Abstract:This special issue in FEBS Open Bio was conceived to highlight some of the current research and future directions regarding research in the field of environmental toxicology of some organic pollutants, in relation to human health and disease. It has long been established that man has been exposed to many new (un-natural) organic chemicals since the beginning of the Industrial Revolution, many of which are found in a vast and diverse range of products, such as agrochemicals, pharmaceuticals, plastics and electronic components, which have been instrumental in driving man's exponential advances in technology over the last 150 years. However, an unforeseen consequence of these advances is that our ecosystem has been exposed to a vast number of these organic chemicals, many of which appear to be persistent within the environment, as well as bioaccumulating in living organisms, including man.

Abstract:The potential for toxicants to harm organisms in the environment is influenced by the physicochemistry of the substances and their environmental behaviors and transformation within ecosystems. This special issue is composed of 20 papers that report on studies which have investigated the fate and toxicity of various toxicants including engineered nanoparticles, pharmaceuticals and personal care products, antibiotics, pathogens, heavy metals, and agricultural nutrients. The environmental transformations of these substances and how these processes affect their toxicity are emphasized. This paper highlights the important findings and perspectives of the selected papers in this special edition, with an aim of providing insights into full-scale evaluation on the toxicity of various contaminants that exist in ecosystems. General suggestions are provided for the future directions of toxicological research.

Abstract:In recent years, studies have shown that exposure to environmental pollutants (e.g., radiation, heavy metal substances, air pollutants, organic pollutants) is a leading cause of human non-communicable diseases. The key to disease prevention is to clarify the harmful mechanisms and toxic effects of environmental pollutants on the body. Metabolomics is a high-sensitivity, high-throughput omics technology that can obtain detailed metabolite information of an organism. It is a crucial tool for gaining a comprehensive understanding of the pathway network regulation mechanism of the organism. Its application is widespread in many research fields such as environmental exposure assessment, medicine, systems biology, and biomarker discovery. Recent findings show that metabolomics can be used to obtain molecular snapshots of organisms after environmental exposure, to help understand the interaction between environmental exposure and organisms, and to identify potential biomarkers and biological mechanisms. This review focuses on the application of metabolomics to understand the biological effects of radiation, heavy metals, air pollution, and persistent organic pollutants exposure, and examines some potential biomarkers and toxicity mechanisms.

Abstract:There has long been a great concern with growing anthropogenic contaminants and their ecological and toxicological effects on living organisms and the surrounding environment for decades. Metabolomics, a functional readout of cellular activity, can capture organismal responses to various contaminant-related stressors, acquiring direct signatures to illustrate the environmental behaviours of anthropogenic contaminants better. This review entails the application of metabolomics to profile metabolic responses of environmental organisms, e.g. animals (rodents, fish, crustacean and earthworms) and microorganisms (bacteria, yeast and microalgae) to different anthropogenic contaminants, including heavy metals, nanomaterials, pesticides, pharmaceutical and personal products, persistent organic pollutants, and assesses their ecotoxicological impacts with regard to literature published in the recent five years. Contaminant-induced metabolism alteration and up/down-regulation of metabolic pathways are revealed in typical organisms. The obtained insights of variations in global metabolism provide a distinct understanding of how anthropogenic contaminants exert influences on specific metabolic pathways on living organisms. Thus with a novel ecotechnique of environmental metabolomics, risk assessments of anthropogenic contaminants are profoundly demonstrated.

Abstract:Pesticides are substances that are meant to control pests or weeds. The production and use of synthetic pesticides have been increasingly growing in recent years worldwide. Pesticides are designed to be toxic to specific groups of organisms, which bring benefits to humans. However, they have considerable adverse effects on other living creatures as well as environmental media including air, water, and soil. Pesticides can enter the human body via the skin, the mouth, and the lungs, thereby leading to diverse toxicity actions on humans. This chapter provides a brief overview of classification, toxicity, health effects, and environmental exposure of pesticides, as well as toxicity and mechanism of three major pesticides (organophosphorus, pyrethroid, and carbamate pesticides).

