Abstract:Chrysin(1) and tectochrysin(2) were respectively synthesized in four steps from 1,3,5-tribromobenzene in an overall yield of 46. 8% and that of 37. 0% with the key step being the Bu4NBr catalyzed hydrolysis of 1-phenyl-3-(2', 4', 6' -trimethoxyphenyl)-1, 3-propanedione (11) under different conditions.

Abstract:A parallel process for the new synthesis of 2,7-dimethyl-2,4,6-octatriene-1,8-dial was investigated.3-(5,5-Dimethyl-1,3-dioxan-2-yl)-2-butenal was obtained by a process involving aldehyde protection,ester exchange and catalytic oxidation from 4-acetoxy-2-methyl-2-butenal.A new catalytic oxidation process was proposed to get 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-butenal by oxidating 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-buten-1-ol with copper(Ⅰ) chloride、2,2,6,6-tetramethyl-1-piperidinyloxy and oxygen.3-(5,5-Dimethyl-1,3-dioxan-2-yl)-2-buten-1-ol was chlorizated with SOCl_2 in DMF to get 2-(3-chloro-1-methyl-1-propenyl)-5,5-dimethyl-1,3-dioxane.2,7-Dimethyl-2,4,6-octatriene-1,8-dial was acquired in an overall yield of 48% by one-pot synthesis method from 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-butenal,2-(3-chloro-1-methyl-1-propenyl)-5,5-dimethyl-1,3-dioxane and triethylphosphite.

Abstract:Benzylation of rutin followed by hydrolysis afforded 3',4',7-O-tribenzylquercetin (3). Compound 3 was condensed with acetylglycosyl bromides in a two-phase system of chloroform-0.25 mol.L-1 aqueous potassium carbonate using tetrabutyl ammonium bromide as catalyst to yield the protected glycosides 4a similar to 4d. Removal of protecting group led to target products 1a similar to 1d and the structures of such products 1a similar to 1d were confirmed by IR, H-1 NMR, MS spectra and elemental analysis.

Abstract:Utilizing aldol condensation and beta-elimination as the key steps, butenolides I and II in excellent enantiomeric purity have been concisely synthesized for the first time. According to the rotation difference between synthetic samples and natural products, the rotations were corrected by the calculation upon HPLC measurements. In addition, an efficient way to synthesize methyl pentadec-14-enoate was developed.

Abstract:Bioprivileged molecules hold great promise for supplementing petrochemicals in sustainable organic synthesis of a diverse bioactive products library. Secologanin, a biorenewable monoterpenoid glucoside with unique structural elements, is the key precursor for thousands of natural monoterpenoid alkaloids. Inspired by its inherent highly congested functional groups, a secologanin-based diversity-oriented synthesis (DOS) strategy for novel pseudo-natural alkaloids was developed. All the reactive units of secologanin were involved in these operation simplicity protocols under mild reaction conditions, including the one-step enantioselective transformation of exocyclic C8, C8/C11, and C8/C9/C10 as well as the chemoenzymatic manipulation of endocyclic C2/C6 via the attack by various nucleophiles. A combinatory scenario of the aforementioned reactions further provided diverse polycyclic products with multiple chiral centers. Preliminary activity screening of these newly constructed molecules led to the discovery of antimalarial and highly potent neuroprotective skeletons. The application of green biorenewable secologanin in diversity-oriented pseudo-natural monoterpenoid alkaloid synthesis might encourage the pursuit of valuable bioactive frameworks.

Abstract:Cyclobutanes with a gem-dimethyl group are common motifs in natural products. However, strategies for constructing enantioenriched gem-dimethyl cyclobutanes are still underdeveloped. Herein, we report an enantioselective approach to synthesize a broad group of chiral 2,3-disubstituted cyclobutanones through sequential 1,4-conjugate addition/trapping/cross-coupling of readily available cyclobutenones. The intermediate 2-bromocyclobutanone provides a valuable synthetic handle for further coupling transformations. In addition, this strategy was successfully utilized to synthesize gem-dimethyl cyclobutane-containing natural products, including (+)-β-caryophyllene, (-)-raikovenal, (-)-1β,9αH-5-linoleoyloxy-4,5-secocaryophyllen-4-one, and (-)-rumphellanones A-C.

