Abstract:The enantioselective functionalization of ubiquitous C( sp 3 )–H bonds is ideally suited for the construction of three-dimensional chiral structures. However, organic molecules often contain multiple C( sp 3 )–H bonds with a similar energy and steric environment, rendering the simultaneous control of site-, chemo- and stereoselectivity extremely challenging. Here we show the merger of molecular photoelectrochemistry with asymmetric catalysis for the highly site- and enantioselective cyanation of benzylic C( sp 3 )–H bonds. This example of photoelectrochemical asymmetric catalysis requires no chemical oxidant and exhibits an exceptional level of site selectivity and functional group tolerance, enabling not only the efficient conversion of feedstock chemicals but also the late-stage functionalization of complex bioactive molecules and natural products, including ones with multiple benzylic sites.

Abstract:The asymmetric cross-coupling reaction is developed as a straightforward strategy toward 1,1-diaryl alkanes, which are a key skeleton in a series of natural products and bioactive molecules in recent years. Here we report an enantioselective benzylic C(sp(3))-H bond arylation via photoredox/nickel dual catalysis. Sterically hindered chiral biimidazoline ligands are designed for this asymmetric cross-coupling reaction. Readily available alkyl benzenes and aryl bromides with various functional groups tolerance can be easily and directly transferred to useful chiral 1,1-diaryl alkanes including pharmaceutical intermediates and bioactive molecules. This reaction proceeds smoothly under mild conditions without the use of external redox reagents.

Abstract:Abstract The selective functionalization of inert CH bonds is regarded as one of the most attractive research field due to the advantages of expanding diversity, reducing waste and synthetic economy in modifying organic compounds. However, because of the high bond energy, similar bond strengths and steric environments, the CH bond functionalization meets the challenge of controlling the regio‐ and stereoselectivity. Due to the efficient electron transfer and energy transfer, the photocatalysis could provide an accessible alternative for the regioselective functionalization of inert CH bonds. In recent years, the enantioselective functionalization of inert CH bonds by photochemistry has been developed. This article summarized this field as photocatalysis with three main strategies: transition‐metal catalysis, chiral Lewis acid catalysis, and chiral organocatalysis.

Abstract:We report a new thiourea-Bronsted acid cooperative catalytic system for the enantioselective cyanosilylation of aldehydes with yields up to 90% and enamioselectivities up to 88%. The addition of an achiral acid was found to be crucial for high asymmetric induction. Mechanistic investigations using a combination of NMR, ESI-MS, and density functional theory computations (including solvent corrections) at the M06/6-31G(d,p) level of theory suggest that the key catalytic species results from the cooperative interaction of bifunctional thioureas and an achiral acid that form well-defined chiral hydrogen-bonding environments.

Abstract:The manipulation of unactivated aliphatic C–H bonds remains one of the most challenging tasks in synthetic chemistry. Direct hydrogen atom transfer (HAT) photocatalysis is an appealing approach to this goal. However, many methods are constrained due to low catalytic efficiency. Here we report the use of a Brønsted acid to enhance the efficiency of an inexpensive organic HAT photocatalyst, eosin Y. This strategy enables valuable transformations, including alkylation, heteroarylation and fluorination, of a wide array of unactivated C( sp 3 )–H bonds, using the alkane substrate as the limiting reagent. The process has been applied to the late-stage functionalization of natural products and pharmaceuticals to selectively form C–H-functionalized analogues. Experimental and computational mechanistic studies show that the HAT reactivity is significantly enhanced when the sp 3 oxygen atoms on eosin Y are protonated. The method has been shown to be general across different types of direct HAT photocatalysts, demonstrating its potential in native C–H bond functionalization.

Abstract:Radical-mediated Hydrogen Atom ion of Csp3-H bonds has become a powerful tool for the asymmetric functionalization of organic feedstocks. Here, we present an asymmetric synthesis of α-aryl amides via carbamoylation of alkylarenes with isocyanates as electrophiles. The synergistic combination of a photoredox and a chiral nickel-catalyst, enables the use of readily available and neutral reagents under mild reaction conditions and provides straightforward access to pharmacologically relevant motifs in enantiomerically pure form.

Abstract:Abstract Methods that enable site selective acylation of sp 3 C-H bonds in complex organic molecules are not well explored, particularly if compared with analogous transformations of aromatic and vinylic sp 2 C-H bonds. We report herein a direct acylation of benzylic C-H bonds by merging N-heterocyclic carbene (NHC) and photoredox catalysis. The method allows the preparation of a diverse range of benzylic ketones with good functional group tolerance under mild conditions. The reaction can be used to install acyl groups on highly functionalized natural product derived compounds and the C-H functionalization works with excellent site selectivity. The combination of NHC and photoredox catalysis offers options in preparing benzyl aryl ketones.

