Abstract:The separation mechanisms in capillary electromatography (CEC) of bare silica - packed column was investigated under aqueous - organic mobile phases. The separations of five typical solutes, including apolar compound (benzene), polar compounds (formamide and dimethyl formamide) and basic compounds (pyridine and quinoline), showed that reverse phase retention, normal phase retention, ion-exchange and electrophoresis mechanisms were involved. The effect of the concentration of organic modifier, the concentration of buffer and the pH value on separation were investigated. Due to the hydrophilicity of bare silica, the silica-packed column exhibits the following merit, i. e., the mobile phase composition can change in rather large range of organic modifier content, buffer concentration and pH value. This means that the bubble problem that are usually met in ODS-packed column can be minimized with the silica-packed column. Additionally, we presented a novel model of CEC, termed as dynamically modified silica column CEC. Five alkyl substitued benzenes were baseline separated with a mobile phase containing cetyltrimethylammonium bromide.
Preparation and characterization of inorganic silica-organic polymer mixture based C16 capillary monolithic column

Abstract:A novel inorganic silica-organic polymer mixed-based capillary electrochromatography monolithic column was prepared by using precursor 3-(trimethoxysilyl) propyl methacrylate with double-functional group hydrolysis/polycondensation in acidic aqueous solution, followed by double-bond co-polymerization with organic monomer hexadecyl mathacrylate under the thermal initiated condition, and the combination of preparation of silica-based monolithic column and organic polymer monolithic column was suggested. A simple operation of controlling sol mixture using mercury infusion when the mixture infused into capillary was raised to acquire smooth interface of stationary phase. The preparation conditions of monolithic column were optimized. The structure of the monolithic columns was characterized by SEM images and nitrogen absorption method, and the results showed that the median pore diameter of the materials was in the range of 3 to 7 nm with average surface area of 103.4 M2 g−1. The influence of buffer concentration, pH value, and organic modifier content on electroosmotic flow (EOF) velocity has been investigated. Seven neutral aromatic hydrocarbons were baseline separated with column efficiency of naphthalene up to 54000 plates m−1. The reproducibility for separation time and retention factor was given in terms of RSD values at 0.9% and 0.6% (n = 5) for run-to-run injections and 1.2% and 0.8% (n = 3) for day-to-day operations. The RSD values of separation time and capacity factor of different columns (n = 3) were 8.7 and 5.7%, respectively. The studies on performance of columns indicated that the columns possess typical reverse retention mechanism.

Abstract:The feasibility of capillary electrochromatography (CEC) was shown by demonstrating on 75 μm (i. d. )capillary columns packed with 3 μm silica packings using buffered aqueous acetonitrile mobile phase. The retention mechanisms of this seperation system were discussed. The results indicated that the retention on silica column was determined by both cation ion exchange and normal phase retention mechanisms. The factors affected on electroosmotic flow in a silica packed columns were also investigated.
Capillary Electrochromatography with A Silica Column with A Dynamically Modified Cationic Surfactant

Abstract:A novel mode of capillary electrochromatography (CEC), called dynamically modified silica-capillary electrochromatography, is described in this paper. The column packed with bare silica was dynamically modified with long chain quaternary ammonium salt, cetyltrimethylammonium bromide (CTAB), which was added into the mobile phase. CTAB ions were adsorbed onto the surface of bare silica, and the resulted hydrophobic layer on the silica gel was used as the stationary phase. Using the dynamically modified silica column, neutral solutes were separated by CEC. The highest number of theoretical plates obtained was about 71,500/m and the relative standard deviations for t0 and capacity factor of toluene were 4.7% and 4.9% for 20 consecutive runs, respectively. The separation mechanism of neutral solutes and the influence of mobile phase composition on the separation was investigated. The separation of nitrogen-containing solutes was carried out with this mode and the peak tailing of basic solute was effectively eliminated because the adsorption of basic solute on silica was blocked by the preferred adsorption of CTAB.
Electrochromatographic Performance of Graphene and Graphene Oxide Modified Silica Particles Packed Capillary Columns

