Abstract:Tetracycline (TC) is extensively utilized in livestock breeding, aquaculture, and medical industry. TC residues seriously harm food security, the environment, and human health. There is an urgent need to exploit a highly efficient and sensitive testing method to monitor TC residue levels in aquatic environments. In this study, graphitic carbon nitride quantum dots (g-CNQDs) were successfully synthesized by a one-step microwave-assisted method using citric acid and urea as precursors. The as-prepared g-CNQDs with size of 1.25-3.75 nm exhibited bright yellow fluorescence at 523 nm when excited at 397 nm. Interestingly, this characteristic fluorescence emission of g-CNQDs could be selectively and efficiently quenched by TC. Based on this phenomenon, for TC detection was successfully explored and applied in real water samples. Wide linear scope of 7-100 μM, low detection limit (LOD) of 0.48 μM, satisfactory recovery of 97.77%-103.4%, and good relative standard deviation (RSD) of 1.05-5.87% were obtained. Mechanism investigations revealed that the static quenching and the inner filter effect (IFE) were responsible for this fluorescence quenching between g-CNQDs and TC. This work not only provided a facile approach for g-CNQDs synthesis but also constructed a g-CNQDs-based fluorescent sensor platform for the highly sensitive and selective detection of TC in aquatic environments.
Ultrasensitive Detection of Tetracycline Using Boron and Nitrogen Co-Doped Graphene Quantum Dots from Natural Carbon Source as the Paper-Based Nanosensing Probe in Difference Matrices

Abstract:Herein, the boron and nitrogen co-doped 0-dimensional graphene quantum dots (B,N-GQDs) with high quantum yield (QY) were synthesized via microwave-assisted hydrothermal method at 170 °C for 20 min using fresh passion fruit juice and boric acid as the starting materials. The 3–6 layers of B,N-GQDs with mean particle size of 9 ± 1 nm were then used for ultra-sensitive and selective detection of tetracycline in aqueous and biological media. The hybridization of boron and nitrogen atoms into the GQD structures increases the intensity of electronegative, resulting in the enhancement of QY to 50 ± 1%. The B,N-GQDs show their excellent analytical performance on tetracycline determination after 2 min of reaction under an optimal condition at pH 5. The linear range of 0.04–70 μM and with limits of detection (LOD) of 1 nM in phosphate buffer saline (PBS), 1.9 nM in urine and 2.2 nM in human serum are obtained. Moreover, the high selectivity of tetracycline by B,N-GQDs over the other 23 interferences is observed. The π-π interaction and electron donor-acceptor principle play pivotal roles in enhancing the ultra-sensitivity and selectivity of B,N-GQDs toward TC detection. Moreover, the B, N-GQD based paper nanosensor exhibits an excellent analytical performance on visual detection of 0.1–30 μM TC in human serum. Results of this study clearly indicate the feasibility of synthesis of B,N-GQDs derived from passion fruit juice for ultrasensitive tetracycline detection, which can open an avenue to use natural products for the preparation of environmentally benign and biocompatible carbon nanomaterials for highly sensitive detection of drugs, antibiotics, organic compounds and biomarkers.

Abstract:In this study, a novel fluorescent "turn-on" aptasensor was developed for sensitive and rapid detection of tetracycline (TC) in animal-derived food. It is based on aptamer-functionalized nitrogen-doped graphene quantum dots (N-GQDs-aptamer) coupled with cobalt oxyhydroxide (CoOOH) nanoflakes. The CoOOH nanoflakes are efficient fluorescence quenchers in homogeneous solutions, and this is due to their advantages of excellent optical properties, superior flexibility, and water dispersibility. The proposed method's mechanism is driven by quenching based on the fluorescence resonance energy transfer (FRET) between the donor (N-GQDs) and the acceptor (CoOOH nanoflakes). On the other hand, fluorescence recovery is caused by the structure switching behavior of the aptamer. Compared with previous methods, our developed method exhibits better behavior in terms of being easy to fabricate and being simple in detection procedure and maintains the detection limit low enough in TC determination: a linear range from 1 to 100 ng mL-1 and a detection limit of 0.95 ng mL-1 (S/N = 3). Furthermore, the proposed method was applied to five animal-derived food samples (milk, honey, fish, eggs, and chicken muscle) and demonstrated practical applicability. As well, the method has the advantages of simplicity in pre-treatment and convenience in instruments, saves times, and is cost-effective. Finally, the proposed method demonstrates significant potential for sensitive and rapid detection of specific components in real samples. Graphical abstract.

