Abstract:The life style of honey bees requires them to learn a variety of specific things in order to navigate and forage. The need for learning is sufficiently predictable with regard to the behavioral contexts and useful cues that it is highly structured and relatively easily studied. Although each cue seems to be learned independently, the set of cues memorized in each context appears to be stored as a unit. The exact nature of the storage system for flower shape, landmarks near the food, and navigational landmarks, as well as certain navigational parameters, is not yet perfectly clear. The nature of these ambiguities is explored. The possibility that some form of a synaptic selection strategy might underlie bee learning is also explored in a preliminary way.
Flexible visual learning in nectar-foraging hornets

Abstract:Pollinators, such as bees, develop flexible memories of colors, patterns, and shapes, for efficient flower recognition. Here we tested whether other flower-foraging insects have evolved similar cognitive abilities underpinning flexible visual learning. We trained wild hornets from two species commonly found in Europe, the invasive yellow-legged hornet ( Vespa velutina nigrithorax ) and the European hornet ( Vespa crabro ), to associate sucrose solution rewards to color stimuli in a Y-maze. Hornets from both species succeeded in differential and reversal learning and developed short-term memories of the learnt associations. Thus, just like bees, hornets can learn various visual cue-reward associations and remember them for at least 1 h for selecting flowers. Our study in non-model species illustrates how standard conditioning approaches can be used to explore and compare the cognitive abilities of animals sharing similar foraging ecologies.

Abstract:Since the demonstration of color vision in honey bees 100 years ago by Karl von Frisch, appetitive conditioning to color targets has been used as the principal way to access behavioral aspects of bee color vision. Yet, analyses on how conditioning parameters affect color perception remained scarce. Conclusions on bee color vision have often been made without referring them to the experimental context in which they were obtained, and thus presented as absolute facts instead of realizing that subtle variations in conditioning procedures might yield different results. Here, we review evidence showing that color learning and discrimination in bees are not governed by immutable properties of their visual system, but depend on how the insects are trained and thus learn a task. The use of absolute or differential conditioning protocols, the presence of aversive reinforcement in differential conditioning and the degrees of freedom of motor components determine dramatic variations in color discrimination. We, thus, suggest top-down attentional modulation of color vision to explain the changes in color learning and discrimination reviewed here. We discuss the possible neural mechanisms of this modulation and conclude that color vision experiments require a careful consideration of how training parameters shape behavioral responses.

Here we evaluate the role of the unconditioned stimulus (US) in producing conditioned inhibition by using Proboscis Extension Response conditioning of the honey bee ( ). Specifically, using variations of a ‘feature-negative’ experimental design, we employ downshifts in US intensity relative to US intensity used during initial excitatory conditioning, to show that an odorant in an odor-odor mixture can become a conditioned inhibitor. We argue that some alternative interpretations to conditioned inhibition are unlikely. However, we show variation across individuals in how strongly they show Conditioned Inhibition, with some individuals possibly revealing a different means of learning about changes in reinforcement. We discuss how resolution of these differences is needed to fully understand whether and how Conditioned Inhibition is manifested in the honey bee, and whether it can be extended to investigate how it is encoded in the CNS. It is also important for extension to other insect models. In particular, work like this will be important as more is revealed of the complexity of the insect brain from connectome projects.

Individual consistency in the learning abilities of honey bees: cognitive specialization within sensory and reinforcement modalities

Abstract:The question of whether individuals perform consistently across a variety of cognitive tasks is relevant for studies of comparative cognition. The honey bee ( Apis mellifera ) is an appropriate model to study cognitive consistency as its learning can be studied in multiple elemental and non-elemental learning tasks. We took advantage of this possibility and studied if the ability of honey bees to learn a simple discrimination correlates with their ability to solve two tasks of higher complexity, reversal learning and negative patterning. We performed four experiments in which we varied the sensory modality of the stimuli (visual or olfactory) and the type (Pavlovian or operant) and complexity (elemental or non-elemental) of conditioning to examine if stable correlated performances could be observed across experiments. Across all experiments, an individual's proficiency to learn the simple discrimination task was positively and significantly correlated with performance in both reversal learning and negative patterning, while the performances in reversal learning and negative patterning were positively, yet not significantly correlated. These results suggest that correlated performances across learning paradigms represent a distinct cognitive characteristic of bees. Further research is necessary to examine if individual cognitive consistency can be found in other insect species as a common characteristic of insect brains.

