Graphdiyne-supported palladium-iron nanosheets: A dual-functional peroxidase mimetic nanozyme for glutathione detection and antibacterial application

Abstract:Multifunctional enzymatic nanomaterials (nanozymes) with good biocompatibility, strong enzymatic activity, and substrate specificity are the desired alternatives to natural enzymes. Here, the design of a peroxidase mimetic nanozyme capable of detecting and depleting glutathione (GSH) in the bacterial cells is presented. The nanozyme palladium-iron nanostructure decorated graphdiyne nanosheet (PdFe/GDY) was prepared by a facile hydrothermal approach. It can efficiently catalyze the decomposition of H2O2 to produce •OH by a peroxidase-like mechanism. Surprisingly, the PdFe/GDY nanozymes can detect GSH in bacteria with excellent specificity and ultra-low detection limit of 24.45 nM. Moreover, the PdFe/GDY nanozymes can consume GSH for bacterial disinfection with killing efficacy > 99.99%, promote wound healing and do not cause significant toxicity in vitro and in vivo. To our knowledge, this is the first example of a GDY-based nanozyme biosensor and antibacterial agent. Collectively, this work would be indicative to construct GDY-based nanozymes in clinical diagnosis and sterilization.

The results show that the PdFe/GDY nanozyme can effectively induce the generation of •OH, which is mainly attributed to the strong interaction between the graphdiyne layer and the PdFe nanostructure, which promotes electron transfer and substrate adsorption. ### Key technical details - **Synthesis of nanozyme**: The PdFe/GDY nanocomposite was prepared by a simple hydrothermal method. The PdFe nano - alloy is evenly distributed on the graphdiyne nanosheets, forming a unique structure. - **Peroxidase - like activity**: The PdFe/GDY nanozyme can efficiently catalyze the oxidation of TMB, producing an obvious blue change, and the maximum absorption peak is located at 652 nm. Its catalytic performance is better than other reported peroxidase - mimicking nanozymes. - **GSH detection**: By adding different concentrations of GSH, it was observed that the blue color of the PdFe/GDY - TMB - H₂O₂ system gradually faded and the absorbance decreased. The linear range is from 50 to 10,000 nM, and the detection limit is 24.45 nM. - **Antibacterial effect**: The PdFe/GDY nanozyme can not only efficiently kill bacteria, but also promote wound healing, and no significant toxicity has been shown in both in vitro and in vivo experiments. - **Stability and sustainability**: The PdFe/GDY nanozyme still maintains about 90% of its initial activity after being exposed to air for 7 days, and the activity can still be maintained at about 90% after multiple cycles of use. ### Conclusion This study successfully developed a multi - functional nanozyme based on graphdiyne, which can not only detect GSH with high sensitivity in complex biological systems, but also achieve a highly efficient antibacterial effect by consuming GSH.
