Abstract:Carbon dots have been widely focused on the field of sensing and detection due to their excellent optical property. Herein, novel orange fluorescent nitrogen and chlorine co-doped carbon dots (N,Cl-CDs) are obtained by one-pot hydrothermal method using o-phenylenediamine and neutral red. Based on the inner filter effect, the prepared N,Cl-CDs can be innovatively developed as an effective "signal-off" multifunctional sensing platform for sensitive determination of tetracycline. The proposed sensor was utilized to realize the determination of tetracycline in Rirver water samples/milk samples (λex = 390 nm, λem = 606 nm) with satisfactory recoveries and relative standard deviations. The linear range of are 0.05 to 45 μM and 45 to135 μM, and detection limit is 3.9 nM (3σ/m). Meanwhile, the luminescent intensity of N,Cl-CDs was reduced gradually when pH changed continuously from 12 to 2, showing a pH-responsive fluorescence property with two linear ranges of pH 3–7 and pH 7–10. In addition, due to the characteristics of low toxicity and excellent biocompatibility, the N, Cl-CDs were also used in the imaging of oocystis cells, which is hopeful to realize the detection of tetracycline in living cells.

Abstract:This work synthesized the ethylenediamine-doped puffer fish skin carbon dots using biomass as a green precursor. Three tetracycline antibiotics (TCs) were detected fluorescence and colorimetrically using carbon dots as probes; on this basis, hydrogels were used to carry out visual identification and quantification. In the presence of TCs, the nitrogen-doped carbon dots (N-CDs) displayed a quenching impact on fluorescence because of fluorescence resonance energy transfer (FRET), dynamic quenching, and band gap transition. As a result, it was possible to identify the precise concentrations of tetracycline (TC), oxytetracycline (OTC), and chlortetracycline (CTC). The assay has great biocompatibility, which was supported by cytotoxicity, a wide linear range (0.5–500 M), a low detection limit (0.15 M), and outstanding selectivity. With a high recovery rate, the approach has been successfully used to identify TCs in food and water samples such milk, honey, and tap water.

Abstract:Tetracyclines residues, particularly oxytetracycline (OTC) and tetracycline (TC), have raised extensive concern because of their serious adverse effects on human health. Herein, a dual-response fluorescent probe based on nitrogen-doped carbon dots (N-CDs) and Eu3+ hybrid (N-CDs-Eu3+) was developed to selectively determine OTC and TC. The N-CDs act as ancillary ligands of Eu3+ and recognition units of OTC/TC, while the Eu3+ ions chelated with N-CDs can also specifically recognize OTC/TC. Upon inclusion of OTC/TC, an enhancement in Eu3+ emission occurs due to the energy transfer from OTC/TC to Eu3+ and the efficient elimination of quenching effect caused by H2O molecule, which is attributed to the incorporation of N-CDs; while the blue fluorescence emitted by the N-CDs decreases under the inner filter effect and static quenching effect caused by OTC/TC. Based on the double and reverse response signals, the ratiometric detection of OTC and TC in the range of 0.1-45 μΜ and 0.1-30 μΜ is achieved with a detection limit of 0.017 and 0.041 μM, respectively. In addition, the noticeable variation in fluorescence color of the probe is integrated with a smartphone-assisted analysis device for the rapid on-site quantitative assay of OTC, where the detection limit is 0.15 μΜ. The results show that this probe performs with excellent specificity and anti-interference for both OTC and TC, and satisfactory detection results are obtained in lake water, milk, and honey samples, thereby confirming that the probe exhibits promising application in food safety and environmental monitoring.

Abstract:Rampant use of tetracycline in animal feed is a threat to food security, the environment, and human health because of the risk of drug residues. Therefore, it is necessary to establish a sensitive, efficient, and reliable method for qualitative and quantitative detection of tetracycline. In this paper, we synthesized fluorescent carbon dots (FCDs) by thermal cracking of crab shell waste, and obtained a fluorescence quantum yield of 30%. Characterization of the FCDs by transmission electron microscopy, Fourier-transform infrared spectroscopy, ultraviolet visible absorption spectroscopy, and photoluminescence spectroscopy showed that they were fluorescent and evenly distributed with an average size of approximately 10 nm. We designed a sensitive probe for detecting tetracycline using the fluorescence intensity change of the FCDs. This method is sensitive, inexpensive, and environmentally friendly. The concentration of tetracycline was examined by comparing the fluorescence intensities of the FCDs before and after tetracycline addition. The limit of detection for tetracycline was 0.005 mg/L (signal-to-noise ratio = 3), which is promising for method development.

