Mass transfer characteristics in disposable rocking bioreactors: A critical review and quantitative data catalogue

Abstract:Disposable rocking bioreactors allow for efficient surface aeration of culture medium via 1D, 2D, or 3D oscillatory movements, as opposed to traditional bioreactors equipped with an internal mechanical stirrer(s) supporting aeration of culture medium supplied by spargers or tube diffusers. The bubble-free surface aeration of the culture medium in rocking bioreactors provides gentle conditions for culture system mixing. Because of that, disposable continuously rocking bioreactors are used for in vitro bioprocessing of fragile and shear-sensitive animal or plant cells, aggregates of such cells, tissues, organs, and biomass-biomaterial hybrid structures, with a sufficient aeration intensity and at desirable values of the k L a coefficient. This review is a critical summary of the methods for determining values of the quantitative parameters characterising mass transfer efficiency in rocking bioreactors, i.e., the k L a coefficient, mixing time, and power input per volume. The thematic overview is supported by an extensive catalogue of quantitative data on those parameters achieved for various disposable rocking systems equipped with polymer film-based containers. Mass transfer efficiency in the rocking bioreactors is characterised by the following values: 0.1–20 h −1 for the k L a coefficient, 5–1000 s for mixing time, and 10–700 W·m −3 for power input per volume.
