Abstract:Modifying the drug-release capacity of titanium implants is essential for maintaining their long-term functioning. Titanium dioxide nanotube (TNT) arrays, owing to their drug release capacity, are commonly used in the biomaterial sphere. Their unique half open structure and arrangement in rows increase the drug release capacity. However, their rapid drug release ability not only reduces drug efficiency but also produces excessive local and systemic deposition of antibiotics. In this study, we designed a tantalum-coated TNT system for drug-release optimization. A decreased nanotube size caused by the tantalum nanocoating was observed through SEM and analyzed (TNT: 110 nm, TNT-Ta1: 80 nm, TNT-Ta3: 40 nm, TNT-Ta5: 20 nm, TNT-Ta7: <5 nm). XPS analysis revealed the distribution of the chemical components, especially that of the tantalum element. In vitro experiments showed that the tantalum nanocoating enhanced cell proliferation; in particular, TNT-Ta5 possessed the best cell viability (about 1.18 of TNT groups at 7d). It also showed that the tantalum nanocoating had a positive effect on osteogenesis (especially TNT-Ta5 and TNT-Ta7). Additionally, hydrophilic/hydrophobic drug (vancomycin/raloxifene) release results indicated that the TNT-Ta5 group possessed the most desirable sustained release capacity. Moreover, in this drug release system, the hydrophobic drug showed more sustained release capacity than the hydrophilic drug (vancomycin: sustained release for more than 48 h, raloxifene: sustained release for more than 168 h). More importantly, TNT-Ta5 is proved to be an appropriate drug release system, which possesses cytocompatibility, osteogenic capacity, and sustained drug release capacity.

Abstract:Background For dental implants, early infection and peri-implantitis after implant restoration are major reasons for implant failure. Plaque biofilm formation is the initiator for implant-related infection. Therefore, it is urgent to develop a new antibacterial implant materials or treatment methods. Aim/Hypothesis In this study, Sr- and Zn- incorporated micro nano-structured titanium surface with nanorods (MNT-Sr Zn) was fabricated by a two-step hydrothermal process, aiming to improve osseointegration and reduce implant failure caused by implant-related infection and broaden new ideas for the future research Material and Methods The Mod Ti surfaces were used as a control. The characterization of different surfaces was detected by a field emission scanning electron microscope (FE-SEM) equipped with energy dispersive spectra (EDS) detector, x-ray diffractometer (XRD) and inductively coupled plasma mass spectroscopy (ICP-MS). Meanwhile, Staphylococcus aureus (S. aureus) was selected to evaluate antibacterial properties through Live Dead Staining and watching the adhesive bacterial morphology under FE-SEM. What's more, rat bone marrow-derived mesenchymal stem cells (rBMSCs) were cultured on the surfaces to detect their early osteogenic properties, like the alkaline phosphatase(ALP) activity and the expressions of osteogenesis related genes. Results The results confirmed that nanorod-like particles with a diameter of about 30-50 nm of MNT-Sr Zn composed of well-crystallized ZnO and SrTiO3 phases. In the EDS spectra, the elements of Zn and Sr were evenly distributed, and approximately 1.49 ± 0.16 wt% and 21.69 ± 2.74 wt% respectively. Besides, the release of Sr2+ and Zn2+ was a controlled and long-term process. More importantly, Zn-containing nanorod-like particles and the morphology could work together, slightly decreasing adhesion of S. aureus but increasing the proportion of dead cells, thus inhibiting subsequent biofilm formation, compared with the controlled group. In addition to this, MNT-Sr Zn could enhance the osteogenic differentiation via the upregulation of the transcription of osteogenic related genes and the expression of ALP in vitro. Conclusion and Clinical Implications This research provides a new surface modification method on titanium substrate for multi-functional implant material development. Due to the co-doping of Sr and Zn in the coating, biomechanical stability, antibacterial capability and early osteogenic inductive effect are enhanced, which has the potential to be applied in dental implantation in the future.
Zinc- and Strontium- Co-Incorporated Nanorods on Titanium Surfaces with Favorable Material Property, Osteogenesis, and Enhanced Antibacterial Activity.

