Abstract:The past decade of social insect research has seen rapid development in automated behavioral tracking and molecular profiling of the nervous system, two distinct but complementary lines of inquiry into phenotypic variation across individuals, colonies, populations, and species. These experimental strategies have developed largely in parallel, as automated tracking generates a continuous stream of behavioral data, while, in contrast, 'omics-based profiling provides a single 'snapshot' of the brain. Better integration of these approaches applied to studying variation in social behavior will reveal the underlying genetic and neurobiological mechanisms that shape the evolution and diversification of social life. In this review, we discuss relevant advances in both fields and propose new strategies to better elucidate the molecular and behavioral innovations that generate social life.

Abstract:Humans have been trying to understand animal behavior at least since recorded history. Recent rapid development of new technologies has allowed us to make significant progress in understanding the physiological and molecular mechanisms underlying behavior, a key goal of neuroethology. However, there is a tradeoff when studying animal behavior and its underlying biological mechanisms: common behavior protocols in the laboratory are designed to be replicable and controlled, but they often fail to encompass the variability and breadth of natural behavior. This Commentary proposes a framework of 10 key questions that aim to guide researchers in incorporating a rich natural context into their experimental design or in choosing a new animal study system. The 10 questions cover overarching experimental considerations that can provide a template for interspecies comparisons, enable us to develop studies in new model organisms and unlock new experiments in our quest to understand behavior.

Abstract:Animals′ behaviours can vary greatly between even closely-related species. While changes in the sensory periphery have frequently been linked to species-specific behaviours, very little is known about if and how individual cell types in the central brain evolve. Here, we develop a set of advanced genetic tools to compare homologous neurons in – which specialises on a single fruit – and . Through systematic morphological analysis of olfactory projection neurons (PNs), we reveal that global anatomy of these second-order neurons is conserved. However, high-resolution, quantitative comparisons identify a striking case of convergent rewiring of PNs in two distinct olfactory pathways critical for ′s host location. Calcium imaging and labelling of pre-synaptic sites in these evolved PNs demonstrate that novel functional connections with third-order partners are formed in . This work demonstrates that peripheral sensory evolution is accompanied by highly-selective wiring changes in the central brain to facilitate ecological specialisation, and paves the way for systematic comparison of other cell types throughout the nervous system.

Abstract:A founder of the fields of behavioural ecology and neuroethology, Peter Marler’s research uncovered fundamental principles underlying how animals learn to communicate and employ signals in numerous contexts. Studying animal taxa as diverse as birds and primates, he helped illuminate how general principles underlie the communication of all animals. Further, he illustrated the power of comparative research to help us learn more about human behaviour and language. His approach shows us the importance of careful, curiosity-driven research inspired by natural history. Here, I discuss some of Peter Marler’s influential research findings and their impact on modern research in animal behaviour.

Abstract:‘Accounting for the sensory abilities of animals is critical in experimental design.’ No researcher would disagree with this statement, yet it is often the case that we inadvertently fall for anthropocentric biases and use ourselves as the reference point. This paper discusses the risks of adopting an anthropocentric view when working with non-human animals, and the unintended consequences this has on our experimental designs and results. To this aim, we provide general examples of anthropocentric bias from different fields of animal research, with a particular focus on animal cognition and behaviour, and lay out the potential consequences of adopting a human-based perspective. Knowledge of the sensory abilities, both in terms of similarities to humans and peculiarities of the investigated species, is crucial to ensure solid conclusions. A more careful consideration of the diverse sensory systems of animals would improve many scientific fields and enhance animal welfare in the laboratory.

In recent years, the diversity of rodent behaviors has been leveraged to advance our understanding of neural circuits underlying social behaviors, and in particular those related to acoustic communication. Here we highlight recent work investigating vocal behaviors across social contexts in multiple rodent species and discuss how these studies reveal both shared and distinct neural circuits.