Abstract:Graphene oxide functionalized silica microspheres (GO@SiO2) were synthesized based on condensation reaction between amino from aminosilica particles and carboxyl groups from GO. Reduction of GO@SiO2 with hydrazinium hydroxide generated graphene modified silica particles (G@SiO2). GO@SiO2 and G@SiO2 packed capillary columns for capillary electrochromatography were thereafter fabricated by pressure slurry packing with single‐particle frits. GO of 0.3 mg/mL in dispersion solution for GO@SiO2 synthesis was considered as a compromise between retaining and column efficiency whereas GO@SiO2 of 20 mg/mL in slurries for column packing was chosen for a homogenous and tight bed. Optimum mobile phases were acquired considering both electroosmotic flow and resolution at an applied voltage of −6 kV as the following: acetonitrile/phosphate buffer (10 mM, pH 7.0), 75:25 (v/v) for polycyclic aromatic hydrocarbons and 50:50 (v/v) for aromatic compounds. A comparison was made between electrochromatographic performances for three PAHs (naphthalene, fluorene and phenanthrene) and three aromatic compounds of various polarities (toluene, aniline and phenol) on bare aminosilica, GO@SiO2 and G@SiO2 packed columns, which proved the contribution of alone or combinational actions of solvophobic effect and π‐π electron stacking as well as hydrogen bonds to retaining behaviors by GO@SiO2 and G@SiO2. Well over‐run, over‐day and over‐column precisions (retention time: 0.3–1.4, 1.1–3.8 and 2.8–5.2%, respectively; peak area: 2.6–6.5, 4.8–8.3 and 6.5–12.6%, respectively) of GO@SiO2 packed columns were a powerful proof for good reproducibility. Analytical characteristics of GO@SiO2 packed capillary columns in CEC analysis of fresh water were evaluated with respect to linearity (R2 = 0.9961–0.9989) over the range 0.1 to 100 mg/L and detection limits of 9.5 for naphthalene, 12.6 for fluorene and 16.2 μg/L for phenanthrene. Further application to fresh water increased the visibility of the proposed material, where good spike recoveries in the range 89–96% were offered.

Abstract:An approach of immobilizing mobile crystalline material (MCM)-41 mesoporous silica nanoparticles on the inner wall of an open-tubular (OT) capillary as the support for coating chiral selector of cellulose tris(3,5-dimethylphenyl-carbamate) (CDMPC) was carried out. By taking advantage of the improved phase ratio of OT capillary with the immobilization of MCM-41 mesoporous material, the cellulose derivative of CDMPC as the chiral selector was simply coated on the MCM-41 nanoparticle layer via the hydrogen-bonding interaction, and the enantioseparation was successively carried out. Eight pairs of acidic, neutral and basic enantiomers were resolved in capillary electrochromatography or capillary liquid chromatography mode. The concentration of CDMPC for coating was systematically investigated to obtain the optimized chromatographic properties on enantioseparation by controlling the supposed film thickness of CDMPC on MCM-41 nanoparticle layer. Comparing with a bare fused silica capillary column coated with CDMPC under the same coating procedure as MCM-41-modified capillary did, the MCM-41-modified capillary column offered much higher enantioselectivity. This result indicated the significance of using the mesoporous nanoparticles as the electrochromatographic support to enhance the phase ratio of OT capillary column in capillary electrochromatography and capillary liquid chromatography. For investigating the effect of experimental conditions on the enantioseparation with this prepared OT capillary column, the content of organic modifier acetonitrile in the mobile phase was thus extensively evaluated to achieve a better chiral separation.

