Abstract:FIELD: chemistry. SUBSTANCE: invention relates to agomelatine co-crystall, which is characterised by that it consists of agomelatine, or N-[2-(7-methoxy-1-naphtyl)ethyl]acetamide of formula (I), and organic acid, which is in solid state at ambient temperature, which is selected from a steam-hydroxybenzoic acid, citric acid, oxalic acid, Gallic acid, maleic acid, malonic acid, glutaric acid, glycolic acid or ketoglutaric acid. Agomelatine co-crystall and organic acid are obtained by mixing two components in organic solvent in desirable proportions: 1 agomelatine equivalent at 0.25-4 mol equivalent of organic acid, than obtained solution is stirred and optionally is heated at temperature not exceeding boiling point of selected solvent, then mixture is cooled while stirring, and deposited co-crystall naturally or precipitates after absorption in second solvent, obtained precipitate is filtered and dried. Also co-crystall is obtained by combined grinding of two components. Also co-crystall is obtained by mixing two components in organic or aqueous-organic solvent, followed by freezing and drying at very low temperature. Alternative method of preparing co-crystall involves mixing powders agomelatine and said acid in mixer, and then mixture is extruded by extrusion in twin-screw mixers without matrix to obtain solid granulated product directly at extrusion device outlet. Agomelatine co-crystall and organic acid are applicable for preparing drugs for treating disturbed melatonin aergic system. Development of a Novel Amorphous Agomelatine Formulation With Improved Storage Stability and Enhanced Bioavailability

Abstract:The present work describes the development of a novel formulation of amorphous agomelatine (AGM) that exhibits enhanced in vitro dissolution rate and bioavailability, as well as improved storage stability. AGM was loaded on a mixture of microcrystalline cellulose with a high specific surface area excipient, namely colloidal silicon dioxide, employing a wet granulation method, and the resultant AGM granules were subsequently formulated into immediate release film-coated tablets. Modulated temperature differential scanning calorimetry, hot-state light microscopy, powder X-ray diffraction, attenuated total reflectance FTIR, and micro-Raman spectroscopy revealed that the active pharmaceutical ingredient existed primarily in the amorphous state within the prepared formulations, with some crystals of polymorph I also present. Accelerated stability studies for up to 6 months in alu-alu blisters showed good physicochemical stability during storage. Finally, in vitro dissolution studies and clinical trials in healthy human volunteers showed a remarkable increase in the in vitro dissolution rate and a ∼1.5-fold increase in bioavailability, respectively, compared to the marketed product.

Abstract:Agomelatine is a new and novel non-SSRI potential treatment option for major depressive disorders, discovered and developed by Servier Laboratories with first marketing approval in 2009. In this work, the solution-mediated crystal form I to II transformation process was studied. The influence factors, such as system temperature, crystallization solvents, and some pharmaceutical excipients, were investigated. DSC, XRPD, SEM, react IR, and in situ FBRM were used to monitor the transformation process. Compared with crystal form I of agomelatine, form II was the thermodynamic stable one, therefore, the transformation was a spontaneous process. By increasing the system temperature and/or the volume ratio of IPA in the solvent mixture, the transformation process would be accelerated. Lactose hydrate and HPMC would slightly inhibit the transformation, while PVP K30 would accelerate transformation process by increasing the solubility.

Abstract:Nanocrystal formulations of the BCS class II agomelatine, were developed by wet media milling. The most suitable stabilizer was identified and effects of process and formulation variables on the nanocrystal size and ζ-potential were evaluated employing a Box-Behnken experimental design. The optimized nanosuspensions were dried and subsequently evaluated for redispersibility and physicochemical properties. Computational simulation of solid state properties was applied to rationalize crystal fracture. It was found that low viscosity hydroxypropylcellulose with sodium dodecyl sulfate is the most suitable stabilizer. Stabilizer concentration exerts a statistically significant effect on particle size, which depends on the mill's rotation speed. The milling process induces a polymorphic transition to form II, which could affect size reduction kinetics. The solidified nanosuspensions' redispersibility is deteriorating progressively with storage time, with only minor differences between drying methods, retaining enhanced dissolution rate. Crystal lattice simulations suggest high mechanical anisotropy of form I crystals, which could be an additional reason for fast particle size reduction prior to the polymorphic transformation. Wet media milling, combined with a suitable drying method, can be an efficient technique for the production of stable nanocrystals of agomelatine. Particle informatics methods can enhance our understanding of the mechanisms responsible for agomelatine's nanocomminution.