Abstract:Antibiotics are emerging contaminants in the environment, and their monitoring is important for food safety and human health. Herein, we synthesized a two-photon carbon quantum dots (TP-CQDs) nanoprobe using a simple and green hydrothermal approach and applied it as a chemosensor to detect chlortetracycline (CTC). CTC quenches the intrinsic fluorescence of TP-CQDs, providing a linear response within the 0.5 to 60 μM range and yielding a limit of detection of only 25 nM. Besides excellent stability and good optical properties, we found that TP-CQDs are selective to CTC as structurally similar antibiotics, along with cations, anions, and amino acids, had no significant influence on fluorescence emission. Finally, we successfully applied this sensing platform to quantify CTC in milk, fish, and eggs.

Abstract:Rampant use of tetracycline in animal feed is a threat to food security, the environment, and human health because of the risk of drug residues. Therefore, it is necessary to establish a sensitive, efficient, and reliable method for qualitative and quantitative detection of tetracycline. In this paper, we synthesized fluorescent carbon dots (FCDs) by thermal cracking of crab shell waste, and obtained a fluorescence quantum yield of 30%. Characterization of the FCDs by transmission electron microscopy, Fourier-transform infrared spectroscopy, ultraviolet visible absorption spectroscopy, and photoluminescence spectroscopy showed that they were fluorescent and evenly distributed with an average size of approximately 10 nm. We designed a sensitive probe for detecting tetracycline using the fluorescence intensity change of the FCDs. This method is sensitive, inexpensive, and environmentally friendly. The concentration of tetracycline was examined by comparing the fluorescence intensities of the FCDs before and after tetracycline addition. The limit of detection for tetracycline was 0.005 mg/L (signal-to-noise ratio = 3), which is promising for method development.

Abstract:In this paper, a novel and selective fluorescent probe for nanocomposite materials were developed by in situ growth carbon quantum dots (CQDs) in the channel of mesoporous silica nanospheres (MSNs) for the detection of tetracycline (TC). The CQDs@MSNs nanocomposites were prepared via a facile hydrothermal method. The CQDs incorporated into MSNs can act as the low-toxicity and stable fluorescent probes for the detection of TC. Under optimal conditions, a fluorescence emission peak at 439 nm was observed for the CQDs@MSNs when the excitation wavelength was set as 360 nm, and the fluorescence peak intensity can be gradually quenched by TC via inner filter effect, a good linear relationship between the concentration of TC from 0 to 100 μM, and the fluorescence quenching intensity of CQDs@MSNs was obtained, with a limit detection of 5.19 μM. More importantly, the CQDs@MSNs have been successfully used for TC determination in real samples.

Abstract:Ti 2 C quantum dots (QDs) with rich surface functional groups have been synthesized using a hydrothermal method, and used to detect tetracycline (Tc) based on enhanced fluorescence. The interaction between the surface functional groups of Ti 2 C QDs and Tc enhanced the fluorescence of Tc at 514 nm, which is used to detect Tc quickly and accurately. Under optimal conditions, the fluorescence intensity was linear to the concentration of Tc in the range of 50.0–30.0 μM, with a detection limit of 21.6 nM. Furthermore, the Tc-Ti 2 C QDs detection system was evaluated for detection of Tc in milk and artificial urine. This study demonstrates a new and simple strategy for Tc detection, which is important for food safety and human health.

