Abstract:Nanomagnet-silica shell (Fe3O4@SiO2) decorated with Au@Pd nanoparticles (NPs) were synthesized successfully. The characterization of Fe3O4@SiO2-NH2-Au@PdNPs was achieved using several spectroscopic and microscopic techniques. The quantitative surface analysis was confirmed using X-ray photoelectron spectroscopy. The Fe3O4@SiO2-NH2-Au@Pd0.30NPs exhibited excellent peroxidase-like activity by effectively catalyzing the oxidation of 3,3',5,5'-tetramethylbenzidine (TMB) in the presence of H2O2. The absorption peaks at 370 and 652 nm confirmed the peroxidase-like activity of the Fe3O4@SiO2-NH2-Au@Pd0.30NPs. The Michaelis-Menten constant (Km) of 0.350 and 0.090 mM showed strong affinity toward H2O2 and TMB at Fe3O4@SiO2-NH2-Au@Pd0.30NPs. The mechanism of the peroxidase-like activity was found to proceed via an electron transfer process. A simple colorimetric sensor based on glucose oxidase and Fe3O4@SiO2-NH2-Au@Pd0.30NPs showed excellent selectivity and sensitivity towards the detection of glucose. The fabricated glucose biosensor exhibited a wide linear response toward glucose from 0.010 to 60.0 μM with an limit of detection of 60.0 nM and limit of quantification of 200 nM. The colorimetric biosensor based on Fe3O4@SiO2-NH2-Au@Pd0.30NPs as a peroxidase mimic was also successfully applied for the determination of glucose concentrations in serum samples. The synthesized Fe3O4@SiO2-NH2-Au@Pd0.30NPs nanozymes exhibited excellent potential as an alternative to horseradish peroxidase for low-cost glucose monitoring.

Abstract:New polymer-enzyme-metallic nanoparticle composite films with a high-load and a high-activity of immobilized enzymes and obvious electrocatalysis/ nano-enhancement effects for biosensing of glucose and galactose are designed and prepared by a one-pot chemical pre-synthesis/ electropolymerization (CPSE) protocol. Dopamine (DA) as a reductant and a monomer, glucose oxidase (GOx) or galactose oxidase (GaOX) as the enzyme, and HA(u)Cl(4) or H(2)PtCl(6) as an oxidant to trigger DA polymerization and the source of metallic nanoparticles, are mixed to yield polymeric.. bionanocomposites (PBNCs), which are then anchored on the electrode by electropolymerization of the remaining DA monomer. The prepared PBNC. material has good biocompatibility; a highly uniform dispersion of the nanoparticles with a narrow size distribution, and high load/activity of the immobilized enzymes, as verified by transmission/scanning electron microscopy and electrochemical quartz crystal microbalance. The thus-prepared enzyme electrodes show a largely improved amperometric biosensing performance, e.-g., a very high detection sensitivity (99 or 129 mu A cm(-2) mm(-1) for glucose for Pt PBNCs on bare or platinized Au), a sub-micromolar limit of detection for glucose, and an excellent durability, in comparison with those based on conventional procedures. Also, the PBNC-based enzyme electrodes work well in the second-generation biosensing mode. The proposed one-pot CPSE protocol may be extended to the preparation of many other functionalized PBNCs for wide applications.
A PdPt Nanoparticle-Decorated Thiol-Functionalized MOF with High Peroxidase-Like Activity for Colorimetric Sensing of D-glucose and Chlorophenol Isomers.

