Abstract:A new synthetic method of pioglitazone hydrochloride, a kind of drug for treating diabetes, was reported. With the piperidine as catalyst, p-hydroxybenzaldehyde condensed with 2,4-thiazolidinedione to give 5-(4-hydroxybenzylidene)thiazolidine-2,4-dione, which was reduced by hydrogen/Pd-C to afford 5-(4-hydroxybenzyl)thiazolidine-2,4-dione. The dione subsequently reacted with potassium hydroxide and 2-(5-ethyl-2-pyridyl)ethyl sulfonate to produce pioglitazone, and finally with hydrochloride acid to obtain the hydrochloride salt. Some compounds were confirmed by H-1 NMR and IR.

Abstract:2-Amino-3-methyl pyridine was prepared with 2-cyano-methylpyridine as raw material via hydrolysis in dilute basic solution using H_2O_2 as oxidant and Hofmann degradation with NaBrO.The feasible reaction conditions were as follows:hydrolysis,(50°C),5% NaOH,3.5 h,10% of H_2O_2;Hofmann degradation,(60°C),12% NaOH,0.5 h.The overall yield was 74%(mol).The product obtained was identified by GC-MS、IR、()~1H NMR and elemental analysis.

Abstract:An improved synthetic approach to tenatoprazole 1 is described. It started from 2,3,5-trimethyl-4-nitropyridine-N-oxide 2 with acetic anhydride via rearrangement and hydrolysis to give 3, Chlorination with SOCl2 yielded 2-chloromethyl-3,5-dimethyl-4-nitropyridine hydrochloride 4, then 4 condensed with 2-mercapto-5-methoxy imidazole [4,5-b]pyridine 5 to give 5-methoxy-2-[(4-nitro-3,5-dimethyl-2-pyridinyl)methylthio]imidazole[4,5-b]pyridine 6. At last the title compound 1 was produced by two methods: 6 was oxidized with MCPBA and then methoxylated with CH3ONa to give 1 and 6 was first methoxylated with CH3ONa and then oxidized with MCPBA to give 1. The overall yield is around 26% for both five-step syntheses.

Abstract:The synthetic method on(R)-rolipram was described in detail.Using isoacetovanillone as materials,(R)-rolipram was synthesized by six steps of alkylation,Wittig-Horner,bromo reaction,phthalimide,enantioselective conjugate reduction and deprotection followed by cyclization reaction.The product yield was 34%.The structure of the product was determined by 1H NMR and MS.

Abstract:p-trifluoromethylbenzonitrile was prepared from p-(trifluoromethyl)chlorobenzene and potussium cyanide catalyzed by tris(triphenylphosphine)nickle(0), which was prepared from anhydrous NiCl2 and triphenylphosphine in the presence of zinc.And then, the produced intermideate, p-trifluoromethylbenzonitrile was treated with hydrogen sulfide to give p-(trifluoromethyl)thiobenzamide and the yield of 90% with the purity of 99.7% was obtained for the objective product.The structure is correct through IR and GC-MS testing.The operation process is simple and reaction condition is moderate.

Abstract:A new, practical and cost-effective route for scalable synthesis of 2-[(4-dodecyloxyphenyl)sulfonyl] butanoic acid, a key intermediate of a new cyan dye-forming coupler containing a sulfone group, was developed by adopting phenol as the starting material. The synthesis was accomplished in five steps with etherification, chlorosulfonation, reduction, nucleophilic reaction by C-S coupling and hydrolyzation. An important objective of the new synthetic route was the synthesis of 2-[4-(dodecyloxyphenyl)sulfonyl]butanoate. Overall yield obtained at optimized conditions increased to 66 %. The synthetic strategy was proven to be a process enabling rapid delivery of the target product with high purity.

Abstract:N-Methyl-4-aminobenzyl sulfonamide, a key intermediate for synthesis of the pharmaceutical substance Sumatriptan, was prepared throught four steps. 4-Nitrobenzyl chloride was first converted to 4-nitrobenzyl sulphonic acid by reacting with sodium hydrosulfite, then the sulphonic acid obtained was oxidized to 4-nitrobenzyl sulfonyl chloride by chlorine, and N-methyl-4-nitrobenzyl sulfonamide was produced by reacting with methylamine, in the end, the title product was obtained by reducing the nitro group to amino group with hydrazine hydrate in the presence of iron trichoride on active carbon as the catalyst. The yields of the four steps are 90%, 93%, 60% and 77%, respectively. The intermediates and product were characterized by GC-MS and H~1NMR.

Abstract:Aim To establish a new method for the synthesis of D-prolinamide.Method D-proline was obtained from the asymmetric transformafion of L-proline via the formation of(S)-tartaric acid salt by using butanal as catalyst.Then,D-prolinamide was synthesized by amino protection,esterification,amidation and deprotection.Results and conclusion The yield of final product was 78.4% and optical purity was 99.3%.Comparing with patent synthetic procedure for D-prolinamide,it is a new synthetic procedure with better yield and purity and suitable for industrial manufacture.

