Abstract:The unique enzyme-like properties of metal nanoparticles (NPs) hold great promise for chemical and biomedical applications; implementation relies on improvements in their catalytic efficiency. We have constructed PtCuCo nanostructures using hydrothermal treatment of Co3O4, Cu2+, and Pt2+ in aqueous solutions, resulting in PtCuCo trimetallic alloy NPs having hollow structures, as confirmed by TEM, XRD, XPS, and EDS analyses. These PtCuCo NPs are capable of oxidizing 3,3',5,5'-tetramethyl-benzidine (TMB) in either the absence or presence of hydrogen peroxide, suggesting efficient peroxidase- and oxidase-like activity. These NPs could also catalyze the reduction of 4-nitrophenol by NaBH4. We believe that the combination of Pt and oxidized Cu and Co species results in surfaces with rich active sites possessing a wide range of electronic variation, which promotes both generation of hydroxyl radicals and electron transfer. Consequently, the catalytic capabilities of our PtCuCo NPs are greater than those of either Pt or PtCu NPs. Building upon the peroxidase-like activity of PtCuCo NPs, we developed a colorimetric method not only capable of specifically detecting glucose, but also able to detect rongalite and HS-, on the basis of the inhibitory effect of these substances upon PtCuCo nanoperoxidase.

Abstract:Glutathione (GSH) is an important reducing molecule in organisms, and its abnormal levels are closely related to certain diseases. Thus, developing a fast and simple strategy to detect GSH, especially in complex channels such as blood vessels, is of great significance for disease prevention and diagnosis. In this work, a novel peroxidase-like catalytic activity tubular microrocket (FeO x /Pt/PEDOT) with efficient propulsion and steerable motion has been successfully developed for GSH detection. The microrocket fueled by H 2 O 2 exhibits strong propulsion for solution self-stirring and effective ROS generation to GSH color reaction. Combining these advantages, the catalytically propelled microrocket-treated sample can greatly shorten a half time of detecting reactions and realize efficient GSH detections. In addition, the microrocket shows effective directional detection of GSH in a designed complex channel due to its excellent magnetically navigation, which is of great significance in GSH detection in the microenvironment. This attractive catalytic microrocket has strong propulsion, effective direction control, and excellent GSH detection performance, which shows great potential in biodetection and biomedical applications, especially in visual detection of vessels microenvironment.

Abstract:Nanocarbon oxides have been proved to possess great peroxidase-like activity, catalyzing the oxidation of many peroxidase substrates, such as 3,3',5,5'-tetramethylbenzidine (TMB) and o-phenylenediamine dihydrochloride (OPD), accompanied by a significant color change. This chromogenic reaction is widely used to detect glucose and occult blood. The chromogenic reaction was intensively investigated with density functional theory and molecular-level insights into the nature of peroxidase-like activity were gained. A radical mechanism was unraveled and the carboxyl groups of nanocarbon oxides were identified as the reactive sites. Aromatic domains connected with the carboxyl groups were critical to the peroxidase-like activity.

Abstract:Inspired by the ultrahigh reactivity of the Fe-N4 active site in natural enzyme horseradish peroxidase (HRP), this work designs and fabricates a two-dimensional (2D) metal- organic framework (MOF)-based nanozyme, 2D Cu-TCPP(Fe) MOFs, involving long-ordered arrangement of the Fe-N4 macrocyclic units. The framework has been constructed by the bridging coordination between Cu(II) ions and the peripheral carboxyl groups of a Fe(III) ion-centered metalloporphyrin TCPPFe. Taking H2O2 and 3,3 ',5,5 '-tetramethylbenzidine (TMB) as the substrates, the nanozyme shows an excellent activity toward H2O2 decomposition with a Michaelis constant (Km) of 2.67 mM smaller than that of HRP. The hydroxyl radical (center dot OH)-involved reaction mechanism has been verified experimentally, with the Fe-N4 moieties identified as the active sites. As a proof-of-concept application, the nanozyme-catalyzed TMB + H2O2 system is employed as a colorimetric assay for glutathione detection, which delivers two linear ranges of 0.1-10 and 10-60 mu M, with a lower detection limit of 90.8 nM (S/N = 3). In the meantime, the satisfying peroxidase-like (POD-like) activity of 2D Cu-TCPP(Fe) MOFs holds great potential for wider biochemical and biomedical applications.
Pt nanoparticles dispersed in a metal-organic framework as peroxidase mimics for colorimetric detection of GSH

