thus has substantial fitness costs. In illuminated urban areas, spindle ermine moths Yponomeuta cagnagella were reported to have evolved a reduced flight-to-light response. Yet, the specific mechanism remained unknown, and was hypothesized to involve either changes in visual perception or general flight ability or overall mobility traits. Here, we test whether spindle ermine moths from urban and rural populations—with known differences in flight-to-light responses—differ in flight-related morphological traits. Urban individuals were found to have on average smaller wings than rural moths, which in turn correlated with a lower probability of being attracted to an artificial light source. Our finding supports the reduced mobility hypothesis, which states that reduced mobility in urban areas is associated with specific morphological changes in the flight apparatus.

Abstract:Visual behavior of insects has long been studied, but it is only recently that a wide variety of genetic tools has become available for its analysis. Perhaps the most basic visual behaviour is phototaxis, locomotion towards a source of light. It is known in many insects and has been studied for over a century but the neural network underlying it is little understood. We recently described in the fruit fly Drosophila how different photoreceptor types contribute to phototaxis. By blocking subsets of them we showed that at least four of the five types are involved. In this short review, we compare phototactic behaviour in fruit flies and other insects (especially honeybees), and discuss what is known about the underlying neural circuitry. :

Switching attraction to inhibition: mating-induced reversed role of sex pheromone in an insect

Abstract:In the moth, Agrotis ipsilon, newly mated males cease to be attracted to the female-produced sex pheromone, preventing them from re-mating until the next night, by which time they would have refilled their reproductive glands for a potential new ejaculate. The behavioural plasticity is accompanied by a decrease in neuron sensitivity within the primary olfactory centre, the antennal lobe (AL). However, it was not clear whether the lack of the sexually guided behaviour results from the absence of sex pheromone detection in the ALs, or if they ignore it in spite of detection, or if the sex pheromone itself inhibits attraction behaviour after mating. To test these hypotheses, we performed behavioural tests and intracellular recordings of AL neurons to non-pheromonal odours (flower volatiles), different doses of sex pheromone and their mixtures in virgin and newly mated males. Our results show that, although the behavioural and AL neuron responses to flower volatiles alone were similar between virgin and mated males, the behavioural response of mated males to flower odours was inhibited by adding pheromone doses above the detection threshold of central neurons. Moreover, we show that the sex pheromone becomes inhibitory by differential central processing: below a specific threshold, it is not detected within the AL; above this threshold, it becomes inhibitory, preventing newly mated males from responding even to plant odours. Mated male moths have thus evolved a strategy based on transient odour-selective central processing, which allows them to avoid the risk-taking, energy-consuming search for females and delay re-mating until the next night for a potential new ejaculate.
Shedding light with harmonic radar: Unveiling the hidden impacts of streetlights on moth flight behavior

Abstract:One of the most dramatic changes occurring on our planet is the ever-increasing extensive use of artificial light at night, which drastically altered the environment to which nocturnal animals are adapted. Such light pollution has been identified as a driver in the dramatic insect decline of the past years. One nocturnal species group experiencing marked declines are moths, which play a key role in food webs and ecosystem services such as plant pollination. Moths can be easily monitored within the illuminated area of a streetlight, where they typically exhibit disoriented behavior. Yet, little is known about their behavior beyond the illuminated area. Harmonic radar tracking enabled us to close this knowledge gap. We found a significant change in flight behavior beyond the illuminated area of a streetlight. A detailed analysis of the recorded trajectories revealed a barrier effect of streetlights on lappet moths whenever the moon was not available as a natural celestial cue. Furthermore, streetlights increased the tortuosity of flights for both hawk moths and lappet moths. Surprisingly, we had to reject our fundamental hypothesis that most individuals would fly toward a streetlight. Instead, this was true for only 4% of the tested individuals, indicating that the impact of light pollution might be more severe than assumed to date. Our results provide experimental evidence for the fragmentation of landscapes by streetlights and demonstrate that light pollution affects movement patterns of moths beyond what was previously assumed, potentially affecting their reproductive success and hampering a vital ecosystem service.

