Abstract:Vital physiological processes that drive the insect molt represent areas of interest for the development of alternative control strategies. The western tarnished plant bug (Lygus hesperus Knight) is a pest of numerous agronomic and horticultural crops but the development of novel control approaches is impeded by limited knowledge of the mechanisms regulating its molt. To address this deficiency, we examined the fundamental relationship underlying the hormonal and molecular components of ecdysis. At 27°C L. hesperus exhibits a temporally controlled nymph-adult molt that occurs about 4 days after the final nymph-nymph molt with ecdysteroid levels peaking 2 days prior to the final molt. Application of exogenous ecdysteroids when endogenous levels had decreased disrupted the nymphal-adult molt, with treated animals exhibiting an inability to escape the old exoskeleton and resulting in mortality compared to controls. Using accessible transcriptomic data, we identified 10 chitinase-like sequences (LhCht), eight of which had protein motifs consistent with chitinases. Phylogenetic analyses revealed orthologous relationships to chitinases critical to molting in other insects. RT-PCR based transcript profiling revealed that expression changes to four of the LhChts was coordinated with the molt period and ecdysteroid levels. Collectively, our results support a role for ecdysteroid regulation of the L. hesperus molt and suggest that cuticle clearance is mediated by LhCht orthologs of chitinases that are essential to the molt process. These results provide the initial hormonal and molecular basis for future studies to investigate the specific roles of these components in molting.

Abstract:Many holo-and hemimetabolous insects enhance their eggshells during embryogenesis by forming a serosal cuticle(SC).To date,scholarly understanding of the SC composition and SC-related gene functions has been limited,especially for hemimetabolous insects.In this study,we initially performed transmission electron mi-croscopic(TEM)observation and chitin staining of the SC in Nilaparvata lugens,a hemimetabolous rice pest known as the brown planthopper(BPH).We confirmed that the SC was a chitin-rich lamellar structure deposited gradually during the early embryo-genesis.Parental RNA interference(RNAi)against Nilaparvata lugens chitin synthase 1(NlCHS1)in newly emerged and matured females resulted in decreases of egg hatchabil-ity by 100％and 76％,respectively.Ultrastructural analyses revealed loss of the lamellar structure of the SC in dsNlCHS1-treated eggs.According to temporal expression pro-files,five cuticle protein coding genes,NlugCpr1/2/3/8/90,were specifically or highly expressed during the SC formation period,and NlugCpr1/2/3/90 were further detected in 72 h eggshells extract by ultra-performance liquid chromatography-tandem mass spec-trometry/mass spectrometry.NlugCpr2/3/90 were likely three SC-specific cuticle proteins.TEM observations of the SC following parental RNAi against NlugCpr1/2/3/8/90 demon-strated that NlugCpr3/8/90 were essential for SC formation.The study provided an under-standing of the SC formation process and SC-related cuticle proteins in BPHs,which offer potential targets for pest control in the egg stage as well.

Abstract:Abstract Background The black soldier fly ( Hermetia illucens ) has significant economic potential. The larvae can be used in financially viable waste management systems, as they are voracious feeders able to efficiently convert low-quality waste into valuable biomass. However, most studies on H. illucens in recent decades have focused on optimizing their breeding and bioconversion conditions, while information on their biology is limited. Methods About 200 fifth instar well-fed larvae were sacrificed in this work. The liquid chromatography-tandem mass spectrometry and scanning electron microscopy were employed in this study to perform a proteomic and ultrastructural analysis of the peritrophic matrix (PM) of H. illucens larvae. Results A total of 565 proteins were identified in the PM samples of H. illucen , of which 177 proteins were predicted to contain signal peptides, bioinformatics analysis and manual curation determined 88 proteins may be associated with the PM, with functions in digestion, immunity, PM modulation, and others. The ultrastructure of the H. illucens larval PM observed by scanning electron microscopy shows a unique diamond-shaped chitin grid texture. Conclusions It is the first and most comprehensive proteomics research about the PM of H. illucens larvae to date. All the proteins identified in this work has been discussed in details, except several unnamed or uncharacterized proteins, which should not be ignored and need further study. A comparison of the ultrastructure between H. illucens larval PM and those of other insects as observed by SEM indicates that the PM displays diverse textures on an ultra-micro scale and we suscept a unique diamond-shaped chitin grid texture may help H. illucens larval to hold more food. This work deepens our understanding of the molecular architecture and ultrastructure of the H. illucens larval PM.