Abstract:Early infection and peri-implantitis after implant restoration are major reasons for dental implant failure. Implant-associated infections are majorly attributed to biofilm formation. In this study, co-incorporated zinc- (Zn-) and strontium- (Sr-) nanorod coating on sandblasted and acid-etched (SLA) titanium (SLA-Zn/Sr) was fabricated by hydrothermal synthesis. It was aimed at promoting osteogenesis while inhibiting biofilm formation. The nanorod-like particles (phi 30-50 nm) were found to be evenly formed on SLA-Zn/Sr (Zn: 1.49 +/- 0.16 wt%; Sr: 21.69 +/- 2.74 wt%) that was composed of well-crystallized ZnTiO3 and SrTiO3 phases. With a sufficient interface bonding strength (42.00 +/- 3.00 MPa), SLA-Zn/Sr enhanced the corrosion resistance property of titanium. Besides, SLA-Zn/Sr promoted the cellular initial adhesion, proliferation and osteogenic differentiation of rBMSCs in vitro while inhibiting the adhesion of Staphylococcus aureus and Porphyromonas gingivalis . In addition, through down-regulating icaA gene expression, this novel surface reduced the secretion of polysaccharide intercellular adhesion (reduced by 87.9% compared to SLActive) to suppress the S. aureus biofilm formation. We, therefore, propose a new chemical modification on titanium for multifunctional implant material development. Due to the Zn/Sr co-doping in coating, material properties, early osteogenic effect and antibacterial ability of titanium can be simultaneously enhanced, which has the potential to be applied in dental implantation in the future.
Research on the surface characterization, corrosion and bioactivity of nano-featured tantalum coating on selective electron beam melted Ti6Al4V alloy

Abstract:A nano-featured tantalum coatings with an average thickness of 800¬†nm and grain size of 50¬†nm were successfully prepared by Selective Electron Beam Melted (SEBM) Ti6Al4V substrates by DC magnetron sputtering technology. The details of the physicochemical properties, corrosion resistance and in vitro biocompatibility of the coating were systematically investigated. Elastic modulus of the nano tantalum coatings was around 123.8¬†GPa and the hardness was 8.08¬†GPa. Compared with uncoated Ti6Al4V matrix, the corrosion potential of the Ta-Ti64 samples was positively shifted and the corrosion current density decreased from 2.07¬†√ó¬†10 ‚ąí7 to 2.85¬†√ó¬†10 ‚ąí8 A¬∑cm ‚ąí2 (0.1¬†mV/s). EIS and PDP results show that the coating enhances the humoral corrosion resistance of Ti6Al4V, which can provide active protection for the substrate to prevent the release of toxic ions caused by long-term corrosion. The mouse osteoblast cell line (MC3T3-E1) is applied in this study for biological evaluation. The optical density at 450¬†nm results showed that the cell proliferation rate of tantalum coated surface was faster than that of Ti6Al4V sheet and pure tantalum sheet. The 5-day cell adhesion density is 1.26 and 1.32 times higher than uncoated Ti6Al4V and pure tantalum respectively. The nano-structured Ta layer significantly improved the initial adhesion of MC3T3-E1 cells and promoted cell proliferation. The corrosion resistance and biocompatibility of Ta coating are not only better than titanium matrix, but also better than pure tantalum. The results prove that tantalum coating is an effective method to modify titanium alloy and holds a great potential value on 3D printed personalized implants application.

