Abstract:Gramine reacted with diethyl acetamidomalonate in the presence of sodium methoxide,the intermediate was selectively hydrolyzed in 4% aqueous solution of sodium hydroxide to give N-acetyl-DL-tryptophane,then DL-tryptophane was obtained through hydrolyzation using 10% aqueous solution of sodium hydroxide.The resolution was carried out with D-tartaric acid to give D-tryptophane D-tartaric acid salt,and benzaldehyde was used as catalyst to make asymmetric transformation.At last the salt was neutralized with triethylamine to give D-tryptophane.The optimum reaction conditions for condensation and resolving reaction were studied.For condensation reaction the molar ratio of sodium methoxide and gramine was 20%,and gramine∶diethyl acetamidomalonate= 1∶1.2(mol);For resolving reaction acetic acid was used as solvent,70℃,the molar ratio of benzaldehyde and DL-tryptophan was 10%,and DL-tryptophane∶D-tartaric acid= 1∶2(mol).

Abstract:Major depression is one of the high prevalent and leading disable mental disorders,and the available drugs are mainly focusing on the monoamine systems.Agomelatine is a novel antidepressant,possessing both melatonergic agonist and 5-hydroxytryptamine 2C(5-HT_2C) antagonist properties.Several lines of evidence from the animal model and clinical studies have showed that agemelatine has definite antidepressant efficacy and has some superiorities such as quick onset time,good tolerance.Agomelatine has been a new choice for treatment of depression.This review summarized the pharmacological action of agemelatine,aimed to help the clinical practictioner understanding this drug more clearly and use it reasonably.

Abstract:A scaffold-hopping strategy toward Agomelatine based on in silico screening and knowledge analysis was employed to design novel antidepressant agents. A series of 3, 4-dihydroisoquinoline compounds were selected for chemical synthesis and biological assessment. Three compounds (6a-1, 6a-2, 6a-9) demonstrated protective effects on corticosterone-induced lesion of PC12 cells. Compound 6a-1 also displayed low inhibitory effects on the growth of HEK293 and L02 normal cells and it was further evaluated for its potential antidepressant effects in vivo. The forced swim test (FST) results revealed that compound 6a-1 remarkably reduced the immobility time of rats and the open field test (OFT) results indicated a better general locomotor activity of the rats treated with compound 6a-1 than those with Agomelatine or Fluoxetine. Mechanism studies implied that compound 6a-1 can significantly reduce PC12 cell apoptosis by up-regulation of GSH and down-regulation of ROS in corticosterone-induced lesion of PC12 cells. Meanwhile, the down-regulation of calcium ion concentration and up-regulation of BDNF level in PC12 cells may account for the neuroprotective effects. Furthermore, compound 6a-1 can increase cell survival and cell proliferation, promote cell maturation in the rat hippocampus after chronic treatment. The acute toxicity data in vivo indicated compound 6a-1 exhibited less hepatotoxicity than Agomelatine.

Abstract:Background: Agomelatine is a novel antidepressant with agonist activity at melatonin receptors (MT(1) and MT(2)), and antagonistic effects at the 5HT(2c) serotonin receptor. Scope: This is a review of current evidence for clinical efficacy and safety, drawn from a literature review using Medline and PsycInfo in 2010 using search terms including agomelatine, melatonergic antidepressant and Valdoxan. Findings: Agomelatine has antidepressant properties and from early on in its prescription agomelatine can improve sleep architecture and efficiency. Licensed for the treatment of depression in adults, other potential indications include seasonal affective disorder and anxiety disorders. Early controlled short-term studies of agomelatine as an antidepressant indicate good efficacy and tolerability. There are fewer side effects on sexual health than with SSRI antidepressants. No withdrawal or discontinuation syndrome has yet been observed. Agomelatine is contraindicated in patients with impaired liver function and repeated liver function tests are suggested during prescription. Conclusion: Agomelatine is a useful and novel addition to the range of avialble antidepressants. Longer term post-marketing studies as to the effects of chronic exposure are required.

Abstract:The synthetic method on(R)-rolipram was described in detail.Using isoacetovanillone as materials,(R)-rolipram was synthesized by six steps of alkylation,Wittig-Horner,bromo reaction,phthalimide,enantioselective conjugate reduction and deprotection followed by cyclization reaction.The product yield was 34%.The structure of the product was determined by 1H NMR and MS.

