Abstract:Peroxidase mimics made of inorganic nanomaterials as alternatives to natural peroxidases have been extensively developed over the past few decades, owing to their superior properties relative to their natural counterparts. Nevertheless, it has been still a challenge to substantially enhance the catalytic efficiency of peroxidase mimics. In this work, we report a type of highly efficient peroxidase mimics that are made of iridium nanoparticles (Ir NPs) with rough surfaces. The Ir NPs possess an ultrahigh catalytic efficiency with a catalytic constant (K-cat) at the regime of 10(10) s(-1), orders of magnitude higher than the K-cat of horseradish peroxidase as a natural peroxidase. As a proof-of-concept demonstration, the Ir NPs were applied to colorimetric lateral flow assay (CLFA). Using carcinoembryonic antigen as a model disease biomarker, the Ir NPs-based CLFA achieved a low limit of detection (LoD) of 39 pg/mL, which was similar to 28 times lower than the LoD of conventional gold nanoparticles-based CLFA using the same antibodies.

Abstract:The unique enzyme-like properties of metal nanoparticles (NPs) hold great promise for chemical and biomedical applications; implementation relies on improvements in their catalytic efficiency. We have constructed PtCuCo nanostructures using hydrothermal treatment of Co3O4, Cu2+, and Pt2+ in aqueous solutions, resulting in PtCuCo trimetallic alloy NPs having hollow structures, as confirmed by TEM, XRD, XPS, and EDS analyses. These PtCuCo NPs are capable of oxidizing 3,3',5,5'-tetramethyl-benzidine (TMB) in either the absence or presence of hydrogen peroxide, suggesting efficient peroxidase- and oxidase-like activity. These NPs could also catalyze the reduction of 4-nitrophenol by NaBH4. We believe that the combination of Pt and oxidized Cu and Co species results in surfaces with rich active sites possessing a wide range of electronic variation, which promotes both generation of hydroxyl radicals and electron transfer. Consequently, the catalytic capabilities of our PtCuCo NPs are greater than those of either Pt or PtCu NPs. Building upon the peroxidase-like activity of PtCuCo NPs, we developed a colorimetric method not only capable of specifically detecting glucose, but also able to detect rongalite and HS-, on the basis of the inhibitory effect of these substances upon PtCuCo nanoperoxidase.

Abstract:In this work, we report a peroxidase mimic composed of iridium (Ir) nanoparticles with an average size of only 1.1 nm that are coupled to a tungsten suboxide (WO2.72) nanorod as support. Such a peroxidase mimic exhibited an area-specific catalytic efficiency as high as 1.5 x 10(4) s(-1) nm(-2), outperforming most existing peroxidase mimics of noble metals. As proof-of-concept demonstrations, the peroxidase mimic was applied to colorimetric immunoassays of carcinoembryonic antigen (a cancer biomarker) and aflatoxin B1 (a potent carcinogen), which provided substantially enhanced detection sensitivities compared to conventional enzyme-based assays.

Abstract:Nanozymes with high catalytic efficiency have been widely investigated and studied in various fields. Compared with natural enzymes, nanozymes with enzyme-mimicking properties possess the advantages of low-cost, scalability, high stability, and tunable catalytic performance. In this paper, platinum nanoparticles loaded on oxygenated nanodiamonds (Pt-ONDs) were synthesised through a one-step method as a highly efficient nanozyme to mimic the function of peroxidase. Based on this, a novel sensing strategy was developed by combining Pt-ONDs and gold bipyramids (AuBPs) for sensitive detection of glucose. The detection limit of this method was established at 0.2 μM (a few thousand times before the lowest detection limit of commercial glucometers), with a wide detection range from 0.2 to 400 μM, highlighting its potential for ultra-high sensitive glucose detection. This study may provide a novel strategy for the application of nanozymes and gold nanostructures for the sensitive detection of glucose.