Abstract:The sensory drive hypothesis of animal signal evolution describes how animal communication signals and preferences evolve as adaptations to local environments. While classical approaches to testing this hypothesis often focus on preference for one aspect of a signal, deep learning techniques like generative models can create and manipulate stimuli without targeting a specific feature. Here, we used an artificial intelligence technique called neural style transfer to experimentally test preferences for color patterns in a fish. Findings in empirical aesthetics show that humans tend to prefer images with the visual statistics of the environment because the visual system is adapted to process them efficiently, making those images easier to process. Whether this is the case in other species remains to be tested. We therefore manipulated how similar or dissimilar male body patterns were to their habitats using the Neural Style Transfer (NST) algorithm. We predicted that males whose body patterns are more similar to their native habitats will be preferred by conspecifics. Our findings suggest that both males and females are sensitive to habitat congruence in their preferences, but to different extents, requiring additional investigation. Nonetheless, this study demonstrates the potential of artificial intelligence for testing hypotheses about animal communication signals.

On the value of diverse organisms in auditory research: From fish to flies to humans

Abstract:Historically, diverse organisms have contributed to our understanding of auditory function. In recent years, the laboratory mouse has become the prevailing non-human model in auditory research, particularly for biomedical studies. There are many questions in auditory research for which the mouse is the most appropriate (or the only) model system available. But mice cannot provide answers for all auditory problems of basic and applied importance, nor can any single model system provide a synthetic understanding of the diverse solutions that have evolved to facilitate effective detection and use of acoustic information. In this review, spurred by trends in funding and publishing and inspired by parallel observations in other domains of neuroscience, we highlight a few examples of the profound impact and lasting benefits of comparative and basic organismal research in the auditory system. We begin with the serendipitous discovery of hair cell regeneration in non-mammalian vertebrates, a finding that has fueled an ongoing search for pathways to hearing restoration in humans. We then turn to the problem of sound source localization - a fundamental task that most auditory systems have been compelled to solve despite large variation in the magnitudes and kinds of spatial acoustic cues available, begetting varied direction-detecting mechanisms. Finally, we consider the power of work in highly specialized organisms to reveal exceptional solutions to sensory problems - and the diverse returns of deep neuroethological inquiry - via the example of echolocating bats. Throughout, we consider how discoveries made possible by comparative and curiosity-driven organismal research have driven fundamental scientific, biomedical, and technological advances in the auditory field.

Abstract:Many animals show an aversion to bright, open spaces, with significant variability seen across species, populations and individuals within populations. Although there is much interest in the underlying causes of this behaviour, few studies have been able to systematically isolate the role of heritable and environmental effects. Here, we addressed this gap using a common garden experiment with cavefish. Specifically, we bred and cross-bred cave loaches ( Barbatula barbatula ), Europe's only known cavefish, in the laboratory, raised the offspring in complete darkness or normal light conditions, and studied their light avoidance behaviour. Cavefish spent much more time in a light area and ventured further out, while surface fish spent considerable time in risk-assessment behaviour between the light and dark areas. Hybrids behaved most similarly to cavefish. Light treatment and eye quality and lens size only had a modest effect. Our results suggest light avoidance behaviour of cavefish has a heritable basis and is fundamentally linked to increased boldness rather than reduced vision, which is likely adaptive given the complete lack of macropredators in the cave environment. Our study provides novel experimental insights into the behavioural divergence of cavefish and contributes to our broader understanding of the evolution of boldness and behavioural adaptation.

Abstract:Social challenges like territorial intrusions evoke behavioral responses in widely diverging species. Recent work has showed that evolutionary "toolkits"-genes and modules with lineage-specific variations but deep conservation of function-participate in the behavioral response to social challenge. Here, we develop a multispecies computational-experimental approach to characterize such a toolkit at a systems level. Brain transcriptomic responses to social challenge was probed via RNA-seq profiling in three diverged species-honey bees, mice and three-spined stickleback fish-following a common methodology, allowing fair comparisons across species. Data were collected from multiple brain regions and multiple time points after social challenge exposure, achieving anatomical and temporal resolution substantially greater than previous work. We developed statistically rigorous analyses equipped to find homologous functional groups among these species at the levels of individual genes, functional and coexpressed gene modules, and transcription factor subnetworks. We identified six orthogroups involved in response to social challenge, including groups represented by mouse genes Npas4 and Nr4a1, as well as common modulation of systems such as transcriptional regulators, ion channels, G-protein-coupled receptors and synaptic proteins. We also identified conserved coexpression modules enriched for mitochondrial fatty acid metabolism and heat shock that constitute the shared neurogenomic response. Our analysis suggests a toolkit wherein nuclear receptors, interacting with chaperones, induce transcriptional changes in mitochondrial activity, neural cytoarchitecture and synaptic transmission after social challenge. It shows systems-level mechanisms that have been repeatedly co-opted during evolution of analogous behaviors, thus advancing the genetic toolkit concept beyond individual genes.

