Abstract:Scalable computational modelling tools are required to guide the rational design of complex hierarchical materials with predictable functions. Here, we utilize mesoscopic modelling, integrated with genetic block copolymer synthesis and bioinspired spinning process, to demonstrate de novo materials design that incorporates chemistry, processing and material characterization. We find that intermediate hydrophobic/hydrophilic block ratios observed in natural spider silks and longer chain lengths lead to outstanding silk fibre formation. This design by nature is based on the optimal combination of protein solubility, self-assembled aggregate size and polymer network topology. The original homogeneous network structure becomes heterogeneous after spinning, enhancing the anisotropic network connectivity along the shear flow direction. Extending beyond the classical polymer theory, with insights from the percolation network model, we illustrate the direct proportionality between network conductance and fibre Young's modulus. This integrated approach provides a general path towards de novo functional network materials with enhanced mechanical properties and beyond (optical, electrical or thermal) as we have experimentally verified.

Abstract:Spider silk is a tough and versatile biological material combining high tensile strength and extensibility through nanocomposite structure and its nonlinear elastic behaviour. Notably, spiders rarely use single silk fibres in isolation, but instead process them into more complex composites, such as silk fibre bundles, sheets and anchorages, involving a combination of spinneret, leg and body movements. While the material properties of single silk fibres have been extensively studied, the mechanical properties of silk composites and meta-structures are poorly understood and exhibit a hereto largely untapped potential for the bio-inspired design of novel fabrics with outstanding mechanical properties. In this study, we report on the tensile mechanics of the adhesive capture threads of the Southern house spider (Kukulcania hibernalis), which exhibit extreme extensibility, surpassing that of the viscid capture threads of orb weavers by up to tenfold. By combining high-resolution mechanical testing, microscopy and in silico experiments based on a hierarchical modified version of the Fibre Bundle Model, we demonstrate that extreme extensibility is based on a hierarchical loops-on-loops structure combining linear and coiled elements. The stepwise unravelling of the loops leads to the repeated fracture of the connected linear fibres, delaying terminal failure and enhancing energy absorption. This principle could be used to achieve tailored fabrics and materials that are able to sustain high deformation without failure.
Recombinant Spidroins Fully Replicate Primary Mechanical Properties of Natural Spider Silk.

Abstract:Despite significant efforts to engineer their heterologous production, recombinant spider silk proteins (spidroins) have yet to replicate the unparalleled combination of high strength and toughness exhibited by natural spider silks, preventing their use in numerous mechanically demanding applications. To overcome this long-standing challenge, we have developed a synthetic biology approach combining standardized DNA part assembly and split intein-mediated ligation to produce recombinant spidroins of previously unobtainable size (556 kDa), containing 192 repeat motifs of the Nephila clavipes dragline spidroin. Fibers spun from our synthetic spidroins are the first to fully replicate the mechanical performance of their natural counterparts by all common metrics, i.e., tensile strength (1.03 +/- 0.11 GPa), modulus (13.7 +/- 3.0 GPa), extensibility (18 +/- 6%), and toughness (114 +/- 51 MJ/m(3)). The developed process reveals a path to more dependable production of high-performance silks for mechanically demanding applications while also providing a platform to facilitate production of other high-performance natural materials.
Strain softening and stiffening responses of spider silk fibers probed using Micro-Extension Rheometer

Abstract:Spider silk possesses unique mechanical properties like large extensibility, high tensile strength, super-contractility, etc. Understanding these mechanical responses require characterization of the rheological properties of silk beyond the simple force-extension relations which are widely reported. Here we study the linear and non-linear viscoelastic properties of dragline silk obtained from social spiders Stegodyphus sarasinorum using a Micro-Extension Rheometer that we have developed. Unlike continuous extension data, our technique allows for the probing of the viscoelastic response by applying small perturbations about sequentially increasing steady-state strain values. In addition, we extend our analysis to obtain the characteristic stress relaxation times and the frequency responses of the viscous and elastic moduli. Using these methods, we show that in a small strain regime (0-4%) dragline silk of social spiders shows strain-softening response followed by strain-stiffening response at higher strains (> 4%). The stress relaxation time, on the other hand, increases monotonically with increasing strain for the entire range. We also show that silk stiffens while ageing within the typical lifetime of a web. Our results demand the inclusion of the kinetics of domain unfolding and refolding in the existing models to account for the relaxation time behaviour.