Abstract:The black soldier fly (BSF) Hermetia illucens (Diptera: Stratiomyidae) plays a significant role at the larval stage in the circular economy due to its ability to convert organic waste into valuable products for energy, food, feed, and agricultural applications. Many data are available on larval development and biomass generation, but basic research on this species is lacking and little is known about adult biology, in particular about the cues involved in sexual recognition. In the present study, using various instruments (stereomicroscope, scanning and transmission electron microscope, hyperspectral camera and spectrophotometer), wing ultrastructure of both sexes was analysed, reflectance and transmission spectra of the wings were measured and behavioural bioassays were carried out to measure male response to specific visual stimuli. The collected data showed the existence of sexual dimorphism in the wings of H. illucens due to iridescent structural colouration generated by a multilayer of melanin located in the dorsal lamina of the central part of the wing. Wing sexual dimorphism is particularly evident regarding the strong emission of blue light of female wings. Blue colour induces in males a strong motivation to mate. The obtained results can help to improve and optimize the breeding techniques of BSF.

Abstract:The black soldier fly (BSF) Hermetia Illucens can grow rapidly and on a wide variety of organic materials, and it is extensively used as a means of disposing of household organic waste. Different phases of the life cycle of BSF larvae (BSFL) are used in this work to extract chitin after the removal of lipids, mineral salts, and proteins. Multiple techniques, such as X-ray diffractometry, infrared spectroscopy, solid-state Nuclear Magnetic Resonance (13C ss-NMR) and thermogravimetric analysis, are used to investigate the chemical and physical characteristics of the extracted samples of chitin, which shows a high degree of acetylation (from 78% to 94%). The extracted chitin shows an increase of the thermal stability of 20 °C in the initial stage of life and 35 °C at the end of the life cycle if compared with a commercial standard. Moreover, the extracted chitin shows an increase in the crystallinity degree during the BSFL growth time (from 72% to 78%).

Abstract:Random plasma somatomedin-C levels were measured in 101 children aged 7 months to 18 years with growth at or below the 5th percentile. 87 of 101 had short stature due to constitutional delay or genetic predisposition and 14 were growth hormone deficient. In the short normal group mean somatomedin-C levels were: 0-2 years: 0.22±0.02U/ml; 2-5 years: 0.28±0.06U/ml; 5-12 years: 0.78±0.1U/ml; 12-18 years: 1.22±0.16U/ml; compared with 0.23±0.04U/ml in the growth hormone deficient children. 55 of 101 had somatomedin-C levels of 0.5U/ml, and had one or more growth hormone stimulation tests. All 14 growth hormone deficient patients fell into this group (range: 0.09-0.47U/ml). The 41 normal patients in this group (growth hormone 6ng/ml) were distibuted among the entire 87 normals as follows: 0-2 years: 10/10; 2-5 years: 19/23; 5-12 years: 11/30; 12-18 years: 1/24. There was considerable overlap of values among growth hormone deficient and short normal children under 12 years of age, but a clear separation above 12 years. In 5 of 6 patients in whom 2 measurements were made, there were major differences in these levels. 1) Random somatomedin-C levels lack specificity in screening for growth hormone deficiency under the age of 12 years. 2) The variation of levels in individual patients necessitates further study to identify a possible diurnal or pulsatile secretory pattern. 3) A standardized time of sampling should be considered.
A multilayer micromechanical model of the cuticle of Curculio longinasus Chittenden, 1927 (Coleoptera: Curculionidae)

Abstract:Curculio longinasus Chittenden, 1927 (Coleoptera: Curculionidae), is a weevil species common throughout the southwestern United States that uses its rostrum - a very slender, curved, beak-like projection of the head - to excavate tunnels in plant organs (such as acorns) for egg laying (oviposition). Once the apical portion of the rostrum has been inserted into the preferred substrate for oviposition, the female begins rotating around the perimeter of the hole, elevating her head by extending the fore-legs, and rotating the head in place in a drilling motion. This action causes significant elastic deformation of the rostrum, which will bend until it becomes completely straight. To better understand the mechanical behavior of the cuticle as it undergoes deformation during the preparation of oviposition sites, we develop a comprehensive micro/macro model of the micromechanical structure and properties of the cuticle, spanning across all cuticular regions, and reliably mirroring the resultant macroscale properties of the cuticle. Our modeling approach relies on the use of multi-scale, hierarchical biomaterial representation, and employs various micromechanical schemata - e.g., Mori-Tanaka, effective field, and Maxwell - to calculate the homogenized properties of representative volume elements at each level in the hierarchy. We describe the configuration and behavior of this model in detail, and discuss the theoretical implications and limitations of this approach with emphasis on future biomechanical and comparative evolutionary research. Our detailed account of this approach can thereby serve as a methodological template for exploring the biomechanical behavior of new insect structures.
The appearance and histological structure of the cuticle of the desert locust, Schistocerca gregaria (Forskål)