Abstract:Microbiologically induced corrosion (MIC) of metallic devices/implants in the oral region is one major cause of implant failure and metal allergy in patients. Therefore, it is crucial to develop practical approaches which can effectively prevent MIC for broad clinical applications of these materials. In the present work, tantalum nitride (TaN)-decorated titanium with promoted bio-corrosion and mechanical property was firstly developed via depositing TaN layer onto pure Ti using magnetron sputtering. The microstructure and chemical constituent of TaN coatings were characterized, and were found to consist of a hard fcc-TaN outer layer. Besides, the addition of TaN coatings greatly increased the hardness and modulus of pristine Ti from 2.54 ± 0.20 to 29.88 ± 2.59 GPa, and from 107.19 ± 6.98 to 295.46 ± 19.36 GPa, respectively. Potentiodynamic polarization and electrochemical impedance spectroscopy studies indicated that TaN coating exhibited higher MIC resistance in comparison to bare Ti and TiN-coated coating in two bacteria-containing artificial saliva solutions. Moreover, the biofilm experiment showed that the TaN-decorated Ti sample possessed good antibacterial performance. The SEM and XPS results after biofilm removal demonstrated that TaN film remained its integrity and stability, while TiN layer detached from Ti surface in the bio-corrosion tests, demonstrating the anti-MIC behavior and the strong binding property of TaN coating to Ti substrate. Considering all these results, TaN-decorated Ti material exhibits the optimal comprehensive performance and holds great potential as implant material for dental applications.
Biocompatibility and Microstructure-Based Stress Analyses of TiNbZrTa Composite Films

Abstract:Background: the clinical application of orthopedic or dental implants improves the quality of the lives of patients. However, the long-term use of implants may lead to implant loosening and related complications. The purpose of this study is to deposit titanium (Ti)-niobium (Nb)-zirconium (Zr)-tantalum (Ta) alloys on the surface of Ti-6Al-4V to increase structural strength and biocompatibility for the possible future application of implants. Materials and methods: Ti, Nb, Zr, and Ta served as the materials for the surface modification of the titanium alloy. TiNbZr and TiNbZrTa coatings were produced using cathodic arc evaporation, and a small amount of nitrogen was added to produce TiNbZrTa(N) film. Annealing and oxidation were then conducted to produce TiNbZrTa-O and TiNbZrTa(N)-O coatings. In this study, biological tests and finite element analyses of those five alloy films, as well as uncoated Ti-6Al-4V, were performed. Human osteosarcoma cells (MG-63) and mouse fibroblast cells (L-929) were used to analyze cytotoxicity, cell viability, and cell morphology, and the bone differentiation of MG-63 was evaluated in an alkaline phosphatase experiment. Furthermore, for measuring the gene expression level of L-929, reverse transcription quantitative real-time polymerase chain reaction (RT-qPCR) was conducted. The three-dimensional (3D) computational models of the coated and uncoated sample films were constructed using images of transmission electron microscopy and computer-aided design software and, then, the stress distributions of all models were evaluated by finite element analysis. Result: the cytotoxicity test revealed that the surface treatment had no significant cytotoxic effects on MG-63 and L-929 cells. According to the results of the cell viability of L-929, more cell activity was observed in the surface-treated experimental group than in the control group; for MG-63, the cell viability of the coated samples was similar to that of the uncoated samples. In the cell morphology analysis, both MG-63 and L-929 exhibited attached filopodia and lamellipodia, verifying that the cells were well attached. The alkaline phosphatase experiment demonstrated that the surface treatment did not affect the characteristics of early osteogenic differentiation, whereas RT-qPCR analysis showed that surface treatment can promote better performance of L-929 cells in collagen, type I, α1, and fibronectin 1. Finally, the results of the finite element analysis revealed that the coated TiNb interlayer can effectively reduce the stress concentration inside the layered coatings. Conclusions: TiNbZrTa series films deposited using cathodic arc evaporation had excellent biocompatibility with titanium alloys, particularly in regard to soft tissue cells, which exhibited an active performance. The finite element analysis verified that the TiNb interlayer can reduce the stress concentration inside TiNbZrTa series films, increasing their suitability for application in biomedical implants in the future.

