Abstract:An integrated approach to enhance and maintain high anammox activity and abundance in an upflow anaerobic sludge blanket (UASB) treating low strength wastewater under moderate and low temperatures was developed. A quantitative PCR assay showed the abandance of anammox bacteria to be 1.68±0.08×109copies/ml in mixed liquor when the temperature was 30°C and was maintained at the level of 1.93±0.41×109copies/ml in mixed liquor at 16°C. A nitrogen removal rate (NRR) of up to 5.72kgN/m3/d was achieved with a hydraulic retention time (HRT) of 0.12h at 30°C, while nitrite and ammonium removal efficiencies were 94.35% and 92.81%, respectively. NRR decreased with a decrease in temperature and was maintained at 2.28kgN/m3/d with an HRT of 0.28h when at 16°C, while nitrite and ammonium removal efficiencies were 92.31% and 78.45%, respectively. The emission rate of the greenhouse gas N2O was below 0.006% of the NRR in the anammox UASB reactor treating low strength wastewater.

Abstract:The impact of temperature on nitrification rate was evaluated in this study through experimental tests, mathematical modeling and sensitivity analysis. The results show that the impact of temperature on fixed film nitrification rate is less significant than that predicted by the van't Hoff–Arrhenius equation. In a fixed film biofilter, the impact of temperature on nitrification rate due to DO (dissolved oxygen) limitation is different from that due to TAN (total ammonia nitrogen) limitation. Sensitivity analysis indicated that a temperature increment at 20°C resulted in nitrification rate increase of 1.108% per °C and 4.275% per °C under DO and TAN limited conditions, respectively. Diffusion mass transport plays an important role in fixed film nitrification processes. Consequently, the effect of temperature on nitrification rate due to bacterial growth rate change in fixed film processes is greatly reduced compared with that of suspended growth processes. When oxygen is limited, the decrease in saturation DO concentration as temperature increases results in a negative temperature impact upon the nitrification rate.

Abstract:Effects of dissolved oxygen on accumulation rate of nitrite under different temperatures were investigated.The results showed significant different effects under temperatures of 30,25,and 20 ℃.Meanwhile,it was observed that the initial accumulation of nitrite,as an important initiator of the partial nitrification process,had a certain degree of particularity,where the pH value was an important factor for the initial accumulation of nitrite.The definitions of the "complying point" and two types of "least ammonia point" were also explained,and analyzed the relationship between the three points.It was able to achieve efficient partial nitrification on the premise that the effluent ammonia was in compliance with national standards.

Abstract:Partial nitrification to nitrite is technically feasible and economically favourable, especially when wastewaters contained high ammonium concentrations or low C/N ratios. Partial nitrification can be obtained by selectively inhibiting nitrite-oxidizing bacteria (NOB) through appropriate regulation of the pH, temperature and dissolved oxygen (DO) concentrations. The effect of pH, DO levels and temperature on ammonia oxidation rate and nitrite accumulation was investigated in order to determine the optimal conditions for partial nitrification of synthetic wastewater with high ammonia concentration. The experiments performed at low DO levels to lower the total oxygen needed in the nitrification step, which means great saving in aeration. During the start-up stage pH and DO were set at 7.0–7.4 and 0.5mg/l, respectively. The reactor was operated until complete partial nitrification was achieved. The effect of pH, DO on partial nitrification was studied, as pH was kept at 6.5, 7.5, 8.5, 9.5 and DO at 0.5±0.2, 1.5±0.2 and 2.5±0.2mg/l, and temperature at 30°C. The influence of temperature on ka value was studied by keeping pH=7.5, DO=1.5mg/l and temperature was controlled at 12, 20 and 30°C, respectively. The results showed that partial nitrification to nitrite was steadily obtained and the optimal operational parameters were pH=7.5, DO=1.5mg/l, T=30°C based on ammonia oxidation rate and nitrite accumulation rate. The maximum ka was achieved and to be 115.1×10−3mg NH4+–N(mg VSSh)−1 under this condition.