Abstract:2,3-Dihydrobenzofurans are crucial building blocks in the synthesis of natural products and pharmaceutical molecules. However, their asymmetric synthesis has been a long-standing formidable challenge so far. In this work, we developed a highly enantioselective Pd/TY-Phos-catalyzed Heck/Tsuji-Trost reaction of o-bromophenols with various 1,3-dienes, allowing expedient access to chiral substituted 2,3-dihydrobenzofurans. This reaction features excellent regio- and enantiocontrol, high functional group tolerance, and easy scalability. More importantly, the demonstration of this method as a highly valuable tool for the construction of optically pure natural products (R)-tremetone and fomannoxin is highlighted.

Abstract:The first enantioselective total syntheses of prenylflavonoid Diels-Alder natural products (-)-kuwanon I, (+)-kuwanon J, (-)-brosimone A, and (-)-brosimone B have been accomplished from a common intermediate based on a concise synthetic strategy. Key elements of the synthesis include a biosynthesis-inspired asymmetric Diels-Alder cycloaddition mediated by a chiral ligand/boron Lewis acid, as well as a process involving regioselective Schenck ene reaction, reduction, and dehydration to realize a biomimetic dehydrogenation for generation of the required diene precursor. Furthermore, a remarkable tandem inter-/intramolecular asymmetric Diels-Alder cycloaddition process was applied for the synthesis of (-)-brosimone A.

Abstract:2-Methylenecyclobutanones (2-MCBones), used to be difficult to access, but can now be easily achieved by a green and stereospecific Ca(OH)(2)-catalyzed direct and simple aldol condensation of cyclobutanone and aldehydes under mild conditions. The obtained (E)-2-MCBones should be a class of potentially useful building blocks in synthesis as they could readily undergo an interesting (PhSe)(2)-catalyzed Baeyer-Villiger (BV) oxidation with H2O2 at room temperature to give the versatile 4-methylenebutanolides. Mechanistic studies revealed that the BV reaction most possibly proceeded via the initial formation of benzeneseleninoperoxoic anhydride [PhSe(O)O](2)O, which then converted to benzeneseleninoperoxoic acid PhSe(O)OOH as the active oxidant, followed by its selective addition to the C=O bond of 2-MCBones and then a selective C-C bond cleavage and rearrangement to give 4-methylenebutanolides.

Abstract:The first enantioselective total syntheses of (-)-kuwanon X, (+)-kuwanon Y, and (+)-kuwanol A have been accomplished by using asymmetric Diels-Alder cyclo-addition promoted by chiral VANOL or VAPOL/boron Lewis acid. The biosynthesis-inspired asymmetric Diels-Alder cyclo-addition shows high exo selectivity (exo/endo = 13/1), which was unprecedented in the previous total syntheses of related prenylflavonoid Diels-Alder natural products. An acid catalyzed intramolecular ketalization process enabled a biomimetic transformation to construct the polycyclic skeleton of kuwanol A efficiently.

Abstract:Three sets of convenient catalytic systems have been developed for the oxidative dimeric cyclization coupling of differently substituted 2,3-allenoic acids catalyzed by Pd(II), affording bibutenolides that are not otherwise readily available. The advantages and disadvantages of these systems are discussed. Although the diastereoselectivity for the bicyclization of racemic 2,3-allenoic acids is low, excellent diastereoselectivity was realized in the bicyclization reaction of optically active 2,3-allenoic acids, leading to the optically active bibutenolides in high yields and ee. Based on a mechanistic study, it is believed that the reaction may proceed by means of a double oxypalladation and reductive elimination to yield butenolide 3 and Pd(0) species, which may be reoxidized to the catalytically active Pd(II) species in the presence of alkyl iodide/air, metallic iodide/air, or benzoquinone.

