Abstract:The unique enzyme-like properties of metal nanoparticles (NPs) hold great promise for chemical and biomedical applications; implementation relies on improvements in their catalytic efficiency. We have constructed PtCuCo nanostructures using hydrothermal treatment of Co3O4, Cu2+, and Pt2+ in aqueous solutions, resulting in PtCuCo trimetallic alloy NPs having hollow structures, as confirmed by TEM, XRD, XPS, and EDS analyses. These PtCuCo NPs are capable of oxidizing 3,3',5,5'-tetramethyl-benzidine (TMB) in either the absence or presence of hydrogen peroxide, suggesting efficient peroxidase- and oxidase-like activity. These NPs could also catalyze the reduction of 4-nitrophenol by NaBH4. We believe that the combination of Pt and oxidized Cu and Co species results in surfaces with rich active sites possessing a wide range of electronic variation, which promotes both generation of hydroxyl radicals and electron transfer. Consequently, the catalytic capabilities of our PtCuCo NPs are greater than those of either Pt or PtCu NPs. Building upon the peroxidase-like activity of PtCuCo NPs, we developed a colorimetric method not only capable of specifically detecting glucose, but also able to detect rongalite and HS-, on the basis of the inhibitory effect of these substances upon PtCuCo nanoperoxidase.

Abstract:In this study, Cu-Cu2O/PtPd nanocomposites were synthesized and characterized for their peroxidase-like enzyme activity. X-ray diffraction and energy dispersive X-ray spectroscopy analyses confirmed the successful synthesis of the nanocomposites, which exhibited a flower-like morphology and a more uniform dispersion than Cu-Cu2O. The catalytic activity of Cu-Cu2O/PtPd was evaluated using the chromogenic substrate 3,3',5,5'-tetramethylbenzidine (TMB), finding that Cu-Cu2O/PtPd outperformed Cu-Cu2O. The optimal temperature and pH for the catalytic activity of Cu-Cu2O/PtPd were determined to be 40 °C and pH 4.0, respectively. A kinetic analysis revealed that Cu-Cu2O/PtPd followed Michaelis-Menten kinetics and exhibited a higher affinity toward TMB than the horseradish peroxidase enzyme. The catalytic mechanism of Cu-Cu2O/PtPd involved the generation of hydroxyl radicals, which facilitated the oxidation of TMB. Furthermore, the Cu-Cu2O/PtPd nanocomposite was successfully applied for the colorimetric detection of glucose, demonstrating a linear range of 8-90 μM, a detection limit of 2.389 μM, and high selectivity for glucose over other sugars.

Abstract:A hybrid of metal-organic frameworks (Fe, Mn) and Au nanoparticles anchored carbon nanotubes (Au/MOFs(Fe, Mn)/CNTs) was fabricated by a facile one-step hydrothermal process. The Au/MOFs(Fe, Mn)/CNTs exhibit highly enhanced peroxidase-like activity, because of the increased active sites, the enhanced partial charge density and electron transfer among the Fermi level of MOFs, Au and CNTs, as well as the synergistic action of Fe and Mn. The hybrid nanomaterials with high catalytic velocity (Vmax = 17.65 × 10-7 M s-1) and affinity (Km = 0.33 mM) for substrates of H2O2 display a great detection performance in the range of 0.34-53.05 nM with a limit of detection (LOD) of 0.18 nM (S/N = 3). Based on the cascade reaction of the artificial peroxidase and glucose oxidase (GOx), the glucose was further better detected in a linear range of 0.005-0.3 μM with a LOD of 0.002 μM (S/N = 3). Notably, considering the regulated peroxidase-like activity by sulfadimethoxine (SDM) aptamer, the concentration of SDM ranging from 0.54 to 41.58 μg L-1 was also detected with a LOD of 0.35 μg L-1 (S/N = 3). The as-synthesized hybrid nanozymes hold promising applications as the multifunctional sensing platform in environmental and biologic target detection with the combination of corresponding enzyme or specific aptamer.