Abstract:Organismal behavior, with its tremendous complexity and diversity, is generated by numerous physiological systems acting in coordination. Understanding how these systems evolve to support differences in behavior within and among species is a longstanding goal in biology that has captured the imagination of researchers who work on a multitude of taxa, including humans. Of particular importance are the physiological determinants of behavioral evolution, which are sometimes overlooked because we lack a robust conceptual framework to study mechanisms underlying adaptation and diversification of behavior. Here, we discuss a framework for such an analysis that applies a "systems view" to our understanding of behavioral control. This approach involves linking separate models that consider behavior and physiology as their own networks into a singular vertically integrated behavioral control system. In doing so, hormones commonly stand out as the links, or edges, among nodes within this system. To ground our discussion, we focus on studies of manakins (Pipridae), a family of Neotropical birds. These species have numerous physiological and endocrine specializations that support their elaborate reproductive displays. As a result, manakins provide a useful example to help imagine and visualize the way systems concepts can inform our appreciation of behavioral evolution. In particular, manakins help clarify how connectedness among physiological systems-which is maintained through endocrine signaling-potentiate and/or constrain the evolution of complex behavior to yield behavioral differences across taxa. Ultimately, we hope this review will continue to stimulate thought, discussion, and the emergence of research focused on integrated phenotypes in behavioral ecology and endocrinology.

Abstract:When animals, including humans, communicate, they convey information and express their perceptions of the world. Because different organisms are able to produce and perceive different signals, the animal world contains a diversity of communication systems. Based on the approach laid out in the 1950s by Nobel laureate Nikolaas Tinbergen, this book looks at animal communication from the four perspectives of mechanisms, ontogeny, function, and phylogeny. The book's great strength is its broad comparative perspective, which enables the reader to appreciate the diversity of solutions to particular problems of signal design and perception. For example, although the neural circuitry underlying the production of acoustic signals is different in frogs, songbirds, bats, and humans, each involves a set of dedicated pathways designed to solve particular problems of communicative efficiency. Lessons learned in animal acoustic cognition through comparisons with humans

Abstract:Humans are an interesting subject of study in comparative cognition. While humans have a lot of anecdotal and subjective knowledge about their own minds and behaviors, researchers tend not to study humans the way they study other species. Instead, comparisons between humans and other animals tend to be based on either assumptions about human behavior and cognition, or very different testing methods. Here we emphasize the importance of using insider knowledge about humans to form interesting research questions about animal cognition while simultaneously stepping back and treating humans like just another species as if one were an alien researcher. This perspective is extremely helpful to identify what aspects of cognitive processes may be interesting and relevant across the animal kingdom. Here we outline some examples of how this objective human-centric approach has helped us to move forward knowledge in several areas of animal acoustic cognition (rhythm, harmonicity, and vocal units). We describe how this approach works, what kind of benefits we obtain, and how it can be applied to other areas of animal cognition. While an objective human-centric approach is not useful when studying traits that do not occur in humans (e.g., magnetic spatial navigation), it can be extremely helpful when studying traits that are relevant to humans (e.g., communication). Overall, we hope to entice more people working in animal cognition to use a similar approach to maximize the benefits of being part of the animal kingdom while maintaining a detached and scientific perspective on the human species.

Abstract:Animal behaviour is an inherently fascinating topic. For many of us, watching animals is what sparked our initial curiosity and eventually led us to become zoologists. Pondering why one animal behaves one way, while another animal behaves differently – is a simple thing to wonder. But at the same time, answering that question is incredibly difficult. Animal behaviour is among the most complex and challenging of the sciences because it is a broad amalgamation of so many different sub-disciplines. Behaviour spans from the responses of individual cells (e.g. detecting light or sensing temperature) to the function of entire ecosystems, and indeed, via animal movement, it connects ecosystems. Consequently, animal behaviour is central to a wide variety of fields including evolution, population and community ecology, disease ecology, genetics, and environmental science, and it plays a critical role in conservation and biosecurity.