Abstract:Low water temperatures and ammonium concentrations pose challenges for anammox applications in the treatment of low C/N municipal wastewater. In this study, a 10 L-water bath sequencing batch reactor combing biofilm and suspended sludge was designed for low C/N municipal wastewater treatment. The nitrogen removal performance via partial nitrification anammox-(endogenous) denitrification anammox process was investigated with anaerobic-aerobic-anoxic mode at low temperatures and dissolved oxygen (DO). The results showed that with the decrease of temperature from 30 to 15°C, the influent and effluent nitrogen concentrations and nitrogen removal efficiencies were 73.7±6.5 mg/L, 7.8±2.8 mg/L, and 89.4%, respectively, with aerobic hydraulic retention time of only 6 h and DO concentration of 0.2-0.5 mg/L. Among that, the stable anammox process compensated for the inhibitory effects of the low temperatures on the nitrification and denitrification processes. Notably, from 30 to 15°C, the anammox activity and relative abundance of the dominant Brocadia genus were increased from 39.7 to 45.5 mgN/gVSS/d and 7.3 to 12.0%, respectively; the single gene expression level of the biofilm increased 9.0 times. The anammox bacteria showed a good adaptation to temperatures reduction. However, nitrogen removal by anammox was not improved by increasing DO (≥ 4 mg/L) at 8-4°C. Overall, the results of this study demonstrate the feasibility of the mainstream anammox process at low temperatures.

Abstract:In this study, the start of partial nitrification (activated sludge)-anaerobic ammonia oxidation (immobilized fillers)-denitrification (immobilized fillers) and stable nitrogen removal in the treatment of rare earth element tailings wastewater were explored. The results showed that, at 25–30 °C, partial nitrification could be rapidly established by controlling the combination of average free ammonia concentration (25.4–60.2 mg/L) and dissolved oxygen concentration (0.5 mg/L). Adopting the stepwise substrate (three stages) and shortening hydraulic retention time (HRT) methods, the total nitrogen removal rate of anaerobic ammonia oxidation (anammox) immobilized fillers reached up to 0.99 kg N•(m 3 •d) −1 . Adopting the C/N ratio of 4.5:1 and shorter HRT methods, the total nitrogen removal rate of denitrification immobilized fillers reached up to 1.07 kg N•(m 3 •d) −1 . The activities of the anammox and denitrification immobilized fillers were increased by 8.39 and 4.46 times, respectively. At steady state operation, the average effluent ammonia nitrogen concentration was 3.01 mg/L, with an average effluent total nitrogen concentration of 5.11 mg/L, which met the requirements of the "Emission Standard for Pollutants from Rare Earth Industry" GB 26451–2011. High-throughput sequencing technology showed that the relative abundance ratios of Nitrosomonas and Nitrospira changed from 0.36:1–108.05:1. The dominant bacterial genera of the anammox immobilized fillers was Candidatus Kuenenia , and that of denitrification immobilized fillers were Pseudomonas and Thauera . This process provides a new technical approach for rare earth element tailings wastewater treatment, with high significance in engineering.

Abstract:In this paper, the bio-activity of activated sludge was investigated using Oxygen Uptake Rate (OUR) as an indicator. The activity of ammonium oxidation and COD removal of both attached- and suspended-growth sludge was determined under different temperature and organic loading rates. The experimental results indicated that at 5 degrees C. the nitrification activity was about 60% of that at 18 degrees C. The nitrification activity of attached sludge was 20-30% lower in comparison with suspended sludge. The denitrification rate of sludge in zones I and II was 0.44 mg NO3-/g MLSS.h and 0.63 mg NO3-/g MLSS.h, respectively, which revealed that the denitrification activity of sludge-II was higher compared with sludge-I, the results also indicated that the accumulation of nitrite was very low.

Abstract:Dissolved organic nitrogen (DON) formed by microbial metabolism in wastewater treatment processes adversely impacts wastewater reuse and receiving waters quality, and microbial metabolism is greatly influenced by temperatures. However, little is known about the effect of microorganisms on DON and bioavailable DON (ABDON) formation under low temperatures. In this study, six reactors were operated at low (8 °C and 15 °C) and room (25 °C) temperatures to evaluate the relationship between microbial activity, microbial communities, and DON and ABDON. Results showed that DON and ABDON concentrations significantly increased at low temperatures (p < 0.05, t-test). DON formation was significantly correlated to microbial activity only, with adenosine triphosphate (negative, r = -0.64) and polysaccharide (positive, r = 0.61) as key indicators; however, ABDON formation was influenced by both microbial activity (polysaccharide > triphenyltetrazolium chloride-dehydrogenases > adenosine triphosphate) and microbial community structure. Short-term tests using the biomass from six reactors were performed at room temperature to further validate the relationship. The distinct differences between these results provide a basis for different strategies on reducing effluent DON and ABDON under low temperatures.
Effect of Temperature on Fungal Nitrification in Simulated In-Situ Aeration of Aged MSW Landfill.