Abstract:No essential structural differences are encountered between the tergites and intersegmental membranes into which the fully developed dorsal abdominal cuticle of Schistocerca is differentiated. An exocuticle and an endocuticle are the principal layers seen throughout the cuticle, but a variation in their relative thickness allows of considerable flexibility in the intersegmental membranes. Both layers are virtually colourless, but owing to their transparency many of the pigments located in the underlying epidermis are visible through them. Still closer resemblance between the tergite and intersegmental membrane regions is encountered in the newly moulted cuticle, in which no hard exocuticle is yet developed. In this stage, however, the epicuticle, which is not readily recognized histologically in the fully hardened cuticle, is clearly seen in sections. It is shown to consist of the four well-defined layers—cuticulin, paraffin, wax and cement—already recognized in the epicuticle of Periplaneta (Dennell & Malek 1955 a ) . The structural differences between the cuticles of Schistocerca and of Periplaneta are considered. It is suggested, in conclusion, that the exocuticle of Schistocerca , in spite of being colourless, is more similar to the amber-coloured zone of the exocuticle of Periplaneta than to the underlying colourless zone.

Abstract:In holometabolous insects, extensive reorganisation of tissues and cells occurs at the pupal stage. The remodelling of the external exoskeleton and internal organs that intervenes during metamorphosis has been traditionally studied in many insect species based on histological or ultrastructural methods. This study demonstrates the use of synchrotron X-ray phase-contrast micro-computed tomography as a powerful, non-destructive tool for in situ morphological observation of anatomical structures at the pupal stage in two Tenebrionid beetles, i.e. Tribolium castaneum and Tenebrio molitor , known as important pests, as well as emerging and promising models in experimental biology. Virtual sections and three-dimensional reconstructions were performed on both males and females at early, intermediate, and late pupal stage. The dataset allowed us to observe the remodelling of the gut and nervous system as well as the shaping of the female and male reproductive system at different pupal ages in both mealworm and red flour beetles. Moreover, we observed that the timing and duration pattern of organ development varied between the species analysed, likely related to the species-specific adaptations of the pre-imaginal stages to environmental conditions, which ultimately affect their life cycle. This research provides new knowledge on the morphological modifications that occur during the pupal stage of holometabolous insects and provides a baseline set of information on beetle metamorphosis that may support future research in forensics, physiology, and ecology as well as an image atlas for educational purposes.

Abstract:Insect cuticles with nano-level structures exhibit functional surface properties such as structural color and superhydrophobicity. Despite the enormous influence the cuticle has had on biomimetic industrial applications, molecular and cellular mechanisms of cuticular extracellular matrix (ECM) assembly into nanoscale structures remain poorly understood in insects and other taxa. Ghiradella (1989) described how a crystallin-like lattice of endoplasmic reticulum (ER) prefigures the patterning of the porous cuticle of the butterfly wing scale with structural color1. Building on that insight, we show that the nanopore structure of the olfactory (olf) organs, which serve as molecular filters in Dro-sophila, is built through a novel process in which ER material is trafficked to the plasma membrane mediated by the autophagy pathway. The process is controlled by the insect-specific protein Gore-tex/Osiris23 (Gox)2, which is localized to the tubular ER of olf hair cells. Gox recruits Ref(2)P, the fly counterpart of mammalian p62/SQSTM13, to initiate ER scission. The excised ER fragments are processed by autophagy to gain access to the plasma membrane and trigger membrane invagination, which plays a role in remodeling the cuticular envelope layer to the nanopore formation. This repurposing of the ER phagy for machinery to support the fabrication of nanoscale ECM by the Gox protein sheds light on the nanopatterning of insect cuticles and their genetic control.