Abstract:Polyvinylpyrrolidone-functionalized IrPt alloy nanostructures (PVP/IrPt ANs) exhibited triple enzyme mimetic activities.Enzyme-like activities of PVP/IrPt ANs were employed for colorimetric and fluorescent detection of H2O2.PVP/IrPt ANs were coupled with the glucose oxidase for rapid, sensitive, and selective detection of glucose.The developed sensor demonstrated good selectivity and recovery of glucose from spiked biological and food samples.Nanomaterials that demonstrate multi-enzyme activities have increasingly gained interest due to their potential applications in point-of-care diagnostics. In the present work, polyvinylpyrrolidone-functionalized bimetallic IrPt alloy nanostructures (PVP/IrPt ANs) exhibited excellent peroxidase-, oxidase-, and catalase-like activities were synthesized with a reflux-assisted one-pot wet chemical process. The use of PVP as stabilizer and surfactant during the synthesis produced monodisperse nanoparticles (NPs) with average diameter of 4.1 nm. The PVP/IrPt ANs catalyze the oxidation of H2O2, TMB, and amplex red through the peroxidase and oxidase biomimetic activities. The kinetic parameters of PVP/IrPt ANs were measured to demonstrate their enhanced enzyme-like behavior relative to previously reported bimetallic nanozymes. The oxidase- and peroxidase-like activities of PVP/IrPt ANs were employed for efficient colorimetric and fluorescent detection of H2O2 concentrations. Moreover, the multi-enzyme activity of PVP/IrPt ANs was coupled with the glucose oxidase activity to fabricate colorimetric and fluorometric glucose sensors. The colorimetric glucose sensor revealed a broad detection range of 2.5 µM – 1.5 mM, while the fluorometric sensor showed excellent limit of detection (LOD) of 177.5 nM. Abstract:Au nanorods @ Pt nanodots core/shell nanostructures, prepared by the Au nanorods (NRs)-mediated growth, exhibit dual functional enzyme-like (peroxidase and oxidase-like) activities. From the viewpoint of enzyme mimics, the relationship between apparent enzyme kinetic parameters and nanomaterials structure is investigated in order to rationally design the catalytic activity. Using peroxidase-like properties of the Au@Pt NRs, the determination of hydrogen peroxide (H2O2) was demonstrated with a limit of detection (LOD) of 4.5×10−5M and a linear range of 4.5×10−5–1×10−3M using o-phenylenediamine (OPD) as chromogenic substrate. Furthermore, in combination with highly specific reactions provided by natural enzymes, selective detections of glucose and lipophilic cholesterol were demonstrated with similar LODs and linear ranges. Additionally, owing to the specific oxidase-like activity of the Au@Pt NRs (ascorbate oxidase), interference of ascorbic acid in the detection of glucose could be eliminated. In conclusion, considering the flexibility in the design of nanomatererials, there is a lot of space to improve their activity and explore their potential applications, especially in relatively harsh conditions.

Abstract:Glucose monitoring is essential to evaluate the degree of glucose metabolism disorders. The enzymatic determination has been the most widely used method in glucose detection because of its high efficiency, accuracy, and sensitivity. Noble metal nanomaterials (NMs, i.e., Au, Ag, Pt, and Pd), inheriting their excellent electronic, optical, and enzyme-like properties, are classified as noble metal nanozymes (NMNZs). As the NMNZs are often involved in two series of reactions, the oxidation of glucose and the chromogenic reaction of peroxide, here the chemical mechanism by employing NMNZs with glucose oxidase (GOx) and peroxidase (POD) mimicking activities is briefly summarized first. Subsequently, the regulation strategies of the GOx-like, POD-like and tandem enzyme-like activities of NMNZs are presented in detail, including the materials, size, morphology, composition, and the reaction condition of the representative NMs. In addition, in order to further mimic the enantioselectivity of enzyme, the design of NMNZs with enantioselective recognition of d-glucose and l-glucose by using different chiral compounds (DNA, amino acids, and cyclodextrins) and molecular imprinting is further described in this review. Finally, the feasible solutions to the existing challenges and a vision for future development possibilities are discussed.

Abstract:Abstract Au nanoparticles (NPs) have been found to be excellent glucose oxidase mimics, while the catalytic processes have rarely been studied. Here, we reveal that the process of glucose oxidation catalyzed by Au NPs is as the same as that of natural glucose oxidase, namely, a two-step reaction including the dehydrogenation of glucose and the subsequent reduction of O 2 to H 2 O 2 by two electrons. Pt, Pd, Ru, Rh, and Ir NPs can also catalyze the dehydrogenation of glucose, except that O 2 is preferably reduced to H 2 O. By the electron transfer feature of noble metal NPs, we overcame the limitation that H 2 O 2 must be produced in the traditional two-step glucose assay and realize the rapid colorimetric detections of glucose. Inspired by the electron transport pathway in the catalytic process of natural enzymes, noble metal NPs have also been found to mimic various enzymatic electron transfer reactions including cytochrome c, coenzymes as well as nitrobenzene reductions.