Abstract:Concentration-dependent photoluminescence carbon dots (CDs) have been successfully synthesized through the one-step hydrothermal treatment of o-phthalic acid and ethylenediamine. The CDs possessed higher fluorescence quantum yield, up to 39.22%, exhibiting distinguished optical property, water solubility, and stability. The CDs that emit strong blue-green fluorescence can visually identify and determine tetracycline (TC), oxytetracycline (OTC), and chlortetracycline (CTC). TC quenched the fluorescence of CDs at 500 nm owing to the inner filter effect; OTC behaved similarly, but the emission wavelength of CDs was red-shifted to 515 nm. Inversely, once CTC was introduced to CDs solution, the fluorescence increased and the emission peak was blue-shifted to 450 nm. Bandgap transition and electrostatic interaction were proposed to be the mechanisms for the detection of OTC and CTC by CDs. Wide linear relationships were established for TC, OTC, and CTC with the limits of detection to be 50 nM, 36 nM, and 373 nM, respectively. Furthermore, the nanoscale probe constructed by this system has been applied to detect tetracyclines (TCs) in complex samples with satisfying recoveries (93.2–114%) and was designed as a portable test strip sensor for visually on-site TCs of honey sample screening. Accordingly, the preparation process of the nano fluorescent probe is simple and environmentally friendly, and the probe has a specific recognition ability for tetracyclines. The synthesized CDs in this work provide a new orientation for fast, effective, and visual real-time detection of tetracycline in actual samples.

Abstract:Tetracycline (TC) is a typical emerging pharmaceuticals and personal care products (PPCPs) pollutants in the environment. However, TC residues cause severe effects on human health. It is crucial to exploit simple, rapid and sensitive methods to detect TC in water samples. A ratiometric fluorescent probes (CDs-Eu 3+ ) is constructed using carbon dots (CDs) and europium ions (Eu 3+ ) for the visual detection of tetracycline (TC). Under 390 nm excitation, CDs-Eu 3+ shows a strong yellow fluorescence at 545 nm and a weak red fluorescence at 619 nm, respectively. In the presence of TC, the fluorescence intensity at 545 nm decreased while the fluorescence intensity at 619 nm increased, which attributed to the internal filtering effect (IFE) and "antenna effect", respectively. Sensing performance of CDs-Eu 3+ probes is further evaluated with respect to various conditions, including salt concentration, Eu 3+ concentration, pH, and reaction time. Results reveal that the proposed ratiometric fluorescent probes exhibits a specific recognition ability toward TC. An excellent linear relationship is observed between the fluorescence intensity ratio (F 619 /F 545 ) and TC concentrations (1⁓100 μM), with a low detection limit (LOD) of 31 nM. The solution (CDs-Eu 3+ -TC) color gradually changes from yellow to red after adding TC. The probes are successfully applied to determine TC in water with recoveries of 85.43⁓97.53%. Finally, a fluorescent test strip is prepared and used for rapid visual semi-quantitative TC detection.

Abstract:The paper-based ratiometric fluorescence sensor is normally prepared by two or more types of fluorescent materials on paper chip for simple, low-cost and fast detection. However, the choice of multi-step and one-step modification on paper chip affects the analytical performance. Herein, a novel paper-based dual-emission ratiometric fluorescence sensor was designed for the selective detection of tetracycline (TC). Carbon dots (CDs) modified with Eu3+ were combined with a sealed paper-based microfluidic chip by two methods, one-step grafting CDs-Eu3+ on paper and step-by-step grafting CDs and Eu3+ on paper. The analytical performance was studied and optimized respectively. The red fluorescence of Eu3+ at 450 nm is enhanced and the blue fluorescence of CDs at 617 nm is quenched by energy transfer in the presence of TC. Under optimal conditions, TC is selectively determined in the linear range from 0.1 μM to 100 μM with the detection limit of 0.03 μM by the step-by-step grafting method. In addition, the sealed paper chip can effectively prevent the pollution and volatilization from reagent. This technique has been used to analyze TC in seafood aquaculture water with satisfactory results.

