Abstract:Herein, a direct, metal-free, and site-selective electrochemical C-H carboxylation of arenes by reductive activation using CO2 as the economic and abundant carboxylic source was reported. The electrocarboxylation was carried out in an operationally simple manner with high chemo- and regioselectivity, setting the stage for the challenging site-selective C-H carboxylation of unactivated (hetero)arenes. The robust nature of the electrochemical strategy was reflected by a broad scope of substrates with excellent atom economy and unique selectivity. Notably, the direct and selective C-H carboxylation of various challenging arenes worked well in this approach, including electron-deficient naphthalenes, pyridines, simple phenyl derivatives, and substituted quinolines. The method benefits from being externally catalyst-free, metal-free and base-free, which makes it extremely attractive for potential applications.

Abstract:Carbonylation reactions have been widely used to construct carbonyl-containing molecules or carbon enhancement reactions, which are mostly catalyzed by noble metals (Pd, Rh, Ru, Ir). In this review, we introduce the copper-catalyzed carbonylation reactions that have been developed in our group. Abstract:In this work, we have achieved the arylation and carbonylation of activated alkenes without transition metal catalysis. This protocol not only exhibits broad substrate scope and functional group tolerance, but also achieves high chemo‐ and regio‐selectivity. Comprehensive Summary The simultaneous construction of two vicinal C—C bonds in a molecule remains a significant challenge. In this work, we disclose an electroreductive carboarylation of activated alkenes under mild, transition metal‐free conditions. Utilizing readily available starting materials (electron‐deficient aryl bromides, activated alkenes, and CO2), this method demonstrates broad substrate scope and good functional group tolerance. Notably, this strategy enables the addition of two distinct electrophiles across an alkene in a highly chemo‐ and regioselective manner.

Abstract:A visible-light-catalyzed arylcarboxylation of allenes with CO 2 was developed using [Ir(ppy) 2 (dtbbpy)]PF 6 (ppy = 2-phenylpyridine; dtbbpy = 4,4′-di- tert -butyl-2,2′-bipyridine) as a photocatalyst to synthesis β-aryl β,γ-unsaturated carboxylic acids. This multicomponent protocol proceeds in an atom-economical way with exclusive regioselectivity. Preliminary mechanistic experiments suggested that allylic carbanion species are the key intermediates. Abstract:Abstract Electrochemical catalytic reductive cross couplings are powerful and sustainable methods to construct C−C bonds by using electron as the clean reductant. However, activated substrates are used in most cases. Herein, we report a general and practical electro-reductive Ni-catalytic system, realizing the electrocatalytic carboxylation of unactivated aryl chlorides and alkyl bromides with CO 2 . A variety of unactivated aryl bromides, iodides and sulfonates can also undergo such a reaction smoothly. Notably, we also realize the catalytic electrochemical carboxylation of aryl (pseudo)halides with CO 2 avoiding the use of sacrificial electrodes. Moreover, this sustainable and economic strategy with electron as the clean reductant features mild conditions, inexpensive catalyst, safe and cheap electrodes, good functional group tolerance and broad substrate scope. Mechanistic investigations indicate that the reaction might proceed via oxidative addition of aryl halides to Ni(0) complex, the reduction of aryl-Ni(II) adduct to the Ni(I) species and following carboxylation with CO 2 .

Abstract:Herein, we report an electrochemical protocol for the dicarboxylation of aryl alkynes using CO(2). With a graphite rod as the cathode and Al as the sacrificial anode, a series of valuable butenedioic acids are obtained in moderate to excellent yields with an E/Z ratio up to 50:1. This method features high E-selectivity, high step and atom economy, easy scalability, and a nice substrate scope, which renders it appealing for promising applications in organic synthesis and materials chemistry.

Abstract:The mechanisms and chemo- and regioselectivities of Ru(II)-catalyzed decarboxylative C-H alkenylation of aryl carboxylic acids with alkynes were investigated with density functional theory (DFT) calculations. The catalytic cycle involves sequential carboxylate-directed C-H activation, alkyne insertion, decarboxylation and protonation. The facile tether-assisted decarboxylation step directs the intermediate toward the desired decarboxylative alkenylation, instead of typical annulation and double alkenylation pathways. The decarboxylation barrier is very sensitive to the tether length, and only the seven-membered ring intermediate can selectively undergo the designed decarboxylation, suggesting a tether-dependent chemoselectivity. This tether-dependent chemoselectivity also applies to the alkyl tethers. In addition, the polarity of solvent is found to control the chemoselectivity between the decarboxylative alkenylation and [4 + 2] annulation. Solvent with low polarity (toluene) favors the decarboxylation pathway, leading to the decarboxylative alkenylation. Solvent with high polarity (methanol) favors the ionic stepwise C-O reductive elimination pathway, leading to the [4 + 2] annulation. To understand the origins of regioselectivity with asymmetric alkynes, the distortion/interaction analysis was applied to the alkyne insertion transition states, and led to a predictive frontier molecular orbital model. The asymmetric alkynes selectively use the terminal with the larger HOMO orbital coefficient to form the C-C bond in the insertion step.

Abstract:Precise synthesis of carboxylic acids via catalytic carboxylation with CO2 is highly appealing. Although considerable advancements have been achieved in difunctionalizing carboxylation of unsaturated hydrocarbons, the asymmetric variants are conspicuously underdeveloped, particularly in addressing axially chiral alkenes. Herein, we report the first catalytic atroposelective carboxylation of alkynes with CO2. A variety of valuable axially chiral carboxylic acids are obtained with good yields and high chemo-, regio-, E/Z and enantio-selectivities. Notably, an unexpected anti-selective carbo-carboxylation is observed in the sp2-hybrid carbo-electrophile-initiated reductive carboxylation of alkynes. Mechanistic studies including DFT calculation elucidate the origin of chiral induction and anti-selectivity in vinyl-carboxylation of alkynes.

A range of alkynes were transformed to the corresponding alpha, beta-unsaturated carboxylic acids in high yields with high regioselectivity. Abstract:Pyridine motifs are widespread pharmacophores in many drugs. Installing various substituents through pyridine C−H bond functionalization is significant for new drug design and discovery. Developments of late-stage functionalization reactions enrich the strategies for selective functionalization of pyridines. However, late-stage C−H carboxylation of pyridines is a long-standing challenge, especially selectively carboxylation with CO2 on pyridine motifs. Herein, we describe a practical method for C4−H carboxylation of pyridines via one-pot C−H phosphination and copper-catalyzed carboxylation of the resulted phosphonium salts with CO2. The reaction is conducted under mild conditions and compatible with multiple active groups and several pyridine drugs, providing diverse valuable isonicotinic acid compounds, demonstrating the application potential of this strategy.

Abstract:A steric effect-controlled, zinc-mediated carboxylation of different 2-alkynyl bromides under an atmospheric pressure of CO2 has been developed by careful tuning of different reaction parameters, including the metal, solvent, temperature, and additive. 2-Substituted 2,3-allenoic acids were afforded from primary 2-alkynyl bromides, whereas the carboxylation of secondary 2-alkynyl bromides yielded 3-alkynoic acids in decent yields. A rationale for the observed regioselectivity has been proposed.
