Abstract:Spider silk has been an attractive biopolymer since ancient times. Learning from both its excellent properties and spinning process, silk provides people with inspiration to develop functional fibers. Recently, inspired by shiny water droplets on a spider's web, we revealed that the capture silk of the cribellate spider would deform to have a special periodic spindle‐knots structure and hence displayed unique wettability, making it efficient at directional water‐collecting. This provides insights in designing functional fibers with unique wettability, by either creating special structures on the fiber surface, or modifying it with responsive molecules. These bioinspired functional fibers may find applications in many fields, such as water collection, smart catalysis, filtration, and sensing.

Abstract:Scalable computational modelling tools are required to guide the rational design of complex hierarchical materials with predictable functions. Here, we utilize mesoscopic modelling, integrated with genetic block copolymer synthesis and bioinspired spinning process, to demonstrate de novo materials design that incorporates chemistry, processing and material characterization. We find that intermediate hydrophobic/hydrophilic block ratios observed in natural spider silks and longer chain lengths lead to outstanding silk fibre formation. This design by nature is based on the optimal combination of protein solubility, self-assembled aggregate size and polymer network topology. The original homogeneous network structure becomes heterogeneous after spinning, enhancing the anisotropic network connectivity along the shear flow direction. Extending beyond the classical polymer theory, with insights from the percolation network model, we illustrate the direct proportionality between network conductance and fibre Young's modulus. This integrated approach provides a general path towards de novo functional network materials with enhanced mechanical properties and beyond (optical, electrical or thermal) as we have experimentally verified.
Recombinant Spidroins Fully Replicate Primary Mechanical Properties of Natural Spider Silk.

Abstract:Despite significant efforts to engineer their heterologous production, recombinant spider silk proteins (spidroins) have yet to replicate the unparalleled combination of high strength and toughness exhibited by natural spider silks, preventing their use in numerous mechanically demanding applications. To overcome this long-standing challenge, we have developed a synthetic biology approach combining standardized DNA part assembly and split intein-mediated ligation to produce recombinant spidroins of previously unobtainable size (556 kDa), containing 192 repeat motifs of the Nephila clavipes dragline spidroin. Fibers spun from our synthetic spidroins are the first to fully replicate the mechanical performance of their natural counterparts by all common metrics, i.e., tensile strength (1.03 +/- 0.11 GPa), modulus (13.7 +/- 3.0 GPa), extensibility (18 +/- 6%), and toughness (114 +/- 51 MJ/m(3)). The developed process reveals a path to more dependable production of high-performance silks for mechanically demanding applications while also providing a platform to facilitate production of other high-performance natural materials.

Abstract:A variety of artificial spinning methods have been applied to produce regenerated silk fibers; however, how to spin regenerated silk fibers that retain the advantages of natural silks in terms of structural hierarchy and mechanical properties remains challenging. Here, we show a bioinspired approach to spin regenerated silk fibers. First, we develop a nematic silk microfibril solution, highly viscous and stable, by partially dissolving silk fibers into microfibrils. This solution maintains the hierarchical structures in natural silks and serves as spinning dope. It is then spun into regenerated silk fibers by direct extrusion in the air, offering a useful route to generate polymorphic and hierarchical regenerated silk fibers with physical properties beyond natural fiber construction. The materials maintain the structural hierarchy and mechanical properties of natural silks, including a modulus of 11 ± 4 GPa, even higher than natural spider silk. It can further be functionalized with a conductive silk/carbon nanotube coating, responsive to changes in humidity and temperature.