Abstract:A colorimetric assay based on an enzyme-inhibition strategy is promising for the on-site detection of pesticide residues. Due to the high cost and low stability of enzymes, nanozymes (nanomaterials with enzyme-like activities) are widely developed as substitutes of enzymes. However, the inhibition of pesticides toward enzymes and nanozymes generally lacks selectivity. It is of great significance and challenge to design a specific pesticide assay based on an activity-inhibition strategy. Here, we discovered that iridium nanoparticles possess both peroxidase-like and oxidase-like activities under the same conditions, and their catalytic mechanisms are different. The synergistic effect of dual enzyme-like activities enhanced the colorimetric signal. Interestingly, the dual enzyme-mimicking activities could be simultaneously inhibited, and the inhibition effect exhibited high selectivity toward malathion. Considering the popularity and the hazards of malathion, a malathion assay method based on activity inhibition was established without enzymes and a redundant process. The synergistic effect of the selective inhibition of dual enzyme-like activities enhanced the selectivity and sensitivity. Abstract:The development of novel catalyst with high catalytic activity is important for electrochemical non-enzymatic glucose sensing. Here, iridium single-atom/nickel oxide nanoparticle/N-doped graphene nanosheet (Ir 1 /NiO/NG) with the loading of 1.13 wt% Ir was successfully synthesized for constructing electrochemical non-enzymatic glucose sensor for the first time. The morphology and structure of Ir 1 /NiO/NG were characterized by XRD, SEM, TEM, HRTEM, and XPS, and the presence of Ir SAs was confirmed by AC-HAADF-STEM. The Ir 1 /NiO/NG shows 65 mV lower oxidation potential and 3.3 times higher response current than Ni(OH) 2 /NG. In addition, Ir 1 /NiO/NG exhibits high sensitivity (70.09 μA mM −1 cm −2 ), excellent selectivity, low detection limit (2.00 μM), and great stability (91.53% current remaining after 21 days) for electrochemical non-enzymatic glucose sensing. The outstanding catalytic and sensing performance of Ir 1 /NiO/NG is mainly attributed to synergistic effect of Ir SAs, NiO nanoparticles, and highly conductive NG, which modulate the electronic and geometric structure of Ir 1 /NiO/NG. This work shows the promising potential of SACs in electrochemical sensing.

Highly dispersed Pt nanoparticles on ultrasmall EMT zeolite: A peroxidase-mimic nanoenzyme for detection of H2O2 or glucose

Abstract:In past decade, Pt-based nanomaterials as peroxidase mimics have attracted much attention for H2O2 and glucose detection. However, easy aggregation of Pt nanoparticles (Pt NPs) greatly decreases their peroxidase-like activity. In this work, novel Pt/EMT nanocomposites were prepared by uniformly loading Pt NPs (5-8 nm) onto the support of ultrasmall EMT zeolite (15-20 nm), a kind of low-silica microporous aluminosilicate material. The hybrid Pt/EMT nanomaterials could be well dispersed in water to form a homogeneous suspension, and were then utilized as a superior peroxidase-like catalyst for oxidation of 3,3',5,5'-tetramethylbenzidine (TMB) in the presence of hydrogen peroxide (H2O2). The optimal catalyst of 2.6Pt/EMT nanocomposite exhibited excellent catalytic performance toward H2O2 and TMB than natural enzyme of horseradish peroxidase (HRP) by using a steady-state kinetic analysis based on the typical Michaelis-Menten kinetics theory. The peroxidase-like catalyst showed a promising activity in a wide pH and temperature range as well as the long-term stability. A facile and reliable colorimetric assay based on the peroxidase mimic of Pt/EMT nanocomposite was constructed for precise detection of H2O2 and glucose in a wide linear range, with low limits of detection of 1.1 μM and 13.2 μM, respectively. Due to high selectivity to glucose against other sugars on the catalyst, the method was demonstrated to accurately measure the concentration of glucose in real samples including human blood serum and fruit juices, indicating a potential application of the Pt/EMT nanocomposites as a robust peroxidase mimic and a reliable biosensor in the fields of clinical diagnosis, pharmaceutical, food research and so on.