Abstract:The invention provides a method for preparing dimemorfan phosphate crystal form II, which comprises the following steps: adding a crude dimemorfan phosphate product into methanol, heating under reflux to dissolve the crude dimemorfan phosphate product, cooling to precipitate crystals, filtering, and drying to obtain the solid dimemorfan phosphate crystal form II. The dimemorfan phosphate crystal form has the following X-diffractometer diagram characteristics; and in the X-ray powder diffractogram, interplanar distance d and Bragg angle 2theta represent the relative intensity relative to the percentage of the strongest line. The invention also provides a pharmaceutical composition containing the dimemorfan phosphate crystal form II as an active component and one or more proper auxiliary materials, such as tablets, capsules, granules, oral liquids, syrups, suspensoids and injections. The dimemorfan phosphate crystal form II has favorable pharmaceutical values, and has the characteristics of favorable purity, definite crystal form, excellent reproducibility and the like; and the dimemorfan phosphate crystal form II has valuable physiological activities in the field of preparations, and can be stored for a long time while remaining high stability.

Abstract:Ezetimibe is a novel selective cholesterol absorption inhibitor which was developed by both Schering-Plough and Merck. An improved process for the preparation of ezetimibe was studied and was described as follows. N-(4-(benzyloxy)benzylidene)-4-fluorobenzenamine (Compound 2) was prepared from 4-hyd-roxybenzaldehyde, 4-fluorobenzenamine and benzyl chloride in one pot. 5-(4-Fluorophenyl)-5-oxopentanoic acid was reacted with pivaloyl chloride and directly condensed with (S)-4-phenyloxazolidin-2-one promoted with LiCl, and then reduced by CBS/BH3 to get (4S)-3-[(5S)-5-(4-fluoror)-5-hydrox-1-oxo-pentyl]-4-phenyl-oxazolidin-2-one (Compound 4). It was condensed with compound 2 catalyzed with Lewis acid after being protected by TMS to afford the condensation intermediate. Finally it was cyclized by TBAF·3H2O and deprotected with Pd/C to get the title compound with a total yield of 56%. Compared with the current technology, the present process shows advantage in high yield, less raw material consumed, and easy production on industrial scale. The specific rotation and melting point of the prepared title compound is the same as that reported in literature,and the structure of the prepared title compound and some intermediates were confirmed by MS and 1H-NMR.

Abstract:An improved, scalable, industrially feasible telescopic process for multigram synthesis of highly commercialized antidepressant drug, moclobemide has been developed. Unlike the conventional approaches, this process enjoys the privilege of being an economical, operationally simple, effectively scalable, green and quantitative method for the synthesis of amide‐containing drug moclobemide, showcasing easy isolation and purification without the aid of column chromatography. An environmentally benign reagent combination of TBN and NHS very effectively converts the acyl source, 4‐chlorobenzaldehyde into its N‐hydroxysuccinimide ester intermediate, which simply affords the drug molecule after nucleophilic substitution with the amine source. The synthetic process has successfully been scaled‐up upto ~ 145 g scale with 75% overall yield and >95% first crop HPLC purity.

Abstract:Carbohydrate-based low molecular weight gelators (LMWGs) exhibit many desirable properties making them useful in various fields including applications as drug delivery carriers. In order to further understand the structural connection to gelation properties, especially the influence of halide substitutions, we have designed and synthesized a series of para-chlorobenzylidene acetal protected D-glucosamine amide derivatives. Fifteen different amides were synthesized, and their self-assembling properties were assessed in multiple organic solvents, as well as mixtures of organic solvents with water. All derivatives were found to be gelators for at least one solvent and majority formed gels in multiple solvents at concentrations lower than 2 wt%. A few derivatives rendered remarkably stable gels in aqueous solutions at concentrations below 0.1 wt%. The benzamide 13 formed gels in water and in EtOH/H2O (v/v 1:2) at 0.36 mg/mL. The gels were characterized using optical microscopy and atomic force microscopy, and the self-assembly mechanism was probed using variable temperature 1H-NMR spectroscopy. Gel extrusion studies using H2O/DMSO gels successfully printed lines of gels on glass slides, which retained viscoelasticity based on rheology. Gels formed by the benzamide 13 were used for encapsulation and the controlled release of chloramphenicol and naproxen, as well as for dye removal for toluidine blue aqueous solutions.
Scaffold Hopping Toward Agomelatine: Novel 3, 4-Dihydroisoquinoline Compounds as Potential Antidepressant Agents