Temporal changes in the physical and mechanical properties of beetle elytra during maturation

Abstract:Changes in physical properties of Tenebrio molitor and Tribolium castaneum elytra (hardened forewings) were studied to understand how the development of microstructure and chemical interactions determine cuticle mechanical properties. Analysis of these properties supports a model in which cuticular material is continuously secreted from epidermal cells to produce an extracellular matrix so that the outermost layers mature first. It is hypothesized that enzymatic crosslinking and pigmentation reactions along with dehydration help to stabilize the protein-chitin network within the initial layers of cuticle shortly after eclosion. Mature layers are proposed to bear most of the mechanical loads. The frequency dependence of the storage modulus and the tan δ values decreased during the beginning of maturation, reaching constant values after 48 h post-eclosion. A decrease of tan δ indicates an increase in crosslinking of the material. The water content declined from 75% to 31%, with a significant portion lost from within the open spaces between the dorsal and ventral cuticular layers. Dehydration had a less significant influence than protein crosslinking on the mechanical properties of the elytron during maturation. When Tribolium cuticular protein TcCP30 expression was decreased by RNAi, the tan δ and frequency dependence of E' of the elytron did not change during maturation. This indicates that TcCP30 plays a role in the crosslinking process of the beetle's exoskeleton. This study was inspired by previous work on biomimetic multicomponent materials and helps inform future work on creating robust lightweight materials derived from natural sources. STATEMENT OF SIGNIFICANCE: Examination of changes in the physical properties of the elytra (hardened forewings) of two beetle species advanced understanding of how the molecular interactions influence the mechanical properties of the elytra. Physical characterization, including dynamic mechanical analysis, determined that the outer portion of the elytra matured first, while epidermal cells continued to secrete reactive components until the entire structure reached maturation. RNA interference was used to identify the role of a key protein in the elytra. Suppression of its expression reduced the formation of crosslinked polymeric components in the elytra. Identifying the molecular interactions in the matrix of proteins and polysaccharides in the elytra together with their hierarchical architecture provides important design concepts in the development of biomimetic materials.

Abstract:Recently, much research has been oriented towards the influence of different food wastes and agricultural by-products on the final larval biomass and chemical composition of the insect species Hermetia illucens L. (Diptera: Stratiomyidae). However, there is a gap in the literature regarding the possible relationship between the feeding substrate of H. illucens larvae and chitin. In this context, in the present study, larvae of H. illucens derived from two populations (i.e., UNIPI and UTH), were reared on different diets composed of fruits, vegetables, and meat. Based on the results, the larval survival was high for all diets tested. Larval growth in terms of weight gain, larval length, and feed conversion ratio (FCR) depended on the composition of each diet. The chitin and chitosan composition of larvae, reared on different substrates, did not reveal significant differences. Given the fact that the feeding substrate represent a significant cost in the industrial production of insects, its correlation with a high value product (i.e. chitosan) is important. On the other hand, as the prepupal stage of H. illucens is currently used as animal feed, the metabolization of chitin by farmed animals when the larvae or prepupae were offered as feed could have adverse effects. Thus, depending on the final product that is to be produced, industries could benefit from the establishment of a suitable diet.

Biodiversity and Activity of the Gut Microbiota Across the Life History of the Insect Herbivore Spodoptera Littoralis

Abstract:Microbes that live inside insects play critical roles in host nutrition, physiology, and behavior. Although Lepidoptera (butterflies and moths) are one of the most diverse insect taxa, their microbial symbionts are little-studied, particularly during metamorphosis. Here, using ribosomal tag pyrosequencing of DNA and RNA, we investigated biodiversity and activity of gut microbiotas across the holometabolous life cycle of Spodoptera littoralis, a notorious agricultural pest worldwide. Proteobacteria and Firmicutes dominate but undergo a structural "metamorphosis" in tandem with its host. Enterococcus, Pantoea and Citrobacter were abundant and active in early-instar, while Clostridia increased in late-instar. Interestingly, only enterococci persisted through metamorphosis. Female adults harbored high proportions of Enterococcus, Klebsiella and Pantoea, whereas males largely shifted to Klebsiella. Comparative functional analysis with PICRUSt indicated that early-instar larval microbiome was more enriched for genes involved in cell motility and carbohydrate metabolism, whereas in late-instar amino acid, cofactor and vitamin metabolism increased. Genes involved in energy and nucleotide metabolism were abundant in pupae. Female adult microbiome was enriched for genes relevant to energy metabolism, while an increase in the replication and repair pathway was observed in male. Understanding the metabolic activity of these herbivore-associated microbial symbionts may assist the development of novel pest-management strategies.

