Abstract:To detect and escape looming threats, night-flying insects must rely on other senses than vision alone. Nocturnal mosquitoes can evade looming objects in the dark, but how they achieve this is still unknown. Here, we show how night-active female malaria mosquitoes escape from rapidly looming objects that simulate defensive actions of blood-hosts. First, we quantified the escape performance of flying mosquitoes from an event-triggered mechanical swatter, showing that mosquitoes use swatter-induced airflow to increase their escape success. Secondly, we used high-speed videography and deep-learning-based tracking to analyze escape flights in detail, showing that mosquitoes use banked turns to evade the threat. By combining escape kinematics data with numerical simulations of attacker-induced airflow and a mechanistic movement model, we unraveled how mosquitoes control these banked evasive maneuvers: they actively steer away from the danger, and then passively travel with the bow wave produced by the attacker. Our results demonstrate that night-flying mosquitoes can detect looming objects when visual cues are minimal, suggesting that they use attacker-induced airflow both to detect the danger and as a fluid medium to move with away from the threat. This shows that escape strategies of flying insects are more complex than previous visually induced escape flight studies suggest. As most insects are of similar or smaller sizes than mosquitoes, comparable escape strategies are expected among millions of flying insect species. The here-observed escape maneuvers are distinct from those of mosquitoes escaping from odor-baited traps, thus providing new insights for the development of novel trapping techniques for integrative vector management.

Abstract:Male mosquitoes form aerial aggregations, known as swarms, to attract females and maximize their chances of finding a mate. Within these swarms, individuals must be able to recognize potential mates and navigate the social environment to successfully intercept a mating partner. Prior research has almost exclusively focused on the role of acoustic cues in mediating the male mosquito's ability to recognize and pursue females. However, the role of other sensory modalities in this behavior has not been explored. Moreover, how males avoid collisions with one another in the swarm while pursuing females remains poorly understood. In this study, we combined free-flight and tethered-flight simulator experiments to demonstrate that swarming Anopheles coluzzii mosquitoes integrate visual and acoustic information to track conspecifics and avoid collisions. Our tethered experiments revealed that acoustic stimuli gated mosquito steering responses to visual objects simulating nearby mosquitoes, especially in males that exhibited a strong response toward visual objects in the presence of female flight tones. Additionally, we observed that visual cues alone could trigger changes in mosquitoes' wingbeat amplitude and frequency. These findings were corroborated by our free-flight experiments, which revealed that Anopheles coluzzii modulate their thrust-based flight responses to nearby conspecifics in a similar manner to tethered animals, potentially allowing for collision avoidance within swarms. Together, these results demonstrate that both males and females integrate multiple sensory inputs to mediate swarming behavior, and for males, the change in flight kinematics in response to multimodal cues might allow them to simultaneously track females while avoiding collisions.
The complex swarming dynamics of malaria mosquitoes emerges from simple minimally-interactive behavioral rules

Abstract:Complex coordinated group behaviors such as bird flocking and fish schooling often rely on intricate interactions among individuals. In these groups, neighbors exhibit strong mutual attraction, alignment, and collision avoidance. Here, we tested whether insect swarms arise from similar simple behavioral rules. Using high-speed videography and agent-based modeling, we identified the behavioral rules governing mating swarms of flying malaria mosquitoes, marking the first such analysis for any insect species. We found that, unlike birds and fish, mosquito swarming behavior is driven primarily by environmental cues like the sunset horizon and ground objects. In fact, interactions between conspecifics are limited to only close-range collision avoidance behaviors. Thus, insect swarming seems fundamentally distinct from bird flocking and fish schooling.