Abstract:A 3D hollow CoZn-LDH@CuO nanosheet array was prepared in situ by using CoZn-ZIF skeleton grown on Cu(OH) 2 nanorods with copper foam as precursor. The catalyst materials are characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM) and energy dispersive X-ray photoelectron spectroscopy (XPS). The synergistic effect between the three metals and the hollow layered nanosheet array structure make the catalyst have larger specific surface area and more exposed active sites, which can effectively improve the electron transfer rate. Using CoZn-LDH@CuO NSA/CF as the working electrode, glucose and H 2 O 2 can be detected with sensitivity of 11,200 (μA mM −1 cm −2 ) and 4585 (μA mM −1 cm), respectively. The linear ranges were 2 μM − 3 mM and 0.8 μM − 3.5 mM and the detection limits is 0.17 μM (S/N = 3). The sensor had excellent stability, selectivity, fast response time (≤3 s), and the recoveries in the actual sample detection is 99.6 %-103.1 %. This strategy provides a new idea for the design and preparation of catalytic electrodes in the field of electrochemical sensing.

Abstract:Herein, glutathione-capped copper nanoclusters (CuNCs) and graphitic carbon nitride nanosheets (g-C3N4 NSs) were synthesized by a facile one-pot chemical reduction and directly thermal pyrolysis following ultrasonic exfoliation approaches, respectively. The introduction of Ce(III) (Ce3+) played dual functions in constructing a fluorescence-enhanced ratiometric nanoprobe (g-C3N4 NSs-Ce3+-CuNCs), i.e., triggering aggregation-induced emission of CuNCs and conjugating g-C3N4 NSs with CuNCs by virtue of electrostatic and coordination interactions. The as-fabricated nanohybrid displayed 460 and 625 nm dual-emitting peaks, attributing to the emission of g-C3N4 NSs and CuNCs, respectively. Upon addition of H2O2, the 625 nm emission was dramatically quenched, whereas the 460 nm emission remained nearly unchanged, thereby causing obvious color changes from purple to blue under a 365-nm UV lamp. A ratiometric fluorescent assay, based on g-C3N4 NSs-Ce3+-CuNCs, was devised for sensitive and visual detection of H2O2, which spanned the linear range of 2–100 μM with a detection limit of 0.6 μM. In the presence of glucose oxidase, the ratiometric nanoprobe could be simultaneously employed to detect glucose across the linear range of 1.6–320 μM with a detection limit of 0.48 μM. In milk and human serum samples, the fortified recoveries for H2O2 and glucose by the nanoprobe were in the range of 95.5–103.6% with RSDs <3.8%. The real detection levels for glucose are consistent with those by a standard glucometer. As such, the ratiometric nanoprobe offers a promising methodology for several practical applications, such as point-of-care diagnosis and workplace health evaluations.

Abstract:Bimetallic copper oxide-gold nanoalloys (CuO-Au nanoalloys) have been prepared and characterized using various techniques confirming their successful synthesis. The peroxidase-like activity of CuO-Au nanoalloys have been investigated and applied towards the colorimetric detection of glucose. CuO-Au nanoalloys as nanoenzymes exhibited peroxidase-like properties which occurred by generating the reactive oxygen species (ROS) due to the catalytic properties of CuO-Au nanoalloys.The generation of ROS was confirmed using radical and/or reactive oxygen species quenching properties of 1,3‑diphenylisobenzofuran (DPBF). The rate of production of ROS was higher when CuO-Au nanoalloys were present in a solution containing DPBF and H2O2 (0.1092 min-1). The absorption peak of DPBF at 430 nm decreased confirming the generation of ROS. The CuO-Au nanoalloys were evaluated their potential use for colorimetric detection of glucose. The detection limit for glucose was obtained to be 6.75 μM. The detection of glucose took place in two steps and at different pH conditions, first glucose oxidation by glucose oxidase occurred in physiological pH (7.4) at 37 °C. The optimum conditions for the CuO-Au nanoalloys peroxidase-like activity were obtained at pH 4 and 25 °C. The chromogen, TMB (3,3',5,5'‑tetramethylbenzidine) oxidation to blue coloured product TMBDI (3,5,3',5'‑tetrabenzidinediimine) occurred with high intensity obtained after 6 min.
Facile Synthesis of Ag@Cu2O Heterogeneous Nanocrystals Decorated N-doped Reduced Graphene Oxide with Enhanced Electrocatalytic Activity for Ultrasensitive Detection of H2O2