Abstract:OBJECTIVE To assess the usefulness of high-frequency diagnostic ultrasonography for evaluation of changes of skin thickness in relation to hydration status and fluid distribution at various cutaneous sites in dogs. ANIMALS 10 clinically normal adult dogs (6 males and 4 females) of various breeds. PROCEDURES Ultrasonographic examination of the skin was performed before and after hydration via IV administration of an isotonic crystalloid solution (30 mL/kg/h for 30 minutes). A 13-MHz linear-array transducer was used to obtain series of ultrasonographic images at 4 different cutaneous sites (the frontal, sacral, flank, and metatarsal regions). Weight and various clinicopathologic variables (PCV; serum osmolality; and serum total protein, albumin, and sodium concentrations) were determined before and after the infusion. These variables and ultrasonographic measurements of skin thickness before and after hydration were compared. RESULTS Among the 10 dogs, mean preinfusion skin thickness ranged from 2,211 microm (metatarsal region) to 3,249 microm (sacral region). Compared with preinfusion values, weight was significantly increased, whereas PCV; serum osmolality; and serum total protein, albumin, and sodium concentrations were significantly decreased after infusion. After infusion, dermal echogenicity decreased and skin thickness increased significantly by 21%, 14%, 15%, and 13% in the frontal, sacral, flank, and metatarsal regions, respectively. CONCLUSIONS AND CLINICAL RELEVANCE Cutaneous site and hydration were correlated with cutaneous characteristics and skin thickness determined by use of high-frequency ultrasonography in dogs. Thus, diagnostic ultrasonography may be a useful tool for the noninvasive evaluation of skin hydration in healthy dogs and in dogs with skin edema.

Abstract:Abstract Diadegma semiclausum (Hellén) (Hymenoptera: Ichneumonidae) is one of the most important biological control agents of the diamondback moth, Plutella xylostella (L.) (Lepidoptera: Plutellidae). Each immature stage of D. semiclausum was studied by dissecting parasitized hosts in the laboratory at 22 ± 1°C and 70–75% RH. Results show that the egg is hymenopteriform, lacking appendages. The larvae seem to be hypermetamorphic, with four instars. The pupal period has two distinct phases: 1) phase I, larval-like with remarkable groups of urate cells; and 2) phase II, exarate pupal appearance with a pigmentation sequence of eyes, ocelli, mandibles, thorax, abdomen, and antennae. The beetle's structural protein CPCFC making elytra tough and rigid

Abstract:The insect cuticle, which serves as both a protective barrier and an efficient lever system for locomotion, is an extracellular matrix primarily composed of chitin and protein. The cuticle protein CPCFC characterized by a "CFC" motif containing 2 Cys split by the insertion of 5 residues is distributed across most insect species and specifically localized in the hard part of the cuticle. However, their physiological function is not fully understood. Here, we report 2 CPCFC proteins, TcCPCFC1 and TcCPCFC2, derived from the Coleopteran insect Tribolium castaneum. We revealed that TcCPCFC1 and TcCPCFC2 were predominantly expressed during the larval and adult stages of T. castaneum, respectively. The transcription downregulation of TcCPCFC1 significantly decreased the modulus and toughness of the elytral cuticle. We found that TcCPCFC proteins have high binding affinity to chitin. We cloned and produced recombinant TcCPCFC proteins and demonstrated that the addition of TcCPCFC proteins to chitin hydrogel greatly enhanced the hydrogel's modulus and toughness by forming denser chitin fibrous networks. Our findings reveal the functional role of CPCFC proteins in enhancing mechanical properties of insect cuticle, and we validate this process in vitro, and offer a protein candidate for fabrication of advanced chitin-based materials.

Abstract:and fish-liver oils for vitamin D, must be treated with reserve. The experiments which have been described have shown that the treatment is ineffective in quantitatively removing vitamin A or, alternatively, in removing enough vitamin A to eliminate interference in the vitamin D determination. The procedure of Milas et al. (1941) completely destroys large amounts of vitamin D. and over 60% of calciferol, even when large quantities of the latter are used. The safest solvent in which to carry out the condensation is ether, but unfortunately, although good recoveries of calciferol and vitamin D, have been obtained with this solvent, complete elimination of vitamin A is not possible within reasonable times of reaction. Condensation in benzene, as used by Pirlot & Rougir (1947), results in up to 20% loss of calciferol and about 40% loss of vitamin D3. The method of selective ultraviolet irradiation to remove vitaminAfrom vitaminD does not appear to be suitable for routine use. It is possible that, under carefully controlled conditions, small amounts of vitamin A (the bulk of which has been removed by other methods) can be successfully eliminated from vitamin D-containing material. Experiments have shown that although vitamin A can be smnoothly destroyed by filtered radiation in the 300-400 m,. range, the irradiation, under similar conditions, of small amounts of vitamin D results in inevitable losses.