Abstract:24 Dragline spider silk is among the strongest and toughest bio-based materials, capable of 25 outperforming most synthetic polymers and even some metal alloys.1,2,3,4 These properties 26 have gained spider silk a growing list of potential applications that, coupled with the 27 impracticalities of spider farming, have driven a decades-long effort to produce 28 recombinant spider silk proteins (spidroins) in engineered heterologous hosts.2 However, 29 these efforts have so far been unable to yield synthetic silk fibers with mechanical 30 properties equivalent to natural spider silk, largely due to an inability to stably produce 31 highly repetitive, high molecular weight (MW) spidroins in heterologous hosts.1,5 Here we 32 address these issues by combining synthetic biology techniques with split intein (SI)33 mediated ligation for the bioproduction of spidroins with unprecedented MW (556 kDa), 34 containing 192 repeat motifs of the Nephila clavipes MaSp1 dragline spidroin. Fibers spun 35 from these synthetic spidroins display ultimate tensile strength (σ), modulus (E), 36 extensibility (ε), and toughness (UT) of 1.03 ± 0.11 GPa, 13.7 ± 3.0 GPa, 18 ± 6%, and 114 ± 37 51 MJ/m3, respectively—equivalent to the performance of natural N. clavipes dragline silk.6 38 This work demonstrates for the first time that microbially produced synthetic silk fibers 39 can match the performance of natural silk fibers by all common metrics (σ, E, ε, UT), 40 providing a more dependable source of high-strength fibers to replace natural spider silks 41 for mechanically demanding applications. Furthermore, our biosynthetic platform can be 42 potentially expanded for the assembly and production of other protein-based materials 43 with high MW and repetitive sequences that have so far been impossible to synthesize by 44 genetic means alone. 45

Abstract:Spiders' webs and gossamer threads are often paraded as paradigms for lightweight structures and outstanding polymers. Probably the most intriguing of all spider silks is the araneid capture thread, covered with tiny glycoprotein glue droplets. Even if compressed, this thread remains surprisingly taut, a property shared with pure liquid films, allowing both thread and web to be in a constant state of tension. Vollrath and Edmonds proposed that the glue droplets would act as small windlasses and be responsible for the tension, but other explanations have also been suggested, involving for example the macromolecular properties of the flagelliform silk core filaments. Here we show that the nanolitre glue droplets of the capture thread indeed induce buckling and coiling of the core filaments: microscopic in-vivo observations reveal that the slack fibre is spooled into and within the droplets. We model windlass activation as a structural phase transition, and show that fibre spooling essentially results from the interplay between elasticity and capillarity. This is demonstrated by reproducing artificially the mechanism on a synthetic polyurethane thread/silicone oil droplet system. Fibre size is the key in natural and artificial setups which both require micrometer-sized fibres to function. The spools and coils inside the drops are further shown to directly affect the mechanical response of the thread, evidencing the central role played by geometry in spider silk mechanics. Beside shedding light on araneid capture thread functionality, we argue that the properties of this biological system provide novel insights for bioinspired synthetic actuators.

Abstract:Axial orientation of molecular chains always results in an increase in fibre strength and a decrease in toughness. Here, taking inspiration from the skin structure, artificial spider silk with a buckled sheath–core structure is developed, with mechanical strength and toughness reaching 1.61 GPa and 466 MJ m−3, respectively, exceeding those of Caerostris darwini silk. The buckled structure is achieved by nano‐pulley combing of polyrotaxane hydrogel fibres through cyclic stretch–release training, which exhibits axial alignment of the polymer chains in fibre core and buckling in fibre sheath. The artificial spider silk also exhibits excellent supercontraction behavior, achieving a work capacity of 1.89 kJ kg−1, and an actuation stroke of 82%. Abstract:Living systems are built of multiscale-composites: materials formed of components with different properties that are assembled in complex micro- and nano-structures. Such biological multiscale-composites often show outstanding physical properties that are unachieved by artificial materials. A major scientific goal is thus to understand the assembly processes and the relationship between structure and function in order to reproduce them in a new generation of biomimetic high-performance materials. Here, we tested how the assembly of spider silk nano-fibres (i.e. glue coated 0.5 micron thick fibres produced by so-called piriform glands) into different micro-structures correlates with mechanical performance by empirically and numerically exploring the mechanical behaviour of line anchors in an orb weaver, a hunting spider and two ancient web builders. We demonstrate that the anchors of orb weavers exhibit outstanding mechanical robustness with minimal material use by the indirect attachment of the silk line to the substrate through a soft domain ('bridge'). This principle can be used to design new artificial high-performance attachment systems.