Abstract:The peroxidase (POD)-like catalytic activity of various nanozymes was extensively applied in many significant fields. In this study, a thiol-functionalized MOF-loaded PdPt nanocomposite (UiO-66-(SH)(2)@PdPt) was fabricated, which possesses superior and selective POD-like activity with strong affinity towards H2O2 and 3,3 & PRIME;,5,5 & PRIME;-tetramethylbenzidine under mild conditions. The POD-like property of UiO-66-(SH)(2)@PdPt was used to sensitively detect the concentration of d-glucose under near-neutral (pH = 6.5) conditions. The detection limit of d-glucose was as low as 2.7 & mu;M, and the linear range of d-glucose was 5-700 & mu;M. In addition, UiO-66-(SH)(2)@PdPt could accelerate the oxidative coupling chromogenic reaction of chlorophenol (CP) and 4-aminoantipyrine (4-AAP) in the presence of H2O2. Based on this phenomenon, a simple and visualized sensing array for the identification of chlorophenol contaminant isomers was further constructed to finally achieve the effective differentiation of three monochlorophenol isomers and six dichlorophenol isomers. Furthermore, a colorimetric detection method for 2-chlorophenol and 2,4-dichlorophenol was established. This work provides an effective means to improve the catalytic activity and selectivity of nanozymes by introducing an ideal carrier, which will be of significant value for the design of efficient nanozymes.
Properties of Dendrimer‐Encapsulated Pt Nanoparticles Doped Polypyrrole Composite Films and Their Electrocatalytic Activity for Glucose Oxidation

Abstract:A type of novel electroanalytical sensing nanobiocomposite material was prepared by electropolymerization of pyrrole containing poly(amidoamine) dendrimers-encapsulated platinum nanoparticles (Pt-PAMAM), and glucose oxidase (GOx). The Pt nanoparticles encapsulated in PAMAM are nearly monodisperse with an average diameter of 3 nm, and they provide electrical conductivity. Polypyrrole acts as a polymer backbone to give stable and homogeneous cast thin films, and it also defines the electrical conductivity. Both Polypyrrole and PAMAM can provide a favorable microenvironment to keep the bioactivity of enzymes such as glucose oxidase. The homogeneity of GOx/Pt-PAMAM-PPy nanobiocomposite films was characterized by atomic force microscopy (AFM). Ampero-metric biosensors fabricated with these materials were characterized electrochemically using cyclic voltammetry (CV) I electrochemical impedance spectra (EIS) and amperometric measurements in the presence of hydrogen peroxide or glucose. All those show the resultant biosensor sensitivity was strongly enhanced within the nanobiocomposite film. The optimized glucose biosensor displayed a sensitivity of 164 mu A mM(-1) cm(-1), a linear range of 0.2 to 600 mu M, a detection limit of 10 nM, and a response time of < 3s.

Abstract:The carbon-based nanomaterials with peroxidase-like property show huge potential as candidates for natural enzymes. The design of carbon nanozymes with high activity can expand their potential application. Herein, the carbon nanozymes with enhanced catalytic property by simultaneous phosphorus, oxygen dual-doping and nanosheet structure construction are fabricated through one-step pyrolysis process. The dual-doped phosphorus and oxygen element is conducive to accelerate electron transfer and form active site with moderate free energy for activation of H2O2 to ·OH. Besides, the nanosheet structure with enlarged surface area is beneficial to expose more catalytic sites. Benefiting from its great peroxides-like activity, the phosphorus, oxygen dual-doped carbon nanosheet materials (POCNS) are used in colorimetric determination of H2O2 and glucose with cascade reaction of glucose oxidase and POCNS. And the sensitive colorimetric detection assay for organophosphorus pesticide of chlorpyrifos was constructed with regulation of peroxidase-like activity by aptamer. The system of POCNS and H2O2 further displays great antibacterial property in inactivation of Escherichia coli. This work provides an avenue to construct high-efficient carbon nanozymes and develop their potential application in environmental and biological field.

