Abstract:Some insect species have gained attention as efficient bioconverters of low-value organic substrates (i.e., residual streams) into high-value biomass. Black soldier fly (BSF) (Hermetia illucens) larvae are particularly interesting for bioconversion due to their ability to grow on a wide range of substrates, including low-value industrial residual streams. This is in part due to the plasticity of the gut microbiota of polyphagous insects, like BSF. Gut microbiota composition varies depending on rearing substrates, via a mechanism that might support the recruitment of microorganisms that facilitate digestion of a specific substrate. At the same time, specific microbial genera do persist on different substrates via unknown mechanisms. This study aimed to offer insights on this microbial plasticity by investigating how the composition of the bacterial community present in the gut of BSF larvae responds to two industrial residual streams: swill (a mixture of catering and supermarket leftovers) and distiller's dried grains with solubles. The bacterial biota composition of substrates, whole larvae at the beginning of the rearing period and at harvest, rearing residues, and larval gut regions were investigated through 16S rRNA gene sequencing. It was observed that both substrate and insect development influenced the bacterial composition of the whole larvae. Zooming in on the gut regions, there was a clear shift in community composition from a higher to a lower diversity between the anterior/middle midgut and the posterior midgut/hindgut, indicating a selective pressure occurring in the middle midgut region. Additionally, the abundance of the bacterial biota was always high in the hindgut, while its diversity was relatively low. Even more, the bacterial community in the hindgut was found to be relatively more conserved over the different substrates, harboring members of the BSF core microbiota. We postulate a potential role of the hindgut as a reservoir for insect-associated microbes. This warrants further research on that underexplored region of the intestinal tract. Overall, these findings contribute to our understanding of the bacterial biota structure and dynamics along the intestinal tract, which can aid microbiome engineering efforts to enhance larval performance on (industrial) residual streams.

Abstract:Fairy shrimps (Crustacea, Anostraca) are specialized inhabitants of inland water bodies that periodically dry or freeze over. Here we report the first observation since 1997 of a member of this basal crustacean order in Belgium and the first sighting of the species Branchipus schaefferi Fischer, 1834 since 1930. Nineteen populations were found in a restricted area located 55 km SE of Brussels in the Province of Hainaut. Based on a field survey, we discuss the habitat characteristics of these populations. We discuss also the distribution and habitat requirements of the species based on literature and formulate a number of guidelines for the conservation of this species as well as other large branchiopods in densely settled areas with intensive agriculture such as Belgium. Finally, we formulate a number of likely explanations for the lack of recent observations of these organisms in Western Europe and in Belgium.

Abstract:Nowadays, biodegrading organic waste, as a solution to confront environmental challenges, has attracted wide attention. A dipteran insect, black soldier fly (BSF), exhibits outstanding capability to convert organic waste into proteins and lipid resources, and thus, much interest has been shown in it. However, information of fundamental biology of BSF is still limited besides its recycling efficiency. In this work, we present a complete proteomic database of BSF at all instars (before prepupa). We further formulated the pathways corresponding to BSF development and built a relationship with the current genetic database. To achieve this, we investigated the proteomics of BSF during different periods. We identified 5036 proteins, and among them, 3905 proteins were annotated in the protein function database. illustrated three pathways related to major physiological processes including the insulin signaling pathway for feeding and growth, fatty acid biosynthesis pathway for fatty acid using, and toll/immune deficiency pathway for immune behavior. The proteins in these three pathways were matched with a published genetic database, and this reference library could be used for future BSF genetic engineering. In conclusion, this work provided a comprehensive protein library of BSF and expands the basic knowledge of BSF for future research.

Abstract:The interest towards the black soldier fly (BSF), Hermetia illucens, has grown impressively in the last few years, fostered by the legislative changes in the European landscape that have lifted the ban regarding the use of BSF larvae as feedstuff. In addition, bioconversion mediated by the larvae of the BSF is viewed as one of the most promising technologies for organic waste processing and valorisation. Finally, new, alternative applications to exploit various larval products such as lipids, chitin, antimicrobial peptides, and frass are being explored. However, this positive trend, confirmed by the increasing number of companies that deal with BSF mass rearing and processing, is in sharp contrast with the limited information on the biology of this insect, in particular on aspects related to its digestive features. This lack of knowledge needs to be carefully considered and filled in coming years, as a deep characterisation of the morphology, physiology, transcriptomics, and proteomics of the digestive system of the insect, as well a fine dissection of related aspects as gut microbiota and pathogens, is a prerequisite to improve the amazing bioconversion capabilities of this dipteron. So far, the larval stages received the most attention in research, but there might still be a lot to win by focusing more on the adult stage. Further expanding the basic knowledge on both the larval and the adult gut could lead to unexpected findings and open new perspectives to produce value-added bioproducts.