Abstract:A nanohybrid composed of Ag@Cu2O heterogeneous nanocrystals supported on N-doped reduced graphene oxide (Ag@Cu2O/N-RGO) has been synthesized by a simple wet-chemical method. The resultant composite consists of N-RGO sheets fully and homogeneously coated with a dense layer of Ag@Cu2O nanocrystals. Both Ag and N-RGO are in direct contact with Cu2O, and Ag nanoparticles with sizes of 2-5 nm are mainly deposited on the surface of Cu2O cubes (edge length of 500 nm). The electrochemical studies reveal that the ternary Ag@Cu2O/N-RGO composite exhibit significantly enhanced electrocatalytic activity for H2O2 sensing compared with either the single component (N-RGO) or two component systems (Cu2O/N-RGO and Ag/N-RGO), which is mainly due to the synergetic catalysis of the ternary system. The nonenzymatic sensor based on Ag@Cu2O/N-RGO composite shows overwhelmingly superior comprehensive performance for the H2O2 detection over the documented Ag-based sensors. More specifically, it displays a rapid response (10 s) to H2O2 over a wide linear range of 54-700 nM with a high sensitivity of 1298.3 mu A mM(-1) cm(-2) and a low detection limit of 10 nM. Moreover, the sensor also exhibits the preferable selectivity in the presence of biologically coactive compounds accompanied with long-term stability and good reproducibility. (C) 2018 Elsevier B.V. Abstract:Due to the unique advantages, developing a rapid, simple and economical synthetic strategy for porous nanomaterials is of great interest. In this work, for the first time, using sodium hypochlorite as a green oxidant, urea was oxidized to CO2 as a carbon source to prepare the fine-particle crosslinked Cu-precursors, which could be further reduced by sodium ascorbate into pure Cu2O nanospheres (NPs) with a porous morphology at room temperature. Interestingly, our study reveals that introduction of an appropriate amount of MgCl2 into the raw materials can tune the pore sizes and surface area, but has no influence on the phase purity of the resulting Cu2O NPs. Significantly, all the synthesized Cu2O NPs exhibited intrinsic peroxidase-like activity with higher affinity towards both 3,3,5,5-tetramethylbenzidine (TMB) and H2O2 than horseradish peroxidase (HRP) due to the highly porous morphology and the electrostatic attraction towards TMB. The colorimetric detection of glucose based on the resulting porous Cu2O NPs presented a limit of detection (LOD) of 2.19 μM with a broad linear range from 1-1000 μM, much better than many recently reported composite-based nanozymes. Meanwhile, this nanozyme system was utilized to detect l-cysteine, exhibiting a LOD value as low as 0.81 μM within a linear range from 0 to 10 μM. More interesting, this sensing system shows high sensitivity and excellent selectivity in determining glucose and l-cysteine, which is suitable for detecting serum samples with reliable results. Therefore, the present study not only develops a simple strategy to prepare Cu2O NPs with controllable porous structure, but also indicates its promising applications in bioscience and disease diagnosis.

Abstract:In past decade, Pt-based nanomaterials as peroxidase mimics have attracted much attention for H2O2 and glucose detection. However, easy aggregation of Pt nanoparticles (Pt NPs) greatly decreases their peroxidase-like activity. In this work, novel Pt/EMT nanocomposites were prepared by uniformly loading Pt NPs (5-8 nm) onto the support of ultrasmall EMT zeolite (15-20 nm), a kind of low-silica microporous aluminosilicate material. The hybrid Pt/EMT nanomaterials could be well dispersed in water to form a homogeneous suspension, and were then utilized as a superior peroxidase-like catalyst for oxidation of 3,3',5,5'-tetramethylbenzidine (TMB) in the presence of hydrogen peroxide (H2O2). The optimal catalyst of 2.6Pt/EMT nanocomposite exhibited excellent catalytic performance toward H2O2 and TMB than natural enzyme of horseradish peroxidase (HRP) by using a steady-state kinetic analysis based on the typical Michaelis-Menten kinetics theory. The peroxidase-like catalyst showed a promising activity in a wide pH and temperature range as well as the long-term stability. A facile and reliable colorimetric assay based on the peroxidase mimic of Pt/EMT nanocomposite was constructed for precise detection of H2O2 and glucose in a wide linear range, with low limits of detection of 1.1 μM and 13.2 μM, respectively. Due to high selectivity to glucose against other sugars on the catalyst, the method was demonstrated to accurately measure the concentration of glucose in real samples including human blood serum and fruit juices, indicating a potential application of the Pt/EMT nanocomposites as a robust peroxidase mimic and a reliable biosensor in the fields of clinical diagnosis, pharmaceutical, food research and so on.