Abstract:Nanozymes are of particular interest due to their enzyme-mimicking activity and high stability that are favorable in biomedical sensing and immunoassays. In this work, we report a highly specific N-doped nanozyme through pyrolysis of framework-confined bovine serum albumin (BSA). This strategy allows one to translate the low-cost and featureless BSA into a highly active enzyme mimic. The obtained carbon nanozyme (denoted as HBF-1-C800) displays 3- to 7-fold enhancement on peroxidase (POD) activity compared with the conventional carbon nanozymes and also shows ca. 5-fold activity enhancement compared to the reported N-doping graphene. Such excellent POD activity originates from high N-doping efficiency, protein-induced defective sites, and the intrinsic porous structure of HBF-1-C800, which provides abundantly accessible active sites and accelerates substrate diffusion simultaneously. Importantly, the HBF-1-C800 nanozyme has highly specific POD activity and also enables resistance to several harsh conditions that should denature natural enzymes. These features allow it with high accuracy, stability, and sensitivity for biosensing applications. Development of an Ultrasmall and Biocompatible Platinum Nanozyme Encapsulated by Zwitterionic Dendrimer for Highly Sensitive Detection of Glucose

Abstract:Many kinds of noble metal nanoparticles can mimic the peroxidase-like function of horseradish peroxidase, which results in their wide applications in bio-related detection and drug delivery. However, those metal nanoparticles usually have low stability and reduced catalytic activity in biological complex medium. Herein, a zwitterionic peroxidase-like enzyme has been developed, which has high stability in fibrinogen solutions and high sensitivity for glucose detection. Maleic anhydride, cysteamine, and zwitterionic peptide EKEKC (EK-5) were used to modify generation 5 poly(amido amine) dendrimers (G5 PAMAM) to prepare zwitterionic dendrimer G5MEKnC with nonfouling properties. Finally, the G5MEKnC-encapsulated platinum nanoparticles (Ptn-G5MEK50C) were prepared by entrapping the platinum nanoparticles (1.40 nm) in the catalytic centers in the interior of G5MEK50C. Pt55-G5MEK50C showed high stability in the buffer solution and the fibrinogen solution within 4 days. They also displayed high biocompatibility toward HeLa cells based on cytotoxicity results and morphological observations. Furthermore, the catalytic oxidation of 3,3',5,5'-tetramethylbenzidine with H2O2 by Pt55-G5MEK50C followed the Michaelis-Menten equation, which confirmed their peroxidase-like properties. The catalytic mechanism was due to the generation of •OH from H2O2. More importantly, the peroxidase-like ability of Pt55-G5MEK50C was successfully used to establish a method for the determination of glucose concentration with a broad linear range of 1-2000 μM and a low detection limit of 0.1 μM. This method was highly accurate for the determination of glucose concentration in plasma. The zwitterionic dendrimer template enhanced the properties of Pt55-G5MEK50C. Taken together, a new kind of biocompatible nanozyme has been developed and successfully used for the sensitive detection of glucose in bio-related medium.

Abstract:With the rapid development of nanotechnology and catalysis science, numerous enzyme-mimicking nanomaterials have been rationally designed as the substitutes for natural enzymes. Herein, the Pt-loaded hollow mesoporous carbon nanospheres (Pt-HMCNs) were proposed as the peroxidase mimic for the first time. The monodispersed HMCNs with huge hollow cavity and mesoporous carbon skeleton were first synthesized via high-temperature carbonization and NaOH etching of core-shell structured SiO2@RF/SiO2 (RF = resorcinol-formaldehyde resin) nanospheres, which were prepared through a simple surfactant-free strategy in a one-pot system only containing three main raw chemicals as tetraethyl orthosilicate, resorcinol, and formaldehyde. And then the Pt-HMCNs were obtained through surface graft of -NH2 groups and loading of as-synthesized ultrasmall Pt nanoparticles (NPs) in size of 1-3 nm on the HMCNs. The prepared Pt-HMCNs nanocomposites exhibited superior peroxidase-like catalytic activity toward the oxidation of 3,3',5,5'-tetramethylbenzidine (TMB) substrate, which owned stronger affinity to H2O2 and TMB as substrates than the natural enzyme (horseradish peroxidase). The outstanding peroxidase-mimic performance of the Pt-HMCNs is due to the unique hollow mesoporous structure as well as the interfacial effect between Pt NPs and carbon skeleton. A simple and general colorimetric approach was established to accurately detect ascorbic acid (AA) and glucose, and their detection limits were calculated to be 3.29 and 35.4 mu M, respectively. Besides, wide linear range and high selectivity against interfering substances were also achieved in this assay during AA and glucose detection based on the Pt-HMCNs catalyst. The colorimetric method built on Pt-HMCNs in this work could be applied to biomedicine, food science, environment monitoring, and other fields for accurately detecting ascorbic acid, glucose, and other important substances in some complex systems.
Peroxidase-mimetic activity of a nanozyme with uniformly dispersed Fe3O4 NPs supported by mesoporous graphitized carbon for determination of glucose

