Abstract:OBJECTIVE:Drug therapy is essential for cancer treatment. The molecular targeted anticancer drugs develop rapidly in recent years, since the effectiveness of traditional chemotherapy is unsatisfactory and the adverse reactions are high. However, molecular targeted anticancer drugs would damage the function of heart, liver or lung, and may cause adverse effects such as hand-foot syndrome, which restrains their clinical application. Therefore, it is critical for pharmaceutical toxicology to study the toxicity, the related mechanisms and the preventive measures of molecular targeted anticancer drugs.

Abstract:Nanotechnology has been a rapidly developing field in the past few decades, resulting in the more and more exposure of nanomaterials to human. The increased applications of nanomaterials for industrial, commercial and life purposes, such as fillers, catalysts, semiconductors, paints, cosmetic additives and drug carriers, have caused both obvious and potential impacts on human health and environment. Nanotoxicology is used to study the safety of nanomaterials and has grown at the historic moment. Molecular toxicology is a new subdiscipline to study the interactions and impacts of materials at the molecular level. To better understand the relationship between the molecular toxicology and nanomaterials, this review summarizes the typical techniques and methods in molecular toxicology which are applied when investigating the toxicology of nanomaterials and include six categories: namely; genetic mutation detection, gene expression analysis, DNA damage detection, chromosomal aberration analysis, proteomics, and metabolomics. Each category involves several experimental techniques and methods.

Abstract:Environmental carcinogens increase cancer incidence via both mutagenic and non-mutagenic mechanisms. There are over 500 known or suspected carcinogens classified by the International Agency for Research on Cancer. Sequencing of both cancerous and histologically non-cancerous tissue has been instrumental in improving our understanding of how environmental carcinogens cause cancer. Understanding how and defining which environmental or lifestyle exposures drive cancer will support cancer prevention. Recent research is revisiting the mechanisms of early tumorigenesis, paving the way for an era of molecular cancer prevention. Significance: Recent data have improved our understanding of how carcinogens cause cancer, which may reveal novel opportunities for molecular cancer prevention.

Abstract:The aim of this review is to comprehensively summarize the recent achievements in the field of toxicogenomics and cancer research regarding genetic-environmental interactions in carcinogenesis and detection of genetic aberrations in cancer genomes by next-generation sequencing technology. Cancer is primarily a genetic disease in which genetic factors and environmental stimuli interact to cause genetic and epigenetic aberrations in human cells. Mutations in the germline act as either high-penetrance alleles that strongly increase the risk of cancer development, or as low-penetrance alleles that mildly change an individual's susceptibility to cancer. Somatic mutations, resulting from either DNA damage induced by exposure to environmental mutagens or from spontaneous errors in DNA replication or repair are involved in the development or progression of the cancer. Induced or spontaneous changes in the epigenome may also drive carcinogenesis. Advances in next-generation sequencing technology provide us opportunities to accurately, economically, and rapidly identify genetic variants, somatic mutations, gene expression profiles, and epigenetic alterations with single-base resolution. Whole genome sequencing, whole exome sequencing, and RNA sequencing of paired cancer and adjacent normal tissue present a comprehensive picture of the cancer genome. These new findings should benefit public health by providing insights in understanding cancer biology, and in improving cancer diagnosis and therapy.

Abstract:The link between inflammation and cancer has been recognized for a long time. Instead of a simple summation of molecular events in single cells, carcinogenesis results from the complex interaction of cancer-stem cells and tumor-permissive microenvironment. The tumor-permissive microenvironment is featured by the suppression of cell-mediated immune response, pro-angiogenesis and inhibition of apoptosis. A number of soluble bioactive factors including various cytokines and growth factors, produced by tumor cells or non-tumor cells, contribute to the formation of such an inflammatory microenvironment by the persistent activation of multiple intracellular signaling pathways such as NF-kappa B and STAT3 pathways.Carcinogenesis is a multi-stage process resulting from the accumulation of many genetic and epigenetic changes. However, it is more than a simple summation of molecular events in single cells. The role of inflammatory microenvironment in carcinogenesis was well recognized, eventually revolutionizing our strategies to cancer treatment and prevention.