Abstract:Spider dragline silk is a remarkable material that shows excellent mechanical properties, diverse applications, biocompatibility and biodegradability. Transgenic silkworm technology was used to obtain four types of chimeric silkworm/spider (termed composite) silk fibres, including different lengths of recombinant Major ampullate Spidroin1 (re-MaSp1) or recombinant Major ampullate Spidroin2 (re-MaSp2) from the black widow spider, Latrodectus hesperus. The results showed that the overall mechanical properties of composite silk fibres improved as the re-MaSp1 chain length increased, and there were significant linear relationships between the mechanical properties and the re-MaSp1 chain length (p < 0.01). Additionally, a stronger tensile strength was observed for the composite silk fibres that included re-MaSp1, which only contained one type of repetitive motif, (GA)(n)/A(n), to provide tensile strength, compared with the silk fibres that includedre-MaSp2, which has the same protein chain length as re-MaSp1 but contains multiple types of repetitive motifs, GPGXX and (GA)(n)/A(n). Therefore, the results indicated that the nature of various repetitive motifs in the primary structure played an important role in imparting excellent mechanical properties to the protein-based silk fibres. A silk protein with a single type of repetitive motif and sufficiently long chains was determined to be an additional indispensable factor. Thus, this study forms a foundation for designing and optimizing the structure of re-silk protein using a heterologous expression system.
The Cribellate Nanofibrils of the Southern House Spider: Extremely Thin Natural Silks with Outstanding Extensibility

Abstract:Nanofibrils are considered the universal building blocks of spider silks. In this work, the thinnest natural spider silk nanofibrils are imaged, measured, and mechanically tested for the first time. Gaining insight into the mechanics of silk at the nanoscale is important. Here, it reveals that nanoscale silk is even stretchier than larger fibers but equally as strong. Cribellate silks, produced by ancient spiders, are fascinating because they feature a highly sophisticated, 3D hierarchical structure consisting of filaments with different diameters and shapes. Here, the smallest and thinnest constituents of the cribellate silk are investigated: nanofibrils that form a dense mesh that is supported by larger fibers. Analysis of their structure via atomic force and transmission electron microscopies shows that they are flattened fibrils, only ≈5 nm thick — thinner than any other natural spider silk fibrils previously reported. In this work, the first mechanical tensile testing experiments on these fibrils are carried out, which reveals that the fibrils show an outstanding extensibility of at least 1100%, almost twice as much as the most stretchable spider silk previously reported. Based on these extraordinary findings, this work significantly expands the parameter space of materials properties attainable by spider silks and provides further insights into their nanomechanics.

Abstract:Here, we report the production of silk incorporating graphene and carbon nanotubes directly by spider spinning, after spraying spiders with the corresponding aqueous dispersions. We observe a significant increment of the mechanical properties with respect to the pristine silk, in terms of fracture strength, Young's and toughness moduli. We measure a fracture strength up to 5.4 GPa, a Young's modulus up to 47.8 GPa and a toughness modulus up to 2.1 GPa, or 1567 J/g, which, to the best of our knowledge, is the highest reported to date, even when compared to the current toughest knotted fibres. This approach could be extended to other animals and plants and could lead to a new class of bionic materials for ultimate applications.

Abstract:The European Community Respiratory Health Survey (ECRHS) was planned to answer specific questions about the distribution of asthma and health care given for asthma in the European Community. Specifically, the survey is designed to estimate variations in the prevalence of asthma, asthma-like symptoms and airway responsiveness; to estimate variations in exposures to known or suspected risk factors for asthma, and assess to what extent these variations explain the variations in the prevalence of disease; and to estimate differences in the use of medication for asthma. The protocol provides specific instructions on the sampling strategy adopted by the survey teams, as well as providing instructions on the use of questionnaires, the tests for allergy, lung function measurements, tests of airway responsiveness, and blood and urine collection. The principal data collection sheets and questionnaires are provided in the appendices, together with information on coding and quality control. The protocol is published as a reference for those who wish to know more of the methods used in the study, and also to give other groups who wish to collect comparable data access to the detailed methodology.

