Abstract:High-resolution imaging with compositional and chemical sensitivity is crucial for a wide range of scientific and engineering disciplines. Although synchrotron X-ray imaging through spectromicroscopy has been tremendously successful and broadly applied, it encounters challenges in achieving enhanced detection sensitivity, satisfactory spatial resolution, and high experimental throughput simultaneously. In this work, based on structured illumination, we develop a single-pixel X-ray imaging approach coupled with a generative image reconstruction model for mapping the compositional heterogeneity with nanoscale resolvability. This method integrates a full-field transmission X-ray microscope with an X-ray fluorescence detector and eliminates the need for nanoscale X-ray focusing and raster scanning. We experimentally demonstrate the effectiveness of our approach by imaging a battery sample composed of mixed cathode materials and successfully retrieving the compositional variations of the imaged cathode particles. Bridging the gap between structural and chemical characterizations using X-rays, this technique opens up vast opportunities in the fields of biology, environmental, and materials science, especially for radiation-sensitive samples.

Abstract:Resolving morphological chemical phase transformations at the nanoscale is of vital importance to many scientific and industrial applications across various disciplines. The TXM-XANES imaging technique, by combining full field transmission X-ray microscopy (TXM) and X-ray absorption near edge structure (XANES), has been an emerging tool which operates by acquiring a series of microscopy images with multi-energy X-rays and fitting to obtain the chemical map. Its capability, however, is limited by the poor signal-to-noise ratios due to the system errors and low exposure illuminations for fast acquisition. In this work, by exploiting the intrinsic properties and subspace modeling of the TXM-XANES imaging data, we introduce a simple and robust denoising approach to improve the image quality, which enables fast and high-sensitivity chemical imaging. Extensive experiments on both synthetic and real datasets demonstrate the superior performance of the proposed method.

Abstract:Abstract We report multimodal scanning hard x-ray imaging with spatial resolution approaching 10 nm and its application to contemporary studies in the field of material science. The high spatial resolution is achieved by focusing hard x-rays with two crossed multilayer Laue lenses and raster-scanning a sample with respect to the nanofocusing optics. Various techniques are used to characterize and verify the achieved focus size and imaging resolution. The multimodal imaging is realized by utilizing simultaneously absorption-, phase-, and fluorescence-contrast mechanisms. The combination of high spatial resolution and multimodal imaging enables a comprehensive study of a sample on a very fine length scale. In this work, the unique multimodal imaging capability was used to investigate a mixed ionic-electronic conducting ceramic-based membrane material employed in solid oxide fuel cells and membrane separations (compound of Ce 0.8 Gd 0.2 O 2− x and CoFe 2 O 4 ) which revealed the existence of an emergent material phase and quantified the chemical complexity at the nanoscale.

Micro-XANES: Chemical Contrast in the Scanning Transmission X-ray Microscope

Abstract:The Scanning Transmission X-ray Microscope (STXM) on the X1A undulator beamline, at the National Synchrotron Light Source, BNL, has been used for imaging various wet, unstained biological samples at 55 nm Rayleigh resolution. The microscope's Modulation Transfer Function was measured and agrees well with theoretical calculations, confirming the performance of the microscope as designed. The microscope is operated mainly in direct imaging mode where x-rays of fixed energy are diffractively focused to a microprobe and the sample is scanned in two dimensions. By varying the x-ray energy while keeping the beam focused to one spot on the sample enables us to get localized chemical composition of the sample from the absorption spectra in an area smaller than 0.2 by 0.2 micron at present. The spatial distribution of the chemical compositions can be obtained with a spacial resolution of 55 nm by imaging at absorption maxima of specific chemical bonds.The chemical sensitivity of the X-ray Absorption Near Edge Structure (XANES) provides a new contrast mechanism for organic system imaging.