Abstract:Background: The honeybee has to detect, process and learn numerous complex odours from her natural environment on a daily basis. Most of these odours are floral scents, which are mixtures of dozens of different odorants. To date, it is still unclear how the bee brain unravels the complex information contained in scent mixtures. Methodology/principal findings: This study investigates learning of complex odour mixtures in honeybees using a simple olfactory conditioning procedure, the Proboscis-Extension-Reflex (PER) paradigm. Restrained honeybees were trained to three scent mixtures composed of 14 floral odorants each, and then tested with the individual odorants of each mixture. Bees did not respond to all odorants of a mixture equally: They responded well to a selection of key odorants, which were unique for each of the three scent mixtures. Bees showed less or very little response to the other odorants of the mixtures. The bees' response to mixtures composed of only the key odorants was as good as to the original mixtures of 14 odorants. A mixture composed of the other, non-key-odorants elicited a significantly lower response. Neither an odorant's volatility or molecular structure, nor learning efficiencies for individual odorants affected whether an odorant became a key odorant for a particular mixture. Odorant concentration had a positive effect, with odorants at high concentration likely to become key odorants. Conclusions/significance: Our study suggests that the brain processes complex scent mixtures by predominantly learning information from selected key odorants. Our observations on key odorant learning lend significant support to previous work on olfactory learning and mixture processing in honeybees.

Abstract:Honey bees are important model systems for the investigation of learning and memory and for a better understanding of the neuronal basics of brain function. Honey bees also possess a rich repertoire of tones and sounds, from queen piping and quacking to worker hissing and buzzing. In this study, we tested whether the worker bees' sounds can be used as a measure of learning. We therefore conditioned honey bees aversively to odours in a walking arena and recorded both their sound production and their movement. Bees were presented with two odours, one of which was paired with an electric shock. Initially, the bees did not produce any sound upon odour presentation, but responded to the electric shock with a strong hissing response. After learning, many bees hissed at the presentation of the learned odour, while fewer bees hissed upon presentation of another odour. We also found that hissing and movement away from the conditioned odour are independent behaviours that can co-occur but do not necessarily do so. Our data suggest that hissing can be used as a readout for learning after olfactory conditioning, but that there are large individual differences between bees concerning their hissing reaction. The basis for this variability and the possible ecological relevance of the bees' hissing remain to be investigated.

Abstract:ABSTRACT Learning visual cues is an essential capability of bees for vital behaviors such as orientation in space and recognition of nest sites, food sources and mating partners. To study learning and memory in bees under controlled conditions, the proboscis extension response (PER) provides a well-established behavioral paradigm. While many studies have used the PER paradigm to test olfactory learning in bees because of its robustness and reproducibility, studies on PER conditioning of visual stimuli are rare. In this study, we designed a new setup to test the learning performance of restrained honey bees and the impact of several parameters: stimulus presentation length, stimulus size (i.e. visual angle) and ambient illumination. Intact honey bee workers could successfully discriminate between two monochromatic lights when the color stimulus was presented for 4, 7 and 10 s before a sugar reward was offered, reaching similar performance levels to those for olfactory conditioning. However, bees did not learn at shorter presentation durations. Similar to free-flying honey bees, harnessed bees were able to associate a visual stimulus with a reward at small visual angles (5 deg) but failed to utilize the chromatic information to discriminate the learned stimulus from a novel color. Finally, ambient light had no effect on acquisition performance. We discuss possible reasons for the distinct differences between olfactory and visual PER conditioning.

Abstract:Significance Do animals encode statistical information about visual patterns the same way as humans do? If so, humans’ superior visual cognitive skills must depend on some other factors; if not, the nature of the differences can provide hints about what makes human learning so versatile. We provide a systematic comparison of automatic visual learning in humans and honey bees, showing that while bees do extract statistical information about co-occurrence contingencies of visual scenes, in contrast to humans, they do not automatically encode conditional information. Thus, acquiring implicit knowledge about the statistical properties of the visual environment may be a general mechanism in animals, but the richer representation developed automatically by humans might require specific probabilistic computational faculties.

Learning modifies attention during bumblebee visual search

Abstract:The role of visual search during bee foraging is relatively understudied compared to the choices made by bees. As bees learn about rewards, we predicted that visual search would be modified to prioritise rewarding flowers. To test this, we ran an experiment testing how bee search differs in the initial and later part of training as they learn about flowers with either higher- or lower-quality rewards. We then ran an experiment to see how this prior training with reward influences their search on a subsequent task with different flowers. We used the time spent inspecting flowers as a measure of attention and found that learning increased attention to rewards and away from unrewarding flowers. Higher quality rewards led to decreased attention to non-flower regions, but lower quality rewards did not. Prior experience of lower rewards also led to more attention to higher rewards compared to unrewarding flowers and non-flower regions. Our results suggest that flowers would elicit differences in bee search behaviour depending on the sugar content of their nectar. They also demonstrate the utility of studying visual search and have important implications for understanding the pollination ecology of flowers with different qualities of reward.