Abstract:Fungal nitrification is one kind of heterotrophic nitrification that involves certain species of fungi promoting the transformation of organic nitrogen and ammonia nitrogen to nitrite/nitrate. In this study, simulated aerated landfill reactors (SALRs) were constructed to investigate fungal nitrification in aged municipal solid refuse, with a focus on understanding the effect of temperature on the performance of fungal nitrification as well as fungal contribution to ammonia nitrogen transformation. Different nitrogen metabolism patterns have been observed in the system with fungi only (SALRF) and complete microbial consortium, i.e., bacteria + fungi (SALRC). At a temperature of 35 °C, autotrophic nitrification dominated the ammonia nitrogen transformation, while fungal nitrification did not significantly contribute to ammonia removal. However, at elevated temperatures (i.e., 45 °C and 55 °C), fungi played a crucial role in ammonia transformation through fungal assimilation and fungal nitrification, with bacterial function suppressed. Furthermore, 45 °C was found to be the optimal temperature for fungal nitrification, exhibiting the highest nitrification rate (13.98 mg L−1 d−1) which accounted for 49.80% of total nitrification rate in the aerated landfill. High throughput sequencing revealed reshaping of fungal community in response to temperature variation. The abundance of Aspergillus fumigatus, with a relative abundance ranging from 67.13% to 92.71% at elevated temperatures, suggested its significant potential for fungal nitrification. These findings have implications for the promotion of nitrogen cycle through strengthening fungal nitrification in aerated landfill sites which often operate at high temperatures.
Deciphering the effect of temperature reduction on the biofilm in partial nitritation-anammox system: Insights from microbial community to metabolic pathways

Abstract:The concept of partial nitritation-anammox (PN/A) process has been proposed for many years, however seasonal temperatures and low ammonia concentrations under mainstream conditions always challenge the application in practical engineering. Thus, we operated a PN/A moving bed biofilm reactor (PN/A-MBBR) for treating 50 mg-NH4/L wastewater at various temperatures of 35 °C, 25 °C, and 15 °C. The effect of temperature reduction on biofilm system was investigated from process performance, microbial community and activity, to metabolic pathways. The results showed that total nitrogen removal efficiency declined from 76.6 % to 48.6 % as the temperature was reduced from 35 °C to 15 °C, while nitrogen removal rate decreased to 0.28 kg-N/m3/d. Low temperature stimulated the secretion of polysaccharide in loosely bound-EPS and protein in tightly bound-EPS. Meanwhile, specific activity of anammox declined from 1.53 to 0.56 g-N/g-VSS/d, and the relative abundance of Candidatus Brocadia dropped from 20.1 % to 9.4 %. Temperature reduction had significant effect on the expression of functional genes (hzsA and hdh) for anammox reaction. Nitrification-denitrification process was enhanced during low temperatures and recovery phases. Biofilm system might respond to temperature reduction by the decline of substance transport and energy consumption, as well as the enhancement of biofilm formation and diversity of nitrogen removal pathways. The findings of this study provide a deep insight into the effect of temperature reduction on PN/A process, also promote the practical applications of mainstream PN/A in the future.

Effect of the form of granular sludge and temperature on anammox immobilized fillers: From performance to microbial community analysis

Abstract:The immobilized carrier was prepared with complete anaerobic ammonia oxidation granular sludge (AnGS) and crushed AnGS, respectively. We evaluated the effects of granular form and continuous temperature changes on nitrogen removal by immobilized anaerobic ammonium oxidation (anammox) filler. The results showed that the rate of nitrogen removal of crushed and encapsulated AnGS was 20% higher than that of direct encapsulated AnGS. However, the latter had higher thresholds of tolerance to Fe2+ and Cu2+. In addition, the immobilization reduced the activation energy of anammox. Above 12.5 °C, the immobilized filler was efficient at removing nitrogen removal through the dual adjustment of temperature-hydraulic retention time. From 12.5 °C to 23 °C, the temperature had a greater influence on the nitrogen removal effect than the HRT. In contrast, HRT had a dominant influence from 23 °C to 32 °C. Anammox activity was severely inhibited below 12.5 °C. High-throughput sequencing analysis showed that the community structure migrated with the changes in temperature. The anammox functional bacteria Candidatus Kuenenia (18.31–39.73%) were the dominant genus at medium and high temperatures, and it was replaced by Chryseobacterium (24.19%) at 8.5 °C. In addition, an RDA analysis showed that Candidatus Brocadia was more adaptable to low temperatures than Candidatus Kuenenia. In addition, Bellilinea was more sensitive to temperature than Candidatus Kuenenia. Thus, the temperature could be appropriately lowered to avoid overbreeding. The results of this study optimized the operation of an anammox immobilized system and promote its further application.