Dynamics of mosquito swarms over a moving marker

Abstract:Insect swarms are a model system for understanding collective behavior where the collective motion appears in disorder. To initiate and maintain a swarm in place, flying insects often use a visual external cue called a marker. In mosquitoes, understanding the swarming behavior and its relation to the marker has an additional medical relevance since swarming often precedes mating in the wild, thus constituting an important stage to intercept for controlling mosquito population. In this paper, we conduct preliminary experiments to characterize the visual coupling between a swarm of mosquitoes and a marker. A laboratory microcosm with artificial lighting was built to stimulate consistent swarming in the malarial mosquito Anopheles stephensi. The experimental setup was used to film a mosquito swarm with a stereo camera system as a marker was moved back-and-forth with different frequencies. System identification analysis of the frequency response shows that the relationship between the swarm and the marker can be described by delayed second order dynamics in a feedback loop. Further, the length of the internal time delay appears to correlate with the number of mosquitoes swarming on the marker indicating that such a delay may be able capture social interactions within swarming systems. For insect swarms, model fitting of trajectory data provides a way to numerically compare swarming behaviors of different species with respect to marker characteristics. These preliminary results motivate investigating linear dynamic system in feedback as a framework for modeling insect swarms and set the stage for future studies.

Abstract:Insects and small animals capable of adhering reversibly to a variety of surfaces employ the unique design of the distal part of their legs. In the case of mosquitoes, their feet are composed of thousands of micro- and nanoscale protruding structures, which impart superhydrophobic properties. Previous research has shown that the superhydrophobic nature of the feet allows mosquitoes to land on water, which is necessary for their reproduction cycle. Here, we show that van der Waals interactions are the main adhesion mechanism employed by mosquitoes to adhere to various surfaces. We further demonstrate that the judicious creation of surface roughness on an opposing surface can increase the adhesion strength because of the increased number of surface elements interacting with the setae through multiple contact points. Although van der Waals forces are shown to be the predominant mechanism by which mosquitoes adhere to surfaces, capillary forces can also contribute to the total adhesion force when the opposing surface is hydrophilic and under humid conditions. These fundamental properties can potentially be applied in the development of superior Long Lasting Insecticidal Nets (LLINs), which represent one of the most effective methods to mitigate mosquito-transmitted infectious diseases such as Malaria, Filaria, Zika, and Dengue.

Abstract:Collisions with raindrops are one of many obstacles insects face during flight. In this fluid dynamics video, we present a series of high-speed films of impacts between mosquitoes and raindrops. We also present drop impacts upon insect mimics, which are unsupported styrofoam balls of the same mass as mosquitoes. High-speed videography and particle tracking during collision are employed to determine the insect position versus time. We determine the magnitude of acceleration by considering the momentum transfer and impact duration. Experiments with live mosquitoes indicate a surprising ability to quickly recover flight post-collision, despite accelerations of 30-300 gravities over durations of 1 ms.

On the Role of Vortical Structures in Aerodynamic Performance of a Hovering Mosquito

Abstract:Mosquitoes have slimmer wings, higher flapping frequencies, and much lower amplitudes than most other insects. These unique features signify special aerodynamic mechanisms. Besides the leading-edge vortex, which is one of the most common mechanisms of flapping-wing flight, mosquitoes have two distinctive mechanisms: trailing-edge vortex and rotational drag. In this study, the three-dimensional flow field around a hovering mosquito is simulated by using the immersed boundary method. The numerical results agree well with previous experimental data. Mechanisms unique to mosquitoes are identified from the instantaneous pressure and vorticity fields. The flow domains, containing several vortical structures produced by the flapping wings, are divided into different regions for quantitatively analyzing the contribution of vortical structures to the lift. Advection of the trailing-edge vortex and production of the leading-edge vortex each contribute peaks in lift. Passive deformation of the wings is also important, as it stabilizes delayed stall and decreases by 26% the maximum aerodynamic power required for hovering flight. In addition, the lift coefficient and power economy are improved as the Reynolds number increases, which explains the better ability of larger mosquitoes to seek and feed on hosts from the aerodynamic point of view. Abstract:High speed video observations of free flying male Aedes aegypti mosquitoes, the dengue and yellow fever vector, along with custom measurement methods, enable measurement of wingbeat frequency, body position and body orientation of mosquitoes during flight. We find these mosquitoes flap their wings at approximately 850 Hz. We also generate body yaw, body pitch and wing deviation measurements with standard deviations of less than 1 degree and find that sideways velocity and acceleration are important components of mosquito motion. Rapid turns involving changes in flight direction often involve large sideways accelerations. These do not correspond to commensurate changes in body heading, and the insect's flight direction and body heading are decoupled during flight. These findings call in to question the role of yaw control in mosquito flight. In addition, using orientation data, we find that sideways accelerations are well explained by roll-based rotation of the lift vector. In contrast, the insect's body pitch angle does not correlate with its forward acceleration. This implies that controlling body roll is important to mosquito dynamics. The dynamic importance of stabilizing body pitch and body yaw is less clear.