Abstract:Multifunctional catalytic performance plays a crucial role in bio-applications through the diversity and durability of artificial nanozymes. An effective synergy with sufficient accessible active sites and high specific surface area is a challenge for composite catalysts, especially to avoid uncontrollable aggregation and structural instability. Here, we fabricated a Cu n O/Au heterostructure dendrimer on copper foam (Cu n O/Au HD/CF) as dual functional catalytic nanozyme to achieve enzyme mimic cascade reactions for efficient colorimetric analysis. A highly porous CF skeleton–based CuO nanowire array (CuO NWA) with a large specific surface area supported an efficient load capacity to assemble sufficient Cu n O/Au HD by electrodeposition. The bimetallic Au-Cu nanozyme successfully achieved an oxidase-like and peroxidase-like cascade catalysis by a target-responsive sensing mechanism. Due to the confirmed catalytic performance of selectivity, anti-interference ability, and reproducibility, a Cu n O/Au HD/CF-based quantitative analytical method was developed for glucose detection with a wide linear range and considerable detection limit of 8.4 μM. The robust nonenzymatic catalytic strategy for colorimetric detection not only confirmed the dual functional catalytic activity of Cu n O/Au HD/CF, but also showed great potential for applications in clinical diagnostics and biochemical analysis.

Abstract:Nanosized materials acting as substitutes of natural enzymes are currently attracting significant research due to their stable enzyme-like characteristics, but some flaws of these nanozymes, including their limited catalytic rate and efficiency, need to be remedied to enable their wider applications. In this work, we verify for the first time the catalytic behavior of uncapped nanobranch-based CuS clews as a peroxidase mimic. XRD, XPS, SEM, and TEM proofs demonstrate that high-purity CuS clews composed of intertwined wires with abundant nanodendrites outside are successfully produced via a facile one-pot hydrothermal synthesis approach, with thiourea as both the sulfion source and the structure-directing agent. The synthesized CuS can catalytically oxidize 3,3',5,5'-tetramethylbenzidine (TMB) by H2O2 to trigger a visible color reaction with rapid response (reaching a maximum change within 5 min). The proposed CuS nanozyme exhibits preferable catalytic kinetics over natural horseradish peroxidase (HRP). This outstanding activity primarily results from the large surface area and rich sites exposed by the uncapped unique structure. Under optimized conditions, the fabricated sensing system provides linear absorbance (652 nm) changes in the H2O2 concentration range of 0.2˜130 μM, with a detection limit of as low as 63 nM. When coupled with glucose oxidase (GOD), the system is demonstrated to be capable of monitoring glucose in blood samples with excellent performance.
Co₃O₄/Au Hybrid Nanostructures as Efficient Peroxidase Mimics for Colorimetric Biosensing

Abstract:Nanomaterials with enzyme-like characteristics (nanozymes) have emerged as potential replacements for natural enzymes due to their potential to overcome several critical limitations of natural enzymes, including low stability as well as high costs in preparation and purification. Herein, we have developed hybrid nanostructures that incorporate cobalt oxide nanoparticles (Co₃O₄ NPs) and gold nanoclusters (AuNCs) through electrostatic attraction induced by simple incubation in an aqueous buffer for 2 hours. Owing to the synergistic effect of Co₃O₄ NPs and AuNCs, the constructed Co₃O₄/Au hybrid nanostructures yielded highly enhanced peroxidase-like activity and enabled rapid catalytic oxidation of a chromogenic substrate, 3,3',5,5'-tetramethylbenzidine (TMB), producing a blue colored solution depending on the amount of H₂O₂. Moreover, we observed catalytic activity of the Co₃O₄/Au hybrid over a broad pH range, especially at physiologically relevant pH in the range of 5.0-7.4, which is advantageous for applications in biological systems. Using the hybrid as peroxidase mimic, we successfully determined the level of target H₂O₂ or glucose by coupling with glucose oxidase with excellent specificity and sensitivity. Based on this study, we expect that Co₃O₄/Au hybrid nanostructures can serve as potent peroxidase mimics for the detection of clinically important target molecules.