Abstract:A Fe3O4/mesoporous graphitized carbon (Fe3O4/m-GC) composite was prepared through a facile calcination method with iron-based metal–organic frameworks (Fe-MOFs) as a sacrificial template. After carbonization, the Fe3O4 nanoparticles were uniformly dispersed in the mesoporous carbon support, resulting in spatial structural stability. The mesoporous carbon support obtained was highly graphitized and exhibited eminent electrical conductivity, which accelerated the electron transfer between the Fe3O4 nanoparticles by Fe(II)/Fe(III) redox cycles and m-GC by C = Csp2/C-Csp3 redox cycles, leading to the excellent peroxidase-mimetic activity of Fe3O4/m-GC. Km values for tetramethylbenzidine (TMB) and H2O2 were 26.8 and 15.8 times lower than that of natural horseradish peroxidase, respectively. Abstract:The enzyme-like properties of nanozymes may be considerably affected by the structure and surface groups, which thus need to be optimized. Here, through a simple NaOH chemical corrosion method, the chemical structure similar to N-Methylpyrrolidone (NMP), which possessed intrinsic oxidase-like activity, was introduced into polypyrrole (PPy), and then this nanomaterial became oxygen-functionalized PPy (o-PPy) with excellent oxidase-like activity from PPy without this property. Furthermore, after compounding magnetic Fe3O4, the obtained nanocomposites Fe3O4@o-PPy nanoparticles (Fe3O4@o-PPy NPs) showed superiorities in separation during synthesis and real-time control of enzyme catalysis. Studies have found that the enzymatic activity of Fe3O4@o-PPy NPs depended on the amount of functionalized oxygen and the conjugation extent of o-PPy. Fe3O4@o-PPy NPs had efficient oxidase-like activity under a wide range of pH and temperature. Based on the oxidase-like activity of Fe3O4@o-PPy NPs, a colorimetric sensor for glutathione (GSH), which presented rich color changes and satisfactory colorimetric resolution by adding the amaranth, was realized. We believe that the functional modification and structural regulation of PPy can not only realize its wider application but also promote the discovery of novel and efficient nanozymes.
Nanostructures of Prussian blue supported on activated biochar for the development of a glucose biosensor

Abstract:This work emphasizes the utilization of biochar, a renewable material, as an interesting platform for anchoring redox mediators and bioreceptors in the development of economic, environmentally friendly biosensors. In this context, Fe(III) ions were preconcentrated on highly functionalized activated biochar, allowing the stable synthesis of Prussian blue nanostructures with an average size of 58.3 nm. The determination of glucose was carried out by indirectly monitoring the hydrogen peroxide generated through the enzymatic reaction, followed by its subsequent redox reaction with reduced Prussian blue (also known as Prussian white) in a typical electrochemical-chemical mechanism. The EDC/NHS (1-ethyl-3-(3-dimethylaminopropyl)carbodiimide hydrochloride and N-Hydroxysuccinimide) pair was employed for the stable covalent immobilization of the enzyme on biochar. The biosensor demonstrated good enzyme-substrate affinity, as evidenced by the Michaelis-Menten apparent kinetic constant (4.16 mmol L −1 ), and analytical performance with a wide linear dynamic response range (0.05–5.0 mmol L −1 ), low limits of detection (0.94 μmol L −1 ) and quantification (3.13 μmol L −1 ). Additionally, reliable repeatability, reproducibility, stability, and selectivity were obtained for the detection of glucose in both real and spiked human saliva and blood serum samples.