Abstract:A chiral capillary monolithic column for capillary electrochromatography (CEC) was prepared by covalent bonding of cellulose tris(3,5-dimethylphenylcarbamate) (CDMPC) on the silica monolithic matrix within the confine of a 50-μm i.d. bare fused silica capillary. Several pairs of enantiomers including neutral and basic analytes were baseline resolved on the newly prepared chiral capillary monolithic column in CEC with aqueous mobile phases. Fast enantioseparation was achieved due to the favorable dynamic properties of silica monolith. The covalent bonding of CDMPC as the chiral stationary phase for CEC also enabled the use of THF in mobile phase for enantioseparation of prazquantel by overcoming the incompatibility of THF and the physically coated CDMPC on a column.

Abstract:A polymer-based neutral monolithic capillary column was prepared by radical polymerization of glycidyl methacrylate and ethylene dimethacrylate in a 100 lam id fused-silica capillary, and the prepared monolithic column was subsequently modified based on a ring opening reaction of epoxide groups with 1 M lysine in solution (pH 8.0) at 75 degrees C for 10 h to produce a lysine chemically bonded stationary phases in capillary column. The ring opening reaction conditions were optimized so that the column could generate substantial EOF Due to the zwitterionic functional groups of the lysine covalently bonded on the polymer monolithic rod, the prepared column can generate cathodic and anodic EOF by varying the pH values of running buffer during CEC separation. EOF reached the maximum of -2.0 x 10(-8) m(2)v(-1)s(-1) and 2.6 x 10(-8) m(2)v(-1)s(-1) with pH of the running buffer of 2.25 and 10, respectively. As a consequence, neutral compounds, ionic solutes such as phenols, aromatic acids, anilines, and basic pharmaceuticals were all successfully separated on the column by CEC. Hydrophobic interaction is responsible for separation of neutral analytes. In addition, the electrostatic and hydrophobic interaction and the electrophoretic migration play a significant role in separation of the ionic or ionizable analytes.
Optimization of the single-step synthesis of hybrid C(8) silica monoliths dedicated to nano-liquid chromatography and capillary electrochromatography

Abstract:Hybrid silica monoliths functionalized with octyl groups and dedicated to chromatographic separations in the reversed-phase mode were directly synthesized within capillaries according to the protocol described by Yan et al. [L.J. Yan, Q.H. Zhang, Y.Q. Feng, W.B. Zhang, T. Li, L.H. Zhang, Y.K. Zhang, J. Chromatogr. A 1121 (2006) 92]. Although these monoliths allowed reaching high efficiencies in capillary electrochromatography (CEC), serious limitations prohibited their application in nano-liquid chromatography (nano-LC). Such limitations observed as poor performances in the nano-LC mode and the lack of reproducibility of the synthesis were related to the longitudinal morphological inhomogeneities of the hybrid material along the capillary. Thus, several modifications were conducted in the synthesis protocol in order to improve the resulting morphology of the monolith making it suitable for nano-LC separations. The influence of several critical parameters (such as the addition temperature of the basic catalyst and the hydrolysis duration) on the textural and chromatographic properties had been extensively studied. It was found that a decrease (i.e. 0 degrees C) of the temperature addition of the basic catalyst associated with a shorter hydrolysis duration (1h instead of 6h) allowed (i) delaying the gelation time and consequently facilitating the capillary filling step, (ii) increasing the structural homogeneity of the hybrid monoliths, i.e. their chromatographic performances in nano-liquid chromatography also (iii) greatly improving the reproducibility of the synthesis within the capillary without impairing the material's carbon load, i.e. the incorporation of the less hydrolysable C(8) precursor. The resulting hybrid monoliths afforded retention factors comparable to that previously obtained for C(18) grafted silica monoliths and efficiencies that are the best ever recorded in nano-LC with hybrid monoliths and that are close to the ones achieved with grafted silica monoliths. In fact, this modified protocol allowed a significant improvement of the performances in nano-LC which could be observed by the decrease of the mean value of H(min) going from 123 microm (Yan's protocol) to 24 microm (modified protocol) for a same length of capillary (l = 8.5 cm). In addition, the reproducibility of the synthesis was greatly improved through a factor six of reduction on the calculated standard deviation of these efficiencies. The high permeability and longitudinal homogeneity of the synthesized monolith allowed increasing the capillary length (for example, a 75-cm capillary was conveniently filled with hybrid silica monolith) and the column could be eluted at a very low backpressure leading to chromatographic performances up to 40,000 plates. Finally, the good efficiencies in the nano-LC mode combined with the excellent performances already present in the CEC mode led to fast (less than 1 min) and high efficient separations in the pressurized capillary electrochromatography (p-CEC) mode.