Abstract:Artificial chemical products are widely used and ubiquitous worldwide and pose a threat to the environment and human health. Accumulating epidemiological and toxicological evidence has elucidated the contributions of environmental chemical contaminants to the incidence and development of chronic diseases that have a negative impact on quality of life or may be life-threatening. However, the pathways of exposure to these chemicals and their involvements in chronic diseases remain unclear. We comprehensively reviewed the research progress on the exposure risks of humans to environmental contaminants, their body burden as indicated by blood monitoring, and the correlation of blood chemical contaminants with chronic diseases. After entering the human body through various routes of exposure, environmental contaminants are transported to target organs through blood circulation . The application of the modern analytical techniques based on human plasma or serum specimens is promising for determining the body burden of environmental contaminants, including legacy persistent organic pollutants , emerging pollutants, and inorganic elements. Furthermore, their body burden, as indicated by blood monitoring correlates with the incidence and development of metabolic syndromes, cancers, chronic nervous system diseases, cardiovascular diseases, and reproductive disorders. On this basis, we highlight the urgent need for further research on environmental pollution causing health problems in humans.

Abstract:The aquatic environment has long been a sink for diverse chemicals released as a result of human activities. In order to assess the associated risks for the aquatic life, both the chemicals’ occurrences (exposure) as well as their effects in the living organisms (hazard) need to be known. These two aspects are studied by environmental chemistry and toxicology, respectively. Mass spectrometry has been successfully applied in both fields, as it can be used not only for measuring the pollutants in different environmental compartments, but also for gaining novel insights into the molecular mechanisms of toxicity. We here discuss the application of mass spectrometry in environmental chemistry and toxicology, illustrated by several case studies drawn from research carried out at our institute.

Abstract:Toxic substances in the environment generate adverse effects at all levels of biological organization from the molecular level to community and ecosystem. Given this complexity, it is not surprising that ecotoxicologists have struggled to address the full consequences of toxic substance release at ecosystem level, due to the limits of observational and experimental tools to reveal the changes in deep structure at different levels of organization. -Omics technologies, consisting of genomics and ecogenomics, have the power to reveal, in unprecedented detail, the cellular processes of an individual or biodiversity of a community in response to environmental change with high sample/observation throughput. This represents a historic opportunity to transform the way we study toxic substances in ecosystems, through direct linkage of ecological effects with the systems biology of organisms. Three recent examples of -omics advance in the assessment of toxic substances are explored here: (1) the use of functional genomics in the discovery of novel molecular mechanisms of toxicity of chemicals in the environment; (2) the development of laboratory pipelines of dose-dependent, reduced transcriptomics to support high-throughput chemical testing at the biological pathway level; and (3) the use of eDNA metabarcoding approaches for assessing chemical effects on biological communities in mesocosm experiments and through direct observation in field monitoring. -Omics advances in ecotoxicological studies not only generate new knowledge regarding mechanisms of toxicity and environmental effect, improving the relevance and immediacy of laboratory toxicological assessment, but can provide a wholly new paradigm for ecotoxicology by linking ecological models to mechanism-based, systems biology approaches.

Abstract:The harmful impacts of pollution on atmospheric, terrestrial and aquatic environments can cause extreme temperature changes, clean water shortage, and increase toxicity. This book studies environmental pollution impacts, from the mechanism of toxic nanoparticles on the molecular level to the detection of trace gas on the satellite perspective. It presents recent developments in advanced monitoring techniques, efficient process technologies and health impact assessment tools, from environmental scientists of diversified fields. The three sections highlight important aspects of emerging monitoring technologies in Atmospheric Environment, Contaminants Control Process and Environmental Toxicity Assessment. Observational tools presented in the first section range from in-situ analytical to digital techniques for atmospheric monitoring. Highlighted in the second section is the recently developed water quality monitoring system for lake stratification and membrane technologies for detection and removal of contaminants. Lastly, toxicity mechanisms of endocrine disruptors and nanoparticles are highlighted on the third section, with important new discoveries. The major topics addressed are: atmospheric environmental monitoring, including monitoring and modeling of trace air pollutants, contaminants control process monitoring, environmental toxicity monitoring and assessments, biological and chemical monitoring for environmental toxicity, monitoring technology for desalination water quality and toxicity measurement of desalted and concentrated water.

Abstract:This overview discusses the ideas behind a collection of articles by invited authors on a diverse range of complex chemical mixtures and analytical methods which represent some of the most relevant and interesting issues on emerging contaminants of environmental health concerns. These chemicals, including those in high volume production, are released on an ongoing basis as a result of various human activities. Moreover, these emerging contaminants are highly toxic to humans and ubiquitous in the environment. The terms "environmental pollution" and "food web contaminant" became widely known from the first decades of the XX century and as a historical background, they have an early link to the agricultural use of mercury fungicides and DDT. Surprisingly for chemists and society, the 1960s saw the emergence of the global problem of environmental and food contamination with polychlorinated biphenyls (PCBs). The environmental science paradigm has lost none of its importance in the meantime, although it has evolved to encompass a vast number of environmentally hazardous chemicals, many of which are termed as "emerging". To put it simply, air-water-soil/sediment are the first compartments affected and become reservoirs or sinks for environmental pollutants, the next is the food web, topped by the top animal predators and humans. Understanding the occurrence, environmental fate and biotoxicity of emerging contaminants is of interest to many areas of science.