Abstract:γ-Butenolide and γ-butyrolactone scaffolds are two types of important core structures in numerous natural products and bioactive targets. However, methods to construct the chiral quaternary arylated γ-butenolide are rarely explored. We herein report an efficient Pd-catalyzed enantioselective γ-arylation of β,γ-unsaturated butenolides with aryl bromides, which shows high γ-selectivity, good functional group tolerance and excellent enantioselectivity. Notably, this protocol also allows for facile construction of tricyclic tetrahydroindolines and tetrahydroisoquinolinones in one step. DFT calculations are consistent with the experimental results, suggesting that the γ-arylation is favoured over the α-arylation. Finally, this method is applied to the rapid synthesis of natural product (R)-(+)-boivinianin A.

Abstract:Mimicking biosynthetic pathways of hongkonoids led to the development of a new Cu(I)-catalyzed [3 + 2] cycloaddition of α-hydroxyketone and β-keto enol ethers, affording tetrahydrofuran acetals in a highly diastereoselective manner and 100% atom economy. Computational studies on the mechanism disclosed a concerted but asynchronous Michael addition/aldol reaction. Of the same importance, this methodology provides a practical biomimetic approach for one-step construction of the dibenzylbutyrolactol lignan backbone starting from two phenyl propane derivatives, opening up a powerful new approach for lignan synthesis, which is showcased by succinct total syntheses of two biologically important aryltetralin-type lignans, β-apopicropodophyllin and cycloolivil. Given the mild and operationally simple conditions, the developed chemistry might have a promising prospect in potential industrial applications.

Abstract:A total synthetic route of two new dihydrostilbenes 5-(2-benzo[1,3]dioxole-5-ylethyl)-6-methoxy benzo [ 1,3]dioxole-4-ol (1) and 5-(2-benzo[1,3]dioxole-5-ylethyl)benzo[1,3]dioxole-4,7-diol (2), which were isolated from Bulbophyllum odoratissimum Lindl. with significant cytotoxicity toward human cancer cell lines, was developed via Horner reaction etc. The natural products 1 and 2 were obtained in 10.5% and 3.3% overall yield, respectively.

Abstract:In this paper, we report the concise total syntheses of three botryane sesquiterpenoids: dehydrobotrydienal, dehydrobotrydienol, and 10-oxodehydrodihydrobotrydial. Abstract:The combination of zirconocene-mediated coupling of silylated alkynes with a protonation-desilylation or bromination-desilylation process afforded otherwise unavailable butadiene derivatives. When (E,E)-2,3-dialkyl-1,4-bis(trimethylsilyl)-1,3-butadienes were treated with 3 equiv of Br-2 in CH2Cl2, (E)-2,3-dialkyl-1,1,4,4-tetrabromo-2-butenes were obtained in excellent yields with perfect stereoselectivity.

Abstract:AbstractHerein we report the first enantioselective total synthesis of 3,5‐dimethylorsellinic acid‐derived meroterpenoids (−)‐berkeleyone A and its five congeners ((−)‐preaustinoids A, A1, B, B1, and B2) in 12–15 steps, starting from commercially available 2,4,6‐trihydroxybenzoic acid hydrate. Based upon the recognition of latent symmetry within D‐ring, our convergent synthesis features two critical reactions: 1) a symmetry‐breaking, diastereoselective dearomative alkylation to assemble the entire carbon core, and 2) a Sc(OTf)3‐mediated sequential Krapcho dealkoxycarbonylation/carbonyl α‐tert‐alkylation to forge the intricate bicyclo[3.3.1]nonane framework. We also conducted our preliminary biomimetic investigations and uncovered a series of rearrangements (α‐ketol, α‐hydroxyl‐β‐diketone, etc.) responsible for the biomimetic diversification of (−)‐berkeleyone A into its five preaustinoid congeners.