Abstract:The black soldier fly (BSF), Hermetia illucens (Diptera: Stratiomyidae), has considerable global interest due to its outstanding capacity in bioconverting organic waste to insect biomass, which can be used for livestock, poultry, and aquaculture feed. Mass production of this insect in colonies requires the development of methods concentrating oviposition in specific collection devices, while the mass production of larvae and disposing of waste may require substrates that are more palatable and more attractive to the insects. In insects, chemoreception plays an essential role throughout their life cycle, responding to an array of chemical, biological and environmental signals to locate and select food, mates, oviposition sites and avoid predators. To interpret these signals, insects use an arsenal of molecular components, including small proteins called odorant binding proteins (OBPs). Next generation sequencing was used to identify genes involved in chemoreception during the larval and adult stage of BSF, with particular attention to OBPs. The analysis of the de novo adult and larval transcriptome led to the identification of 27 and 31 OBPs for adults and larvae, respectively. Among these OBPs, 15 were common in larval and adult transcriptomes and the tertiary structures of 8 selected OBPs were modelled. In silico docking of ligands confirms the potential interaction with VOCs of interest. Starting from the information about the growth performance of H. illucens on different organic substrates from the agri-food sector, the present work demonstrates a possible correlation between a pool of selected VOCs, emitted by those substrates that are attractive for H. illucens females when searching for oviposition sites, as well as phagostimulants for larvae. The binding affinities between OBPs and selected ligands calculated by in silico modelling may indicate a correlation among OBPs, VOCs and behavioural preferences that will be the basis for further analysis.

Abstract:Diadromus collaris (Gravenhorst) is an important pupal parasitoid of the diamondback moth, Plutella xylostella (L.), a major insect pest of cruciferous vegetables worldwide. We studied the development of immature stage of D. collaris by dissecting parasitized hosts in the laboratory at 25 +/- 1 degrees C and 50-80% relative humidity. The results show that all immature stages complete their development within the same host in 11 approximate to 12 d. The egg is hymenopteriform and appears to be anhydropic. There are four larval instars. The first instar is transparent with a sclerotized rectangular chitinous head capsule and distinct mandibles. Head capsule of the second instar turns into more isosceles trapezium-shaped. The third instar looks similar to the second instar but proportionally much larger. The sclerization and dimension of the fourth instar increases significantly. The pupa is a typical exarate form and lacks a cocoon. All life history was fully documented with detailed photomicrographs. Our study will be useful for understanding the physiological interactions between D. collaris and P. xylostella.

Abstract:Beetles possess a set of highly modified and tanned forewings, elytra, which are lightweight yet rigid and tough. Immediately after eclosion, the elytra are initially thin, pale and soft. However, they rapidly expand and subsequently become hardened and often dark, resulting from both pigmentation and sclerotization. Here, we identified changes in protein composition during the developmental processes of the elytra in the Japanese rhinoceros beetle, Trypoxylus dichotomus. Using mass spectrometry, a total of 414 proteins were identified from both untanned and tanned elytra, including 31 cuticular proteins (CPs), which constitute one of the major components of insect cuticles. Moreover, CPs containing Rebers and Riddiford motifs (CPR), the most abundant CP family, were separated into two groups based on their expression and amino acid sequences, such as a Gly-rich sequence region and Ala-Ala-Pro repeats. These protein groups may play crucial roles in elytra formation at different time points, likely including self-assembly of chitin nanofibers that control elytral macro and microstructures and dictate changes in other properties (i.e., mechanical property). Clarification of the protein functions will enhance the understanding of elytra formation and potentially benefit the development of lightweight materials for industrial and biomedical applications. STATEMENT OF SIGNIFICANCE: The beetle elytron is a light-weight natural bio-composite which displays high stiffness and toughness. This structure is composed of chitin fibrils and proteins, some of which are responsible for architectural development and hardening. This work, which involves insights from molecular biology and materials science, investigated changes in proteomic, architectural, and localized mechanical characteristics of elytra from the Japanese rhinoceros beetle to understand molecular mechanisms driving elytra development. In the present study, we identified a set of new protein groups which are likely related to the structural development of elytra and has potential for new pathways for processing green materials.