Abstract:A scaffold-hopping strategy toward Agomelatine based on in silico screening and knowledge analysis was employed to design novel antidepressant agents. A series of 3, 4-dihydroisoquinoline compounds were selected for chemical synthesis and biological assessment. Three compounds (6a-1, 6a-2, 6a-9) demonstrated protective effects on corticosterone-induced lesion of PC12 cells. Compound 6a-1 also displayed low inhibitory effects on the growth of HEK293 and L02 normal cells and it was further evaluated for its potential antidepressant effects in vivo. The forced swim test (FST) results revealed that compound 6a-1 remarkably reduced the immobility time of rats and the open field test (OFT) results indicated a better general locomotor activity of the rats treated with compound 6a-1 than those with Agomelatine or Fluoxetine. Mechanism studies implied that compound 6a-1 can significantly reduce PC12 cell apoptosis by up-regulation of GSH and down-regulation of ROS in corticosterone-induced lesion of PC12 cells. Meanwhile, the down-regulation of calcium ion concentration and up-regulation of BDNF level in PC12 cells may account for the neuroprotective effects. Furthermore, compound 6a-1 can increase cell survival and cell proliferation, promote cell maturation in the rat hippocampus after chronic treatment. The acute toxicity data in vivo indicated compound 6a-1 exhibited less hepatotoxicity than Agomelatine.

Abstract:GLYX-13, a NMDAR glycine-site partial agonist, was discovered as a promising antidepressant with rapidly acting effects but no ketamine-like side effects. However, the reported synthetic process route had deficiencies of low yield and the use of unfriendly reagents. Here, we report a scaled-up synthesis of GLYX-13 with an overall yield of 30% on the hectogram scale with a column chromatography-free strategy, where the coupling and deprotection reaction conditions were systematically optimized. Meanwhile, the absolute configuration of precursor compound of GLYX-13 was identified by X-ray single crystal diffraction. Finally, the activity of GLYX-13 was verified in the cortical neurons of mice through whole-cell voltage-clamp technique.

Abstract:OBJECTIVETo establish a new approach to synthesis of diethyl N-[4-[(2,4-diaminopyrido[3,2-d]pyrimidin-6-yl)methylamino]benzoyl]-L-glutamate.METHODSTarget compound (5) was synthesized by the use of (2,4-dioxo-tetrahydropyridopyrimidin-6-yl)methyl acetate (1) as starting material via hydrolysis, chlorination, condensation with diethyl (p-aminobenzoyl)glutamate and aminolysis.RESULTSA new approach to synthesis of diethyl N-[4-[(2,4-diaminopyrido[3,2-d]pyrimidin-6-yl)methylamino]benzoyl]-L-glutamate was established. This synthetic route has hydrolysis reaction, chlorination, diethyl N-(p-aminobenzoyl)-L-glutamate condensation reaction and ammonolysis reaction. The total yield is 36.7%.The structures of those compounds have identified by 1H nuclear magnetic resonance, 13C nuclear magnetic resonance and mass spectrometry. This synthetic route avoid the unstable brominated reaction product and improves the harsh condition of ammonolysis reaction.CONCLUSIONThe new synthetic route has improved the reaction condition and the stability of the intermediate, and increased the extent of the derivative compounds, which has great significance to anti-folic acid of anti-tumor inhibitor synthesis.

Abstract:A new and efficient method has been reported for the synthesis of 2-amino-9-[4-acetoxy-3-(acetoxymethyl)butyl-1-yl]purine (famciclovir) starting from guanine. The route involves chlorination of guanine, optimized Mitsunobu reaction, coupling with diethyl malonate, hydrogenation, reduction and esterification, and the overall yield is about 29%. This method does not require any form of chromatographic purification to give pure famciclovir, and it is an industrially viable manufacturing process for this drug.
A Novel Synthetic Process for Zolpidem Tartrate

Abstract:A new synthetic process of zolpidem tartrate (1) was reported.3-Bromo-4-(4-methylphenyl)-4-oxobutanoic acid (4) was synthesized via Friedel-Crafts reaction and bromonation from toluene (2),than the crude product 4 reacted with 2-amino-5-methylpyridine (5) to give 2-[6-methyl-2-(p-tolyl) imidazo [1,2-a] pyridin-3-yl] acetic acid (6).The latter reacted with dimethylamine to afford zolpidem (7).Finally,the target compound 1 was obtained via a salification of 7 and L-(+)-tartaric acid with a total yield of 36％ (based on 2) and a purity of 99.8％.The synthesis of compound 6 from 4 and 5 was a new method which have not yet been reported in literature.