Abstract:Metal-organic skeleton materials have been widely used in catalysis with their porous structure and adsorption properties. Precious metal nanoparticles have good catalytic properties. If the noble metal nanoparticles are adsorbed on the MOFs surface, the active sites can be increased and the catalytic effect of the materials can be greatly improved. We successfully synthesized Pt@ZIF-8 in two steps, the average particle size of Pt nanoparticles is about 3 nm. Pt@ZIF-8 possesses peroxidase activity and can oxidize colorless TMB to oxTMB in the presence of hydrogen peroxide. The peroxide-like nature of Pt@ZIF-8 is consistent with Michaelis-Menten kinetics. Glutathione is a reducing substance that reduces blue oxTMB to colorless oxTMB. This colorimetric method achieves a simple, sensitive and intuitive detection of glutathione. The detection limit of this experiment is low, which is promising in biomolecular detection.

Abstract:Artificial peroxisome plays an important part in protocell system construction and disease therapy. However, it remains an enormous challenge to exploit a practicable artificial peroxisome with multiple and stable activities. Nanozymes with multienzyme mimetic activities stand out for artificial peroxisome preparation. Herein, a novel nanozyme─Co-nanoparticle-embedded N-enriched carbon nanocubes (Co,N-CNC) decorated by hollow NiPt nanospheres (hNiPt@Co-NC) with featured tetra-enzyme mimetic activities of natural peroxisome─was prepared. Due to the synergistic effect of hollow NiPt nanospheres (hNiPtNS) and cubic porous Co,N-CNC support, hNiPt@Co-NC exhibited oxidase (OXD), peroxidase (POD), catalase (CAT), and superoxide dismutase (SOD)-like activities with comparable catalytic efficiency, enabling it to be a competitive candidate for artificial peroxisome investigation. Based on the high OXD-mimetic activity of hNiPt@Co-NC, a facile colorimetric platform was proposed for reduced glutathione (GSH) detection with a wide linear range (0.1-5 μM, 5-100 μM) and a low detection limit (27 nM). Thus, the hNiPt@Co-NC with tetra-enzyme mimetic activities possessed bright prospects in diversified biotechnological applications, including artificial organelles, biosensing, and medical diagnostics.

Abstract:To strengthen the enzyme-mimic catalytic activity of classical transition metal copper and iron oxide, bimetal Cu-Fe oxide mesoporous nanoparticles (Cu-Fe MNPs) were prepared in one pot under the microwave-assisted conditions. The proposed Cu-Fe MNPs possessed a well-distributed mesoporous structure with an average aperture of 3.91 nm and a high BET specific surface area of 197.93 m2g−1. Its peroxidase-like kinetic parameters (Km/Vmax) to catalyze the redox of TMB and H2O2 were 0.011 mM/3.01×10−8 M·s−1 and 57.44 mM/4.12×10−8 M·s−1, respectively. In particular, either toxic o-aminophenol (o-AP) or bioactive glutathione (GSH) could deteriorate its peroxidase-like property with obvious color changing from blue to colorless for the proposed Cu-Fe MNPs-TMB-H2O2 system. Under the optimized conditions, Cu-Fe MNPs-TMB-H2O2 could selectively response o-AP and GSH with quite low detection limits of 1.57×10−8 mol/L (S/N=3) for o-AP and 3.35×10−8 mol/L (S/N=3) for GSH respectively. When applied for visual monitoring of o-AP and GSH in real aqueous, fruit juice and medicinal samples, the detection recovery (%) is between 96.8~101.4 % for o-AP and 97.1~105.8 % for GSH. The mechanism for synergic-enhancement peroxidase-like activity of Cu-Fe MNP and these for selective recognition of o-AP and GSH were investigated in detail.