Abstract:Distinguishing between nectar and non-nectar odors is challenging for animals due to shared compounds and changing ratios in complex mixtures. Changes in nectar production throughout the day and potentially many times within a forager's lifetime add to the complexity. The honeybee olfactory system, containing less than 1000 principal neurons in the early olfactory relay, the antennal lobe (AL), must learn to associate diverse volatile blends with rewards. Previous studies identified plasticity between AL neurons but its role in odor learning remains poorly understood. We used a computational network model and live imaging of the honeybee's AL to explore the neural mechanisms and functions of plasticity in the early olfactory system. Our findings revealed that when trained with a set of rewarded and unrewarded odors, the AL inhibitory network suppresses shared chemical compounds while enhancing responses to distinct compounds. This results in improved pattern separation and a more concise neural code. Our Calcium imaging data support these predictions. Analysis of a Graph Convolutional Network in machine learning performing an odor categorization task revealed a similar mechanism of contrast enhancement. Our model provides insights into how inhibitory plasticity in the early olfactory network reshapes coding for efficient learning of complex odors.

Collective defense of honeybee colonies: experimental results and theoretical modeling

Abstract:Social insect colonies routinely face large vertebrate predators, against which they need to mount a collective defense. To do so, honeybees use an alarm pheromone that recruits nearby bees into mass stinging of the perceived threat. This alarm pheromone is carried directly on the stinger, hence its concentration builds up during the course of the attack. Here, we investigate how individual bees react to different alarm pheromone concentrations, and how this evolved response-pattern leads to better coordination at the group level. We first present an individual dose-response curve to the alarm pheromone, obtained experimentally. Second, we apply Projective Simulation to model each bee as an artificial learning agent that relies on the pheromone concentration to decide whether to sting or not. If the emergent collective performance benefits the colony, the individual reactions that led to it are enhanced via reinforcement learning, thus emulating natural selection. Predators are modeled in a realistic way so that the effect of factors such as their resistance, their killing rate or their frequency of attacks can be studied. We are able to reproduce the experimentally measured response-pattern of real bees, and to identify the main selection pressures that shaped it. Finally, we apply the model to a case study: by tuning the parameters to represent the environmental conditions of European or African bees, we can predict the difference in aggressiveness observed between these two subspecies.

Abstract:Honey bee ecology demands they make both rapid and accurate assessments of which flowers are most likely to offer them nectar or pollen. To understand the mechanisms of honey bee decision-making, we examined their speed and accuracy of both flower acceptance and rejection decisions. We used a controlled flight arena that varied both the likelihood of a stimulus offering reward and punishment and the quality of evidence for stimuli. We found that the sophistication of honey bee decision-making rivalled that reported for primates. Their decisions were sensitive to both the quality and reliability of evidence. Acceptance responses had higher accuracy than rejection responses and were more sensitive to changes in available evidence and reward likelihood. Fast acceptances were more likely to be correct than slower acceptances; a phenomenon also seen in primates and indicative that the evidence threshold for a decision changes dynamically with sampling time. To investigate the minimally sufficient circuitry required for these decision-making capacities, we developed a novel model of decision-making. Our model can be mapped to known pathways in the insect brain and is neurobiologically plausible. Our model proposes a system for robust autonomous decision-making with potential application in robotics.
Application of a Prediction Error Theory to Pavlovian Conditioning in an Insect

Abstract:Elucidation of the conditions in which associative learning occurs is a critical issue in neuroscience and comparative psychology. In Pavlovian conditioning in mammals, it is thought that the discrepancy, or error, between the actual reward and the predicted reward determines whether learning occurs. This theory stems from the finding of Kamin's blocking effect, in which after pairing of a stimulus with an unconditioned stimulus (US), conditioning of a second stimulus is blocked when the two stimuli are presented in compound and paired with the same US. Whether this theory is applicable to any species of invertebrates, however, has remained unknown. We first showed blocking and one-trial blocking of Pavlovian conditioning in the cricket <i>Gryllus bimaculatus</i>, which supported the Rescorla-Wagner model but not attentional theories, the major competitive error-correction learning theories to account for blocking. To match the prediction error theory, a neural circuit model was proposed, and prediction from the model was tested: the results were consistent with the Rescorla-Wagner model but not with the retrieval theory, another competitive theory to account for blocking. The findings suggest that the Rescorla-Wagner model best accounts for Pavlovian conditioning in crickets and that the basic computation rule underlying Pavlovian conditioning in crickets is the same to those suggested in mammals. Moreover, results of pharmacological studies in crickets suggested that octopamine and dopamine mediate prediction error signals in appetitive and aversive conditioning, respectively. This was in contrast to the notion that dopamine mediates appetitive prediction error signals in mammals. The functional significance and evolutionary implications of these findings are discussed.