Abstract:Spider silks are among the toughest known materials and thus provide models for renewable, biodegradable, and sustainable biopolymers. However, the entirety of their diversity still remains elusive, and silks that exceed the performance limits of industrial fibers are constantly being found. We obtained transcriptome assemblies from 1098 species of spiders to comprehensively catalog silk gene sequences and measured the mechanical, thermal, structural, and hydration properties of the dragline silks of 446 species. The combination of these silk protein genotype-phenotype data revealed essential contributions of multicomponent structures with major ampullate spidroin 1 to 3 paralogs in high-performance dragline silks and numerous amino acid motifs contributing to each of the measured properties. We hope that our global sampling, comprehensive testing, integrated analysis, and open data will provide a solid starting point for future biomaterial designs.

Abstract:Spider silk is a remarkable example of bio-material with superior mechanical characteristics. Its multilevel structural organization of dragline and viscid silk leads to unusual and tunable properties, extensively studied from a quasi-static point of view. In this study, inspired by the Nephila spider orb web architecture, we propose a novel design for mechanical metamaterials based on its periodic repetition. We demonstrate that spider-web metamaterial structure plays an important role in the dynamic response and wave attenuation mechanisms. The capability of the resulting structure to inhibit elastic wave propagation in sub-wavelength frequency ranges is assessed and parametric studies are performed to derive optimal configurations and constituent mechanical properties. The results show promise for the design of innovative lightweight structures for tunable vibration damping and impact protection, or the protection of large scale infrastructure such as suspended bridges.

Abstract:Raman spectroscopy is used to elucidate the effect of spinning conditions upon the structure and mechanical properties of silk spun by Nephila spiders from the major ampullate gland. Silk fibers produced under natural spinning conditions with spinning rates between 2 and 20 mm s −1 differed in microstructure and mechanical properties from fibers produced either more slowly or more rapidly. The data support the “uniform strain” hypothesis that the reinforcing units in spider silk fibers are subjected to the same strain as the fiber, to optimize the toughness. In contrast, in the case of synthetic high-performance polymer fibers, the both units and the fiber experience uniform stress, which maximizes stiffness. The comparison of Nephila major and minor ampullate silks opens an intriguing window into dragline silk evolution and the first evidence of significant differences between the two silks providing possibilities for further testing of hypotheses concerning the uniform strain versus uniform stress models. Impact statement It is well established that the microstructure and mechanical properties of engineering materials are controlled by the conditions employed to both synthesize and process them. Herein, we demonstrate that the situation is similar for a natural material, namely spider silk. We show that for a spider that normally produces silk at a reeling speed of between 2 and 20 mm s −1 , silk produced at speeds outside this natural processing window has a different microstructure that leads to inferior tensile properties. Moreover, we also show that the silk has a generic microstructure that is optimized to respond mechanically to deformation such that the crystals in the fibers are deformed under conditions of uniform strain. This is different from high-performance synthetic polymer fibers where the microstructure is optimized such that crystals within the fibers are subjected to uniform stress. Graphic abstract

Abstract:Spiders use structural conversion of the conserved globular 127‐residue spacer domains to make strong silk fibers. This domain, interestingly containing motifs that are similar to human amyloidogenic motifs, self‐assembles into amyloid‐like fibrils through a non‐nucleation‐dependent pathway. Integrating this spacer into a chimeric spidroin enhances silk‐like fiber assembly, improves molecular homogeneity and significantly strengthens the fibers. Spider silk—an environmentally friendly protein‐based material—is widely recognized for its extraordinary mechanical properties. Biomimetic spider silk‐like fibers made from recombinant spider silk proteins (spidroins) currently falls short compared to natural silks in terms of mechanical performance. In this study, it is discovered that spiders use structural conversion of molecular enhancers—conserved globular 127‐residue spacer domains—to make strong silk fibers. This domain lacks poly‐Ala motifs but interestingly contains motifs that are similar to human amyloidogenic motifs, and that it self‐assembles into amyloid‐like fibrils through a non‐nucleation‐dependent pathway, likely to avoid the formation of cytotoxic intermediates. Incorporating this spacer domain into a recombinant chimeric spidroin facilitates self‐assembly into silk‐like fibers, increases fiber molecular homogeneity, and markedly enhances fiber mechanical strength. These findings highlight that spiders employ diverse strategies to produce silk with exceptional mechanical properties. The spacer domain offers a way to enhance the properties of recombinant spider silk‐like fibers and other functional materials.