Abstract:Measuring the three-dimensional (3D) distribution of chemistry in nanoscale matter is a longstanding challenge for metrological science. The inelastic scattering events required for 3D chemical imaging are too rare, requiring high beam exposure that destroys the specimen before an experiment completes. Even larger doses are required to achieve high resolution. Thus, chemical mapping in 3D has been unachievable except at lower resolution with the most radiation-hard materials. Here, high-resolution 3D chemical imaging is achieved near or below one nanometer resolution in a Au-Fe$_3$O$_4$ metamaterial, Co$_3$O$_4$ - Mn$_3$O$_4$ core-shell nanocrystals, and ZnS-Cu$_{0.64}$S$_{0.36}$ nanomaterial using fused multi-modal electron tomography. Multi-modal data fusion enables high-resolution chemical tomography often with 99\% less dose by linking information encoded within both elastic (HAADF) and inelastic (EDX / EELS) signals. Now sub-nanometer 3D resolution of chemistry is measurable for a broad class of geometrically and compositionally complex materials.

Abstract:Label-free imaging of nanoscale targets with intrinsic properties is crucial for chemistry, physics, and life science to unveil the underlying mechanisms. Plasmonic imaging techniques are particularly attractive because they allow real-time imaging, providing insights into nanoscale detection and nanocatalysis. Here, we present a high-resolution plasmonic imaging method that is capable of imaging nanomaterials with high morphological fidelity and high throughput. We demonstrate that this approach allows for high-resolution plasmonic imaging of various nanomaterials ranging from nanoparticles and nanowires to two-dimensional nanomaterials and accurate tracking of the interfacial dynamics of nanoparticles. Given the experimental simplicity and capacity for label-free and real-time imaging of nanomaterials with high spatial resolution and high throughput, this approach can serve as a promising platform for characterizing nanomaterials at the single-particle level.

Abstract:Non-destructive nano-imaging of the internal structure of solid matter is only feasible using hard X-rays due to their high penetration. The highest resolution images are achieved at synchrotron radiation sources (SRF), offering superior spectral brightness and enabling methods such as X-ray ptychography delivering single-digit nm resolution. However the resolution or field of view is ultimately constrained by the available coherent flux. To address this, the beam's incoherent fraction can be exploited using multiple parallel beams in an approach known as X-ray multibeam ptychography (MBP). This expands the domain of X-ray ptychography to larger samples or more rapid measurements. Both qualities favor the study of complex composite or functional samples, such as catalysts, energy materials, or electronic devices. The challenges of performing ptychography at high energy and with many parallel beams must be overcome to extract the full advantages for extended samples while minimizing beam attenuation. Here, we report the application of MBP with up to 12 beams and at photon energies of 13 and 20 keV. We demonstrate performance for various samples: a Siemens star test pattern, a porous Ni/\ce{Al2O3} catalyst, a microchip, and gold nano-crystal clusters, exceeding the measurement limits of conventional hard X-ray ptychography without compromising image quality.

Abstract:Efforts to map atomic-scale chemistry at low doses with minimal noise using electron microscopes are fundamentally limited by inelastic interactions. Here, fused multi-modal electron microscopy offers high signal-to-noise ratio (SNR) recovery of material chemistry at nano- and atomic- resolution by coupling correlated information encoded within both elastic scattering (high-angle annular dark field (HAADF)) and inelastic spectroscopic signals (electron energy loss (EELS) or energy-dispersive x-ray (EDX)). By linking these simultaneously acquired signals, or modalities, the chemical distribution within nanomaterials can be imaged at significantly lower doses with existing detector hardware. In many cases, the dose requirements can be reduced by over one order of magnitude. This high SNR recovery of chemistry is tested against simulated and experimental atomic resolution data of heterogeneous nanomaterials.

Abstract:Improving the spatial and spectral resolution of 2D X-ray near-edge absorption structure (XANES) has been a decade-long pursuit to probe local chemical reactions at the nanoscale. However, the poor signal-to-noise ratio in the measured images poses significant challenges in quantitative analysis, especially when the element of interest is at a low concentration. In this work, we developed a post-imaging processing method using deep neural network to reliably improve the signal-to-noise ratio in the XANES images. The proposed neural network model could be trained to adapt to new datasets by incorporating the physical features inherent in the latent space of the XANES images and self-supervised to detect new features in the images and achieve self-consistency. Two examples are presented in this work to illustrate the model's robustness in determining the valence states of Ni and Co in the LiNi x Mn y Co 1-x-y O 2 systems with high confidence.