Abstract:Tantalum (Ta) has been used to modify the surface of titanium (Ti) due to its superior biological response under some clinically compromised situations such as osteoporosis and diabetes; however, the techniques used presented technical disadvantages. The currently study prepared Ta2O5 and Mg-incorporated Ta2O5 coatings on Ti6Al4V by a sol-gel method and compared in vitro response of rabbit bone marrow mesenchymal stem cells. Both coatings were submicrometer-thick and comprised nano-sized particles, the former being crystalline whereas the latter amorphous. Compared with uncoated Ti6Al4V, both coatings increased the corrosion resistance and water wettability, and improved the viability, proliferation, osteoblastic differentiation of the cells. Moreover, they also upregulated the expression of an angiogenesis-related gene (HIF-1α). With the exception of corrosion rate, greatest improvements were observed from the Mg-incorporated Ta2O5 coating. These improvements were probably due to the nanostructures and elements release of Mg and Ta from the coatings. All above results suggest that, Ta2O5-based coatings are promising tools for improving the biological response of Ti-based implants under compromised conditions.

Micro-nano porous structured tantalum-coated dental implants promote osteogenic activity in vitro and enhance osseointegration in vivo

Abstract:Due to its excellent biocompatibility and corrosion resistance, tantalum demonstrates versatility as an implant material. However, limited studies investigated the role of tantalum coated titanium-based dental implants. This study aimed to investigate the potential application of micro-nano porous structured tantalum coating on the surface of titanium dental implant. In the present study, micro-nano porous structured tantalum coating was prepared by vacuum plasma spraying (VPS) under selected optimum parameters, various characteristics of tantalum coating (Ta/Ti), including the morphology, potential, constituent, and hydrophilia, were investigated in comparison with its respective control groups, sandblasted titanium (Ti) and titanium coating (Ti/Ti). The adhesion, proliferation, and osteogenic differentiation ability of rat bone marrow mesenchymal cells (BMSCs) on different materials were assessed in vitro. Then the osseointegration capacity of Ti, Ti/Ti, Ta/Ti, and Straumann implants in canine mandible was evaluated with micro-CT, histological sections, and energy dispersive X-ray spectroscopy. These results demonstrated that micro-nanostructured, uneven, and granular tantalum coating was successfully prepared on titanium substrate by VPS with pore size ranging from 50 nm to 5 μm and thickness ranging from 80 to 100 μm. Tantalum coating revealed the highest surface potential, best hydrophilia, and most protein adsorption among Ta/Ti, Ti/Ti, and Ti. Furthermore, Ta/Ti surfaces significantly promoted the adhesion, proliferation, and osteogenic differentiation of BMSCs. In vivo, Ta/Ti implants displayed positive osseointegration capability associated with increased bone mineral density and formation of new bone around implants without tantalum particles released. Together, these findings indicate that tantalum-coated titanium dental implants may serve as a new type of dental implant.
Tantalum-incorporated hydroxyapatite coating on titanium implants: its mechanical and in vitro osteogenic properties