Abstract:Spider silk is a remarkable example of bio-material with superior mechanical characteristics. Its multilevel structural organization of dragline and viscid silk leads to unusual and tunable properties, extensively studied from a quasi-static point of view. In this study, inspired by the Nephila spider orb web architecture, we propose a novel design for mechanical metamaterials based on its periodic repetition. We demonstrate that spider-web metamaterial structure plays an important role in the dynamic response and wave attenuation mechanisms. The capability of the resulting structure to inhibit elastic wave propagation in sub-wavelength frequency ranges is assessed and parametric studies are performed to derive optimal configurations and constituent mechanical properties. The results show promise for the design of innovative lightweight structures for tunable vibration damping and impact protection, or the protection of large scale infrastructure such as suspended bridges.

Increasing silk fibre strength through heterogeneity of bundled fibrils

Abstract:Can naturally arising disorder in biological materials be beneficial? Materials scientists are continuously attempting to replicate the exemplary performance of materials such as spider silk, with detailed techniques and assembly procedures. At the same time, a spider does not precisely machine silk-imaging indicates that its fibrils are heterogeneous and irregular in cross section. While past investigations either focused on the building material (e.g. the molecular scale protein sequence and behaviour) or on the ultimate structural component (e.g. silk threads and spider webs), the bundled structure of fibrils that compose spider threads has been frequently overlooked. Herein, I exploit a molecular dynamics-based coarse-grain model to construct a fully three-dimensional fibril bundle, with a length on the order of micrometres. I probe the mechanical behaviour of bundled silk fibrils with variable density of heterogenic protrusions or globules, ranging from ideally homogeneous to a saturated distribution. Subject to stretching, the model indicates that cooperativity is enhanced by contact through low-force deformation and shear 'locking' between globules, increasing shear stress transfer by up to 200 per cent. In effect, introduction of a random and disordered structure can serve to improve mechanical performance. Moreover, addition of globules allows a tuning of free volume, and thus the wettability of silk (with implications for supercontraction). These findings support the ability of silk to maintain near-molecular-level strength at the scale of silk threads, and the mechanism could be easily adopted as a strategy for synthetic fibres.

Abstract:Axial orientation of molecular chains always results in an increase in fibre strength and a decrease in toughness. Here, taking inspiration from the skin structure, artificial spider silk with a buckled sheath–core structure is developed, with mechanical strength and toughness reaching 1.61 GPa and 466 MJ m−3, respectively, exceeding those of Caerostris darwini silk. The buckled structure is achieved by nano‐pulley combing of polyrotaxane hydrogel fibres through cyclic stretch–release training, which exhibits axial alignment of the polymer chains in fibre core and buckling in fibre sheath. The artificial spider silk also exhibits excellent supercontraction behavior, achieving a work capacity of 1.89 kJ kg−1, and an actuation stroke of 82%. Spinning Conditions Affect Structure and Properties of Nephila Spider Silk

Abstract:Raman spectroscopy is used to elucidate the effect of spinning conditions upon the structure and mechanical properties of silk spun by Nephila spiders from the major ampullate gland. Silk fibers produced under natural spinning conditions with spinning rates between 2 and 20 mm s −1 differed in microstructure and mechanical properties from fibers produced either more slowly or more rapidly. The data support the “uniform strain” hypothesis that the reinforcing units in spider silk fibers are subjected to the same strain as the fiber, to optimize the toughness. In contrast, in the case of synthetic high-performance polymer fibers, the both units and the fiber experience uniform stress, which maximizes stiffness. The comparison of Nephila major and minor ampullate silks opens an intriguing window into dragline silk evolution and the first evidence of significant differences between the two silks providing possibilities for further testing of hypotheses concerning the uniform strain versus uniform stress models. Impact statement It is well established that the microstructure and mechanical properties of engineering materials are controlled by the conditions employed to both synthesize and process them. Herein, we demonstrate that the situation is similar for a natural material, namely spider silk. We show that for a spider that normally produces silk at a reeling speed of between 2 and 20 mm s −1 , silk produced at speeds outside this natural processing window has a different microstructure that leads to inferior tensile properties. Moreover, we also show that the silk has a generic microstructure that is optimized to respond mechanically to deformation such that the crystals in the fibers are deformed under conditions of uniform strain. This is different from high-performance synthetic polymer fibers where the microstructure is optimized such that crystals within the fibers are subjected to uniform stress. Graphic abstract
Robust substrate anchorages of silk lines with extensible nano-fibres