Abstract:A novel type of inorganic silica gel-organic polymer mixture based capillary monolithic column was prepared using methacryloxypropyltrimethoxysilane hydrolyzing and co-condensation polymerization in the presence of an acid solution, and the product reacted with hexadecyl methacrylcetyl ester by double bond polymerization in the presence of a heat initiator. The method proposed combined of the preparation of silica-based monolithic column with organic polymer-based monolithic column. A technique introducing sol mixture into capillary by suction mercury was carried out to obtain a flat and orderly interface of stationary phase. The influences of running voltage, organic additive, concentration and pH of buffer on electroosmotic flow(EOF) were investigated respectively. The structure and pore size distribution of the monolithic material were characterized by scanning electronmicroscopy and nitrogen adsorption method. The pore size distribution profile of mesopores of the material was around 3-7 nm with average specific surface area of material was 103.4 M2/g. Chromatographic characteristics of the monolithic column prepared with the optimized fabrication condition were studied. Seven neutral hydrocarbon, such as alkylbenzenes and polycyclic aromatic hydrocarbons (PAHs), were baselinely separated using the mixture based monolithic column. For naphthalene, the separation efficiency of 54000 theoretical plates/m was achieved, the RSD values of retention times and capacity factor were 0.9% and 0.6% for five consecutive runs, 1.2% and 0.8% for day-to-day(n=3), 8.7% and 5.7% for column-to-column(n=3), respectively.
Surfactant-Assisted Ethane-Bridged Silica Coating for Capillary Electrochromatography

Abstract:With surfactant P123 as structure directing reagent, 1, 2-bis (trimethoxysilyl) ethane was hydrolyzed under acid condition. The resulting ethane-bridged silica was coated onto the inner walls of fused silica capillaries and used as the stationary phase for capillary electrochromatography. The bridged ethyl silica provided hydrophobic groups for reversed-phase separation. A comparative coated capillary was fabricated without the use of surfactant in the preparation of the bonded silica. Separation of model neutral compounds was compared between these two kinds of capillaries. Surfactant-assisted organosilica-coated capillaries displayed much superior retention efficiency without obviously decreased electroosmotic flow. The existence of surfactant in the synthesis of the sol results in higher surface areas of the coating. Such ethane-bridged organosilica stationary phases can be used under basic conditions.

Abstract:Octadecyl silyl silica (ODS) phase coated with immobilized polysiloxanes (OV1701, SE-54, SE-30) were synthesized, their characteristics as capillary electrochromatography (CEC) column packing materials were studied. It was found that, although the polysiloxane coatings were different in polarity, the resulting packing materials showed the highest efficiencies when the respective coating ratios (polysiloxane:ODS, w/w) were all 20-30%. As expected, packing materials coated with different polysiloxanes resulted in different selectivity on solute pairs. Separations on these stationary phases were studied with different factors such as pH values and acetonitrile contents of the mobile phases. It was found that all these kind of stationary phases could resist basic mobile phase with a pH value as high as 11.6. Tests were made to analyze polar, basic drugs with CEC using the stationary phases.