Abstract:Background: Agomelatine is a novel antidepressant with agonist activity at melatonin receptors (MT(1) and MT(2)), and antagonistic effects at the 5HT(2c) serotonin receptor. Scope: This is a review of current evidence for clinical efficacy and safety, drawn from a literature review using Medline and PsycInfo in 2010 using search terms including agomelatine, melatonergic antidepressant and Valdoxan. Findings: Agomelatine has antidepressant properties and from early on in its prescription agomelatine can improve sleep architecture and efficiency. Licensed for the treatment of depression in adults, other potential indications include seasonal affective disorder and anxiety disorders. Early controlled short-term studies of agomelatine as an antidepressant indicate good efficacy and tolerability. There are fewer side effects on sexual health than with SSRI antidepressants. No withdrawal or discontinuation syndrome has yet been observed. Agomelatine is contraindicated in patients with impaired liver function and repeated liver function tests are suggested during prescription. Conclusion: Agomelatine is a useful and novel addition to the range of avialble antidepressants. Longer term post-marketing studies as to the effects of chronic exposure are required.

Abstract:ABSTRACT Introduction Under the treatment of commonly used antidepressants, many patients with major depressive disorder (MDD) do not achieve remission. All previous first-line treatments for depression have focused on the enhancement of monoaminergic activity. Agomelatine was the first antidepressant with a mechanism of action extending beyond monoaminergic neurotransmission. Areas covered The aim of this case history is to describe the discovery strategy and development of agomelatine. The pharmacodynamic profile of the drug is briefly presented. The article summarizes (a) the preclinical behavioral data on agomelatine’s effects on depressive-like behavior, anxiety, and circadian rhythmicity disruptions, and (b) the results of early preclinical studies on safety, efficacy in MDD, and the risk-benefit pharmacological profile. Furthermore, the article examines findings of post-marketing research on safety, efficacy, and cost-effectiveness of the drug. Expert opinion There is now evidence supporting the clinical efficacy and safety profile of agomelatine in the acute-phase treatment of MDD. Agomelatine may be more effective in specific subgroups of MDD patients, those with severe anxiety symptoms or disturbed circadian profiles. Its antidepressant and anxiolytic activities are due to synergy between its melatonergic and 5-hydroxytryptaminergic effects. Since its discovery, novel compounds acting on the melatonergic system have been under investigation for the treatment of MDD.

Abstract:Key PointsMajor depression is a serious and debilitating disease for which current pharmacotherapy is inadequate in terms of limited efficacy, a long delay to onset of action and induction of side effects.All currently available antidepressants act by monoaminergic mechanisms of action, generally through the suppression of monoamine reuptake.Circadian rhythms, which are under the control of the suprachiasmatic nucleus, are often disrupted in depressed states. Accordingly, suprachiasmatic nucleus-localized melatonergic receptors, which are known to synchronize circadian rhythms, are an attractive target for the treatment of depression.Agomelatine, the first melatonergic antidepressant, was designed to improve depressed states by resynchronizing perturbed biological rhythms. Its 'synergistic' agonist properties at melatonin receptors plus antagonist properties at 5-hydroxytryptamine 2C (5-HT2C) receptors account for its beneficial influence on depressed states.In extensive preclinical studies using cellular, neurochemical and behavioural procedures, agomelatine displayed a broad-based profile of actions that were predictive of antidepressant activity.Therapeutic trials showed that agomelatine displays both short-term and long-term efficacy in major depression, including an early improvement in sleep quality and daytime functioning, preservation of sexual function, lack of weight gain, lack of withdrawal symptoms after discontinuation, and good tolerability.On the basis of the above data, agomelatine was granted marketing authorization in 2009 for the treatment of major depression in Europe.