Abstract:Living systems are built of multiscale-composites: materials formed of components with different properties that are assembled in complex micro- and nano-structures. Such biological multiscale-composites often show outstanding physical properties that are unachieved by artificial materials. A major scientific goal is thus to understand the assembly processes and the relationship between structure and function in order to reproduce them in a new generation of biomimetic high-performance materials. Here, we tested how the assembly of spider silk nano-fibres (i.e. glue coated 0.5 micron thick fibres produced by so-called piriform glands) into different micro-structures correlates with mechanical performance by empirically and numerically exploring the mechanical behaviour of line anchors in an orb weaver, a hunting spider and two ancient web builders. We demonstrate that the anchors of orb weavers exhibit outstanding mechanical robustness with minimal material use by the indirect attachment of the silk line to the substrate through a soft domain ('bridge'). This principle can be used to design new artificial high-performance attachment systems.

Abstract:In this letter, we adopt a new approach combining theoretical modeling with silk stretching measurements to explore the mystery of the structures between silkworm and spider silks, leading to the differences in mechanical response against stretching. Hereby the typical stress-strain profiles are reproduced by implementing the newly discovered and verified "$\beta$-sheet splitting" mechanism, which primarily varies the secondary structure of protein macromolecules; our modeling and simulation results show good accordance with the experimental measurements. Hence, it can be concluded that the post-yielding mechanical behaviors of both kinds of silks are resulted from the splitting of crystallines while the high extensibility of spider dragline is attributed to the tiny $\beta$-sheets solely existed in spider silk fibrils. This research reveals for the first time the structural factors leading to the significant difference between spider and silkworm silks in mechanical response to the stretching force. Additionally, the combination of theoretical modeling with experiments opens up a completely new approach in resolving conformation of various biomacromolecules.

Abstract:Here, we report the production of silk incorporating graphene and carbon nanotubes directly by spider spinning, after spraying spiders with the corresponding aqueous dispersions. We observe a significant increment of the mechanical properties with respect to the pristine silk, in terms of fracture strength, Young's and toughness moduli. We measure a fracture strength up to 5.4 GPa, a Young's modulus up to 47.8 GPa and a toughness modulus up to 2.1 GPa, or 1567 J/g, which, to the best of our knowledge, is the highest reported to date, even when compared to the current toughest knotted fibres. This approach could be extended to other animals and plants and could lead to a new class of bionic materials for ultimate applications.

Abstract:Spider silk is attractive because it is strong and tough. Moreover, an enormous range of mechanical properties can be achieved with only small changes in chemical structure. Our research shows that the full range of thermo-mechanical properties of silk fibres can be predicted from mean field theory for polymers in terms of chemical composition and the degree of order in the polymer structure. Thus, we can demonstrate an inherent simplicity at a macromolecular level in the design principles of natural materials. This surprising observation allows in depth comparison of natural with man-made materials.

Abstract:Spider dragline silk is distinguished through the highest toughness of all natural as well as artificial fiber materials. To unravel the toughness’s molecular foundation and to enable manufacturing biomimetic analogues, we investigated the morphological and functional structure of recombinant fibers, which exhibit toughness similar to that of the natural template, on the molecular scale by means of vibrational spectroscopy and on the mesoscale by X-ray scattering. Whereas the former was used to identify protein secondary structures and their alignment in the natural as well as artificial silks, the latter revealed nanometer-sized crystallites on the higher structural level. Furthermore, a spectral red shift of a crystal-specific absorption band demonstrated that macroscopically applied stress is directly transferred to the molecular scale, where it is finally dissipated. Concerning this feature, both the natural as well as the biomimetic fibers are almost indistinguishable, giving rise to the toughness of both fiber materials.