Abstract:This study evaluated the effectiveness of embedding immobilization technology in wastewater treatment and its capacity to enhance the hydrolysis acidification process. Based on this technology, a stable anaerobic environment has been maintained. Results showed that the rates of dissolved organic nitrogen (DON) and dissolved organic phosphorus (DOP) conversion both exceeded 98 % under short hydraulic retention time (HRT = 2h) and ambient temperature. Notably, acetic acid and propionic acid comprised up to 90.9 % of the total volatile fatty acids in the effluent, providing suitable carbon sources for downstream denitrification. 16S rRNA gene sequencing indicated that biofillers effectively enriched and retained functional bacteria, causing norank_Anaerolineaceae (11.6 %-29.7 %) and norank_Bacteroidetes_vadinHA17 (10.8 %-14.9 %) as the dominant genera in the reactor, which were crucial for refractory organic matter degradation. Immobilized biofillers effectively improved wastewater biodegradability, supporting a stable microbial community with high DON and DOP conversion rates as well as increased VFA accumulation.

Abstract:To solve the problems of a slow start, easy loss, and easily inhibited activity of the Anammox bacteria suspension culture, polyvinyl alcohol-polypropylene (PVA-PP) was used to prepare the Anammox immobilized filler. To improve the activity of Anammox bacteria and ensure stable operation of the reaction system, the effects of COD interference, change in pH value, and rotating speed on the nitrogen removal characteristics of the immobilized filler were determined in batch tests. Changes in the structure and diversity of the bacteria in the filler were analyzed by a high-throughput sequencing technique. The results showed that the activity of Anammox bacteria could recover to 100% on the 30th day, and the total nitrogen removal rate was 87.7% when the total nitrogen volume load (NLR) was 0.69 kg·(m3·d)-1 at the stage of 99 days. After 140 days of long-term operation, the total nitrogen removal rate (NRR) reached 1.83 kg·(m3·d)-1, which was 9.4 times the suspended sludge before immobilization. The diversity of the population was maintained in the inclusion carrier, and the effective enrichment of Candidatus Kuenenia (AF375995.1), which performs anaerobic ammonia-oxidization, increased from 11.06% to 32.55%. The influence of COD interference and changes in the pH value of Anammox bacteria was significantly weakened, and the PVA-PP entrapped carrier could achieve the coupling removal of nitrogen by Anammox and denitrification. Appropriate external hydraulic disturbance would promote the Anammox reaction in immobilized systems.
Synergistic denitrification mechanism of domesticated aerobic denitrifying bacteria in low-temperature municipal wastewater treatment

Abstract:To address the problems of low efficacy and low microbial activity in low-temperature municipal wastewater treatment, this study utilized an air-lift micro-pressure internal circulation integrated reactor (AMICIR). Through controlling the amount of aeration and dissolved oxygen (DO) in the reactor, AMICIR creates alternating aerobic and anaerobic environments, explores the enrichment conditions of aerobic denitrifying bacteria, examines the changes in pollutant removal efficiency and the characteristics of bacterial colony structure during the process of enrichment of aerobic denitrifying bacteria in the system, and reveals the mechanism of nitrogen removal by aerobic denitrifying bacteria cooperating with anaerobic denitrifying bacteria in the low-temperature municipal wastewater treatment system. Experimental results showed average removal rates of NH 4 + -N, chemical oxygen demand (COD), total phosphorus (TP), and total nitrogen (TN) reaching 93.85%, 89.30%, 92.75%, and 75.4%, respectively. The microorganisms secreted large amounts of proteins and polysaccharides, forming zoogloea and anaerobic microenvironments conducive to traditional denitrification reactions. IlluminaMiSeq sequencing analysis revealed the presence of anaerobic phyla. The system was enriched with a large number of microorganisms, and aerobic denitrifying bacteria ( Flavobacterium , Rhodoferax , and Pseudomonas ) were successfully cultured. Flavobacterium emerged as the dominant species, with relative abundance ranging from 18.56% to 22.60%. Functional gene prediction indicated high abundance of aerobic denitrification genes, such as napA . Aerobic denitrifying bacteria were successfully enriched in the system to improve nitrogen removal from municipal wastewater at low temperatures.