Abstract:Some flying animals use active sensing to perceive and avoid obstacles. Nocturnal mosquitoes exhibit a behavioral response to divert away from surfaces when vision is unavailable, indicating a short-range, mechanosensory collision-avoidance mechanism. We suggest that this behavior is mediated by perceiving modulations of their self-induced airflow patterns as they enter a ground or wall effect. We used computational fluid dynamics simulations of low-altitude and near-wall flights based on in vivo high-speed kinematic measurements to quantify changes in the self-generated pressure and velocity cues at the sensitive mechanosensory antennae. We validated the principle that encoding aerodynamic information can enable collision avoidance by developing a quadcopter with a sensory system inspired by the mosquito. Such low-power sensing systems have major potential for future use in safer rotorcraft control systems.

Abstract:Mosquitoes are known to choose oviposition habitats actively based on their suitability for survival of their offspring. They thus avoid laying eggs in dangerous habitats with aquatic predators through detection of their cues. Visual, tactile, and chemical cues emitted by certain predators have been shown to reduce oviposition in some mosquitoes, which in turn may affect their adult population sizes. However, those tests tend to be limited to species from temperate regions. Moreover, mosquito oviposition response to predator cues have been shown to be predator specific, but this has not been investigated for many potential predators worldwide. In the African subtropics, temporary ponds can be important mosquito breeding habitats and the same habitats are also used by a group of specialized killifish that can survive dry periods using drought resistant eggs. Although they are reported as mosquito predators, it is unknown whether mosquitoes actively avoid laying eggs in ponds with annual killifish. We investigated this using the mosquito Culex quinquefasciatus and the free and caged killifish Nothobranchius neumanni in the outdoor mesocosm experiments in Tanzania. We found that Cx. quinquefasciatus had 71% lower oviposition in mesocosms with free swimming killifish and 68% lower oviposition in mesocosms with caged killifish. We conclude that predator-released chemical cues alone are enough to explain the observed responses. If the cues can be isolated and chemically identified can be developed into a biological mosquito control strategy and used to safeguard public health in areas where mosquito borne diseases are endemic.

Abstract:Mosquito-borne viral diseases are a major concern of global health and result in significant economic losses in many countries. As natural vectors, mosquitoes are very permissive to and allow systemic and persistent arbovirus infection. Intriguingly, persistent viral propagation in mosquito tissues neither results in dramatic pathological sequelae nor impairs the vectorial behavior or lifespan, indicating that mosquitoes have evolved mechanisms to tolerate persistent infection and developed efficient antiviral strategies to restrict viral replication to nonpathogenic levels. Here we provide an overview of recent progress in understanding mosquito antiviral immunity and advances in the strategies by which mosquitoes control viral infection in specific tissues.
Acoustotactic response of mosquitoes in untethered flight to incidental sound