Abstract:This work presents an adaptive structure inspired by spider webs' behavior. To investigate the dynamic properties and performance of this system, numerical models are developed to examine the effects of pretension in radial strings, and Young's modulus, and damping ratio on the natural frequency and total energy of the system. An experimental study was conducted to validate theoretical results. Stepper motors controlled by a microcontroller are utilized to increase the pretension in the radial strings of the web in order to tune the web's energy absorption ability. It is demonstrated that the pretension, Young's modulus, and damping ratio in the radial strings can significantly affect the natural frequency and total energy of full and damaged webs. It is also shown that increasing the pretension in the radial strings compensates for the loss of stiffness due to the damaged strings. Finally, it is shown that controlling the pretension in radial strings can provide higher energy absorption capability for the spider web.
In-drop capillary spooling of spider capture thread inspires highly extensible fibres

Abstract:Spiders' webs and gossamer threads are often paraded as paradigms for lightweight structures and outstanding polymers. Probably the most intriguing of all spider silks is the araneid capture thread, covered with tiny glycoprotein glue droplets. Even if compressed, this thread remains surprisingly taut, a property shared with pure liquid films, allowing both thread and web to be in a constant state of tension. Vollrath and Edmonds proposed that the glue droplets would act as small windlasses and be responsible for the tension, but other explanations have also been suggested, involving for example the macromolecular properties of the flagelliform silk core filaments. Here we show that the nanolitre glue droplets of the capture thread indeed induce buckling and coiling of the core filaments: microscopic in-vivo observations reveal that the slack fibre is spooled into and within the droplets. We model windlass activation as a structural phase transition, and show that fibre spooling essentially results from the interplay between elasticity and capillarity. This is demonstrated by reproducing artificially the mechanism on a synthetic polyurethane thread/silicone oil droplet system. Fibre size is the key in natural and artificial setups which both require micrometer-sized fibres to function. The spools and coils inside the drops are further shown to directly affect the mechanical response of the thread, evidencing the central role played by geometry in spider silk mechanics. Beside shedding light on araneid capture thread functionality, we argue that the properties of this biological system provide novel insights for bioinspired synthetic actuators.
Nanoconfinement Controls Stiffness, Strength and Mechanical Toughness of Β-Sheet Crystals in Silk

Abstract:Silk features exceptional mechanical properties such as high tensile strength and great extensibility, making it one of the toughest materials known. The exceptional strength of silkworm and spider silks, exceeding that of steel, arises from β-sheet nanocrystals that universally consist of highly conserved poly-(Gly-Ala) and poly-Ala domains. This is counterintuitive because the key molecular interactions in β-sheet nanocrystals are hydrogen bonds, one of the weakest chemical bonds known. Here we report a series of large-scale molecular dynamics simulations, revealing that β-sheet nanocrystals confined to a few nanometres achieve higher stiffness, strength and mechanical toughness than larger nanocrystals. We illustrate that through nanoconfinement, a combination of uniform shear deformation that makes most efficient use of hydrogen bonds and the emergence of dissipative molecular stick–slip deformation leads to significantly enhanced mechanical properties. Our findings explain how size effects can be exploited to create bioinspired materials with superior mechanical properties in spite of relying on mechanically inferior, weak hydrogen bonds.
Generative modeling, design and analysis of spider silk protein sequences for enhanced mechanical properties

Abstract:Spider silks are remarkable materials characterized by superb mechanical properties such as strength, extensibility and lightweightedness. Yet, to date, limited models are available to fully explore sequence-property relationships for analysis and design. Here we propose a custom generative large-language model to enable design of novel spider silk protein sequences to meet complex combinations of target mechanical properties. The model, pretrained on a large set of protein sequences, is fine-tuned on ~1,000 major ampullate spidroin (MaSp) sequences for which associated fiber-level mechanical properties exist, to yield an end-to-end forward and inverse generative strategy. Performance is assessed through: (1), a novelty analysis and protein type classification for generated spidroin sequences through BLAST searches, (2) property evaluation and comparison with similar sequences, (3) comparison of molecular structures, as well as, and (4) a detailed sequence motif analyses. We generate silk sequences with property combinations that do not exist in nature, and develop a deep understanding the mechanistic roles of sequence patterns in achieving overarching key mechanical properties (elastic modulus, strength, toughness, failure strain). The model provides an efficient approach to expand the silkome dataset, facilitating further sequence-structure analyses of silks, and establishes a foundation for synthetic silk design and optimization.