Abstract:<h3 class="Heading">Objective</h3><p class="Para">The fabrication of bioactive coatings on metallic implants to enhance osseointegration has become a topic of general interest in orthopedics and dentistry. Hydroxyapatite (HA) coating has been shown to induce bone formation and promote bone-implant integration. Unfortunately, poor mechanical performance has hindered this from becoming a favorable coating material. The majority of present studies have focused in incorporating different elements into HA coatings to improve mechanical properties. In recent years, tantalum (Ta) has received increasing attention due to its excellent biocompatibility and corrosion resistance. The aim of on the present study was to investigate the fabrication and biological performance of Ta-incorporated HA coatings.</p><h3 class="Heading">Methods</h3><p class="Para">Ta-incorporated HA coatings were fabricated using the plasma spray technique on a titanium substrate, and the surface characteristics and mechanical properties were examined. In addition, the effects of Ta-incorporated HA coatings on the biological behavior of mesenchymal stem cells (BMSCs) were investigated.</p><h3 class="Heading">Results</h3><p class="Para">Ta-incorporated HA coatings with microporous structure had higher roughness and wettability. In addition, the bonding strength of Ta/HA coatings with the substrate was substantially superior to HA coatings. Furthermore, Ta-incorporated HA coatings not only facilitated initial cell adhesion and faster proliferation, but also promoted the osteogenic differentiation of BMSCs.</p><h3 class="Heading">Conclusion</h3><p class="Para">These results indicate that the incorporation of Ta could improve mechanical performance and increase the osteogenic activity of HA coatings. Abstract:Plasma alloying technique capable of producing metallic coatings with metallurgical bonding has attracted much attention in dental and orthopedic fields. In this study, the effects of temperature and time of plasma tantalum (Ta) alloying technique on the mechanical, electrochemical, and osteoblastic properties of Ta coatings were systematically investigated. Ta coatings prepared at 800 °C possess better interfacial strengths than those prepared at 750 and 850 °C, and the interfacial strength increases with prolonged alloying time (30–120 min). At 800 °C, however, the increased proportion of the soft Ta deposition layer with alloying time in the whole coating impairs the surface mechanical properties of the entire coating, as convinced by decreased microhardness and wear resistance. Moreover, Ta coatings exhibit better corrosion resistance than the Ti6Al4V substrate in Dulbecco’s modified Eagle medium. The enhanced adhesion and extracellular matrix mineralization level of osteoblasts demonstrate the better cytocompatibility and osteogenic activity of the Ta coating. Ta30 (Ta coating prepared at 800 °C for 30 min) exhibits excellent mechanical, electrochemical, and osteoblastic behaviors and is promising in biomedical applications.

Abstract:Although titanium alloy (Ti–6Al–4V) has good mechanical and biological properties as a bone implant material, it still has defects of biologically inertness and poor antibacterial ability. In this study, a multi-target magnetron sputtering method was used to prepare Ta(Cu)–O composite films with three different ratios (Ta(Cu 10.82 )-O; Ta(Cu 5.68 )-O; Ta(Cu 3.43 )-O) on the surface of Ti–6Al–4V to solve the problems above. In vitro osteoblast proliferation experiments showed that the Ta(Cu)–O composite films significantly accelerated the proliferation, adhesion and spreading of osteoblasts, especially for Ta(Cu 3.43 )-O. And due to the excellent antibacterial activity of Cu, the Ta(Cu)–O composite films also showed good antibacterial effect. In conclusion, the results showed that the Ta(Cu)–O films can enhance the antibacterial properties of Ti–6Al–4V while accelerating the growth of osteoblasts, displaying great clinical potential in bone-implants applications.

Abstract:Bone defects caused by tumors, trauma, or other bone diseases are common in clinical practice. Titanium alloy (TC4) and tantalum (Ta) are the primary metallic materials for bone repair. However, TC4 frequently suffers from poor osseointegration and implant loosening, while the high cost of Ta limits its widespread use for reconstructing large bone defects. To address this issue, this research employs magnetron sputtering (MSP) to deposit a nano-thin amorphous Ta-coating on the 3D-printed TC4 implant. SEM examinations of compressed samples show robust interfacial bonding without shedding, while the presence of Ta within the porous structure indicates high coverage. High-resolution TEM analysis first reveals a distinct multilayer amorphous zone consisting of surface TaO x , Ta-coating, and hybrid TaO x and TiO x , along with an impact-refinement nanocrystalline TC4 zone. Insufficient energy leads to the formation of amorphous layers. Meanwhile, interfacial bonding is strengthened through the solid solution of oxygen and interfacial densification of amorphous-crystalline bonding and impact compression. Biological evaluations demonstrate that the Ta-coating exhibits superior cell proliferation and bone ingrowth effects compared to TC4 and pure Ta implants, primarily attributed to the amorphous oxide composition on the surface. This study offers novel, cost-efficient, and excellent biological performance materials for bone defect implants.

