Abstract:OBJECTIVE To study and improve the synthesis of donepezil.METHODS 3, 4-dimethoxy-benzaldehyde was condensed with malomc acid in the presence of pyridine, then hydrogenated with Raney Ni to get3-(3,4-dimethoxy-phenyl)-propanic acid, which was cy-clized with poly-phosphoric acid (PPA) to yield 5,6-dimetrioxy-l-indenone (intermediate 1). N-benzyl piperidone was epoxidated with (CH3)2 SOCH2- to get epoxide, which was rearranged with magnesium bromide-etherate to get N-benzyl-4-formyl-piperidine (intermediate 2) . The two intermediates were condensed with NaOH, followed by selective hydrogenation with 5% Pd-C and hydrochlorination to yield donepezil. RESULTS The structures of intermediates and donepezil were identified by IR,1H-NMR and MS.CONCLUSION This method avoided using expensive reagents such as n-BuLi, IDA, and there was no need to use column chromatograph in all steps.