Abstract:A hydrophilic chiral capillary monolithic column for enantiomer separation in CEC was prepared by coating cellulose tris(3,5-dimethylphenyl-carbamate) (CDMPC) on porous hydrophilic poly(acrylamide-co-N,N'-methylene-bisacrylamide) (poly(AA-co-MBA)) monolithic matrix with confine of a fused-silica capillary. The coating conditions were optimized to obtain a stable and reproducible chiral stationary phase for CEC. The effect of organic modifier of ACN in aqueous mobile phase for the enantiomer separation by CEC was investigated, and the significant influence of ACN on the enantioresolution and electrochromatographic retention was observed. Twelve pairs of enantiomers including acidic, neutral, and basic analytes were tested and nine pairs of them were baseline-enantioresolved with acidic and basic aqueous mobile phases. A good within-column repeatability in retention time (RSD = 2.4%) and resolution (RSD = 3.2%) was obtained by consecutive injections of a neutral compound, benzoin, on a prepared chiral monolithic column, while the between-column repeatability in retention time (RSD = 6.4%) and resolution (RSD = 9.6%) was observed by column-to-column examination. The prepared monolithic stationary phase showed good stability in either acidic or basic mobile phase.
Prepared Polymethacrylate-Based Monoliths for the Separation of Cations by Non-Suppressed Capillary Ion Chromatography.

Abstract:This paper describes a novel analytical system for non-suppressed capillary ion chromatography. Methacrylate monolithic columns were prepared from silanized fused-silica capillaries of 320 mu m i.d. by in situ polymerization of glycidyl methacrylate and ethylene dimethacrylate in the presence of 1,4-butanediol, 1-propanol and water as the porogen solvents. The introduction of cation-exchange sites was achieved by sulfonating the matrix with sodium sulfite to produce total cation-exchange capacities in the range of 45-105 mu equiv/mL for a 25 cm column. The conditions (concentrations of sodium sulfite solution, reacting time and modified flow rate) of sulfonation were optimized. The hydrodynamic and chromatographic performances were estimated. Coupled with a conductivity detector, a capillary ion chromatography system was set up with the prepared column. Finally, the resultant column was used for the separations of five common univalent cations (Li+, Na+, NH4+, K+ and Cs+) using methanesulfonic acid as the eluent and four divalent cations (Mg2+, Ca2+, Sr2+ and Ba2+) by non-suppressed capillary ion chromatography; the chromatographic parameters were further researched.

Abstract:Monolithic capillary columns based on polyacrylamides with UV initiated technology were made,and the length and boundaries of the column bed were made precisely through adjustment of UV radiation.β-cyclodextrin(β-CD) was bonded to the porous polymers of columns,which served as chiral selector.FITC labeled chiral amino acids,which had important impacts on metabonomics in vivo,were separated and detected by laser induced fluorescence(LIF) detection using liquid chromatography mode under an alkaline condition.Theoretical number of the column was about 125 000 plates/m.The influence of the mobile phase on the chromatographic performances was analyzed.

Abstract:Columns for open tubular capillary electrochromatography, coated with a mixed-mode (RP/ion-exchange) stationary phase, were prepared by using the sol-gel method. The synthetic procedure was optimized by changing the ratios of tetraethoxysilane, octyltriethoxysilane, and 3-aminopropyltriethoxysilane in the initial sol. SEM studies reveal that a coating with about 400 nm thickness can be obtained. The inner surface properties of these capillaries were probed by measuring the EOF as a function of pH. The surface of this stationary phase contains octyl, amine, and residual silanol moieties; the amine and silanol groups determine the net charge on the inner surface of the capillary and can produce a switchable EOF (anodal/cathodal). The performances of the columns were evaluated by open tubular capillary electrochromatography using a wide range of compounds (polycyclic aromatic hydrocarbons, aromatic acids, and aromatic amines).