Abstract:Nanomaterials with enzyme-like characteristics (nanozymes) have emerged as potential replacements for natural enzymes due to their potential to overcome several critical limitations of natural enzymes, including low stability as well as high costs in preparation and purification. Herein, we have developed hybrid nanostructures that incorporate cobalt oxide nanoparticles (Co₃O₄ NPs) and gold nanoclusters (AuNCs) through electrostatic attraction induced by simple incubation in an aqueous buffer for 2 hours. Owing to the synergistic effect of Co₃O₄ NPs and AuNCs, the constructed Co₃O₄/Au hybrid nanostructures yielded highly enhanced peroxidase-like activity and enabled rapid catalytic oxidation of a chromogenic substrate, 3,3',5,5'-tetramethylbenzidine (TMB), producing a blue colored solution depending on the amount of H₂O₂. Moreover, we observed catalytic activity of the Co₃O₄/Au hybrid over a broad pH range, especially at physiologically relevant pH in the range of 5.0-7.4, which is advantageous for applications in biological systems. Using the hybrid as peroxidase mimic, we successfully determined the level of target H₂O₂ or glucose by coupling with glucose oxidase with excellent specificity and sensitivity. Based on this study, we expect that Co₃O₄/Au hybrid nanostructures can serve as potent peroxidase mimics for the detection of clinically important target molecules.

Abstract:Nanomaterials with enzyme-mimicking characteristics have engaged great awareness in various fields owing to their comparative low cost, high stability, and large-scale preparation. However, the wide application of nanozymes is seriously restricted by the relatively low catalytic activity and poor specificity, primarily because of the inhomogeneous catalytic sites and unclear catalytic mechanisms. Herein, a support-sacrificed strategy is demonstrated to prepare a single iron site nanozyme (Fe SSN) dispersed on the porous N-doped carbon. With well-defined coordination structure and high density of active sites, the Fe SSN performs prominent peroxidase-like activity by efficiently activating H2 O2 into hydroxyl radical (•OH) species. Furthermore, the Fe SSN is applied in colorimetric detection of glucose through a multienzyme biocatalytic cascade platform. Moreover, a low-cost integrated agarose-based hydrogel colorimetric biosensor is designed and successfully achieves the visualization evaluation and quantitative detection of glucose. This work expands the application of single-site catalysts in the fields of nanozyme-based biosensors and personal biomedical diagnosis.

