Abstract:Recent advances in microemulsion electrokinetic chromatography (MEEKC) are reviewed. After brief introduction to the principles of MEEKC, various influence factors including the compositions and types of microemulsion system, pH of the buffer and the addition of organic modifier on MEEKC separation are discussed in details and the applications in different areas are summarized. Abstract:Microemulsion electrokinetic chromatography (MEEKC) and solvent modified micellar electrokinetic chromatography (MEKC) were investigated with the goal of the rapid separation of complex heroin and amphetamine samples. The rapid simultaneous separation of 17 species of heroin, amphetamine and their basic impurities and adulterants was performed within about 10min using MEEKC for the first time, whereas solvent modified MEKCs were unable to resolve all the components. The comparisons between MEEKC and solvent modified MEKC proved internal lipophilic organic phase in microemulsions played an important role in improving the separation performance with respect to efficiency. However, the role of internal lipophilic organic phase in MEEKC was disgusted at high concentrations of cosurfactant, and the separations of MEEKC and 1-butanol modified MEKC became similar at high concentrations of 1-butanol. The evaluation of reproducibility, linearity and detection limit of optimized MEEKC method provided good results for all the analytes investigated, thus allowing its application to real controlled drug preparation analysis.

Abstract:A new mode of the scanning electrochemical microscope (SECM) operation was developed that combines reagent delivery from the nanopipette with electron transfer at the conductive substrate and ion transfer across the liquid/liquid interface supported at the nanopipette tip. This approach offers potential advantages for measurements of heterogeneous electron transfer kinetics and reaction rate imaging by enabling straightforward separation of the contributions of surface topography and reactivity features to the tip current. It addresses some other long-standing technical issues, including sensitive probing of low signal sources (e. g., immobilized enzymes or catalyst particles) without diffusional broadening and the elimination of the elevated background signal in generation/collection-type experiments. The high spatial resolution attainable in electron transfer/ion transfer mode experiments and the absence of redox mediator species in the bulk solution are advantageous for studies of biological cells.

Abstract:Micellar electrokinetic chromatography (MEKC) and microemulsion electrokinetic chromatography (MEEKC) have been investigated for the separation of nine xanthones from Securidaca inappendiculata, based on the systematic optimization of such parameters as pH, concentration of borate buffer, and sodium dodecyl sulfate (SDS) concentration. The study indicated that, in MEKC, although the addition of modifiers including organic solvents and cyclodextrins (CDs) could greatly affect the separation selectivity of xanthones, the baseline separation for nine xanthones could not be completed. As a special variant of electrokinetic chromatography (EKC), MEEKC successfully achieved the baseline separation of nine xanthones with satisfactory repeatability. Compared with MEKC, MEEKC showed better separation selectivity and higher separation efficiency.

Abstract:A simple and sensitive micellar electrokinetic capillary chromatography (MEKC) method was developed for the separation and determination of six flavonoids in Epimedium brevicornum Maxim. Field-enhanced sample injection with reverse migrating micelles (FESI-RMM) was used for on-line concentration of the flavonoids. An electrolyte containing 20 mM H3PO4, 100 mM SDS, 20% acetonitrile and 2% 2-propanol (pH 2.0) was chosen as the electrophoretic buffer. By optimizing the stacking conditions, about 40-360-fold improvement in the detection sensitivity was obtained for the flavonoids.

Abstract:A comparison between chiral cyclodextrin-modified microemulsion electrokinetic chromatography (CD-MEEKC) and cyclodextrin-modified micellar electrokinetic chromatography (CD-MEKC) for the enantiomeric separation of esbiothrin was carried out. For both methods, the separation conditions were optimized by varying CD types and concentration, running buffer pH and compositions, organic modifiers, and temperature. The optimal CD-MEEKC conditions were 0.8% n-heptane, 2.3% SDS, 6.6% n-butanol, 90.3% 10 mM sodium tetraborate containing 3% (w/v, the ratio of CD mass to microemulsion volume) methyl-beta-cyclodextrin, pH 10, 25 degrees C. The optimized CD-MEKC conditions were 3.3% SDS, 96.7% 10 mM sodium tetraborate containing 5% (w/v) beta-CD, pH 10, 25 degrees C. The difference in physicochemical properties of the buffer and CDs resulted in different optimal CD type. The competitive distribution between the microemulsion (or micelle) and chiral CD contributed to the chiral separation. Both methods provided excellent separation (R-s approximate to 3) with similar migration time (ca. 15 min). CD-MEEKC provided higher separation efficiencies (> 300000) than CD-MEKC (> 200000). The LODs for CD-MEEKC and CD-MEKC were 4.7 mu g/ml, and 3.2 mu g/mL, respectively. The RSDs of migration time and peak area for CD-MEEKC were slightly higher than for CD-MEKC. Both the demonstrated CD-MEEKC and CD-MEKC methods provided high efficiencies, low LODs, and reproducible enantioseparations of esbiothrin.

