Abstract:Confocal laser scanning microscopy (CLSM) as an innovative technique has been widely used in biological research and it offers unique opportunities to visualize distribution and transportation of proteins in various media. For protein chromatographic purification studies, CLSM can further provide direct and detailed information about protein adsorption behaviors. This review highlighted the application of CLSM in investigating protein adsorption processes including single-component and multi-component studies in finite bath adsorption and real-time observation of protein adsorption in the micro-column with various adsorbents. Factors such as liquid phase conditions and the limitation of CLSM application in protein adsorption were discussed. The adsorption profiles in different adsorbents, such as the widely observed shrinking core profile, the overshoot adsorption profile as well as some special profiles were compared and discussed. Abstract:Recent advances in microemulsion electrokinetic chromatography (MEEKC) are reviewed. After brief introduction to the principles of MEEKC, various influence factors including the compositions and types of microemulsion system, pH of the buffer and the addition of organic modifier on MEEKC separation are discussed in details and the applications in different areas are summarized. Abstract:This work reviews the literature of chiral capillary electrokinetic chromatography from January 2016 to March 2021. This is done to explore the state-of-the-art approach and recent developments carried out in this field. The separation principle of the technique is described and supported with simple graphical illustrations, showing migration under normal and reversed polarity modes of the separation voltage. The most relevant applications of the technique for enantioseparation of drugs and other enantiomeric molecules in different fields using chiral selectors in single, dual, or multiple systems are highlighted. Measures to improve the detection sensitivity of chiral capillary electrokinetic chromatography with UV detector are discussed, and the alternative aspects are explored, besides special emphases to hyphenation compatibility to mass spectrometry. Partial filling and counter migration techniques are described. Indirect identification of the separated enantiomers and the determination of enantiomeric migration order are mentioned. The application of Quality by Design principles to facilitate method development, optimization, and validation is presented. The elucidation and explanation of chiral recognition in molecular bases are discussed with special focus on the role of molecular modeling.

Abstract:The ease of varying the conditions of electrophoretic separation and the possibility of using a wide variety of chiral selectors make the capillary electrophoresis method a promising alternative to high-performance liquid chromatography for the separation of racemic drugs. This review summarizes the main directions of development of electrophoretic methods for enantiomeric analysis of biologically active compounds, including the use of various modifiers of the quartz capillary walls and background electrolyte (cyclodextrins, nanoparticles, ionic liquids, etc.), formation of multilayer coatings, and combining of enantiomeric analysis with online derivatization and preconcentration of analytes.

Abstract:The migration mechanism of ionizable compounds in capillary electrochromatography (CEC) is more complicated than in high perfomance liquid chromatography (HPLC) due to the involvement of electrophoresis and the second chemical equilibrium. The separation mcchanism of ionizable compounds in CEC has been studied theoretically. The electrochromatographic capacity factors of ions ( k’) in CEC and in the pressurized CEC are derived by phenomenologicul approach. The influence of pH, voltage, pressure on k’ is discussed. In addition, the k’ of weak acid and weak base are derivcd base on acid-base equilibrium and the influence of pH on k’ is studied theoretically.

Abstract:Microalgae are potential sources for the sustainable production of valuable chemicals including polyphenols, pigments, and ω-3 PUFAs. However, successful exploitation of these high value compounds in the food, healthcare and pharmaceutical sectors depends greatly on their effective separation, identification, and analysis after recovery from the biomass. The findings of this review paper illustrated that chromatographic methods coupled to different types of detectors have been used as a crucial part of research on microalgal polyphenols, Omega-3 Polyunsaturated Fatty Acids (ω-3 PUFAs), and pigments production through identification, measurement, sample preparation, and purification practices. Therefore, it is important to provide a comprehensive review regarding the current research in the field. The basic operating principles, parametric optimisation and detection units of common (liquid chromatography and gas chromatography) and novel chromatographic techniques (counter current chromatography, expanded bed adsorption chromatography and supercritical fluid chromatography) used to separate, identify, and quantify polyphenols, PUFAs and pigments from microalgae matrices are comprehensively reviewed.

Capillary electrochromatography with monolithic silica column: I. Preparation of silica monoliths having surface-bound octadecyl moieties and their chromatographic characterization and applications to the separation of neutral and charged species

Abstract:Monolithic silica columns with surface-bound octadecyl (C18) moieties have been prepared by a sol-gel process in 100 microm ID fused-silica capillaries for reversed-phase capillary electrochromatography of neutral and charged species. The reaction conditions for the preparation of the C18-silica monoliths were optimized for maximum surface coverage with octadecyl moieties in order to maximize retention and selectivity toward neutral and charged solutes with a sufficiently strong electroosmotic flow (> 2 mm/s) to yield rapid analysis time. Furthermore, the effect of the pore-tailoring process on the silica monoliths was performed over a wide range of treatment time with 0.010 M ammonium hydroxide solution in order to determine the optimum time and conditions that yield mesopores of narrow pore size distribution that result in high separation efficiency. Under optimum column fabrication conditions and optimum mobile phase composition and flow velocity, the average separation efficiency reached 160 000 plates/m, a value comparable to that obtained on columns packed with 3 microm C18-silica particles with the advantages of high permeability and virtually no bubble formation. The optimized monolithic C18-silica columns were evaluated for their retention properties toward neutral and charged analytes over a wide range of mobile phase compositions. A series of dimensionless retention parameters were evaluated and correlated to solute polarity and electromigration property. A dimensionless mobility modulus was introduced to describe charged solute migration and interaction behavior with the monolithic C18-silica in a counterflow regime during capillary electrochromatography (CEC )separations. The mobility moduli correlated well with the solute hydrophobic character and its charge-to-mass ratio.