Abstract:<p>S,N-doped carbon dots (S,N-CDs) were facilely prepared <em>via</em> a hydrothermal method by using thiourea and citric acid (CA) as precursors. The as-prepared S,N-CDs exhibited high stability in aqueous solution and were applied for fluorometric detection of tetracyclines (TCs). The approach was developed on the basic of the fluorescence of S,N-CDs at 440 nm, which was selectively quenched by TCs through the inner filter effect (IFE) mechanism. Moreover, the fluorescence of TCs with maximum at 600 nm could be used as the internal standard signal, resulting in a fluorometric spectral response. The limit of detection (LOD) was 0.25, 0.39 and 0.26 μM for tetracycline (TC), doxycycline (DC) and oxytetracycline (OTC), respectively. Furthermore, S,N-CDs were successfully utilized for the detection of residual TCs in fish and pork samples with acceptable recovery and good RSD.</p>

Abstract:Background Currently, many types of pharmaceuticals are used to treat disease and bacterial infections. Tetracycline (TC) is a widely applied human and veterinary medicine. However, the widespread usage and lack of awareness in the management of TC lead to its unavoidable discharge into the environment. The consequences include the proliferation of resistant strains of bacteria and adverse impacts on nontarget organisms and microbial ecosystems, which means that TC is an emerging substance of concern and that it is crucial to develop determination platforms with cost-effective and fast-response features. Methods Carbon quantum dots (CQDs) have received tremendous attention owing to their great performance in fluorescent probing. In the present work, ammonium sulfate-doped carbon quantum dots (S,NCQDs) are fabricated through a facile hydrothermal synthesis pathway. Significant Findings The results illustrate that the S,NCQDs exhibit sensitive determination ability toward tetracycline via a static quenching mechanism and possess several superior features, including a rapid response, good sensitivity and stability, broad linear detection range (1–100 μM), and low LOD value (0.1203 μM), when applied in the determination of TC. Additionally, the application of S,NCQDs valorizes ammonium sulfate waste from semiconductor manufactory waste and has great potential in fluorescence sensing. Abstract:In this work, a ratiometric fluorometric sensor based on nickel nanoclusters (NiNCs)-europium complex (NiNCs-Eu3+) was constructed for the highly selectivity detection of tetracyclines (TCs) in water samples. In the presence of TCs, the blue fluorescence of the sensor NiNCs-Eu3+ was quenched at 430 nm and the characteristic red fluorescence of Eu3+-TCs appeared at 620 nm because of the combined help of inner filter effect (IFE) and antenna effect. Under the optimized conditions (100 mM Eu3+ (100 µL); temperature (25℃); reaction time (10 min), HEPES buffer solution (pH = 7.0)), the sensor offered a wide detection range of tetracycline (TC) and oxytetracycline (OTC) from 0.1 to 50 μM with the detection limit (LOD) of 25 nM and 21 nM, respectively. Moreover, the sensor was able to detect of TC and OTC in tap and lake water with high recovery rate (89.10%-97.60%). In addition, the portable paper-based sensor was constructed using filter paper embedded with NiNCs-Eu3+. The distinct fluorescent color of the paper-based sensor varied from bright blue to red against different concentrations of TC and OTC. These above findings demonstrated the potential for wide application of as-prepared ratio metric fluorescence sensor for visual detection of TCs in water samples.