Abstract:The separation of enantiomers is of great importance in biology, pharmaceutics, agriculture and environment. The different separation modes (i.e. capillary zone electrophoresis (CZE), micellar electrokinetic chromatography (MEKC), capillary electrochromatography (CEC), etc) and many chiral selectors available make capillary electrophoresis (CE) technique a powerful tool for chiral analysis[1]. On the basis of ligand-exchange (LE) mechanism introduced by Davankov and Rogozhin[2] in the early 1970s for high performance liquid chromatography (HPLC), the first application of CE in chiral separation was reported by Zare's group[3,4]. Using Cu(Ⅱ) complexes of L-histidine or aspartame as chiral selectors, 14 dansyl amino acids (Dns-AAs) were resolved. The authors observed a significant improvement in resolution when a micelle forming surfactant such as sodium dodecyl sulfate (SDS) was added to the electrolyte performing MEKC. The method was called as ligand-exchange micellar electrokinetic capillary chromatography (LE-MEKC). As a hybrid mode of possessing both the advantage of high enantioselectivity in ligand-exchange mechanism and the main advantages of MEKC, LE-MEKC allows the manipulation of the selectivity for large classes of neutral and charged compounds, making possible separation that otherwise are not feasible by using only the mode of LE or MEKC. Using this combined separation mode, hydroxy acids and dipeptides, as well as 16 positional and optical isomers of tryptophan derivatives could be optically resolved[5]. The same group also proposed a method for the determination of the critical micelle concentration (CMC) of anionic surfactants based on LE-MEKC principle[6].

Abstract:Isoquinoline alkaloids and saponins are natural products with significant pharmacological effects. Because of the extreme similarity and diversity of their structures, separation of these compounds by capillary electrophoresis (CE) is difficult. A promising separation technique with good separation selectivity and high efficiency, microemulsion electrokinetic chromatography (MEEKC), has been used to separate ten isoquinoline alkaloids and six saponins from Gleditsia sinensis. Anionic and cationic surfactants such as sodium dodecyl sulfate (SDS) and cetyltrimethylammonium bromide (CTAB) were used as surfactants in MEEKC, on the basis of a series of separation optimizations. For separation of isoquinoline alkaloids comparison of MEEKC with non-aqueous capillary electrophoresis (CE) is discussed with regard to separation efficiency, selectivity, analysis time, and repeatability Study of the separation of the saponins revealed that separation selectivity was different for MEEKC with CTAB and SDS as surfactants; separation with CTAB was much better than that obtained with SDS.

Abstract:Many separation methods have been developed for biomedical analysis, including chromatographic (e.g. high performance liquid chromatography (HPLC) and gas chromatography (GC)) and electrophoretic methods (e.g. gel electrophoresis and capillary electrophoresis (CE)). Among these techniques, CE provides advantages in terms of high separation efficiency, simplicity, low sample and solvent volume consumption, short analysis time and applicability to a wide range of biomedically important substances. Microchip electrophoresis (ME) is a miniaturized platform of CE and is now considered as a simpler and more convenient alternative, which has demonstrated potential in analytical chemistry. High-throughput, cost-effective and portable analysis systems can be developed using ME. The current review describes different separation modes and detectors that have been employed in ME to analyze various classes of biomedical analytes (e.g. pharmaceuticals and related substances, nucleic acids, amino acids, peptides, proteins, antibodies and antigens, carbohydrates, cells, cell components and lysates). Recent applications (during 2010-2014) in these areas are presented in tables and some significant findings are highlighted.

The results indicated that the additions of modifiers could greatly affect the separation selectivity of xanthones, and the migration order of those xanthones depends on their structures and solute–micelle-modifier interactions. As a special variant of electrokinetic chromatography, microemulsion electrokinetic chromatography (MEEKC) has also been utilized for this separation for the comparison with MEKC, showing that MEEKC has better selectivity.

Abstract:An investigation of the basic factors which govern the microemulsion EKC (MEEKC) and MEKC for the separation of four benzoylurea. (BU) insecticides and their four analogs was carried out. In MEEKC, the separation of eight BU compounds was optimized by changing the microemulsion composition, such as concentration of SDS, octane, n-butanol, and isopropanol percentages, as well as capillary temperature. Separation optimization was also carried out for MEKC, showing that ACN and a high level of another additive gamma-CD were needed to achieve effective separation of these analytes. Although separation with baseline resolution was achieved by either MEEKC or MEKC methods, the separation selectivity resulting from the proposed MEEKC method was completely different from that of MEKC. In addition analytical time in MEEKC was longer than that in MEKC, but in view of theoretical plate numbers, detection limits, and reproducibility, both methods were effective for the analysis of BU insecticides and their analogs.