Herein, we proposed fabricating a novel fluoride-doped tantalum oxide (F-TaO x ) coating on titanium by one-step reactive sputtering deposition to endow implants with superior antibacterial and osteogenic ability. By altering the fluorine and oxygen loading amount, the coating's physicochemical properties, corrosion resistance, antibacterial and osteogenic activities are tuned simultaneously. All coatings exhibit nano-morphology, superhydrophilicity and improved surface hardness. Despite containing fluoride, the F-TaO x coating produced with fluorine and oxygen dual-deposition still shows enhanced corrosion resistance due to the protection of existing metal oxidizes. The antibacterial test reveals that the F-TaO x coating exhibits long-lasting and superior antibacterial properties. The microenvironment acidification by F - , the excessive ROS generation and the disruption of ATP by F - , TiF 3 and Ta 2 O 5 , and the repulsive force by superhydrophilic surface synergistically result in bacterial death. The cell experiments indicate that the F-TaO x coating shows good cytocompatibility, promote cell spread and osteogenic differentiation by enhancing ALP activities and ECM mineralization. This work provides a promising strategy for the surface functionalization of metallic implants to produce dual anti-infection and osteogenesis effects for orthopedic and dental applications.

Abstract:Osteogenic differentiation and antibacterial property are two-core requirements for bone implants, but typically they are not compatible. Zinc (Zn) as a common bioactive material is widely used in the medical field. A balance between antibacterial and osteogenic activity can be achieved at appropriate Zn concentration. In this study, the TNTZ/Zn micro/nano-composites were prepared by doping Zn nanoparticles onto Ti-35Nb-2Ta-3Zr (TNTZ) surface through the friction stir processing (FSP) technique. The results of material characterization revealed the homogeneous distribution of Zn, the presence of α′′ martensite, and the increase in microhardness in the stir zone. Due to the solid metallurgical bonding between Zn NPs and substrate, Zn ions were released slowly and may be biologically responsive through body fluid corrosion. In terms of biological activity, the expression of adhesion-related protein vinculin and osteogenic differentiation genes were up-regulated by TNTZ/Zn, which improved the early adhesion and osteogenic differentiation of bone marrow stromal stem cells. Besides, TNTZ/Zn presented excellent antibacterial properties, and direct contact with the fine grain surface containing Zn NPs may be the dominant mechanism. TNTZ/Zn micro/nano-composites have excellent mechanical properties, osteogenic differentiation and antibacterial properties, providing an efficient strategy for titanium alloy surface modification and showing tremendous potential in dental applications.

Abstract:Titanium and titanium alloy materials are often made as medical implants. Surface properties of titanium medical implants play decisive roles for antibacterial and biological response and tissue biocompatibility. In this study, cathode arc evaporation (CAE) technology was used to deposit TiNb–Zr–Ta and nitrogen-doped TiNb–Zr–Ta coatings with nanostructures on the Ti–6Al–4V dental abutment screws. The deposited coatings were then oxidized at high temperature to produce different oxides on the surface of the coatings. The mechanical properties , antibacterial properties and biocompatibilities of the coatings were analyzed. The effect of coatings on the abutment screw loosening and removal torque between the abutment screw and the pure titanium implant fixture was studied by removal torque measurement. In addition, Staphylococcus aureus (S. aureus) was used for evaluating the antibacterial properties of coatings, and mouse fibroblasts (L929) were used for ISO 10993-5 cytotoxicity analyses to determine whether it had good biocompatibility. Nanolayered structures and multiphases of polycrystalline TiNb–Zr–Ta were obtained by co-deposition of Ta , Zr and TiNb. When small content of N was introduced, the TiNb–Zr–Ta(N) showed a polycrystalline TiNb–Zr–Ta where N atoms entered the metallic lattice at interstitial sites. After oxidation, oxide phases of Ta 2 O 5 , TiO 2 , Nb 2 O 5 and ZrO 2 were formed on the surface of TiNb–Zr–Ta and TiNb–Zr–Ta(N). Ta 2 O 5 was obtained on the surface of the coating prior to other oxides such as TiO 2 , Nb 2 O 5 and ZrO 2 . The deposited TiNb–Zr–Ta and TiNb–Zr–Ta(N) coatings had high contact angle and exhibited a hydrophobic feature, while the annealed TiNb–Zr–Ta–O and TiNb–Zr–Ta(N)–O showed hydrophilicity. The deposited TiNb–Zr–Ta had similar hardness and Young's modulus values to that of the uncoated Ti–6Al–4V. When N was doped, the TiNb–Zr–Ta(N) had the highest hardness and Young's modulus. The hardness of the annealed TiNb–Zr–Ta(N) coating only decreased slightly due to the small oxide layer of ∼140 nm. Compared with the uncoated Ti–6Al–4V, TiNb–Zr–Ta serial coatings deposited on abutment screws had higher removal torques, which prevented the abutment screw of dental implant from loosening after long-term use. The biocompatibility of TiNb–Zr–Ta serial coatings was similar to that of the uncoated Ti–6Al–4V. In addition, the oxidized TiNb–Zr–Ta and TiNb–Zr–Ta(N) coatings had excellent antibacterial properties. It is speculated that the resulting Ta 2 O 5 of both coatings may inhibit bacterial growth and improve the antibacterial effect. Based on the above advantages, the TiNb–Zr–Ta serial coatings are expected to have the potential to be applied to biomedical implants.