Abstract:Mosquitoes are vectors for some of the most devastating diseases on the planet. Given the centrality of acoustic sensing in the precopulatory behavior of these vectors, the use of an exogenous acoustic stimulus offers the potential of interfering with the courtship behavior of these insects. Previous research on the acoustotactic response of mosquitoes has been conducted on tethered preparations using low-intensity sound stimuli. To quantify differences in acoustotactic responses between mosquitos of distinct sex and species, we examined the effects of incidental sound stimuli on the flight behavior of free-flying male vs. female Aedes aegypti and Anopheles gambiae mosquitoes. The key variables were sound frequency (100-1000 Hz) and intensity (67-103 dB, measured at 12.5 cm from the source), and the acoustotactic response was measured in terms of the relative increase in flight speed in response to the stimulus. The data show, for the first time, significant sex- and species-specific differences in acoustotactic responses. A. aegypti exhibited a greater response to sound stimulus compared to An. gambiae, and the response also extended over a larger range of frequencies. Furthermore, the males of both species displayed a greater acoustotactic response than females, with An. gambiae females exhibiting minimal response to sound.
Video Recording Can Conveniently Assay Mosquito Locomotor Activity

Abstract:Anopheles gambiae and Aedes aegypti are perhaps the best studied mosquito species and important carriers of human malaria and arbovirus, respectively. Mosquitoes have daily rhythms in behaviors and show a wide range of activity patterns. Although Anopheles is known to be principally nocturnal and Aedes principally diurnal, details of mosquito activity are not easily assayed in the laboratory. We recently described FlyBox, a simple tracking system for assaying Drosophila locomotor activity rhythms and thought that it might also be applicable to monitoring mosquito activity. Indeed, we show here that FlyBox can easily, conveniently, affordably and accurately measure the activity of Anopheles as well as Aedes over several days. The resulting profiles under light-dark as well as constant darkness conditions are compatible with results in the literature, indicating that this or similar systems will be useful in the future for more detailed studies on a range of insect species and under more diverse laboratory conditions.
Visual and thermal stimuli modulate mosquito-host contact with implications for improving malaria vector control tools

Abstract:Malaria prevention relies on mosquito control interventions that use insecticides and exploit mosquito behavior. The rise of insecticide resistance and changing transmission dynamics urgently demand vector control innovation. To identify behavioral traits that could be incorporated into such tools, we investigated the flight and landing response of Anopheles coluzzii to human-like host cues. We show that landing rate is directly proportional to the surface area of thermal stimulus, whereas close-range orientation is modulated by both thermal and visual inputs. We modeled anopheline eye optics to theorize the distance at which visual targets can be detected under a range of conditions, and experimentally established mosquito preference for landing on larger targets, although landing density is greater on small targets. Target orientation does not affect landing rate; however, vertical targets can be resolved at greater distance than horizontal targets of the same size. Mosquito traps for vector control could be significantly enhanced by incorporating these features.

Abstract:The collective motion of groups of animals emerges from the net effect of the interactions between individual members of the group. In many cases, such as birds, fish, or ungulates, these interactions are mediated by sensory stimuli that predominantly arise from nearby neighbors. But not all stimuli in animal groups are short range. Here, we consider mating swarms of midges, which are thought to interact primarily via long-range acoustic stimuli. We exploit the similarity in form between the decay of acoustic and gravitational sources to build a model for swarm behavior. By accounting for the adaptive nature of the midges’ acoustic sensing, we show that our ‘adaptive gravity’ model makes mean-field predictions that agree well with experimental observations of laboratory swarms. Our results highlight the role of sensory mechanisms and interaction range in collective animal behavior. Additionally, the adaptive interactions that we present here open a new class of equations of motion, which may appear in other biological contexts.

Here, we characterized the three-dimensional spatial and temporal flight kinematics of males swarming above a ground marker. We observed that the location, shape and volume of swarms were highly stereotypic, consistent over the swarming duration, regardless of the number of individuals in the swarm. The swarm had an elliptical cone shape, and we observed a differential spatial distribution of flight kinematics parameters within the swarm volume. Among these parameters, only swarm density varied with swarm size. Using a sensory system-informed model, we show that swarm location and shape can accurately be modelled based on visual perception of the marker. To control swarm height, swarming individuals maintain an optical angle of the marker ranging from 24° to 55°. Limiting the viewing angle deviation to 4.5% of the maximum value results in the observed elliptical cone swarm shape. We discuss the implications of these finding in mating success, speciation and for vector control.