Abstract:Achieving ultra-high tensile strength and exceptional toughness is a longstanding goal for structural materials. However, previous attempts using covalent and non-covalent bonds have failed, leading to the belief that these two properties are mutually exclusive. Consequently, commercial fibers have been forced to compromise between tensile strength and toughness, as seen in the differences between nylon and Kevlar. To address this challenge, we drew inspiration from the disparate tensile strength and toughness of nylon and Kevlar, both of which are polyamide fibers, and developed an innovative approach that combines specific intermolecular disulfide bonds and reversible hydrogen bonds to create ultra-strong and ultra-tough polyamide spider silk fibers. Our resulting Supramolecular polyamide spider silk, which has a maximum molecular weight of 1084 kDa, exhibits high tensile strength (1180 MPa) and extraordinary toughness (433 MJ/m 3 ), surpassing Kevlar's toughness 8-fold. This breakthrough presents a new opportunity for the sustainable development of spider silk as an environmentally friendly alternative to synthetic commercial fibers, as spider silk is composed of amino acids. Future research could explore the use of these techniques and fundamental knowledge to develop other super materials in various mechanical fields, with the potential to improve people's lives in many ways. Statement of Significance • By emulating synthetic commercial fibers such as nylon and polyethylene, we have successfully produced supramolecular-weight polyamide spider silk fibers with a molecular weight of 1084 kDa through a unique covalent bond-mediated linear polymerization reaction of spider silk protein molecules. This greatly surpasses the previous record of a maximum molecular weight of 556 kDa. • We obtained supramolecular polyamide spider silk fibers with both high-tensile strength and toughness. The stress at break is 1180 MPa, and the toughness is 8 times that of kevlar, reaching 433 MJ/m 3 . • Our results challenge the notion that it is impossible to manufacture fibers with both ultra-high tensile strength and ultra-toughness, and provide theoretical guidance for developing environmentally friendly and sustainable structural materials that meet industrial needs.

Abstract:Spider dragline silk is distinguished through the highest toughness of all natural as well as artificial fiber materials. To unravel the toughness’s molecular foundation and to enable manufacturing biomimetic analogues, we investigated the morphological and functional structure of recombinant fibers, which exhibit toughness similar to that of the natural template, on the molecular scale by means of vibrational spectroscopy and on the mesoscale by X-ray scattering. Whereas the former was used to identify protein secondary structures and their alignment in the natural as well as artificial silks, the latter revealed nanometer-sized crystallites on the higher structural level. Furthermore, a spectral red shift of a crystal-specific absorption band demonstrated that macroscopically applied stress is directly transferred to the molecular scale, where it is finally dissipated. Concerning this feature, both the natural as well as the biomimetic fibers are almost indistinguishable, giving rise to the toughness of both fiber materials.

Abstract:An essential element in the web-trap architecture, the capture silk spun by ecribellate orb spiders consists of glue droplets sitting astride a silk filament. Mechanically this thread presents a mixed solid-liquid behavior unknown to date. Under extension, capture silk behaves as a particularly stretchy solid, owing to its molecular nanosprings, but it totally switches behavior in compression to now become liquid-like: It shrinks with no apparent limit while exerting a constant tension. Here, we unravel the physics underpinning the unique behavior of this "liquid wire" and demonstrate that its mechanical response originates in the shape-switching of the silk filament induced by buckling within the droplets. Learning from this natural example of geometry and mechanics, we manufactured programmable liquid wires that present previously unidentified pathways for the design of new hybrid solid-liquid materials.