Abstract:Various nanomaterials with peroxidase activity (nanozyme) have been designed for bio catalysis and biosensing, however, most of them need further design and modification of probe molecules for the specific binding reaction with targets. This results in a decrease in catalysis activity and hinders them to be perfect alternatives to natural enzyme in biosensing. In this work, an enhanced nanozyme was synthesized by functionalizing natural microperoxidase-11 (MP-11) on a hybrid graphene oxide-gold (GO-Au) material. The designed nanozyme showed an enhanced catalysis activity and realized a robust and efficient colorimetric detection of cysteine based on specific binding reaction between active iron center from MP-11 and thiol in cysteine. The enhanced properties show promising applications of complex nanozyme and provides a great opportunity for developing efficient sensing systems.

Abstract:The oxidase-mimicking activity of ferroelectric BaTiO 3 nanoparticles (BTO NPs) is reported. In the absence of hydrogen peroxide or oxygen, BTO NPs catalyzed the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) and other chromogenic substrates. The catalytic constant (K cat ) was eight times that of fluorescein and 10 4 times that of bovine serum albumin-protected Se nanozymes. In the mechanism of the chromogenic reaction catalyzed by BTO NPs, the intrinsic electric field of the ferroelectric BTO drove the separation of holes and electrons, and the separated carriers reacted with water to produce reactive oxygen species that oxidized the chromogenic substrates. The BTO NPs-TMB chromogenic system was used to detect glutathione , which restored the blue-colored oxidized TMB to its colorless initial state. There was a good linear relationship between the absorbance change and the GSH concentration over the range 0.50–20 μM, with a 0.2-μM limit of detection. This work not only proposed a new idea of screening and designing nanozymes from ferroelectric materials but also expand the application of ferroelectric nanomaterials to bio-detection.

Abstract:A facile approach is proposed for the growth of platinum nanoparticles on graphene oxide (PtNPs/GO). The resulting PtNPs/GO hybrid has been proved to function as peroxidase mimics that can catalyze the oxidation of peroxidase substrates in the presence of hydrogen peroxide (H2O2). Kinetic studies indicate that the PtNPs/GO nanocomposite has a considerably higher affinity for both 3,3', 5,5'-tetramethyl-benzidine (TMB) and H2O2 than those of other platinum-based peroxidase mimics. Furthermore, colorimetric recognition and sensing of L-cysteine with high sensitivity and selectivity is presented based on target-induced shielding against the peroxidase-like activity of PtNPs/GO. We envision that this material will be an ideal candidate for a wide range of potential applications in the fields of biomedicine and environmental chemistry.

Abstract:Glutathione (GSH), as an significant intracellular antioxidant, plays a vital role in preserving the balance of cellular redox homeostasis. The accurate and sensitive detection of GSH is essential for understanding the cellular redox status and for the diagnosis and treatment of many diseases. Herein, a core–shell structure composed of glucose oxidase (GOx), platinum (Pt) nanoparticles and a zeolitic imidazolate framework (ZIF-8) shell was constructed. Pt NPs in the core–shell showed high peroxidase-like catalytic activity. The as-prepared Pt-GOx@H-ZIF-8 can be utilized for colorimetric detection of GSH, exhibiting a broad linear range from 0.5 to 90 μM, and a low limit of detection (LOD) of 0.4 μM. Pt-GOx@H-ZIF-8 showed excellent sensitivity, selectivity and stability towards glutathione detection, making it a promising candidate for intracellular antioxidants.