Abstract:Abstract Sexual communication in nocturnal fireflies has been presumed to be facilitated exclusively by bioluminescence, which has replaced the pheromones used by diurnal species. Although some fireflies are suspected of using combined signalling, this view has so far not been supported by empirical evidence, and these species have been viewed more as outliers. In this study, we have investigated the visual and olfactory capability of sexual communication of a night-active glow-worm firefly Lamprohiza splendidula (Linnaeus, 1767). We recorded and categorized the behaviour of searching males in reaction to four different experimental treatments, in which the females could be located based solely on visual or olfactory cues or a combination of the two. The pheromone-only treatment attracted significantly more males than the control, thus confirming pheromone utilization in sexual communication of this species. This work represents the first direct evidence of combined pheromone and bioluminescence use in fireflies and refutes the hypothesis of exclusive use of light in courtship of the nocturnal species. We theorize that this life strategy is likely to be inherent in the whole genus and possibly common in all glow-worm firefly species.

Abstract:In spite of the crucial role it is believed to play in nature, fluorescence in natural organisms remains under-investigated from optical, chemical and biological perspectives. One example is the transparent wings of insects from the order Hemiptera: fluorescence emission has so far not been reported in any of the 3,000 described species of the superfamily Cicadoidea. The phenomenon has however been investigated in several butterfly and moth species, in which the coloured wing scales are known to embed fluorophores such as papiliochrome II. Photonic structures present in these scales may mediate the associated light emission. Fluorescence has also been described in the vein joints and the membrane of the transparent wings of damselflies and dragonflies. This emission was attributed to the presence of autofluorescent proteins such as resilin that gives rise to low stiffness and high strain in the biological tissues, allowing passive wing deformations. In this article, we characterised the fluorescence emission from the transparent wings of the grey cicada \textit{Cicada orni} (Linnaeus, 1758) and the common cicada \textit{Lyristes (Tibicen) plebejus} (Scopoli, 1763), using linear and nonlinear optical methods. We compared their properties with the fluorescence emission from two other insects exhibiting transparent wings, namely, \textit{Hemaris fuciformis} (Linnaeus, 1758) commonly known as the broad-bordered bee hawk-moth and the Bornean damselfly \textit{Vestalis amabilis} Lieftinck, 1965. These fluorescent behaviours were optically characterised by one- and multi-photon microscopy and spectrometry, allowing us to infer the symmetry of the material as well as the biological role of the fluorophores in the visual perception of insects and to explain the high absorption by the material in the range 300-400~nm.

Dim Red Light During Scotophase Enhances Mating of a Moth Through Increased Male Antennal Sensitivity Against the Female Sex Pheromone

Abstract:Insects are behaviorally and physiologically affected by different light conditions, including photoperiod, light intensity, and spectrum. Light at night has important influences on nocturnal insects, including most moth species. Moth copulation and mating usually occur at night. Although a few studies examine changes in insect mating under artificial light at night, detailed influences of light, such as that of monochromatic light, on moth mating remain largely unknown. In this study, on the basis of long-term insects rearing experience, dim red light (spectrum range: 610-710nm, with a peak at 660nm; 2.0 Lux) during scotophase was hypothesized to enhance mating in the yellow peach moth, Conogethes punctiferalis. To test the hypothesis, the mating of moths under dim red, blue, and white lights during scotophase was observed. Under the dim red light, the enhancement of mating in C. punctiferalis was observed. In addition, the electroantennografic response of males against the female sex pheromone increased with red light treatment during scotophase. In an analysis of the differentially expressed genes in the antennae of males under red light and dark conditions, the expression levels of two odorant-binding protein (OBP) genes, CpunOBP2 and CpunPBP5, were up-regulated. Two genes were then expressed in Escherichia coli, and the recombinant proteins showed strong binding to female pheromone components in fluorescence-binding assays. Thus, the results of this study indicated that dim red light at night enhanced the mating of C. punctiferalis. One of the mechanisms for the enhancement was probably an increase in the antennal sensitivity of males to the female sex pheromone under red light that was caused by increases in the expression levels of pheromone-binding protein genes in male antennae.
Neural Summation in the Hawkmoth Visual System Extends the Limits of Vision in Dim Light