A thermodynamic analysis of the spider silk and the importance of complexity

Abstract:The spider silk is one of the most interesting bio-materials investigated in the last years. One of the main reasons that brought scientists to study this organized system is its high level of resistance if compared to other artificial materials characterized by higher density. Subsequently, researchers discovered that the spider silk is a complex system formed by different kinds of proteins, organized (or disorganized) to guarantee the required resistance, which is function of the final application and of the environmental conditions. Some spider species are able to make different silks, up to twelve, having a composition that seems to be function of the final use (i.e. dragline web, capture web, etc). The aim of this paper is to analyze the properties of the spider silk by means of a thermodynamic approach, taking advantage of the well-known theories applied to polymers, and to try to underline and develop some intriguing considerations. Moreover, this study can be taken as an example to introduce and discuss the importance of the concept of optionality and of the anti-fragile systems proposed by N. N. Thaleb in his book "Antifragile: Things that gain from disorder".

Abstract:Nanofibrils are considered the universal building blocks of spider silks. In this work, the thinnest natural spider silk nanofibrils are imaged, measured, and mechanically tested for the first time. Gaining insight into the mechanics of silk at the nanoscale is important. Here, it reveals that nanoscale silk is even stretchier than larger fibers but equally as strong. Cribellate silks, produced by ancient spiders, are fascinating because they feature a highly sophisticated, 3D hierarchical structure consisting of filaments with different diameters and shapes. Here, the smallest and thinnest constituents of the cribellate silk are investigated: nanofibrils that form a dense mesh that is supported by larger fibers. Analysis of their structure via atomic force and transmission electron microscopies shows that they are flattened fibrils, only ≈5 nm thick — thinner than any other natural spider silk fibrils previously reported. In this work, the first mechanical tensile testing experiments on these fibrils are carried out, which reveals that the fibrils show an outstanding extensibility of at least 1100%, almost twice as much as the most stretchable spider silk previously reported. Based on these extraordinary findings, this work significantly expands the parameter space of materials properties attainable by spider silks and provides further insights into their nanomechanics.

Abstract:Spider silk is repeatedly stretched while performing biological functions. There is a close relationship between the shape change of the fibre materials and their mechanical properties. However, the effect of the deformation and interval time on the structure and tensile behaviour properties of spider silk after repeatedly stretching by given strain value has been rarely reported. Here we found that major ampullate silk (MAS) can revert its tensile behaviour independent of its previous loading history via intervals of approximately 8 s to 5 min with constant and increased elongation, respectively, after being subjected to yield and hardening regions. The true stress-true strain curve beyond a given value of true strain is independent from the previous loading history of the sample. Even after longer intervals (≥1 h), MAS can reproduce the last tensile behaviour via one stretched. Despite recognizing the development of irreversible deformations in the material when tested in air, the reversible change in tensile behaviour outside the spider silk's elastic region has rarely been observed before. MAS has at least one proper ground state that allows it to present good shape and mechanical behaviour memory in terms of longitudinal stretching, functioning as a new strategy to achieve certain tensile properties. The analysis of the true stress-true strain curves was performed from a series of loading‒unloading tests to evaluate the evolution of those mechanical parameters with the cycle number. The elastic modulus measured in the loading steps increases monotonously with increasing values of true strain reached in the cycles. In contrast, a marginal variation is found in the values of the yield stress measured in the different cycles. The memory and variation in the mechanical behaviour and performance of MAS can be accounted for through the irreversible and reversible deformation micromechanisms and its combination in which the viscoelasticity of the material plays a leading role. These findings may be helpful to guide the biomimetic design of novel fibre materials such as spider silk gut via artificially stretching spider silk glands.