Abstract:In contrast to the rapid growth of electrophotocatalysis in recent years, enantioselective catalytic reactions powered by this unique methodology remain rare. In this work, we report an electrophotochemical metal-catalyzed protocol for direct asymmetric decarboxylative cyanation of aliphatic carboxylic acids. The synergistic merging of electrophotochemical cerium catalysis and asymmetric electrochemical copper catalysis permits mild reaction conditions for the formation and utilization of the key carbon centered radicals by combining the power of light and electrical energy. Electrophotochemical cerium catalysis enables radical decarboxylation to produce alkyl radicals, which could be effectively intercepted by asymmetric electrochemical copper catalysis for the construction of C-CN bonds in a highly stereoselective fashion. This environmentally benign method smoothly converts a diverse array of arylacetic acids into the corresponding alkyl nitriles in good yields and enantioselectivities without using chemical oxidants or pre-functionalization of the acid substrates and can be readily scaled up.

Abstract:Enantioselective catalytic processes are promoted by chiral catalysts that can execute a specific mode of catalytic reactivity, channeling the chemical reaction through a certain mechanistic pathway. Here, we show how by simply using visible light we can divert the established ionic reactivity of a chiral allyl–iridium(iii) complex to switch on completely new catalytic functions, enabling mechanistically unrelated radical-based enantioselective pathways. Photoexcitation provides the chiral organometallic intermediate with the ability to activate substrates via an electron-transfer manifold. This redox event unlocks an otherwise inaccessible cross-coupling mechanism, since the resulting iridium(ii) centre can intercept the generated radicals and undergo a reductive elimination to forge a stereogenic centre with high stereoselectivity. This photochemical strategy enables difficult-to-realize enantioselective alkyl–alkyl cross-coupling reactions between allylic alcohols and readily available radical precursors, which are not achievable under thermal activation.

Abstract:Regioselective C-H bond functionalization is pivotal in modern scientific exploration, offering solutions for achieving novel synthetic methodologies and pharmaceutical development. In this aspect, achieving alternative regioselective functionalization, like para-selective products in electron-poor aromatics, diverges from traditional methods. Leveraging the advantages of atomically dispersed photocatalysts, we designed a robust photocatalyst for an unconventional regioselective aromatic C–H bond functionalization. This innovation enabled para-selective trifluoromethylations of electron-deficient meta-directing aromatics (-NO2, -CF3, -CN, etc.), which is entirely orthogonal to the traditional approaches. Mechanistic experiments and DFT analysis confirmed the interaction between Cu-metal and the aromatic substrate, alongside the photocatalyst's molecular arrangement, driving selective exposure of the para-selective functionalization. This strategic approach elucidated pathways for precise molecular transformations, advancing the frontier of regioselective C–H bond functionalization by using atomically dispersed photocatalysts in organic synthesis.

Abstract:The Paternò–Büchi reaction is the [2+2] photocycloaddition of a carbonyl with an alkene to afford oxetane products. Enantioselective catalysis of this classical photoreaction, however, has proven to be a long-standing challenge. Many of the best-developed strategies for asymmetric photochemistry are not suitable to address this problem because the interaction of carbonyls with Brønsted or Lewis acidic catalysts can alter the electronic structure of their excited state and divert their reactivity towards alternate photoproducts. We show herein that an alternative triplet rebound strategy enables the stereocontrolled reaction of an excited-state carbonyl compound in its native, unbound state. These studies have resulted in the development of the first highly enantioselective catalytic Paternò–Büchi reaction, catalyzed by a novel hydrogen-bonding chiral Ir photocatalyst.

Abstract:The asymmetric functionalization of C–H is one of the most attractive strategies in asymmetric synthesis. In the past decades, catalytic enantioselective C(sp3)–H functionalization has been intensively studied and successfully applied in various asymmetric bond formations, whereas asymmetric C(sp3)–H alkylation was not well developed. Photoredox catalysis has recently emerged as an efficient way to synthesize organic compounds under mild conditions. Despite many photoinduced stereoselective reactions that have been achieved, the related enantioselective C(sp3)–C(sp3) coupling is challenging, especially of the photocatalytic asymmetric C(sp3)–H radical alkylation. Here, we report a visible light induced Cu catalyzed asymmetric sp3 C–H alkylation, which is effective for coupling with unbiased primary, secondary, and tertiary alkyl fragments in high enantioselectivities.