Abstract:CE has been a continuously evolving analytical methodology since its first introduction in the 1980s of the last century. The development of new CE separation procedures, the coupling of these systems to more sensitive and versatile detection systems, and the advances in miniaturization technology have allowed the application of CE to the resolution of new and complex analytical problems, overcoming the traditional disadvantages associated with this method. In the present work, different recent trends in CE and their application to the determination of high complexity samples (as biological fluids, individual cells, etc.) will be reviewed: capillary modification by different types of coatings, microfluidic CE, and online microextraction CE. The main advantages and disadvantages of the different proposed approaches will be discussed with examples of most recent applications.

Abstract:Biomolecules, specifically proteins, polysaccharides, and secondary metabolites are potential lead compounds due to their remarkable pharmacological properties. However, the complex molecular structure of the biomolecules makes their separation processes of great challenges. The conventional downstream processes require multistep protocols that are less efficient, high solvent consumption, expensive, time-consuming, and laborious. Hence, aqueous two-phase system (ATPS) is a reliable technique for the extraction and purification of biomolecules from a complex mixture. ATPS is an environmentally friendly, simple, cost effective, and easily scalable process. It requires a short processing time to separate biomolecules of industrial values simultaneously in a single process. Modifications have also been performed by introducing deep eutectic solvents, ionic liquids, carbohydrates, amino acids or copolymers to enhance the process efficiency with an increased yield, purity and bioactivity of recovered biomolecules. This review attempts to review the recent developed ATPSs and their efficiency to extract, isolate, and purify biomolecules such as proteins, polysaccharides, secondary metabolites and other biological substances. The review provides insights into the feasibility and reliability of ATPS for biomolecule recovery.

Abstract:In the last decade, the instrumentation improvements in supercritical fluid chromatography (SFC) and the hyphenation to mass spectrometry (MS), have increased the SFC acceptance between scientists, becoming today a valuable tool in analytical chemistry. The unique selectivity, short analysis times, low consumption of organic solvents, and the greener mobile phase, have contributed to expanding its applicability which has led to an increase in the number of publications especially in the bioanalysis area. This work reviews the advantages and main applications of SFC in bioanalysis during the last 5 years. Fundamental aspects concerning mobile phase composition, stationary phase, hyphenation to MS as well as matrix effect have been discussed. Finally, the most relevant applications have been summarized.

Abstract:This review surveys the recent progress in the adsorbed stationary phases for capillary electrochromatography (CEC). Adsorption-based methods for preparation of stationary phase are novel approaches in CEC, which allow rapid and facile preparing stationary phases with desirable selectivity onto an open-tubular fused-silica capillary, a bare-silica or ion-exchange packed column or a monolithic silica or polymer column. A variety of adsorbing agents have been developed as adsorbed stationary phases, including ionic long-chain surfactant, protein, peptide, amino acid, charged cyclodextrin (CD), basic compound, aliphatic ionene, and ion-exchange latex particle. The adsorbed stationary phases have been applied to separation of neutral, basic and acidic organic compounds, inorganic anions and enantiomers. They have also been applied to on-line sample concentration, fast separation and study of the competitive binding of enantiomers with protein.

Abstract:Capillary electrophoresis (CE) has rapidly gained great interests among researchers in many different fields. One of these areas is the separation of small ions such as inorganic cations[1-9], anions[10-16], and low Mr organic molecules[17-20]. However, as the separation of ions by CE is depended on the difference in electrophoretic mobilities of the ions, and this difference for some of them are very small. Thus, a background electrolyte (BGE) system must be selected to modify the mobilities of the analyte ions, e.g., the actual mobilities of some alkali, alkaline earth metal and ammonium cations are very similar[21].

Abstract:This review article expands on the previous one (Jmeian, Y. and El Rassi, Z. Electrophoresis 2009, 30, 249-261) by reviewing pertinent literature in the period extending from early 2008 to the present. Similar to the previous review article, the present one is concerned with proteomic sample preparation (e.g. depletion of high-abundance proteins, reduction of the protein dynamic concentration range, enrichment of a particular subproteome), and the subsequent chromatographic and/or electrophoretic prefractionation prior to peptide separation and identification by LC-MS/MS. This review article differs from the first version published in Electrophoresis 2009, 30, 249-261 by expanding on capturing/enriching subglycoproteomics by lectin affinity chromatography. Ninety-eight articles published in the period extending from early 2008 to the present have been reviewed. By no means is this review article exhaustive: its aim is to give a concise report on the latest developments in the field.