Abstract:Most of the world's animals are active in dim light and depend on good vision for the tasks of daily life. Many have evolved visual adaptations that permit a performance superior to that of manmade imaging devices [1]. In insects, a major model visual system, nocturnal species show impressive visual abilities ranging from flight control [2, 3], to color discrimination [4, 5], to navigation using visual landmarks [6-8] or dim celestial compass cues [9, 10]. In addition to optical adaptations that improve their sensitivity in dim light [11], neural summation of light in space and time-which enhances the coarser and slower features of the scene at the expense of noisier finer and faster features-has been suggested to improve sensitivity in theoretical [12-14], anatomical [15-17], and behavioral [18-20] studies. How these summation strategies function neurally is, however, presently unknown. Here, we quantified spatial and temporal summation in the motion vision pathway of a nocturnal hawkmoth. We show that spatial and temporal summation combine supralinearly to substantially increase contrast sensitivity and visual information rate over four decades of light intensity, enabling hawkmoths to see at light levels 100 times dimmer than without summation. Our results reveal how visual motion is calculated neurally in dim light and how spatial and temporal summation improve sensitivity while simultaneously maximizing spatial and temporal resolution, thus extending models of insect motion vision derived predominantly from diurnal flies. Moreover, the summation strategies we have revealed may benefit manmade vision systems optimized for variable light levels [21].

Abstract:Animals employ different sexual signal modes (e.g. visual, acoustic, chemical) in different environments and behavioural contexts. If sensory structures are costly, then evolutionary shifts in primary signal mode should be associated with changes in sensor morphology. Further, sex differences are expected if male and female signalling behaviours differ. Fireflies are known for their light displays, but many species communicate exclusively with pheromones, including species that recently lost their light signals. We performed phylogenetically controlled analyses of male eye and antenna size in 46 North American taxa, and found that light signals are associated with larger eyes and shorter antennae. In addition, following a transition from nocturnal light displays to diurnal pheromones, eye size reductions occur more rapidly than antenna size increases. In agreement with the North American taxa, across 101 worldwide firefly taxa in 32 genera, we found light displays are associated with larger eye and smaller antenna sizes in both males and females. For those taxa with both male and female data, we found sex differences in eye size and, for diurnal species, in antenna size.

Abstract:The animals, in particular insects (Drosophila melanogaster), response towards odor stimuli in nature can be established by measuring the dynamic of the odor response. Such an approach is innovative since responses to odors were tested only at certain time point so far, not in hour intervals for several hours. The odor attraction to 14 natural and ecologically relevant odor stimuli such as fruits- ripe and rotten, yeast and vinegar was tested. A mathematical model to evaluate obtained data is proposed in this study in which the number of flies caught over several time points is presented as one simple parameter showing trapping potential of the trap housing particular odor stimuli. The knowledge concerning dynamic of the odor response in Drosophila melanogaster may enlighten the principles of flies behavior in context of exposure towards odor stimulus.

Exploration-Exploitation Model of Moth-Inspired Olfactory Navigation

Abstract:Navigation of male moths toward females during the mating search offers a unique perspective on the exploration-exploitation (EE) model in decision-making. This study uses the EE model to explain male moth pheromone-driven flight paths. We leverage wind tunnel measurements and 3D tracking using infrared cameras to gain insights into male moth behavior. During the experiments in the wind tunnel, we add disturbance to the airflow and analyze the effect of increased fluctuations on moth flights in the context of the proposed EE model. We separate the exploration and exploitation phases by applying a genetic algorithm to the dataset of moth 3D trajectories. First, we demonstrate that the exploration-to-exploitation rate (EER) increases with distance from the source of the female pheromone, which can be explained in the context of the EE model. Furthermore, our findings reveal a compelling relationship between EER and increased flow fluctuations near the pheromone source. Using the open-source pheromone plume simulation and our moth-inspired navigation model, we explain why male moths exhibit an enhanced EER as turbulence levels increase, emphasizing the agent's adaptation to dynamically changing environments. This research extends our understanding of optimal navigation strategies based on general biological EE models and supports the development of advanced, theoretically supported bio-inspired navigation algorithms. We provide important insights into the potential of bio-inspired navigation models for addressing complex decision-making challenges.