Abstract:In this work, TiN-incorporated graphite-like amorphous carbon (alpha-GLC) bioceramic films were successfully fabricated on Ti alloy (Ti6Al4V) using the magnetron sputtering technique. The biocompatibility and bio-tribological properties of the TiN/alpha-GLC-coated Ti alloy were evaluated. Based on the clinical application, the microstructure, biocompatibility, histomorphological observation, and mechanical properties were characterized by scanning electron microscopy, transmission electron microscopy, X-ray diffraction, nanoindentation test, and in vitro and in vivo tests including MTT, cell adhesion test, and slice observation of hard tissues. These techniques confirmed that there was strong bonding between the Ti alloy and the TiN/alpha-GLC film. In addition, face-centered cubic TiN and hexagonal close-packed Ti were clearly detected in the TiN/alpha-GLC films. A partial nanocomposite structure, comprised of homogeneous nanoclusters and crystalline particles, was detected in the amorphous matrix. Cell adhesion and MTT test results indicated that the TiN/alpha-GLC film could promote cell attachment and proliferation. Phase composition and the film morphology were determined to be the crucial factors that contributed to the improved biocompatibility. The biotribological property test showed that the TiN/alpha-GLC film with a Ti content of 7.8 at% had the lowest coefficient of friction (COF) of 0.076 and the lowest wear rate of 1.76 x 10(-6) mm(3) (N m)(-1). The low COF was attributed to the superior mechanical properties of the TiN/alpha-GLC composite film. The TiN/alpha-GLC-coated Ti alloy implants were placed subcutaneously into rabbits for different time periods, and the tissue response decreased significantly as compared to that of the pure alpha-GLC film. The excellent biocompatibility and function reliability of the TiN/alpha-GLC bioceramic films showed that they are potential candidates for bone tissue implants.
Microstructure and biocompatibility of porous-Ta/Ti-6Al-4 V component produced by laser powder bed fusion for orthopedic implants

Abstract:Tantalum is attracting more attention as an outstanding orthopedic implant material due to its excellent corrosion resistance and biocompatibility. However, the high melting point and strong oxygen affinity of tantalum make its preparation more difficult and costly. In addition, the high cost of raw materials also limits the wider application of tantalum. To break these limitations, porous tantalum coatings with different pore sizes were successfully prepared on Ti-6Al-4 V base by Laser Powder Bed Fusion, constructing an integral porous-Ta/Ti-6Al-4 V component with excellent bioactivity and osteointegration capacity. A strong metallurgical bonding was formed between tantalum and Ti-6Al-4V, and the bonding strength is more than 460 MPa. The porous tantalum coating is designed as diamond-like structure with strong symmetry. The porosity of tantalum coatings is 70%, and the pore size ranges from 447 to 865 μm. Its deformation characteristics are extremely similar to those of natural cancellous bone. The average compressive yield strength of the porous Ta coatings ranges from 5.8 to 24. 2 MPa. And the average compressive elastic modulus of porous Ta coatings (0.6–1.5 GPa) was close to that of cancellous bone. Corrosion resistance tests proved that the Ta-coated Ti-6Al-4 V alloy had an excellent chemical stability in simulated body fluid. In vitro tests showed that Mouse calvarial preosteoblasts presented a good adhesion and growth behavior on porous Ta coatings. Therefore, it is preliminarily determined that the customized porous-Ta/Ti-6Al-4 V component by Laser powder bed fusion can be a candidate material for orthopedic implantation.