Abstract:Key PointsMajor depression is a serious and debilitating disease for which current pharmacotherapy is inadequate in terms of limited efficacy, a long delay to onset of action and induction of side effects.All currently available antidepressants act by monoaminergic mechanisms of action, generally through the suppression of monoamine reuptake.Circadian rhythms, which are under the control of the suprachiasmatic nucleus, are often disrupted in depressed states. Accordingly, suprachiasmatic nucleus-localized melatonergic receptors, which are known to synchronize circadian rhythms, are an attractive target for the treatment of depression.Agomelatine, the first melatonergic antidepressant, was designed to improve depressed states by resynchronizing perturbed biological rhythms. Its 'synergistic' agonist properties at melatonin receptors plus antagonist properties at 5-hydroxytryptamine 2C (5-HT2C) receptors account for its beneficial influence on depressed states.In extensive preclinical studies using cellular, neurochemical and behavioural procedures, agomelatine displayed a broad-based profile of actions that were predictive of antidepressant activity.Therapeutic trials showed that agomelatine displays both short-term and long-term efficacy in major depression, including an early improvement in sleep quality and daytime functioning, preservation of sexual function, lack of weight gain, lack of withdrawal symptoms after discontinuation, and good tolerability.On the basis of the above data, agomelatine was granted marketing authorization in 2009 for the treatment of major depression in Europe.

Abstract:ABSTRACT Introduction Under the treatment of commonly used antidepressants, many patients with major depressive disorder (MDD) do not achieve remission. All previous first-line treatments for depression have focused on the enhancement of monoaminergic activity. Agomelatine was the first antidepressant with a mechanism of action extending beyond monoaminergic neurotransmission. Areas covered The aim of this case history is to describe the discovery strategy and development of agomelatine. The pharmacodynamic profile of the drug is briefly presented. The article summarizes (a) the preclinical behavioral data on agomelatine’s effects on depressive-like behavior, anxiety, and circadian rhythmicity disruptions, and (b) the results of early preclinical studies on safety, efficacy in MDD, and the risk-benefit pharmacological profile. Furthermore, the article examines findings of post-marketing research on safety, efficacy, and cost-effectiveness of the drug. Expert opinion There is now evidence supporting the clinical efficacy and safety profile of agomelatine in the acute-phase treatment of MDD. Agomelatine may be more effective in specific subgroups of MDD patients, those with severe anxiety symptoms or disturbed circadian profiles. Its antidepressant and anxiolytic activities are due to synergy between its melatonergic and 5-hydroxytryptaminergic effects. Since its discovery, novel compounds acting on the melatonergic system have been under investigation for the treatment of MDD.

Abstract:FIELD: chemistry. SUBSTANCE: invention relates to agomelatine co-crystall, which is characterised by that it consists of agomelatine, or N-[2-(7-methoxy-1-naphtyl)ethyl]acetamide of formula (I), and organic acid, which is in solid state at ambient temperature, which is selected from a steam-hydroxybenzoic acid, citric acid, oxalic acid, Gallic acid, maleic acid, malonic acid, glutaric acid, glycolic acid or ketoglutaric acid. Agomelatine co-crystall and organic acid are obtained by mixing two components in organic solvent in desirable proportions: 1 agomelatine equivalent at 0.25-4 mol equivalent of organic acid, than obtained solution is stirred and optionally is heated at temperature not exceeding boiling point of selected solvent, then mixture is cooled while stirring, and deposited co-crystall naturally or precipitates after absorption in second solvent, obtained precipitate is filtered and dried. Also co-crystall is obtained by combined grinding of two components. Also co-crystall is obtained by mixing two components in organic or aqueous-organic solvent, followed by freezing and drying at very low temperature. Alternative method of preparing co-crystall involves mixing powders agomelatine and said acid in mixer, and then mixture is extruded by extrusion in twin-screw mixers without matrix to obtain solid granulated product directly at extrusion device outlet. Therapeutic Advances in Psychopharmacology