Abstract:This review surveys the recent progress in the adsorbed stationary phases for capillary electrochromatography (CEC). Adsorption-based methods for preparation of stationary phase are novel approaches in CEC, which allow rapid and facile preparing stationary phases with desirable selectivity onto an open-tubular fused-silica capillary, a bare-silica or ion-exchange packed column or a monolithic silica or polymer column. A variety of adsorbing agents have been developed as adsorbed stationary phases, including ionic long-chain surfactant, protein, peptide, amino acid, charged cyclodextrin (CD), basic compound, aliphatic ionene, and ion-exchange latex particle. The adsorbed stationary phases have been applied to separation of neutral, basic and acidic organic compounds, inorganic anions and enantiomers. They have also been applied to on-line sample concentration, fast separation and study of the competitive binding of enantiomers with protein.

Abstract:Enantiomeric separations in CEC with the macrocyclic antibiotic vancomycin immobilized silica monolith as a chiral stationary phase are presented. The monolithic silica capillary columns were prepared by a sol-gel process in fused-silica capillaries with an inner diameter of 50 mu m and subsequently in situ immobilization of vancomycin as a chiral selector by reductive amination. Enantioselectivity was obtained for eight pairs of enantiomers in nonaqueous polar organic or aqueous mobile phases and most of them were baseline-separated with high column efficiencies. it was observed that the organic modifier ratio (MeOH/ACN) in the polar organic mobile phase played a significant role in controlling the resolution and efficiency of the enantiomers. In enantiomeric separation of propranolol, repeatability for column efficiency and resolution in the nonaqueous mobile phase was given in terms of RSD values at 1.1 and 2.3% (n = 5) for run-to-run injections and 7.2 and 9.6% (n = 5) for column-to-column testing while repeatability for the separation of thalidomide in the aqueous mobile phase was given in terms of RSD values at 1.5, 2.8% and 6.1, 10.5%, respectively.

Abstract:The physical adsorption method proposed previously has been successfully applied to a monolithic silica column. By virtue of the physical adsorption, a chiral stationary phase of avidin was prepared onto the silica monolith. The phase ratio of resulting stationary phase was evaluated with frontal analysis. The method proved to be comparable in phase ratio to the chemical bonding methods used in high- performance liquid chromatography (HPLC). Enantiomer separations were carried out in capillary electrochromatography (CEC) and capillary liquid chromatography (CLC) modes. Due to its larger phase ratio, the resulting column showed more powerful separation capability as compared to open-tubular CEC (OTCEC). Twelve chiral compounds were baseline-resolved. The resulting column showed high separation efficiency, with average theoretical plate numbers of 66 000/m for CLC and 122 000/m for CEC. Good reproducibility was observed, with RSD value less than 1.3% for retention time, retention factor and separation factor, and less than 6.6% for plate counts and resolution (n = 40). Fast separations were achieved with a short column. The test enantiomers were baseline-resolved within 4 min under CLC and CEC modes. In addition, field-enhanced sample injection (FESI) was coupled to CLC as well as CEC to improve the detection sensitivity.

Abstract:Polymer-based cation exchange capillary monolithic column was prepared for capillary ion chromatography (CIC) by radical polymerization in a 320-mu m-i. d. fused-silica capillary tube, taking glycidyl methacrylate (GMA) as the functional monomer, ethylene dimethacrylate (EDMA) as the cross-linking agent and in the presence of 1,4-butanediol, 1-propanol and water as the porogen solvents and azobisisobutyronitrile (AIBN) as a suitable initiator. Introduction of cation exchange sites was achieved by reacting with Na2SO3. Coupled with the prepared cation exchange capillary monolithic column, a capillary ion chromatography system was constructed with a pump (in mu L/min level), a capillary detection cell and a UV detector, on which more chromatographic characteristics were further studied. With the CIC system, 9 common univalent and divalent cations (Li+, Na+, NH4+, K+, Cs+, Mg2+, Ca2+, Sr2+, Ba2+) were successfully separated and determined by flow rate gradient. Another interesting separation of cations and melamine was also tested on the prepared column.