Abstract:Stag beetles, recognized as common saproxylic insects, are valued for their vibrant coloration and distinctive morphology. These beetles play a crucial ecological role in decomposition and nutrient cycling, serving as a vital functional component in ecosystem functioning. Although previous studies have confirmed that stag beetles are predominantly fungivores, the fluctuations in their intestinal fungal communities at different developmental stages remain poorly understood. In the current study, high-throughput sequencing was employed to investigate the dynamic changes within intestinal fungal communities at various developmental stages in the stag beetle Dorcus hopei . Results showed that microbial diversity was higher during the larval stage than during the pupal and adult stages. Furthermore, significant differences were identified in the composition of the intestinal fungal communities across the larval, pupal, and adult stages, suggesting that developmental transitions may be crucial factors contributing to variations in fungal community composition and diversity. Dominant genera included Candida , Scheffersomyces , Phaeoacremonium , and Trichosporon . Functional predictions indicated a greater diversity and relative abundance of endosymbiotic fungi in the larval gut, suggesting a potential dependency of larvae on beneficial gut fungi for nutrient acquisition. Additionally, the application of abundance-based β-null deviation and niche width analyses revealed that the adult gut exerted a stronger selection pressure on its fungal community, favoring certain taxa. This selection process culminates in a more robust co-occurrence network of fungal communities within the adult gut, thereby enhancing their adaptability to environmental fluctuations. This study advances our understanding of the intestinal fungal community structure in stag beetles, providing a crucial theoretical foundation for the development of saproxylic beetle resources, biomass energy utilization, plastic degradation strategies, and beetle conservation efforts.

Abstract:A survey for slug- and snail-associated nematodes was conducted in forests, parks, botanical gardens, and nature reserves at 13 localities in Belgium to uncover more diversity of gastropod mollusc-associated nematodes and to characterise Pellioditis populations found in the country. A total of 319 slugs and snails belonging to nine species were examined. Arion vulgaris was the most commonly found mollusc species in this study (eight locations), and 19.4% of the examined mollusc specimens were found infected by nematodes. The highest prevalence of nematodes was observed in Cornu aspersum (60%) followed by A. vulgaris (34.8%), Limax maximus (28.6%), and Cepaea sp. (20%). Eleven nematode species belonging to eight families were isolated and identified from the mollusc hosts including Alloionema appendiculatum, Angiostoma dentiferum, A. gandavense, Angiostrongylus vasorum, Cosmocerca longicauda, Panagrolaimus cf. subelongatus, Pellioditis californica, P. hermaphrodita, Rhabditis sp., Tetrameres cf. fissispina, and Troglostrongylus cf. brevior.Pellioditis was the most commonly found nematode genus (at nine localities) and C. longicauda and P. californica were reported in Belgium for the first time. Co-infections of more than one nematode species were observed in eight (2.5%) molluscs specimens. Most co-infections consisted of two nematode species. In one A. vulgaris specimen, a co-infection of three nematode species (A. vasorum, P. hermaphrodita, and Tetrameres cf. fissispina) was observed. Four ex vivo cultures of P. californica and six ex vivo cultures of P. hermaphrodita were established from single hermaphrodites, and both species were described based on light microscopy, scanning electron microscopy, and morphometric, morphological, and molecular data.

Abstract:Although the composition of invertebrate assemblages associated with kelps has motivated several studies in the recent past, little is known about assemblages associated with smaller, understorey macroalgae in these ecosystems. Here, the composition of invertebrate assemblages living within understorey macroalgae of a kelp (Laminaria digitata) forest has been studied over two sampling dates, and the structure of the food web investigated using stable isotopes. A total of 145 species of mobile fauna, mainly amphipods and gastropods, were identified. Although differences were date-dependent, we showed that the three species considered (Palmaria palmata, Mastocarpus stellatus, Ellisolandia elongata) sheltered different associated assemblages, including high host-specificity, which suggests that the animal diversity associated with rocky shores is enhanced by a high algal diversity. Overall, a dominance of gastropods was observed for the two-dimensional, leaf-like P. palmata, while the three-dimensionally structured species (M. stellatus, E. elongata) were characterized by a dominance of amphipods. Stable isotopes highlighted different trophic structures; E. elongata-associated assemblages were most likely relying on a dominant food source, sediment organic matter, while other assemblages were based on a wider diversity of food sources (algae, sediment, suspended organic matter). Our results illustrate the need to consider the microhabitats formed by understorey macroalgae in order to thoroughly assess the diversity and understand the functioning of coastal rocky ecosystems.