Abstract:The toxicity evaluation system of environmental pollutants has undergone numerous changes due to the application of new technologies. Single-cell toxicogenomics is rapidly changing our view on environmental toxicology by increasing the resolution of our analysis to the level of a single cell. Applications of this technology in environmental toxicology have begun to emerge and are rapidly expanding the portfolio of existing technologies and applications. Here, we first summarized different methods involved in single-cell isolation and amplification in single-cell sequencing process, compared the advantages and disadvantages of different methods, and analyzed their development trends. Then, we reviewed the main advances of single-cell toxicogenomics in environmental toxicology, emphatically analyzed the application prospects of this technology in identifying the target cells of pollutants in early embryos, clarifying the heterogeneous response of cell subtypes to pollutants, and finding pathogenic bacteria in unknown microbes, and highlighted the unique characteristics of this approach with high resolution, high throughput, and high specificity by examples. We also offered a prediction of the further application of this technology and the revolution it brings in environmental toxicology. Overall, these advances will provide practical solutions for controlling or mitigating exogenous toxicological effects that threaten human and ecosystem health, contribute to improving our understanding of the physiological processes affected by pollutants, and lead to the emergence of new methods of pollution control.

Abstract:Considering the chemical complexity and toxicity data gaps of environmental mixtures, most studies evaluate the chemical risk individually. However, humans are usually exposed to a cocktail of chemicals in real life. Mixture health assessment remains to be a research area having significant knowledge gaps. Characterization of chemical composition and bioactivity/toxicity are the two critical aspects of mixture health assessments. This review seeks to introduce the recent progress and tools for the chemical and biological characterization of environmental mixtures. The state-of-the-art techniques include the sampling, extraction, rapid detection methods, and the in vitro, in vivo, and in silico approaches to generate the toxicity data of an environmental mixture. Application of these novel methods, or new approach methodologies (NAMs), has increased the throughput of generating chemical and toxicity data for mixtures and thus refined the mixture health assessment. Combined with computational methods, the chemical and biological information would shed light on identifying the bioactive/toxic components in an environmental mixture.

Abstract:Despite extensive evidence showing that exposure to specific chemicals can lead to disease, current research approaches and regulatory policies fail to address the chemical complexity of our world. To safeguard current and future generations from the increasing number of chemicals polluting our environment, a systematic and agnostic approach is needed. The “exposome” concept strives to capture the diversity and range of exposures to synthetic chemicals, dietary constituents, psychosocial stressors, and physical factors, as well as their corresponding biological responses. Technological advances such as high-resolution mass spectrometry and network science have allowed us to take the first steps toward a comprehensive assessment of the exposome. Given the increased recognition of the dominant role that nongenetic factors play in disease, an effort to characterize the exposome at a scale comparable to that of the human genome is warranted.

Abstract:This study emphasizes the importance of considering the metabolic and toxicity mechanisms of environmental concern chemicals in real-life exposure scenarios. Furthermore, environmental chemicals may require metabolic activation to become toxic, and competition for binding sites on receptors can affect the severity of toxicity. The multicomplex process of chemical toxicity is reflected in the activation of multiple pathways during toxicity of which AhR activation is major. Real-life exposure to a mixture of concern chemicals is common, and the composition of these chemicals determines the severity of toxicity. Nutritional essential elements can mitigate the toxicity of toxic heavy metals, while the types and ratio of composition of PAH can either increase or decrease toxicity. The epigenetic mechanisms of heavy metals and PAH toxicity involves either down-regulation or up-regulation of some non-coding RNAs (ncRNAs) whereas specific small RNAs (sRNAs) may have dual role depending on the tissue and circumstance of expression. Similarly, decrease DNA methylation and histone modification are major players in heavy metals and PAH mediated toxicity and FLT1 hypermethylation is a major process in PAH induced carcinogenesis. Overall, this review provides the understanding of the metabolism of environmental concern chemicals, emphasizing the importance of considering mixed compositions and real-life exposure scenarios in assessing their potential effects on human health and diseases development as well as the dual mechanism of toxicity via genetic or epigenetic axis.