Abstract:A combined process comprised of ex-situ nitrification in an aged refuse bioreactor (designated as A bioreactor) and in-situ denitrification in a fresh refuse bioreactor (designated as F bioreactor) was constructed for investigating N2O emission during the stabilization of municipal solid waste (MSW). The results showed that N2O concentration in the F bioreactor varied from undetectable to about 130 ppm, while it was much higher in the A bioreactor with the concentration varying from undetectable to about 900 ppm. The greatly differences of continuous monitoring of N2O emission after leachate cross recirculation in each period were primarily attributed to the stabilization degree of MSW. Moreover, the variation of N2O concentration was closely related to the leachate quality in both bioreactors and it was mainly affected by the COD and COD/TN ratio of leachate from the F bioreactor, as well as the DO, ORP, and NO3(-)-N of leachate from the A bioreactor.

It has been proved that the optimal operational parameters were at 35 degrees C, ammonia loading 0.45 kg x (m3 x d)(-1) and < 0.5 mg/L, respectively, with the effluent NO3(-) -N concentration below 20 mg x L(-1) and rho(NO2(-) -N)/rho(NH4(+) -N) ratio being close to 1.0. It is not observed severe membrane fouling during all the experiment. Fluorescence in situ hybridization analysis indicated that aerobic ammonium oxidizers were the dominant population, and nitrite-oxidizing bacteria were inhibited. The microbiological community analysis further provided the necessary biological information for the realization of partial nitrification.

Abstract:This work compared the efficiencies and internal reasons for aniline removal in a bioaugmented sequencing batch reactor at elevated dissolved oxygen (DO) concentrations. Nearly complete aniline removal was achieved while medium DO of 2.65 mg/L was optimal for subsequent nitrogen removal via heterotrophic nitrification-aerobic denitrification. Apart from the highest bacterial diversity richness, favorable DO condition largely enriched putatively aniline degrader, nitrifiers and denitrifiers. Further evidence from qPCR confirmed that moderate DO significantly stimulated the functional genes including aniline degrading gene tad, nitrifying genes amoA, hao, and denitrifying genes nirS and napA, respectively. This study indicates that the significant enrichment of key microorganisms and effective functional genes under optimal DO is the inner mechanism for reliable aniline degradation and subsequent nitrogen removal in the activated sludge reactor.

Abstract:Temperature is a key element affecting the activity of microorganisms in activated sludge. Low water temperature generally leads to decreasing wastewater treatment efficiency and destroying sludge settleability. In this study, activated sludge samples from a municipal wastewater treatment plant (WWTP) implementing oxidation ditch process was used to investigate the bacterial community characteristics of a system that operates well in a cold region (Xinjiang, China) by high-throughput 16S rRNA gene sequencing. The results showed that the influent temperature was 7-12 °C in winter and 13-17 °C in summer, while the sludge volume index (SVI) of samples was between 51 and 74 mL/g. The average removal efficiencies for chemical oxygen demand (COD), biochemical oxygen demand (BOD5), suspended solid (SS), ammonia nitrogen (NH4+-N), total nitrogen (TN), and total phosphorus (TP) were 94%, 95%, 95%, 91%, 73% and 89%, respectively. The bacteria were distributed in 32 phyla and 559 genera. The dominant phyla were Proteobacteria (28.85-48.45%), Bacteroidetes (20.00-31.22%), Chloroflexi (3.59-12.23%), Actinobacteria (1.58-15.54%) and Firmicutes (1.38-10.49%). The dominant genera were Saprospiraceae_norank (4.41-12.23%), Comamonadaceae_unclassified (3.82-8.83%), Anaerolineaceae_norank (1.39-9.35%), Dokdonella (1.13-11.26%), Candidatus_Microthrix (0.26-7.50%), Flavobacterium (0.32-8.14%), Ferribacterium (0.36-5.19%) and Nitrospira (0.084-5.37%), which were different from those found in warm-region WWTPs. Contrary to previous studies, the relative abundance of ammonia-oxidizing bacteria (AOB; Nitrosomonas and Nitrosomonadaceae) and nitrite-oxidizing bacteria (NOB; Nitrospira) increased when the temperature decreased. The successful operation of this WWTP suggests that cold-region WWTPs can achieve good pollutants removal efficiency by simultaneously maintaining an ultra-low sludge load and high dissolved oxygen concentration in the oxidation ditch. The findings of this study provide fundamental knowledge required for an efficient and stable operation of WWTPs in cold regions.