Abstract:There is great interest in using microfluidic channels packed with a stationary phase for chemical separations of complex mixtures. A key advantage of such techniques is the use of electroosmotic flow (EOF), controlled simply by applying electrical potentials between reservoirs. A disadvantage for this technique, however, is a lack of compatibility with gradient elution separations. This limitation arises from the dependence of EOF velocity on run buffer content (including the concentration of organic modifier). Here, we introduce a method for implementing gradient elution in electrochromatography in which multiple run buffers are velocity-matched, such that the elution profile resembles that found in conventional HPLC. This method is driven entirely with EOF, meaning that pumps, valves, and pressure fittings are not required. The method was validated by application to separations of peptide standards and protein digests. These results suggest that microfluidic electrochromatography may be compatible with a wide range of applications that have previously been unexplored.

Abstract:The selectivity of microemulsion electrokinetic capillary chromatography (MEEKC) was studied for the separation of nine xanthones under various operation variables including pH and concentrations of buffer, surfactant and cosurfactant, oil type, addition of organic solvents and cyclodextrins (CDs), capillary temperature and applied voltage. The selectivity was evaluated using such parameters as logarithm of octanolwater partition coefficient (log P), peak symmetry factor and theoretical plate number (N), showing that the changes in operating conditions had significantly different impacts on the separation selectivity for hydrophobic and hydrophilic analytes. The baseline separation of nine xanthones was achieved with satisfactory repeatability and high separation efficiency using 50 mmol/L borate buffer (pH 95) containing 10% (v/v) nbutanol, 80 mmol/L nheptane, 120 mmol/L SDS and 5 mmol/L sulfated βCD, under 20 kV applied voltage and at 35 ℃.

Abstract:The separation and selectivity of eight aromatic compounds ranging from hydrophilic to hydrophobic properties in micellar electrokinetic chromatography (MEKC) using sodium dodecyl sulfate (SDS) micelles or Tween 20-modified mixed micelles were investigated. The effect of different operation conditions such as SDS and Tween 20 modifier surfactant concentration, buffer pH, and applied voltage was studied. The resolution and selectivity of analytes could be markedly affected by changing the SDS micelle concentration or Tween 20 content in the mixed micelles. Applied voltage and pH of running buffers were used mainly to shorten the separation time. Complete separation of eight analytes could be achieved with an appropriate choice of the concentration of SDS micelles or Tween 20-modified mixed micelles. Quicker elution and better precision could be obtained with SDS–Tween 20 mixed micelles than with SDS micelles. The mechanisms that migration order of those analytes was mainly based on their structures and solute–micelle interactions, including hydrophobic, electrostatic, and hydrogen bonding interactions, were discussed.

Abstract:A new MEEKC method assisted with pressure-driven mobile phase was presented for the separation of felted explosives. Microemulsion solution was composed of 80 mmol/L heptane-120 mmol/L SDS (sodium dodecyl sulphate)-900 mmol/L butanol-10 mmol/L borate at pH 9.4 and a pressure-driven flow of 0.020 mL/min under 1.3 MPa was employed to manipulate the separation. Explosives HMX (1, 3, 5, 7-tetranitro-1, 3, 5, 7-tetrazacyclooctane) and TATB; (triamino-trinitrobenzene), which were felted on fluorine rubber F2311 (polytrifluorochlorethylene and polyvinylidene fluoride 1/1 co-polymerization) and F2314 (polytrifluorochlorethylene and polyvinylidene fluoride 4/1 co-polymerization) were well separated with very good peak shapes.

Abstract:Recently, 1-butanol modified MEKC was proven to be similar to MEEKC in separation performance. In the present work, typical microemulsion containing 0.8% n-octane/3.3% SDS/6.6% 1-butanol/20 mM borax buffer and corresponding swollen micelle without n-octane were used to compare their microdroplet structures including hydrodynamic radius, electrokinetic potential ζ and charge density at the hydrodynamic shear surface, as well as microenvironment polarity in the interior of the microdroplets. Three kinds of corticosteroids were separated with MEEKC and 1-butanol modified MEKC to assess their separation performances. The experiment results showed that both microstructure and separation performance in microemulsion and in swollen micelle systems were alike, no matter whether oil phase n-octane was present. The environment polarity in the core of swollen micelle was slightly higher than in the microemulsions, and both of them were higher than in n-octane medium. Furthermore, the influences of SDS and 1-butanol concentration on microstructures were measured in details. Increasing the amount of SDS, hydrodynamic radius decreased in microemulsion but increased in swollen micelle. On the contrary, ζ and shear surface charge density changed in the reverse trends. With increment of 1-butanol concentration, the hydrodynamic radius increased dramatically in microemulsions, whereas decreased slightly in swollen micelle. Even though using n-octane as oil core was not a key factor, microemulsions and swollen micelle as pseudostationary phase in EKC should not be exactly the same.