Abstract:With superior bone growth and integration abilities, porous tantalum (Ta) implants prepared by depositing Ta coating on foam carbon have been widely used in clinical for two decades. To solve their high cost and high weight disadvantages, the deposition of thin Ta coating on porous titanium (Ti) alloys substrate by chemical vapor deposition (CVD) is expected to be a promising strategy. However, until now realizing the novel strategy is still a big challenge. In this study, by the TaCl5-H2 reaction, we investigated the preparation of Ta coating on Ti6Al4V (TC4) matrix by CVD. The effects of reaction temperature and gas flow on the microstructure, composition, and properties of the Ta coating were systematically investigated. The results indicated that Ta coatings about 5 μm with high quality was successfully prepared on plane and porous TC4 for the first time, and the reaction temperature played a more important role than the gas flow. The optimal preparation parameters were determined to be 950 °C and 200 sccm. Under the condition, Ta coating with the fully α-Ta phase, high purity, high uniformity and compactness was achieved. The Ta coating exhibited excellent binding force, corrosion resistance, hydrophilicity. Through eliminating the toxic V ions dissolution from TC4, the Ta coating also obtained excellent cell compatibility, which is meaningful for porous TC4 implants widely used in clinic with very high specific surface area. Therefore, this research evidenced that, by CVD, the novel thin Ta coated porous TC4 implants with lower cost, lower weight and higher biocompatibility could be achieved.

Abstract:As a biomaterial for bone repair, the application of polyetheretherketone (PEEK) has been limited by its poor wear resistance and cell behaviour. Inspired by the double 'soft-hard' phase structure, tantalum (Ta)/PEEK composite coating was fabricated on a titanium surfac e . All composite coatings had a lower friction coefficient and wear resistance under fetal bovine serum conditions. The adhesion, proliferation, and osteogenic differentiation of rat bone osteoblastic cells (MC3T3-E1) and mononuclear macrophage cells (RAW264.7) on the Ta/PEEK composite coating was enhanced. In particular, composite coatings effectively reduced inflammatory markers, with cytokines TGF-β and IL-10 released. As a result, titanium alloys with Ta reinforced PEEK coatings could exhibit improved bioactivity and tribology.

Abstract:Titanium (Ti) alloys are widely utilized in orthopedics owing to their excellent mechanical properties and biocompatibility. To improve their resistance to corrosion and ion release properties, substrates of Ti alloy have been produced employing powder metallurgy by adding alloying elements (Si and Nb) at 5wt.% along with CP-Ti. Two torch flame sprays have been utilized for coating the Ti-5Nb and Ti-5Si alloys with two kinds of nanocoating: HAp+25%SiC (type-A) and ZSM5+25%ZrO2 (type-B). These nanocoating combinations represented bioactive and bioinert to combine the biological and mechanical properties of the implant surface. Different tests and characterization techniques have been carried out, including SEM, XRD, AFM, AAS, hardness, adhesion strength, and corrosion resistance. The results manifested that the coatings (types A and B) improved the properties of Ti alloys; however, ZSM5+25%ZrO2 has better properties than type-A in terms of less porosity, higher crystallinity%, higher hardness, higher adhesion strength, lower corrosion rate, and less Ti ions release. Comparing the results of the two Ti alloys, Ti-5Si has higher hardness, corrosion resistance, and less ionic release than the Ti-5Nb alloy. Hence, the Ti-5Si coated by ZSM5+25%ZrO2 (B coated Ti-5Si) is the best sample in this study.