Abstract:The Hard X-ray Nanoprobe (HXN) beamline at NSLS-II has been designed and constructed to enable imaging experiments with unprecedented spatial resolution and detection sensitivity. The HXN X-ray Microscope is a key instrument for the beamline, providing a suite of experimental capabilities which includes scanning fluorescence, diffraction, differential phase contrast and ptychography utilizing Multilayer Laue Lenses (MLL) and zoneplate (ZP) as nanofocusing optics. Abstract:We demonstrate a technique that allows highly surface sensitive imaging of nanostructures on planar surfaces over large areas, providing a new avenue for research in materials science, especially for \textit{in situ} applications. The capabilities of hard X-ray grazing incidence ptychography combine aspects from imaging, reflectometry and grazing incidence small angle scattering in providing large field-of-view images with high resolution transverse to the beam, horizontally and along the surface normal. Thus, it yields data with resolutions approaching electron microscopy, in two dimensions, but over much larger areas and with a poorer resolution in the third spatial dimension, along the beam propagation direction. Similar to grazing incidence small angle X-ray scattering, this technique facilitates the characterization of nanostructures across statistically significant surface areas or volumes within potentially feasible time frames for \textit{in situ} experiments, while also providing spatial information.

Abstract:Structure and composition at the nanoscale determine the behavior of biological systems and engineered materials. The drive to understand and control this behavior has placed strong demands on developing methods for high resolution imaging. In general, the improvement of three-dimensional (3D) resolution is accomplished by tightening constraints: reduced manageable specimen sizes, decreasing analyzable volumes, degrading contrasts, and increasing sample preparation efforts. Aiming to overcome these limitations, we present a non-destructive and multiple-contrast imaging technique, using principles of X-ray laminography, thus generalizing tomography towards laterally extended objects. We retain advantages that are usually restricted to 2D microscopic imaging, such as scanning of large areas and subsequent zooming-in towards a region of interest at the highest possible resolution. Our technique permits correlating the 3D structure and the elemental distribution yielding a high sensitivity to variations of the electron density via coherent imaging and to local trace element quantification through X-ray fluorescence. We demonstrate the method by imaging a lithographic nanostructure and an aluminum alloy. Analyzing a biological system, we visualize in lung tissue the subcellular response to toxic stress after exposure to nanotubes. We show that most of the nanotubes are trapped inside alveolar macrophages, while a small portion of the nanotubes has crossed the barrier to the cellular space of the alveolar wall. In general, our method is non-destructive and can be combined with different sample environmental or loading conditions. We therefore anticipate that correlative X-ray nano-laminography will enable a variety of in situ and in operando 3D studies.

Abstract:Modern laser-based XUV light sources provide very high photon fluxes which have previously only been available at large scale facilities. This allows high-performance XUV nanoscale imaging to be implemented in a table-top manner, and thus qualifies XUV imaging as a novel imaging technique complementing electron and visible-light microscopy. This article presents the current state-of-the-art in table-top XUV light sources and matched coherent imaging schemes. Selected experiments demonstrate the unique capabilities of XUV imaging—namely, nanoscale (sub-20 nm) resolution, single shot imaging, imaging of extended samples and 3D imaging of μm-sized objects. In addition, future prospects will be discussed, including scaling to few-nm resolution, extension to the soft x-ray spectral region, chemically-specific imaging at absorption edges and time-resolved imaging on femtosecond time-scales.