Abstract:Polyvinylpyrrolidone-functionalized IrPt alloy nanostructures (PVP/IrPt ANs) exhibited triple enzyme mimetic activities.Enzyme-like activities of PVP/IrPt ANs were employed for colorimetric and fluorescent detection of H2O2.PVP/IrPt ANs were coupled with the glucose oxidase for rapid, sensitive, and selective detection of glucose.The developed sensor demonstrated good selectivity and recovery of glucose from spiked biological and food samples.Nanomaterials that demonstrate multi-enzyme activities have increasingly gained interest due to their potential applications in point-of-care diagnostics. In the present work, polyvinylpyrrolidone-functionalized bimetallic IrPt alloy nanostructures (PVP/IrPt ANs) exhibited excellent peroxidase-, oxidase-, and catalase-like activities were synthesized with a reflux-assisted one-pot wet chemical process. The use of PVP as stabilizer and surfactant during the synthesis produced monodisperse nanoparticles (NPs) with average diameter of 4.1 nm. The PVP/IrPt ANs catalyze the oxidation of H2O2, TMB, and amplex red through the peroxidase and oxidase biomimetic activities. The kinetic parameters of PVP/IrPt ANs were measured to demonstrate their enhanced enzyme-like behavior relative to previously reported bimetallic nanozymes. The oxidase- and peroxidase-like activities of PVP/IrPt ANs were employed for efficient colorimetric and fluorescent detection of H2O2 concentrations. Moreover, the multi-enzyme activity of PVP/IrPt ANs was coupled with the glucose oxidase activity to fabricate colorimetric and fluorometric glucose sensors. The colorimetric glucose sensor revealed a broad detection range of 2.5 µM – 1.5 mM, while the fluorometric sensor showed excellent limit of detection (LOD) of 177.5 nM. Abstract:Platinum nanoparticles (PtNPs) are classical peroxidase-like nanozyme; self-agglomeration of nanoparticles leads to the undesirable reduction in stability and catalytic activity. Herein, a hybrid peroxidase-like nanocatalyst consisting of PtNPs in situ growing on g–C3N4 nanosheets with enhanced peroxidase-mimic catalytic activity (PtNP@g–C3N4 nanosheets) was prepared for H2O2 and oxidase-based colorimetric assay. g–C3N4 nanosheets can be used as carriers to solve the problem of poor stability of PtNPs. We observed that the catalytic ability could be maintained for more than 90 days. PtNP@g–C3N4 nanosheets could quickly catalyze the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB), and the absorbance of blue color oxidized TMB (oxTMB) showed a robust linear relationship with the concentration of H2O2 (the detection limit (LOD): 3.33 μM). By utilizing H2O2 as a mediator, this strategy can be applied to oxidase-based biomolecules (glucose, organophosphorus, and so on, that generate or consume hydrogen peroxide) sensing. As a proof of concept, a sensitive assay of cholesterol that combined PtNP@g–C3N4 nanosheets with cholesterol oxidase (ChOx) cascade catalytic reaction was constructed with an LOD of 9.35 μM in a widespread range from 10 to 800 μM (R2 = 0.9981). In addition, we also verified its ability to detect cholesterol in fetal bovine serum. These results showed application prospect of PtNP@g–C3N4 nanosheets-based colorimetry in sensing and clinical medical detection.

Abstract:In past decade, Pt-based nanomaterials as peroxidase mimics have attracted much attention for H2O2 and glucose detection. However, easy aggregation of Pt nanoparticles (Pt NPs) greatly decreases their peroxidase-like activity. In this work, novel Pt/EMT nanocomposites were prepared by uniformly loading Pt NPs (5-8 nm) onto the support of ultrasmall EMT zeolite (15-20 nm), a kind of low-silica microporous aluminosilicate material. The hybrid Pt/EMT nanomaterials could be well dispersed in water to form a homogeneous suspension, and were then utilized as a superior peroxidase-like catalyst for oxidation of 3,3',5,5'-tetramethylbenzidine (TMB) in the presence of hydrogen peroxide (H2O2). The optimal catalyst of 2.6Pt/EMT nanocomposite exhibited excellent catalytic performance toward H2O2 and TMB than natural enzyme of horseradish peroxidase (HRP) by using a steady-state kinetic analysis based on the typical Michaelis-Menten kinetics theory. The peroxidase-like catalyst showed a promising activity in a wide pH and temperature range as well as the long-term stability. A facile and reliable colorimetric assay based on the peroxidase mimic of Pt/EMT nanocomposite was constructed for precise detection of H2O2 and glucose in a wide linear range, with low limits of detection of 1.1 μM and 13.2 μM, respectively. Due to high selectivity to glucose against other sugars on the catalyst, the method was demonstrated to accurately measure the concentration of glucose in real samples including human blood serum and fruit juices, indicating a potential application of the Pt/EMT nanocomposites as a robust peroxidase mimic and a reliable biosensor in the fields of clinical diagnosis, pharmaceutical, food research and so on.