Abstract:Background: Glycosylamine play an important role in living organisms. Most plants store their chemical resources in the form of inactive glycosides, which are broken down and converted into sugar in the body of herbivores by hydrolyzing enzymes. Glycosylamine are obtained from the secondary metabolism of plants and consist of two parts. One part of it contains sugar like glucose and is inactive in most substances, and has a good effect on the solubility of the glycosylamine derivatives and its absorption and even its transfer from one organ to another. The therapeutic effect is related to the second part, which is called aglycan (or aglucan). Glycosylamine derivatives form a large group of valuable medicinal substances, which at the same time include some of the most dangerous and toxic substances in nature. These substances are present in many groups of flowering plants. Glycosides are made in different ways in different metabolic pathways. These materials have a complex and special chemical structure and leave special effects on the human body. Glycosylamine derivatives are called O-glycosid, N-glycosid, S-glycosid in terms of atoms coupling to anomeric carbon. Carbohydrate esterification reaction to prepare new glycosylamine derivatives is one of the most important carbohydrate reactions. The anomeric position of carbohydrates with strong leaving groups is very important for the preparation of glycosides. Preparation of glycosylamine derivatives based on acetylated carbohydrates is the main purpose of this article. Different carbohydrates were acetylated under mild conditions and high yields. The anomeric position was deacetylated by a magnesium oxide heterogeneous catalyst in methanol solvent. In order to prepare new glycosides acetylated/ deacytylated carbohydrate reacted with N-Methyl-(naphtha-2- ylmethoxy)amine. The final product was identified by various spectroscopic methods. Methods: Anhydrous sodium acetate (4 g) and α-D-glucose (28 mg, 5 g) were mixed. The mixture was transferred to a 200 ml Round-bottom flask . Acetic acid (260 mg, 25 ml) was added to the reaction mixture. The reaction mixture was heated in a boiling water bath for 2 h until complete dissolution of the glucose. Then 100 ml of ice was added. After 1 hour, White crystals formed and were washed with cold water. And then 1 mol of glucose pentaacetate per 50 ml of methanol was dissolved by a magnetic stirrer. After thatMgO (0.2gr) was added to the reaction mixture. The reaction mixture was refluxed at room temperature within 4-5 hours. After 5 hours, the solvent was removed and separated by chromatography. It is then washed with hexane and crystallized by etherhexane. And the final step reacted with N-Methyl- (naphtha-2-ylmethoxy) amine in order to prepare new glycosylamine derivatives. Results: Magnesium oxide in methanol solvent is one of the best effective catalysts in the deacetylation of the anomer position of acetylated carbohydrates. It is done under ambient temperature and in very easy conditions and makes carbohydrates susceptible to extensive chemical reactions. One of these reactions is the formation of glycosides. Various carbohydrates are acetylated by acetic anhydride in the presence of sodium acetate, and the anomeric position is deacetylated by a heterogeneous catalyst. In order to prepare N-glycoside, the glycosides are reacted with N-alkoxy Nmethylglycosyl amine. The final products are identified by various spectroscopic methods. Conclusion: Glucose, mannose, for example, were acetylated by acetic anhydride in the presence of sodium acetate. And the anomeric position of pentaacetate glucose was selectively deacetylated with magnesium oxide in a methanol solvent. Then, the reaction for the preparation of glycosylamine derivatives was carried out under very mild and easy conditions. The aminoglycosides synthesized in this article are used as raw materials for the synthesis of a wide range of antibacterials.

Abstract:The development of a large scale synthesis of the glucocorticoid agonist BI 653048 BS H3PO4 (1 center dot H3PO4) is presented. A key trifluoromethyl ketone intermediate 22 containing an N-(4-methoxyphenyl)ethyl amide was prepared by an enolization/bromine-magnesium exchange/electrophile trapping reaction. A nonselective propargylation of trifluoromethyl ketone 22 gave the desired diastereomer in 32% yield and with dr = 98:2 from a 1:1 diastereomeric mixture after crystallization. Subsequently, an asymmetric propargylation was developed which provided the desired diastereomer in 4:1 diastereoselectivity and 75% yield with dr = 99:1 after crystallization. The azaindole moiety was efficiently installed by a one pot cross coupling/indolization reaction. An efficient deprotection of the 4-methoxyphenethyl group was developed using H3PO4/anisole to produce the anisole solvate of the API in high yield and purity. The final form, a phosphoric acid cocrystal, was produced in high yield and purity and with consistent control of particle size.