Abstract:In 1982 we proposed the concept and a framework for implementing molecular cancer epidemiology. Here, we review progress during the past 17 years in validating and applying this approach to cancer prevention. There have been major advances, notably in the understanding of environment-susceptibility interactions in human cancer. However, a review of major findings to date reveals several urgent research needs to keep pace with the rapid evolution in knowledge of mechanisms in carcinogenesis. Although much valuable progress continues to be made in the study of carcinogens that cause direct DNA damage and are mutagenic, exogenous and endogenous carcinogens can also act by altering gene expression, cell proliferation and differentiation. The mechanisms include aberrant DNA methylation, oxidative damage, effects on metabolism of nitrogen oxide and nitrites, activation of receptors and transcription factors, cyclins and other cell cycle proteins. Sensitive, validated biomarkers are needed to detect these mechanisms in small numbers of cells, tissues or fluids. There is also increasing recognition that individual risk from carcinogen exposure varies as a function of both inherited and acquired factors. Recent advances in genomics, microassay technologies and informatics hold promise for rapid identification of polymorphic variants or changes in expression of genes influencing both response and susceptibility to carcinogens. Another emerging area of molecular epidemiology concerns the role of nutrition and specific dietary factors (including studies on antioxidants, energy metabolism, insulin and various growth factors) and the modulating effect of genetic polymorphisms. Finally, molecular epidemiology has enormous potential in cancer prevention through the early identification of 'at risk' populations and the rapid assessment of intervention efficacy. Its success in fully reaching this potential will depend on the application of validated biomarkers, with adherence to sound epidemiologic and ethical principles.

Abstract:Recent studies have strongly indicated that certain daily-consumed dietary phytochemicals could have cancer protective effects against transgenic mice cancer models and cancers mediated by carcinogens, irradiations and carcinogenic metabolites derived from exogenous or endogenous sources. The cancer-protective effects elicited by these dietary compounds are believed to be due at least in part to the induction of cellular defense systems including the detoxifying and antioxidant enzymes system, as well as the inhibition of anti-inflammatory and anti-cell growth signaling pathways culminating in cell cycle arrest and/or cell-death. In this review, we summarize the potential mechanisms including the modulation of nuclear factor kappaB (NF-κB), cyclooxygenases-2 (COX-2), activator protein-1 (AP-1), mitogen-activated protein kinases (MAPKs) and the induction of phase II cellular detoxifying and antioxidant enzymes mediated mainly by the antioxidant response elements (ARE) within the promoter regions of these genes through nuclear factor-erythroid 2-related factor 2 (Nrf2), a member of the Cap ‘n’ collar (CNC) family of the basic region-leucine zipper transcription factor. In addition, we also review several animal models of carcinogenesis and cancer chemopreventive efficacy studies of these animal models using dietary chemopreventive compounds. Finally, we discuss the cellular signaling cascades mediated by Nrf2, NF-κB, AP-1, MAPKs and COX-2, which have been considered to play pivotal roles in tumor initiation, promotion and progression processes, and could be promising molecular targets for the design of drugs targeting cancer prevention and therapy.

Abstract:Chemotherapy, the most widely accepted treatment for malignant tumors, is dependent on cell death induced by various drugs including antimetabolites, alkylating agents, mitotic spindle inhibitors, antitumor antibiotics, and hormonal anticancer drugs. In addition to causing side effects due to non-selective cytotoxicity, chemotherapeutic drugs can initiate and promote metastasis, which greatly reduces their clinical efficacy. The knowledge of how they induce metastasis is essential for developing strategies that improve the outcomes of chemotherapy. Herein, we summarize the recent findings on chemotherapy-induced metastasis and discuss the underlying mechanisms including tumor-initiating cell expansion, the epithelial-mesenchymal transition, extracellular vesicle involvement, and tumor microenvironment alterations. In addition, the use of combination treatments to overcome chemotherapy-induced metastasis is also elaborated.