Abstract:Nanomaterials with enzyme-mimicking characteristics have engaged great awareness in various fields owing to their comparative low cost, high stability, and large-scale preparation. However, the wide application of nanozymes is seriously restricted by the relatively low catalytic activity and poor specificity, primarily because of the inhomogeneous catalytic sites and unclear catalytic mechanisms. Herein, a support-sacrificed strategy is demonstrated to prepare a single iron site nanozyme (Fe SSN) dispersed on the porous N-doped carbon. With well-defined coordination structure and high density of active sites, the Fe SSN performs prominent peroxidase-like activity by efficiently activating H2 O2 into hydroxyl radical (•OH) species. Furthermore, the Fe SSN is applied in colorimetric detection of glucose through a multienzyme biocatalytic cascade platform. Moreover, a low-cost integrated agarose-based hydrogel colorimetric biosensor is designed and successfully achieves the visualization evaluation and quantitative detection of glucose. This work expands the application of single-site catalysts in the fields of nanozyme-based biosensors and personal biomedical diagnosis.

Abstract:Nanozyme is a promising field that offers the substitution for natural enzymes using various nanomaterials. Various nanomaterials with peroxidase-like activity were investigated. Among them, transition metal chalcogenides were explored as promising nanozymes due to their excellent enzyme-mimicking activities. However, ruthenium selenide has not been studied as a peroxidase mimic because of the difficulty for synthesis. Herein, we prepared ruthenium selenide nanomaterial with ordered mesoporous structure (P-RuSe2) employing KIT-6 silica as the template. The composition and structure of P-RuSe2 were fully characterized. Further, its peroxidase-like activity was investigated. P-RuSe2 possessed excellent peroxidase-mimicking activity, which catalyzed the oxidation of peroxidase substrates, including 3,3′,5,5′-tetramethylbenzidine, o-phenylenediamine, and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) in the presence of H2O2. Moreover, P-RuSe2 exhibited higher peroxidase-like activity when compared with several representative nanozymes as well as bulk RuSe2. To demonstrate its potential applications, the colorimetric detection systems for H2O2 and glucose were successfully constructed based on P-RuSe2 nanozyme.

Abstract:In this work, we report the synthesis of Cu-Ag bimetallic nanopartiles and g-C3N4 nanosheets decorated on zeolitic imidazolate framework-8 (ZIF-8) to form a Cu-Ag/g-C3N4/ZIF hybrid. The hybrid was synthesized and characterized by Transmission electron microscopy (TEM), Fourier transformed infrared (FTIR), the X-ray diffraction (XRD) and X-ray photoelectron spectroscopy (XPS). The Cu-Ag/g-C3N4/ZIF hybrid has intrinsic peroxidaselike catalytic activity towards the oxidation of TMB in the presence of H2O2. The situ synthesis of Cu-Ag bimetallic nanopartiles on 2D support such as g-C3N4 nanosheets would significantly enhance the peroxidaselike catalytic properties of individual Cu-Ag bimetallic nanopartiles and the g-C3N4 nanosheets. After loading of Cu-Ag bimetallic nanopartiles and g-C3N4 nanosheets on the ZIF-8, the hybrids exhibited superior peroxidaselike catalytic activity and good recyclability. Then, this method was applied for detecting glucose in human serum, owing the significant potential for detection of metabolites with H2O2-generation reactions.

Herein, by using benzoic acid as capping agent, we fabricated uniform PCN-224 nanoparticles (NPs), which were composed of Zr6 cluster and tetrakis(4-carboxyphenyl)porphyrin (H2TCPP). Then Fe(Ⅲ) ions were incorporated into the center of H2TCPP producing iron porphyrin unit similar to hemin serving as catalytic active site. The procedure of iron incorporation didn't change the size and morphology of PCN-224 NPs. The newly formed Fe@PCN-224 NPs possessed peroxidase-like activity with much lower Km values and higher Kcat values than most of peroxidase-like nanozymes as well as natural horseradish peroxidase (HRP), indicating their enhanced catalytic activity. Taking advantage of the excellent peroxidase-like activity of Fe@PCN-224 NPs, we successfully developed a colorimetric method for simple and sensitive detection of hydrogen peroxide and glucose in combination with glucose oxidase.
Fe3o4 Magnetic Nanoparticles As Peroxidase Mimetics and Their Applications in H2o2 and Glucose Detection