Abstract:Background Currently, many types of pharmaceuticals are used to treat disease and bacterial infections. Tetracycline (TC) is a widely applied human and veterinary medicine. However, the widespread usage and lack of awareness in the management of TC lead to its unavoidable discharge into the environment. The consequences include the proliferation of resistant strains of bacteria and adverse impacts on nontarget organisms and microbial ecosystems, which means that TC is an emerging substance of concern and that it is crucial to develop determination platforms with cost-effective and fast-response features. Methods Carbon quantum dots (CQDs) have received tremendous attention owing to their great performance in fluorescent probing. In the present work, ammonium sulfate-doped carbon quantum dots (S,NCQDs) are fabricated through a facile hydrothermal synthesis pathway. Significant Findings The results illustrate that the S,NCQDs exhibit sensitive determination ability toward tetracycline via a static quenching mechanism and possess several superior features, including a rapid response, good sensitivity and stability, broad linear detection range (1–100 μM), and low LOD value (0.1203 μM), when applied in the determination of TC. Additionally, the application of S,NCQDs valorizes ammonium sulfate waste from semiconductor manufactory waste and has great potential in fluorescence sensing. Abstract:This work synthesized the ethylenediamine-doped puffer fish skin carbon dots using biomass as a green precursor. Three tetracycline antibiotics (TCs) were detected fluorescence and colorimetrically using carbon dots as probes; on this basis, hydrogels were used to carry out visual identification and quantification. In the presence of TCs, the nitrogen-doped carbon dots (N-CDs) displayed a quenching impact on fluorescence because of fluorescence resonance energy transfer (FRET), dynamic quenching, and band gap transition. As a result, it was possible to identify the precise concentrations of tetracycline (TC), oxytetracycline (OTC), and chlortetracycline (CTC). The assay has great biocompatibility, which was supported by cytotoxicity, a wide linear range (0.5–500 M), a low detection limit (0.15 M), and outstanding selectivity. With a high recovery rate, the approach has been successfully used to identify TCs in food and water samples such milk, honey, and tap water.

Abstract:In this study, a system was established for the detection of tetracyclines (TCs) by simply mixing graphene quantum dots (GQDs) and Eu3+. The GQDs-Eu3+ system exhibits both strong emission of GQDs and weak characteristic emission of Eu3+ upon 360 nm excitation. In the presence of TCs, the luminescence of GQDs was quenched, while the luminescence of Eu3+ was significantly enhanced. It is worth to mention that the GQDs we prepared possess a lot of carboxyl groups on the surface and edge. This carboxyl groups play an important role in fluorescent enhancement of Eu3+. On the basis of this effect, a novel label free ratiometric sensor was developed for the detection of TCs. Under the optimized conditions, linear concentration ranges from 0 to 20 μM for tetracycline was found, and the detection limits was 8.2 nM. This system also exhibited excellent selectivity toward TCs over other antibiotics. In addition, it was successfully applied to the determination of tetracycline in river water and milk samples. The proposed system was simple in design, fast in operation, rapid in response and cost-effective. In view of the advantages, the developed system demonstrates great potential for the detection of TCs in environmental and biological samples.

Abstract:The antibiotic tetracycline can be efficiently used as medicine for the deterrence of bacterial infections in humans, animals, and plants. However, the unprecedented use of tetracycline is of great concern owing to its low biodegradability, extensive usage, and adverse impacts on the environment and water quality. In this study, a sensitive spectrofluorometric method was proposed for the direct determination of tetracycline, based on biocompatible fluorescent carbon dots (CDs). The synthesis of CDs was performed by adopting a green hydrothermal procedure from carrot juice without requiring surface passivation or outflowing any environmentally hazardous waste. X-ray diffraction analysis and transmission electron microscopy revealed amorphous spherical-shaped CDs that exhibited blue emission under blue illumination. The fabricated fluorescent probe directly detected tetracycline in the concentration range of 4.00 × 10-6 to 1.55 × 10-5 mol L-1 with an LOD of 1.33 × 10-6 mol L-1. The performance of the probe was assessed in a tap water sample, with recovery values between 80.70 and 103.60%. The method's greenness was evaluated using the Analytical Green metric approach (AGREE) and confirmed to be within the green range. The developed method is facile, rapid, cost-effective, and offers a wide linear range and satisfactory selectivity, making it potentially suitable for determining tetracycline in water applications.
G-quadruplex Specific Thioflavin T-based Label-Free Fluorescence Aptasensor for Rapid Detection of Tetracycline.