Abstract:The complexity of cancer requires a comprehensive approach to understand its diverse manifestations and underlying mechanisms. The Research Topic begins with the perspective article Senga et al. highlighting the need to integrate clear endpoints that anchor KCCs to the acquisition of a complete malignant phenotype into chemical testing. Thus, an allencompassing strategy that incorporates both evolving KCCs and cancer hallmarks, including the role of the microenvironment, is essential to enable the targeted identification of prevalent carcinogens and facilitate zone-specific prevention strategies. To achieve this goal, collaboration between the KCCs and cancer hallmark communities becomes essential.Schroeder et. al. demonstrated that environmental chemicals with established exposure disparities between non-Hispanic Black women and non-Hispanic White women may influence breast phenotypic plasticity, a new hallmark of cancer. This type of plasticity is associated with basal-like breast cancers typically associated with an aggressive triple-negative subtype which affects African American women at rates of 2-3 times that of White women. These data were largely generated with a high-content imaging microscope demonstrating that innovative techniques will bring us closer to understanding cancer disparities at a molecular level. In addition, to better address and understand breast cancer disparities, it is also important to understand risk among different races. Most epidemiological studies have been performed on non-Hispanic White women. Original research by Patil et. al. studied benign breast disease and the subsequent development of breast cancer in African American women.The identification of extracellular vesicles (EVs) in vivo holds potential for the discovery of early biomarkers of carcinogenesis, and/or toxicity endpoints. EVs from donor cells communicate with recipient cells and/or tissues and have the potential to induce toxicity or promote tumorigenesis. However, much of what is currently known about EVs is from studies that perform exogenous administration. A perspective by Nambiar et. al. reviews methodologies that track and alter EVs directly in vivo, as they are released by donor cells. The authors make the argument that advancements in EV engineering with mouse transgenesis and modern sequencing technologies may provide more insight into this largely unknown area of native EV function and biology. A perspective article by Silver et. al. highlights the significance of detecting and characterizing circulating EVs as biomarkers for chemotherapy-induced cardiotoxicity. There are current capabilities to characterize circulating EVs in mice and human liquid cohorts for translational toxicology and cancer medicine.The perspective article by Hariharan et. al. discusses the significance of using human datadriven approaches to improve wellness and reduce tumor recurrence in cancer survivors. Regaining wellness is challenging due to the presence of a myriad of issues induced by radiation, chemotherapy, immunotherapy and/or targeted interventions. This perspective provides promising evidence that global interventions may be possible with data-driven approaches coupled to quality of life and cognitive measurements as well as biological age measurements.Two original articles examined molecular and carcinogenic effects of a chemical exposure. Carswell et al. used targeted templated Oligo-sequencing and DNA methylation profiles on a genome-wide scale to identify DNA methylation alterations with early-in-life dichloroacetic acid (DCA) exposure to determine potential mechanisms of liver tumorigenesis. This question arose from previous paradoxical findings whereby DCA exposure shunted cellular metabolism from aerobic glycolysis, which is termed the Warburg Effect and is associated with cancer, to oxidative phosphorylation. Gonzalez-Pons et al. examined the interactions between a high-fat diet and benzo(a)pyrene on tumorigenesis in an aggressive mouse model of estrogen receptor negative breast cancer.Cohen provides a comprehensive review of the different modes of action of chemical carcinogenesis and how to screen for human carcinogens using the primary endpoints of DNA reactivity and mutagenesis, immunomodulation, increased estrogenic activity, and cytotoxicity with consequent regeneration. Mechanistic understanding of chemical carcinogenicity and hazard identification will be critical for prevention of future cancers.In conclusion, cancer prevention strategies will significantly contribute to reaching the goal of the White House Cancer Moonshot Initiative of decreasing 50% cancer mortality by the year 2047 (6). This can be only achieved by bringing together novel ideas, multidisciplinary efforts, and cutting-edge technologies. Abstract:Research on molecular targeted therapy of tumors is booming, and novel targeted therapy drugs are constantly emerging. Small molecule targeted compounds, novel targeted therapy drugs, can be administered orally as tablets among other methods, and do not draw upon genes, causing no immune response. It is easily structurally modified to make it more applicable to clinical needs, and convenient to promote due to low cost. It refers to a hotspot in the research of tumor molecular targeted therapy. In the present study, we review the current Food and Drug Administration (FDA)-approved use of small molecule targeted compounds in tumors, summarize the clinical drug resistance problems and mechanisms facing the use of small molecule targeted compounds, and predict the future directions of the evolving field.