Abstract:Multifunctional enzymatic nanomaterials (nanozymes) with good biocompatibility, strong enzymatic activity, and substrate specificity are the desired alternatives to natural enzymes. Here, the design of a peroxidase mimetic nanozyme capable of detecting and depleting glutathione (GSH) in the bacterial cells is presented. The nanozyme palladium-iron nanostructure decorated graphdiyne nanosheet (PdFe/GDY) was prepared by a facile hydrothermal approach. It can efficiently catalyze the decomposition of H2O2 to produce •OH by a peroxidase-like mechanism. Surprisingly, the PdFe/GDY nanozymes can detect GSH in bacteria with excellent specificity and ultra-low detection limit of 24.45 nM. Moreover, the PdFe/GDY nanozymes can consume GSH for bacterial disinfection with killing efficacy > 99.99%, promote wound healing and do not cause significant toxicity in vitro and in vivo. To our knowledge, this is the first example of a GDY-based nanozyme biosensor and antibacterial agent. Collectively, this work would be indicative to construct GDY-based nanozymes in clinical diagnosis and sterilization.

Abstract:Prussian blue nanoparticles (PBNPs) have been certified a kind of mimetic enzyme possesses the advantages of stability, high catalytic activity and low prices. Ferritins are natural nanoscale structures with unique three-dimensional structures and biological functions. In this context, we synthesized PBNPs on the surface of the iron oxide core of ferritin taking use of the ferric iron of the core, we also intended to retain the specificity of ferritin for some biological use. Our results show the resulting nanostructures (Prussian blue-ferritin nanoparticles, PB-Ft NPs) got very small size and relatively high catalytic activity , furthermore, PB-Ft NPs successfully combined the intrinsic enzyme mimetic activity of PBNPs and the specificity to tumor cells of ferritin. The peroxidase-like activity and catalase-like activity of PB-Ft NPs were studied. Peroxidase-like activity which fits well the Michaelis-Menten kinetics was found strongly depending on pH, temperature and the concentration of PB-Ft NPs. Then a sensitive method for glucose detection was developed using glucose oxidase (GOx) and PB-Ft NPs. PB-Ft NPs displayed catalase-like activity in PH higher than 5.0, the generated oxygen was measured by the dissolved oxygen electrode.Enzyme-linked immunosorbent assay (ELISA) shows PB-Ft NPs possess both specificity and peroxidase-like activity.

Abstract:In this work, a kind of biomimetic catalyst was designed as a signal probe by the assembly of porphyrin on porous carbon (PC) derived from metal-organic frameworks for electrochemical biosensing. First, PC was prepared by direct carbonization of a zeolite-type metal-organic framework as both a precursor and a template, and characterized by transmission electron microscopy, nitrogen sorption isotherm, X-ray photoelectron spectroscopy and X-ray diffraction. Then the PC was non-covalently functionalized with iron(III) meso-5,10,15,20-tetrakis(4-carboxyphenyl) porphyrin chloride (FeTCPP) via non-covalent interactions. The resulting nanocomposite of PC/FeTCPP possesses an excellent electrocatalytic activity toward oxygen reduction. Coupling with the enzymatic catalysis of glucose oxidase, a glucose biosensor was constructed on the basis of O-2 consumption. The biomimetic sensor enables a reliable and sensitive determination of glucose with a linear range of 0.5 to 18 mM and a detection limit of 0.08 mM at a signal-to-noise ratio of 3. Moreover, the biosensor exhibits the analytical reproducibility and stability with a lower relative standard deviation of 4.2%. In an application to detect glucose in human serum samples, this glucose biosensor had good detection accuracy with analytical recoveries from 97.3% to 107.7%. Therefore, the porphyrin functionalized PC provides a promising biomimetic platform for constructing the biosensors, and has potential application in bioanalysis and clinical diagnosis.