Abstract:Nanozyme possess the advantages of high enzyme simulation activity, low cost, and excellent stability. As a substitute for natural enzymes, nanozyme have demonstrated significant potential in various fields. In this study, a bimetallic organometallic framework Cu@Co-MOFs was synthesized via the hydrothermal method using terephthalic acid (H2BDC) as a ligand and characterized. Cu@Co-MOFs exhibits outstanding peroxidase activity (POD) by catalyzing the oxidation of 3,3 ',5,5 '-tetramethylbenzidine (TMB) and other color-developing substrates through center dot OH formation. A simple, rapid, and reliable colorimetric method for quantitatively detecting glutathione (GSH) was developed based on the Pod-like activity of Cu@Co-MOFs. The linear range of this method was 1-1200 mu M with a detection limit of 0.72 mu M. This work not only proposes a bimetallic organic framework with high POD as a straightforward and practical platform for visually detecting GSH but also provides a strategy to enhance the performance of nanomaterials.

Abstract:A Fe3O4/mesoporous graphitized carbon (Fe3O4/m-GC) composite was prepared through a facile calcination method with iron-based metal–organic frameworks (Fe-MOFs) as a sacrificial template. After carbonization, the Fe3O4 nanoparticles were uniformly dispersed in the mesoporous carbon support, resulting in spatial structural stability. The mesoporous carbon support obtained was highly graphitized and exhibited eminent electrical conductivity, which accelerated the electron transfer between the Fe3O4 nanoparticles by Fe(II)/Fe(III) redox cycles and m-GC by C = Csp2/C-Csp3 redox cycles, leading to the excellent peroxidase-mimetic activity of Fe3O4/m-GC. Km values for tetramethylbenzidine (TMB) and H2O2 were 26.8 and 15.8 times lower than that of natural horseradish peroxidase, respectively. Abstract:Water-soluble cupric oxide nanoparticles are fabricated via a quick-precipitation method and used as peroxidase mimetics for ultrasensitive detection of hydrogen peroxide and glucose. The water-soluble CuO nanoparticles show much higher catalytic activity than that of commercial CuO nanoparticles due to their higher affinity to hydrogen peroxide. In addition, the as-prepared CuO nanoparticles are stable over a wide range of pH and temperature. This excellent stability in the form of aqueous colloidal suspensions makes the application of the water-soluble CuO nanoparticles easier in aqueous systems. A colorimetric assay for hydrogen peroxide and glucose has been established based on the catalytic oxidation of phenol coupled with 4-amino-atipyrine by the action of hydrogen peroxide. This analytical platform not only confirms the intrinsic peroxidase-like activity of the water-soluble cupric oxide nanoparticles, but also shows its great potential applications in environmental chemistry, biotechnology and medicine.

Herein, a hybrid peroxidase-like nanocatalyst consisting of PtNPs in situ growing on g–C3N4 nanosheets with enhanced peroxidase-mimic catalytic activity (PtNP@g–C3N4 nanosheets) was prepared for H2O2 and oxidase-based colorimetric assay. g–C3N4 nanosheets can be used as carriers to solve the problem of poor stability of PtNPs. We observed that the catalytic ability could be maintained for more than 90 days. PtNP@g–C3N4 nanosheets could quickly catalyze the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB), and the absorbance of blue color oxidized TMB (oxTMB) showed a robust linear relationship with the concentration of H2O2 (the detection limit (LOD): 3.33 μM). By utilizing H2O2 as a mediator, this strategy can be applied to oxidase-based biomolecules (glucose, organophosphorus, and so on, that generate or consume hydrogen peroxide) sensing. As a proof of concept, a sensitive assay of cholesterol that combined PtNP@g–C3N4 nanosheets with cholesterol oxidase (ChOx) cascade catalytic reaction was constructed with an LOD of 9.35 μM in a widespread range from 10 to 800 μM (R2 = 0.9981). In addition, we also verified its ability to detect cholesterol in fetal bovine serum. These results showed application prospect of PtNP@g–C3N4 nanosheets-based colorimetry in sensing and clinical medical detection.