Abstract:Nanozyme possess the advantages of high enzyme simulation activity, low cost, and excellent stability. As a substitute for natural enzymes, nanozyme have demonstrated significant potential in various fields. In this study, a bimetallic organometallic framework Cu@Co-MOFs was synthesized via the hydrothermal method using terephthalic acid (H2BDC) as a ligand and characterized. Cu@Co-MOFs exhibits outstanding peroxidase activity (POD) by catalyzing the oxidation of 3,3 ',5,5 '-tetramethylbenzidine (TMB) and other color-developing substrates through center dot OH formation. A simple, rapid, and reliable colorimetric method for quantitatively detecting glutathione (GSH) was developed based on the Pod-like activity of Cu@Co-MOFs. The linear range of this method was 1-1200 mu M with a detection limit of 0.72 mu M. This work not only proposes a bimetallic organic framework with high POD as a straightforward and practical platform for visually detecting GSH but also provides a strategy to enhance the performance of nanomaterials.
Peroxidase-Like Activity Of Water-Soluble Cupric Oxide Nanoparticles And Its Analytical Application For Detection Of Hydrogen Peroxide And Glucose

Abstract:Water-soluble cupric oxide nanoparticles are fabricated via a quick-precipitation method and used as peroxidase mimetics for ultrasensitive detection of hydrogen peroxide and glucose. The water-soluble CuO nanoparticles show much higher catalytic activity than that of commercial CuO nanoparticles due to their higher affinity to hydrogen peroxide. In addition, the as-prepared CuO nanoparticles are stable over a wide range of pH and temperature. This excellent stability in the form of aqueous colloidal suspensions makes the application of the water-soluble CuO nanoparticles easier in aqueous systems. A colorimetric assay for hydrogen peroxide and glucose has been established based on the catalytic oxidation of phenol coupled with 4-amino-atipyrine by the action of hydrogen peroxide. This analytical platform not only confirms the intrinsic peroxidase-like activity of the water-soluble cupric oxide nanoparticles, but also shows its great potential applications in environmental chemistry, biotechnology and medicine.
Enhanced peroxidase-like activity of Fe@PCN-224 nanoparticles and their applications for detection of H2O2and glucose

Abstract:Inspired by natural hemin structure, the fabrication of metal organic framework (MOF) nanomaterials constructed by metal ions and iron porphyrin for peroxidase mimicking has attracted extensive attention recently due to their high concentration of active sites and great channeling for substrate diffusion. Herein, by using benzoic acid as capping agent, we fabricated uniform PCN-224 nanoparticles (NPs), which were composed of Zr6 cluster and tetrakis(4-carboxyphenyl)porphyrin (H2TCPP). Then Fe(Ⅲ) ions were incorporated into the center of H2TCPP producing iron porphyrin unit similar to hemin serving as catalytic active site. The procedure of iron incorporation didn't change the size and morphology of PCN-224 NPs. The newly formed Fe@PCN-224 NPs possessed peroxidase-like activity with much lower Km values and higher Kcat values than most of peroxidase-like nanozymes as well as natural horseradish peroxidase (HRP), indicating their enhanced catalytic activity. Taking advantage of the excellent peroxidase-like activity of Fe@PCN-224 NPs, we successfully developed a colorimetric method for simple and sensitive detection of hydrogen peroxide and glucose in combination with glucose oxidase.

Abstract:N, N'-di (L-glutamic amine)-perylene-3,4,9,10-tetracarboxylic diimide (GAPDI) functionalized Sb2S3 nanocomposites (GAPDI-Sb2S3) were fabricated using a hydrothermal method and characterized. GAPDI-Sb2S3 nanocomposites can catalyze the oxidation of 3,3 ',5,5 '- tetramethyl benzidine (TMB) by H2O2. The experimental conditions including pH and temperature were optimized. GAPDI-Sb2S3 nanocomposites showed a strong affinity for TMB substrates, and their catalytic activity followed the Michaelis-Menten kinetics. The experimental results showed that GAPDI-Sb2S3 nanocomposites exhibited a low detection limit (LOD = 0.39 mu M) and high sensitivity for H2O2 detection and showed good response to glucose detection with a LOD of 0.42 mu M over arrange of 2-10 mu M. These findings suggest broad application prospects in the fields of environmental chemistry, biotechnology, and biomedicine.