Abstract:X-ray spectroscopy has nanometer spatial resolution, long penetration depth and excellent elemental specificity. The power of synchrotron-based X-ray microscopy (XRM) has been demonstrated in assessing the structure and function of cells at the subcellular level [<a class="link link-ref link-reveal xref-bibr" href="">1</a>]. However, currently available contrast agents or probes for XRM, such as metal or semiconductor nanoparticles, generally lack specificity and biocompatibility and thus limit its utility [<a class="link link-ref link-reveal xref-bibr" href="">2</a>]. Newly developed genetic tags for electron microscopy (EM) provide EM contrast in cellular compartments <em>in situ</em> and enable intracellular specific protein imaging and spatially resolved proteomic mapping [<a class="link link-ref link-reveal xref-bibr" href="">3</a>,<a class="link link-ref link-reveal xref-bibr" href="">4</a>], but use of EM for whole cell imaging remains challenging because of low penetration capability.

Abstract:Hard X‐ray multibeam ptychography paves the path toward high‐resolution, non‐destructive 3D imaging of mm‐sized samples. The present study describes the achievement of imaging of 0.1mm+ samples using up to 12 parallel uniquely coded X‐ray beams with energies of up to 20 keV. Cutting‐edge laser technology enables 3D printing of a high aspect ratio lens array for hard X‐ray parallel focusing. Hard X‐rays are needed for non‐destructive nano‐imaging of solid matter. Synchrotron radiation facilities (SRF) provide the highest‐quality images with single‐digit nm resolution using advanced techniques such as X‐ray ptychography. However, the resolution or field of view is ultimately constrained by the available coherent flux. Abstract:An in-house designed transmission X-ray microscopy (TXM) instrument has been developed and commissioned at beamline BL18B of the Shanghai Synchrotron Radiation Facility (SSRF). BL18B is a hard (5-14 keV) X-ray bending-magnet beamline recently built with sub-20 nm spatial resolution in TXM. There are two kinds of resolution mode: one based on using a high-resolution-based scintillator-lens-coupled camera, and the other on using a medium-resolution-based X-ray sCMOS camera. Here, a demonstration of full-field hard X-ray nano-tomography for high-Z material samples (e.g. Au particles, battery particles) and low-Z material samples (e.g. SiO-2 powders) is presented for both resolution modes. Sub-50 nm to 100 nm resolution in three dimensions (3D) has been achieved. These results represent the ability of 3D non-destructive characterization with nano-scale spatial resolution for scientific applications in many research fields.

Abstract:Spatial resolution defines the physical limit of microscopes for probing biomolecular localization and interactions in cells. Whereas synchrotron-based X-ray microscopy (XRM) represents a unique approach for imaging a whole cell with nanoscale resolution due to its intrinsic nanoscale resolution and great penetration ability, existing approaches to label biomolecules rely on the use of exogenous tags that are multi-step and error-prone. Here, we repurpose engineered peroxidases as genetically encoded X-ray-sensitive tags (GXET) for site-specific labeling of protein-of-interest in mammalian cells. We find that 3,3'-diaminobenzidine (DAB) polymers that are in-situ catalytically formed by fusion-expressed peroxidases are visible under XRM. Using this new tag, we imaged the protein location associated with the alteration of a DNA-methylation pathway with an ultra-high resolution of 30 nanometers. Importantly, the excellent energy resolution of XRM enables multicolor imaging using different peroxidase tags. The development of GXET enlightens the way to nanoscopic imaging for biological studies.

Abstract:Full-field transmission X-ray microscopy (TXM) in conjunction with X-ray absorption near edge structure (XANES) spectroscopy provides two-dimensional (2D) or three-dimensional (3D) morphological and chemical-specific information within samples at the tens of nanometer scale. This technique has a broad range of applications in materials sciences and battery research. Despite its extensive applicability, 2D XANES imaging is subject to the disadvantage of information overlap when the sample thickness is uneven. 3D XANES imaging combines 3D TXM with XANES to obtain 3D distribution information on chemical states. A 3D XANES imaging method has been established at the Shanghai Synchrotron Radiation Facility (SSRF) and has been used to characterize the structure and chemical state of commercial LiNixCoyMnzO2 (NCM, x + y + z = 1) battery powder materials. The imaging results provide a visual representation of the 3D chemical state information of the particles with depth resolution, allowing for the direct observation of 3D nickel oxidation.