Abstract:Even with many antidepressant and anxiolytic drugs available on the market, there are still patients who do not respond well to the standard first or second line treatments for affective or anxiety disorders. The antidepressant agomelatine has been used in Europe for several years. Agomelatine, an agonist at melatonin receptors and an antagonist at serotonin receptors, can be particularly useful in patients suffering from a major depressive disorder associated with insomnia. Some clinical data have shown a limited effect for agomelatine in a subset of patients with major depression. A number of case reports published in 2011-2016 describe the effect of agomelatine in combination with an established antidepressant, such as escitalopram, venlafaxine, duloxetine, moclobemide or bupropion. A successful combination of agomelatine was reported after adjunctive use of agomelatine combined with clomipramine, escitalopram, and venlafaxine in patients with major depression or obsessive–compulsive disorder. Moreover, bupropion or moclobemide augmentation with agomelatine in patients with major depressive disorder led to a significant improvement. Other supportive data have been published, such as analysis of the VIVALDI study, although it should be noted that the study was supported by the manufacturer of agomelatine. In this study, agomelatine in combination with other antidepressants was shown to be effective and well tolerated in practice, although the most effective antidepressant treatment in the study consisted of agomelatine alone and not in combination with other antidepressants. There have also been two published case reports about the concomitant use of duloxetine and agomelatine which were not efficacious. The positive results of agomelatine augmentation with other antidepressants should be confirmed through randomized, double-blind clinical trials.

Abstract:The chlorinated tetracyclic 1,5-dichloro-9,10-dihydro-9,10-ethanoanthracen-12-yl)-N-methylmethanamine 1, a maprotiline analog, has been synthesized via reduction and the Diels–Alder reaction followed by reductive amination of aldehyde 2.1D-NMR (DEPT) and 2D-NMR (HSQC, DQF-COSY) techniques were recruited for structural elucidation in addition to HRMS. Density functional theory calculations were performed to identify the possible isomers of the intermediate compound aldehyde 2; these calculations were in good agreement with experimental results where aldehyde 2 could exist in three isomers with comparable energies. In addition, the side chain of this aldehyde 2 was extended via the Wittig reaction to obtain the unsaturated ester 5 that was subjected to selective olefinic catalytic hydrogenation to obtain the corresponding saturated ester 6. Molecular docking simulation showed that all the compounds (1, 2, 5, and 6) have high antidepressant activities and form stable complexes with LeuT by inhibiting the neurotransmitter reuptake at the synapse and hence are good candidates as antidepressant drugs.

Abstract:In this study, a series of novel arylpropylamine derivatives were designed, synthesized and evaluated as potential multi-target antidepressants. Among them, compound (R)-13j displayed unique pharmacological features, exhibiting excellent inhibitory potency against serotonin and noradrenaline transporters (SERT/NET) and high affinity for 5-HT2A/2C receptor, and showing low affinity for histamine H1, adrenergic α1 receptors and hERG channels (to reduce QT interval prolongation). Molecular docking studies provided a rational binding model of (R)-13j in complex with SERT and 5-HT2A/2C receptor. In animal models, compound (R)-13j dose-dependently reduced the immobility time in the tail suspension test (TST) and the forced swimming test (FST) in mice, with higher efficacy when compared to duloxetine, and showed no stimulatory effect on the locomotor activity. Moreover, compound (R)-13j significantly shortened the immobility time in the ACTH-induced rat model of treatment-resistant depression (TRD). Furthermore, compound (R)-13j also exhibited a higher threshold for acute toxicity than duloxetine. In addition, compound (R)-13j possessed a favorable pharmacokinetic profile in mice. Taken together, compound (R)-13j may constitute a novel class of drugs for the treatment of depression.

Abstract:OBJECTIVE:To synthesize venlafaxine with an improved novel method.METHODS:p-methoxypheny lethyl-acid was reacted with SOCl(2) to produce acyl chloride which was reacted with N,N-dimethylamine solution to get amide; then through Ivanov reaction and reduction by KBH(4)/BF(3).Et(2)O to yield venlafaxine.RESULT:Venlafaxine was successfully synthesized by using this method with an yield rate of 50.3%.CONCLUSION:The improved method is suitable for industrial production of venlafaxine.