Abstract:A label-free fluorescence aptasensor was developed for the rapid detection of tetracycline (TET) based on G-quadruplex structure of TET aptamers and G-quadruplex specific dye Thioflavin T (ThT). The fluorescence of free ThT is essentially weak in aqueous solution, whereas it selectively identifies the G-quadruplex of aptamers to form the G-quadruplex/ThT conjugates, resulting in an enormous increase of the fluorescence intensity. However, the fluorescence intensity of G-quadruplex/ThT conjugates was drastically suppressed due to the release of free ThT from G-quadruplex/ThT conjugates after the addition of TET via specific binding with TET aptamers. The key factors affecting sensitivity and selectivity including the reaction medium, binding time of ThT to TET aptamers, incubation time between TET aptamers and TET, concentration of ThT and TET aptamers were investigated in detail. The optimal conditions were as follows: ultrapure water as reaction medium, binding time of 5min, incubation time of 1 min, 9.0 mu mol/L ThT and 0.03 mu mol/L aptamers. A good linear relationship (correlation coefficient of 0.9973) was obtained between the fluorescence quenching efficiency (F-0 - F) / F0 and the logarithm of TET concentration in the range of 0.01-1.0 mu mol/L. The limit of detection was 0.001 mu mol/L (S/N = 3). The proposed assay was applied for the detection of TET in the spiked honey and milk samples with recoveries ranging from93.5% to 106.9%. The developed label-free fluorescence aptasensor showed advantages of high specificity, low cost and short time-consuming, illustrating potential application for on-site detection of TET in foodstuffs. Sensitive and selective detection of tetracycline using fluorescence-enhanced Eu(III)-functionalized silver nanoparticles with homocysteine

Abstract:Utilizing metal luminescence enhancement to design fluorescent probes is a very sensible strategy. Herein, a fluorescent probe based on europium (III)-functionalized silver nanoparticles-conjugated homocysteine (AgNPs-Hcy-Eu3+) was proposed for the selective and sensitive detection of tetracycline (TC). In this probe, Eu(III) was employed as the detection signal unit for TC, while AgNPs-Hcy was used as the ligand of fluorescence enhancement. When TC exists, it can bind to Eu3+ immobilized in AgNPs-Hcy, leading to an enhanced fluorescence signal from Eu3+ through energy transfer. Under optimal conditions, the fluorescence intensity of AgNPs-Hcy-Eu3+ increased linearly with increasing TC concentration in the range of 0.1-30 μM (R2 = 0.9964). The fluorescent probe own fluorescence enhancement, paving the way for sensitive detection with a low detection limit of 0.083 μM. It also has good selectivity for common antibiotics and anions. This work can be applied to the determination of TC in tap water and milk with recoveries of 94-98.5%. We expect AgNPs-Hcy-Eu3+ to have potential applications in environmental testing and food safety.
One-Pot facile synthesis of fluorescent copper nanoclusters for highly selective and sensitive detection of tetracycline

Abstract:Due to the excellent characteristics, fluorescent copper nanoclusters (Cu NCs) have aroused great interest in recent years. Herein, the simple prepared, environmentally friendly fluorescent Cu NCs were synthesized by using trypsin as the stabilizer and applied for the determination of tetracycline. Uniformly dispersed Try-Cu NCs were obtained with average size of 3.5 ± 0.3 nm and some excellent merits of good water solubility, UV light stability and salt stability. Emission peaks around 460.0 nm were visibly quenched by tetracycline based on static quenching mechanism and inner filter effect (IFE). Two excellent linear relationships were observed between ln(F0/F) and tetracycline concentrations in the range of 1-100 μM and 100-300 μM with limit of detection (LOD) of 0.084 μM. Meanwhile, this nanoprobe exhibited an apparent selectivity for tetracycline detection. Moreover, Try-Cu NCs were successfully employed to determine tetracycline in serum and milk samples after facile pretreatment with satisfactory recovery rates and credible standard deviation. The results suggested that this as-prepared Try-Cu NCs had excellent application prospects in the future.
Zinc-doped carbon quantum dots-based ratiometric fluorescence probe for rapid, specific, and visual determination of tetracycline hydrochloride