Abstract:Learning to anticipate events based on the predictive relationship between an action and an outcome (operant conditioning) is a form of associative learning shared by humans and most of other living beings, including invertebrates. Several behavioral studies on the mechanisms of operant conditioning have included Melipona quadrifasciata, a honey bee that is easily manipulated due to lack of sting. In this work, brain proteomes of Melipona bees trained using operant conditioning and untrained (control) bees were compared by two-dimensional gel electrophoresis analysis within pI range of 3-10 and 4-7; in order to find proteins specifically related to this type of associative learning.One protein was detected with differential protein abundance in the brains of trained bees, when compared to not trained ones, through computational gel imaging and statistical analysis. This protein was identified by peptide mass fingerprinting and MS/MS peptide fragmentation using a MALDI-TOF/TOF mass spectrometer as one isoform of arginine kinase monomer, apparently dephosphorylated. Brain protein maps were obtained by 2-DE (Two-dimensional gel electrophoresis) from a total proteins and phosphoproteins extract of the bee Melipona quadrifasciata. One isoform of arginine kinase, probably a dephosphorylated isoform, was significantly more abundant in the brain of trained bees using operant conditioning. Arginine kinase has been reported as an important enzyme of the energy releasing process in the visual system of the bee, but it may carry out additional and unexpected functions in the bee brain for learning process.

Abstract:Abstract Culture refers to behaviours that are socially learned and persist within a population over time. Increasing evidence suggests that animal culture can, like human culture, be cumulative: characterized by sequential innovations that build on previous ones 1 . However, human cumulative culture involves behaviours so complex that they lie beyond the capacity of any individual to independently discover during their lifetime 1–3 . To our knowledge, no study has so far demonstrated this phenomenon in an invertebrate. Here we show that bumblebees can learn from trained demonstrator bees to open a novel two-step puzzle box to obtain food rewards, even though they fail to do so independently. Experimenters were unable to train demonstrator bees to perform the unrewarded first step without providing a temporary reward linked to this action, which was removed during later stages of training. However, a third of naive observer bees learned to open the two-step box from these demonstrators, without ever being rewarded after the first step. This suggests that social learning might permit the acquisition of behaviours too complex to ‘re-innovate’ through individual learning. Furthermore, naive bees failed to open the box despite extended exposure for up to 24 days. This finding challenges a common opinion in the field: that the capacity to socially learn behaviours that cannot be innovated through individual trial and error is unique to humans.

Abstract:Previous studies suggest that social learning in bumblebees can occur through second-order conditioning, with conspecifics functioning as first-order reinforcers. However, the behavioural mechanisms underlying bumblebees' acquisition of socially learned associations remain largely unexplored. Investigating these mechanisms requires detailed quantification and analysis of the observation process. Here we designed a new 2D paradigm suitable for simple top-down high-speed video recording and analysed bumblebees' observational learning process using a deep-learning-based pose-estimation framework. Two groups of bumblebees observed live conspecifics foraging from either blue or yellow flowers during a single foraging bout, and were subsequently tested for their socially learned colour preferences. Both groups successfully learned the colour indicated by the demonstrators and spent more time facing rewarding flowers—whether occupied by demonstrators or not—compared to non-rewarding flowers. While both groups showed a negative correlation between time spent facing non-rewarding flowers and learning outcomes, the observer bees in the blue group benefited from time spent facing occupied rewarding flowers, whereas the yellow group showed that time facing unoccupied rewarding flowers by the observer bees positively correlated with their learning outcomes. These results suggest that socially influenced colour preferences are shaped by the interplay of different types of observations rather than merely by observing a conspecific at a single colour. Together, these findings provide direct evidence of the dynamical viewing process of observer bees during social observation, opening up new opportunities for exploring the details of more complex social learning in bumblebees and other insects.

Abstract:The European honey bee is a model organism for studying social behaviors. Comprehensive analyses focusing on the differential expression profiles of genes between the brains of nurse bees and foragers, or in the mushroom bodies—the brain structure related to learning and memory, and multimodal sensory integration—has identified candidate genes related to honey bee behaviors. Despite accumulating knowledge on the expression profiles of genes related to honey bee behaviors, it remains unclear whether these genes actually regulate social behaviors in the honey bee, in part because of the scarcity of genetic manipulation methods available for application to the honey bee. In this review, we describe the genetic methods applied to studies of the honey bee, ranging from classical forward genetics to recently developed gene modification methods using transposon and CRISPR/Cas9. We then discuss future functional analyses using these genetic methods targeting genes identified by the preceding research. Because no particular genes or neurons unique to social insects have been found yet, further exploration of candidate genes/neurons correlated with sociality through comprehensive analyses of mushroom bodies in the aculeate species can provide intriguing targets for functional analyses, as well as insight into the molecular and neural bases underlying social behaviors.