Abstract:This study aimed to establish a representative strain collection of dominant aerobic bacteria from black soldier fly larvae ( Hermetia illucens , BSFL). The larvae were fed either chicken feed or fiber-rich substrates to obtain a collection of BSFL-associated microorganisms. Via an approach based on only considering the highest serial dilutions of BSFL extract (to select for the most abundant strains), a total of 172 bacteria were isolated. Identification of these isolates revealed that all bacteria belonged to either the Proteobacteria (66.3%), the Firmicutes (30.2%), the Bacteroidetes (2.9%) or the Actinobacteria (0.6%). Twelve genera were collected, with the most abundantly present ones (i.e., minimally present in at least three rearing cycles) being Enterococcus (29.1%), Escherichia (22.1%), Klebsiella (19.8%), Providencia (11.6%), Enterobacter (7.6%), and Morganella (4.1%). Our collection of dominant bacteria reflects largely the bacterial profiles of BSFL already described in literature with respect to the most important phyla and genera in the gut, but some differences can be noticed depending on substrate, biotic and abiotic factors. Furthermore, this bacterial collection will be the starting point to improve in vitro digestion models for BSFL, to develop mock communities and to find symbionts that can be added during rearing cycles to enhance the larval performances, after functional characterization of the isolates, for instance with respect to enzymatic potential.

Abstract:We characterise in detail the larval and pupal cuticle of the black soldier fly Hermetia illucens L. (Diptera: Stratiomyidae), a key insect species in circular economy. In particular, we focus on ultrastructure using scanning and transmission electron microscopy, material characterization and composition (elements and minerals) with confocal laser scanning microscope, energy dispersive X-ray microanalysis, powder X-ray diffraction and mechanical properties with nanoindentation measurements. Calcium carbonate crystallizes on the epicuticle as blocks of calcite in the pupal cuticle. Calcium carbonate granules are stored in two specialised Malpighian tubules. CaCO3 is already present in the cuticle of young larval instars, but it is mainly in the form of amorphous calcium carbonate while the amount of calcite increases during larval development. The presence of calcite leads to cuticle hardening. Larval and pupal cuticles contain large amounts of resilin which guarantee cuticle flexibility.

Abstract:This study explores the utilisation of European freshwater mussels as hosts by the European bitterling ( Rhodeus amarus) at their current sympatric occurrence range. Our study confirms Pseudanodonta complanata as a suitable, occasional host for bitterling. The spawning relationship of R. amarus with freshwater mussels has physiological costs for the hosts, leading to resource competition and potential negative effects on host fertility. Further research is needed to assess the dynamics of host-parasite interactions, mussel adaptations to prevent parasitism, and the consequences of the bitterling recent and possible future expansion in Europe for mussel populations.

Abstract:Climate driven changes and anthropogenic pressures on the marine environment have been shown to favor the increase in certain potentially harmful species. Among them, Noctiluca scintillans , a common dinoflagellate, often blooms during warm summers and is known to affect plankton communities. In this study, we assessed the dynamics in abundance and cell size of N. scintillans as well as the relationship between N. scintillans and small soft-bodied zooplankton in the Belgian part of the North Sea (BPNS), since negative correlations between these plankton groups have been previously reported for nearby regions. This study is the first to present consistently counted N. scintillans cell numbers and measured cell lengths, through the analysis of ZooScan images from samples taken monthly at stations throughout the coastal zone of the BPNS. The results show that N. scintillans demonstrated clear seasonal dynamics with both high densities and large cell sizes in spring/summer (May-July). The occurrence of N. scintillans in the analyzed plankton samples and the abundance of N. scintillans at the observed peak intensities nearly tripled over a period of 5 years. A zero-inflated model showed a correlation of N. scintillans abundance with temperature as well as with phosphate concentrations, suggesting that anthropogenic influences such as climate change and riverine nutrient inputs could affect the temporal dynamics of the species. The results, on the other hand, did not show any negative impact of N. scintillans on the soft-bodied plankton community.

Abstract:External structures of insects contribute to the ability of herbivores to select and feed on their host plants. The invasive spotted lanternfly, <em>Lycorma delicatula</em> (Hemiptera: Fulgoridae) is an economically important and polyphagous insect pest in the eastern US. The lanternfly causes substantial damage to many woody plants by sucking phloem sap, reducing photosynthesis, causing weeping wounds, and creating conditions for sooty mold. Lanternfly nymphs switch host plants during their development. However, little is known about relationship between the lanternfly and its plant hosts, and particularly about morphological adaptations of the lanternfly to host plant usage at each developmental stage of the pest. In this study, we focused on assessing changes in morphology of (a) the lanternfly mouthparts (stylets and labium), and (b) the lanternfly tarsal tips (arolia and tarsal claws) at each developmental stage. Our study revealed several developmental patterns among which the presence of the indentations on mandibular stylets in late instars and adults, as well as the exponential growth of the labium and stylet length, and the tarsal claw dispersal during the lanternfly development. Our findings are critical for investigating and predicting the lanternfly host range, and the lanternfly dispersal to new host trees at each developmental stage.