Abstract:Simvastatin is a new antihyperlipidaemic agent. It can be synthesized with the material of lovastatin, hydroxy protecting agent of trimethylchlorosilane, and t-butyl lithium instead of pyrrole lithium. The total yield is about 78%.The structure is identified by UV,IR,MS,NMR spectrums.This method is very easy and the cost is very low, but the yield is high. Most of organic solvents can be recovered in order to be used next time. So this method will have perfect industrialization foreground.

Abstract:OBJECTIVE:To modify the synthetic method of fluvoxamine. METHODS:Fluvoxamine was synthesized from 4-trifluoromethylbenzonitrile by the steps of Grignard reaction, hydrolysis and oximation. RESULT:The chemical structure of the synthesized product was confirmed by (1)HNMR, and the total yield reached to 36.16%. CONCLUSION:The results indicate that the synthetic route is practical.

Abstract:Several fluorinated derivatives of the anti-HIV maturation agent bevirimat (1) were synthesized and evaluated for anti-HIV replication activity. The modified positions were the C-2, C-3, C-28, and C-30 positions, either directly on the betulinic acid (2) skeleton or in the attached side chains. Compound 18, which has a trifluoromethyl group added to C-30 of its isopropenyl group, exhibited similar potency to 1 against HIV-1NL4-3. In total, our current studies support our prior conclusion that C-30 allylic modification is unlikely to be a pharmacophore for anti-HIV activity, but could be a meaningful route to manipulate other properties of 2-related compounds.

Abstract:The titled compound (1), which is an important intermediate of metoprolol, was synthesized in overall 29% yield from cheap p-nitrotoluene and paraformaldehyde via condensation, alkylation, reduction, diazotisation and hydrolysis. The key step (p-nitrotoluene condensation with paraformaldehyde) was investigated in detail, including effects of catalysts, solvent, and reactants mole ratio on yield. Thus, 2 mol p-nitrotoluenes reacted with 1 mol paraformaldehyde in DMSO, catalyzed by potassium phenol to give p-nitrophenyl ethanol (4) with the conversion of p-nitrotoluene of 31% and selectivity of 72%. The remaining p-nitrotoluene could be recycled.

Abstract:Ezetimibe is a novel selective cholesterol absorption inhibitor which was developed by both Schering-Plough and Merck. An improved process for the preparation of ezetimibe was studied and was described as follows. N-(4-(benzyloxy)benzylidene)-4-fluorobenzenamine (Compound 2) was prepared from 4-hyd-roxybenzaldehyde, 4-fluorobenzenamine and benzyl chloride in one pot. 5-(4-Fluorophenyl)-5-oxopentanoic acid was reacted with pivaloyl chloride and directly condensed with (S)-4-phenyloxazolidin-2-one promoted with LiCl, and then reduced by CBS/BH3 to get (4S)-3-[(5S)-5-(4-fluoror)-5-hydrox-1-oxo-pentyl]-4-phenyl-oxazolidin-2-one (Compound 4). It was condensed with compound 2 catalyzed with Lewis acid after being protected by TMS to afford the condensation intermediate. Finally it was cyclized by TBAF·3H2O and deprotected with Pd/C to get the title compound with a total yield of 56%. Compared with the current technology, the present process shows advantage in high yield, less raw material consumed, and easy production on industrial scale. The specific rotation and melting point of the prepared title compound is the same as that reported in literature,and the structure of the prepared title compound and some intermediates were confirmed by MS and 1H-NMR.

Abstract:OBJECTIVETo establish a new approach to synthesis of diethyl N-[4-[(2,4-diaminopyrido[3,2-d]pyrimidin-6-yl)methylamino]benzoyl]-L-glutamate.METHODSTarget compound (5) was synthesized by the use of (2,4-dioxo-tetrahydropyridopyrimidin-6-yl)methyl acetate (1) as starting material via hydrolysis, chlorination, condensation with diethyl (p-aminobenzoyl)glutamate and aminolysis.RESULTSA new approach to synthesis of diethyl N-[4-[(2,4-diaminopyrido[3,2-d]pyrimidin-6-yl)methylamino]benzoyl]-L-glutamate was established. This synthetic route has hydrolysis reaction, chlorination, diethyl N-(p-aminobenzoyl)-L-glutamate condensation reaction and ammonolysis reaction. The total yield is 36.7%.The structures of those compounds have identified by 1H nuclear magnetic resonance, 13C nuclear magnetic resonance and mass spectrometry. This synthetic route avoid the unstable brominated reaction product and improves the harsh condition of ammonolysis reaction.CONCLUSIONThe new synthetic route has improved the reaction condition and the stability of the intermediate, and increased the extent of the derivative compounds, which has great significance to anti-folic acid of anti-tumor inhibitor synthesis.

Abstract:The synthetic condition of tulobuterol was optimized to gain lower impurity content. Two intermediates were analyzed, and three degradation impurities were isolated using preparative liquid chromatography for the first time and subsequently characterized by various techniques. Possible degradation impurities were deduced by an oxidative mechanism. Two intermediate impurities were detected: α-bromo-2-chloroacetophenone and 1-(2-chlorophenyl)-2-bromoethanol. Three unreported degradation impurities were found and characterized as N-tert-butyl glycine, o-chloro-benzoic acid and chlorobenzene. The single crystal structure of tulobuterol was firstly reported.