Abstract:Cancer research has advanced throughout the years with respect to the personalization of the treatments and to targeting cancer-related molecular signatures on different organs. Still, the adverse events of the treatments such as radiotherapy are of high concern as they may increase the mortality rate due to their severity. With the improved efficiency of cancer treatments, patient survival has been increasing. Consequently, the number of patients with adverse effects from radiotherapy is also expected to increase in the forthcoming years. Therefore, approaches for personalized treatments include the elimination of adverse events and decreasing the toxicity in healthy tissues while increasing the efficiency of cancer cytotoxicity. In this context, this paper aims to discuss the recent advances in the field of thorax irradiation therapy and its related toxicities leading to radiation pneumonitis in cancer patients. Molecular mechanisms involved in the radiation-induced lung injury and approaches used to overcome this lung injury are discussed. The discourse covers approaches such as therapeutic administration of natural products, current and prospective radioprotective drugs, and applications of mesenchymal stem cells for radiation-induced lung injury.

Abstract:Toxicogenomics measures molecular features, such as transcripts, proteins, metabolites and epigenomic modifications, to understand and predict the toxicological effects of environmental and pharmaceutical exposures. Transcriptomics has become an integral tool in contemporary toxicology research owing to innovations in gene expression profiling that can provide mechanistic and quantitative information at scale. These data can be used to predict toxicological hazards through the use of transcriptomic biomarkers, network inference analyses, pattern-matching approaches and artificial intelligence. Furthermore, emerging approaches, such as high-throughput dose-response modelling, can leverage toxicogenomic data for human health protection even in the absence of predicting specific hazards. Finally, single-cell transcriptomics and multi-omics provide detailed insights into toxicological mechanisms. Here, we review the progress since the inception of toxicogenomics in applying transcriptomics towards toxicology testing and highlight advances that are transforming risk assessment.

Abstract:As non-traditional nutrients, the biological activity of phytochemicals have been extensively studied for their antioxidant, anti-inflammatory, and apoptosis-promoting effects in various diseases. The general anti-cancer benefits of phytochemicals have been demonstrated in both basic researches and clinical trials. However, researchers understanding of how phytochemicals target cancer-related signaling pathways is still in its infancy. Molecular docking simulation analyses have yielded a large amount of cellular target molecules of phytochemicals. Herein, we review the potential signaling pathways which may be involved in the phytochemical-driven cancer benefits. We expect these findings to help in the design of potential cancer treatments designed through manipulating the binding modes and sites of these plant chemicals.

Abstract:Among all malignant tumors in the whole universe, the incidence and mortality of lung cancer disease rank first. Especially in the past few years, the occurrence of lung cancer in the urban population has continued to increase, which seriously threatens the lives and health of people. Among the many treatments for lung cancer, chemotherapy is the best one, but traditional chemotherapy has low specificity and drug resistance. To address the above issue, this study reviews the five biological pathways that common terpenoid compounds in medicinal plants interfere with the occurrence and development of lung cancer: cell proliferation, cell apoptosis, cell autophagy, cell invasion, metastasis, and immune mechanism regulation. In addition, the mechanism of the terpenoid natural traditional Chinese medicine monomer compound combined with Western medicine in the multipathway antilung cancer is summarized.