Abstract:In this work, a rapid, specific, and visual ratiometric fluorescence probe was constructed for tetracycline hydrochloride (TCH) determination based on zinc-doped carbon quantum dots (Zn-CDs). In the presence of TCH, the blue fluorescence at 440 nm originating from Zn-CDs was quenched, and the green fluorescence at 515 nm stemming from TCH was enhanced. The inner filter effect (IFE) and the chelation between Zn and tetracycline are the main mechanisms for the conversion of spectra. The spectrum and color change completed and stabilized within 1 min, indicating the possibility of real-time detection of TCH. The detection range for TCH is 0.1–50 μM, and the low detection limit is 61.1 nM. In addition, Zn-CDs-based test strips were successfully applied to direct visual identification of TCH in actual samples of river water and milk, indicating the possibility of their practical application.

Abstract:Tetracycline (TC) residues are harmful to the environment and human body, so it is necessary to develop a highly sensitive probe for rapid detection of tetracycline residues. In the present paper, a novel dye-doped porous metal–organic framework (UiO-66)-based multi-color fluorescent nano-probe was designed for sensitive ratiometric detection of tetracycline (TC). In this probe, dye-molecules doped UiO-66 was used as a fluorescent internal standard, and the externally grafted lanthanide Eu3+ complex was used as response signals. The fluorescence of the Eu3+ complex was selectively enhanced with increasing concentrations of TC, which was accompanied by a visual blue-to-red color switch. The nano-probe had a linear response between 0.1 and 6 μM with a lowest detection limit of 17.9 nM, which was much lower than the maximum residue limits set by the United States Food and Drug Administration (676 nM) and the European Union (225 nM). The applicability of this method in the analysis of actual samples was evaluated by the determination of TC in honey and milk samples, indicating satisfactory recovery and good reproducibility. In addition, a cost-effective paper-based probe for rapid and visual detection of TC was developed by fixing the nano-probe on filter papers. With the help of a smartphone camera to capture the fluorescence color, and chromaticity analysis software, the calculation and analysis of red (R) and blue (B) values can be realized, which has the potential for real-time visual detection of TC.

Green Preparation of Fluorescent Nitrogen-Doped Carbon Quantum Dots for Sensitive Detection of Oxytetracycline in Environmental Samples

Abstract:Nitrogen-doped carbon quantum dots (N-CQDs) with strong fluorescence were prepared by a one-step hydrothermal method using natural biomass waste. Two efficient fluorescent probes were constructed for selective and sensitive detection of oxytetracycline (OTC). The synthesized N-CQDs were characterized by UV-visible absorption spectra, fluorescence spectra, Fourier transform infrared spectroscopy (FT-IR), X-ray photon spectroscopy (XPS), atomic force microscopy (AFM), and high-resolution transmission electron microscopy (HRTEM), which proved that the synthesized N-CQDs surface were functionalized and had stable fluorescence performance. The basis of N-CQDs detection of OTC was discussed, and various reaction conditions were studied. Under optimized conditions, orange peel carbon quantum dots (ON-CQDs) and watermelon peel carbon quantum dots (WN-CQDs) have a good linear relationship with OTC concentrations in the range of 2–100 μmol L−1 and 0.25–100 μmol L−1, respectively. ON-CQDs and WN-CQDs were both successfully applied in detecting the OTC in pretreated tap water, lake water, and soil, with the recovery rate at 91.724–103.206%, and the relative standard deviation was less than 5.35%. The results showed that the proposed N-CQDs proved to be green and simple, greatly reducing the detection time for OTC in the determination environment.