Abstract:Depression is a common psychiatric disorder with an estimated global prevalence of 4.4 %. Here, we designed a series of new multimodal monoaminergic arylpiperazine derivatives using a pharmacophore hybrid approach and synthesized them for the treatment of depression. Molecular docking was employed to elucidate the differences in activity and selectivity of the corresponding compounds on SERT, NET, and DAT. In vitro experiments demonstrated that compound A3 has a relatively balanced multi-target activity profile with SERT reuptake inhibition (IC50 = 12 nM), NET reuptake inhibition (IC50 = 78 nM), DAT reuptake inhibition (IC50 = 135 nM), and 5-HT1AR agonism (EC50 = 34 nM). Pharmacokinetic experiments revealed that A3 exhibited excellent bioavailability and low clearance in mice. Subsequent behavioral experiments further confirmed its significant antidepressant effects. These results further highlight the rationality of our design strategy.

The drug has a quick onset of action and a long maintenance of the therapeutic effect, also it is well tolerated. Abstract:INTRODUCTION: Agomelatine is an antidepressant with unique pharmacological actions; it is both a melatonin agonist and selective serotonin antagonist. Both actions combined are necessary for antidepressant efficacy. Effects on melatonin receptors enable resynchronisation of disrupted circadian rhythms with beneficial effects on sleep patterns. Areas covered: The issue of use of an antidepressant for depression co-morbid with somatic disorders is covered by the authors. A review of the literature from 2000 to August 2018 was undertaken using Scopus and Web of Science with the key words: agomelatine, depression, medical illness. Depression in Parkinson's disease, cardiovascular illness and type II diabetes is reviewed with evidence of efficacy. Bipolar depression and seasonal affective disorder may also react favourably. Agomelatine may have specific efficacy on symptoms of anhedonia. Expert opinion: Despite approval in some major jurisdictions, the drug has failed to gain registration in the United States. A defining issue may be questions about longer term efficacy: unequivocal effectiveness in placebo-controlled relapse prevention studies has not always been demonstrated. Continuation studies suggest maintenance of clinical responsiveness. A major disadvantage of the drug is its' potential hepatotoxicity and the need for repeated clinical laboratory tests.

Abstract:GLYX-13, a NMDAR glycine-site partial agonist, was discovered as a promising antidepressant with rapidly acting effects but no ketamine-like side effects. However, the reported synthetic process route had deficiencies of low yield and the use of unfriendly reagents. Here, we report a scaled-up synthesis of GLYX-13 with an overall yield of 30% on the hectogram scale with a column chromatography-free strategy, where the coupling and deprotection reaction conditions were systematically optimized. Meanwhile, the absolute configuration of precursor compound of GLYX-13 was identified by X-ray single crystal diffraction. Finally, the activity of GLYX-13 was verified in the cortical neurons of mice through whole-cell voltage-clamp technique.

Abstract:A series of novel isobenzofuran-1(3H)-one derivatives were designed and synthesized as antidepressants. Firstly, the serotonin reuptake inhibition of these compounds was tested in vitro, and most of them exhibited activity. Particularly, compounds 9d, 10a, and 10c demonstrated superior inhibitory effects and possibly avoided addiction via the μ-opioid receptor and CCK-B receptor. Secondly, the antidepressant effect of compound 10a was evaluated using chronic restraint stress (CRS)-induced mice. The results showed that compound 10a significantly improved CRS-induced depression-like behavior by increasing the neurotransmitters 5-HT in the cortex and THP2 expression in the hippocampus. Thirdly, compound 10a was further investigated and found to enhance CRS-induced hippocampal neuron damage recovery and elevate the expression of synaptic-associated proteins such as BDNF, TrkB, PSD95, and Spinophilin in CRS-induced mice. These findings suggested that novel isobenzofuran-1(3H)-one derivative showed efficacy in treating depression, with compound 10a emerging as a potential lead compound warranting further investigation.

Abstract:An improved, scalable, industrially feasible telescopic process for multigram synthesis of highly commercialized antidepressant drug, moclobemide has been developed. Unlike the conventional approaches, this process enjoys the privilege of being an economical, operationally simple, effectively scalable, green and quantitative method for the synthesis of amide‐containing drug moclobemide, showcasing easy isolation and purification without the aid of column chromatography. An environmentally benign reagent combination of TBN and NHS very effectively converts the acyl source, 4‐chlorobenzaldehyde into its N‐hydroxysuccinimide ester intermediate, which simply affords the drug molecule after nucleophilic substitution with the amine source. The synthetic process has successfully been scaled‐up upto ~ 145 g scale with 75% overall yield and >95% first crop HPLC purity.