Abstract:Artificial enzyme mimetics are a current research interest because natural enzymes bear some serious disadvantages, such as their catalytic activity can be easily inhibited and they can be digested by proteases. A very recently study reported by Yan et al. has proven that Fe(3)O(4) magnetic nanoparticles (MNPs) exhibit an intrinsic enzyme mimetic activity similar to that found in natural peroxidases, though MNPs are usually thought to be biological and chemical inert (Gao, L. Z.; Zhuang, J.; Nie, L.; Zhang, J. B.; Zhang, Y.; Gu, N.; Wang, T. H.; Feng, J.; Yang, D. L.; Perrett, S.; Yan, X. Y. Nat. Nanotechnol. 2007, 2, 577-583). In the present work, we just make use of the novel properties of Fe(3)O(4) MNPs as peroxidase mimetics reported by Yan et al. to detect H(2)O(2). The Fe(3)O(4) MNPs were prepared via a coprecipitation method. The as-prepared Fe(3)O(4) MNPs were then used to catalyze the oxidation of a peroxidase substrate 2,2'-azino-bis(3-ethylbenzo-thiazoline-6-sulfonic acid) diammonium salt (ABTS) by H(2)O(2) to the oxidized colored product (see eq 1) which provides a colorimetric detection of H(2)O(2). As low as 3 x 10(-6) mol/L H(2)O(2) could be detected with a linear range from 5 x 10(-6) to 1 x 10(-4) mol/L via our method. More importantly, a sensitive and selective method for glucose detection was developed using glucose oxidase (GOx) and the as-prepared Fe(3)O(4) MNPs. The detection platforms for H(2)O(2) and glucose developed in the present work not only further confirmed that the Fe(3)O(4) MNPs possess intrinsic peroxidase-like activity but also showed great potential applications in varieties of simple, robust, and easy-to-make analytical approaches in the future.

Abstract:Due to possessing an extremely small size and a large surface area per unit of volume, nanomaterials have specific characteristic physical, chemical, photochemical, and biological properties that are very useful in many new applications. Nanoparticles' catalytic activity and intrinsic ability in generating or scavenging reactive oxygen species in general can be used to mimic the catalytic activity of natural enzymes. Many nanoparticles with enzyme-like activities have been found, potentially capable of being applied for commercial uses, such as in biosensors, pharmaceutical processes, and the food industry. To date, a variety of nanoparticles, especially those formed from noble metals, have been determined to possess oxidase-like, peroxidase-like, catalase-like, and/or superoxide dismutase-like activity. The ability of nanoparticles to mimic enzymatic activity, especially peroxidase mimics, can be used in a variety of applications, such as detection of glucose in biological samples and waste water treatment. To study the enzyme-like activity of nanoparticles, the electron spin resonance method represents a critically important and convenient analytical approach for zero-time detection of the reactive substrates and products as well as for mechanism determination.

Abstract:Gold nanoclusters (AuNCs) represent an emerging type of engineered nanomaterials with intrinsic enzymatic activity for both chemical and biological applications, but the catalytic activity of most reported AuNCs remains rather limited. Herein, we report a new, efficient strategy of promoting the peroxidase-mimic activity of AuNCs by tailoring their catalytic interfaces via small molecule-mediated weak interactions. Inspired by the presence of imidazole structures in many biocatalytic centers, we screened a series of imidazole-containing small molecules to evaluate their impact on the enzymatic activity of AuNCs. Through monitoring the absorbance change of 3,3',5,5'-tetramethylbenzidine, 1-methyl-2-imidazolecarboxaldehyde (MCA) was identified to possess the most significant effect on enhancing the peroxidase-mimic activity of glutathione-stabilized AuNCs (GSH-AuNCs) among all the examined molecules. Interestingly, the enhancement effect of MCA on the catalytic activity of these AuNCs was found to be highly reversible and can be switched on/off by simply adding MCA/dialysis treatment. Molecular dynamics simulations and further experimental analysis confirmed that these MCA molecules were adsorbed on the surface of GSH-AuNCs through weak non-covalent interactions. The underlying mechanism analysis suggested that the presence of MCA can efficiently promote the production of •OH in the GSH-AuNC system. As a proof of example, we then demonstrated that the presence of MCA can greatly increase the bioanalytical performance of AuNC-based peroxidase mimics, as evidenced by a 65-fold lower LOD for glucose detection of AuNCs@MCA than that using AuNCs only. Finally, the present system has been successfully applied for sensing the blood glucose level of both healthy people and diabetics with promising results.