Abstract:Utilizing metal luminescence enhancement to design fluorescent probes is a very sensible strategy. Herein, a fluorescent probe based on europium (III)-functionalized silver nanoparticles-conjugated homocysteine (AgNPs-Hcy-Eu3+) was proposed for the selective and sensitive detection of tetracycline (TC). In this probe, Eu(III) was employed as the detection signal unit for TC, while AgNPs-Hcy was used as the ligand of fluorescence enhancement. When TC exists, it can bind to Eu3+ immobilized in AgNPs-Hcy, leading to an enhanced fluorescence signal from Eu3+ through energy transfer. Under optimal conditions, the fluorescence intensity of AgNPs-Hcy-Eu3+ increased linearly with increasing TC concentration in the range of 0.1-30 μM (R2 = 0.9964). The fluorescent probe own fluorescence enhancement, paving the way for sensitive detection with a low detection limit of 0.083 μM. It also has good selectivity for common antibiotics and anions. This work can be applied to the determination of TC in tap water and milk with recoveries of 94-98.5%. We expect AgNPs-Hcy-Eu3+ to have potential applications in environmental testing and food safety.
Nitrogen-doped cyan-emissive carbon quantum dots for fluorescence tetracycline detection and lysosome imaging

Abstract:Tetracyclines (TCs) prevent the growth of peptide chains and the synthesis of proteins, and they are widely used to inhibit Gram-positive and -negative bacteria. For the detection of tetracyclines in cell and in vitro , a convenient and simple detection system based on nitrogen-doped cyan carbon quantum dots (C-CQDs) was developed. C-CQDs have excellent excitation-independent properties, the best optimal excitation peak is 360 nm and the best emission peak is 480 nm. Based on the inner filter effect (IFE), the fluorescence intensity of C-CQDs in solution decreases with the increase of tetracyclines. In the range of 0–100 μM, C-CQDs present a good linear relationship with three tetracyclines (CTC, TET, OCT), with R 2 all greater than 0.999. C-CQDs can detect tetracycline in milk samples with recovery in the range of 98.2–103.6%, which demonstrates their potential and broad application in real samples. Furthermore, C-CQDs exhibit excellent lysosomal targeting, as indicated by a Pearson's coefficient of 0.914 and an overlap of 0.985. The internalisation of C-CQDs was mainly affected by lipid raft-mediated endocytosis in endocytic pathway experiments. These experiments indicate that C-CQDs can be effectively used to detect TC content and target lysosomes as an alternative to commercial dyes.

Abstract:Lanthanide coordination polymer nanoparticles (LML-Eu3+-GMP CPNPs) were fabricated as a ratiometric fluorescence sensor for real-time and visual assays of tetracycline (TC). The LML-Eu3+-GMP CPNPs were prepared by spontaneous self-assembly of Eu3+, luminol (LML) and guanosine 5'-monophosphate (GMP) at room temperature, which only emitted blue fluorescence dominated by LML, and no red characteristic fluorescence of Eu3+ was observed. In the presence of TC, TC can trigger the antenna effect (AE) and inner filter effect (IFE), the blue fluorescence of LML-Eu3+-GMP CPNPs is quenched by TC via IFE, and the red characteristic fluorescence of Eu3+ can be turned on through AE. When sodium citrate (Cit) is added to the sensing system, it can further enhance the red fluorescence of Eu3+. The dual-ligand CPNP sensor has a rapid fluorescence response, noticeable fluorescence color change, good selectivity, and high sensitivity with a detection limit of 3.4 nM. It was successfully applied to detect TC in honey, milk, lake water and tap water. Meanwhile, the portable smartphone and test paper were applied to develop POCT devices for real-time and visual detection of TC, which could offer a promising application for real-time and semiquantitative assays of TC.

Abstract:Tetracycline (TC) residues are harmful to the environment and human body, so it is necessary to develop a highly sensitive probe for rapid detection of tetracycline residues. In the present paper, a novel dye-doped porous metal–organic framework (UiO-66)-based multi-color fluorescent nano-probe was designed for sensitive ratiometric detection of tetracycline (TC). In this probe, dye-molecules doped UiO-66 was used as a fluorescent internal standard, and the externally grafted lanthanide Eu3+ complex was used as response signals. The fluorescence of the Eu3+ complex was selectively enhanced with increasing concentrations of TC, which was accompanied by a visual blue-to-red color switch. The nano-probe had a linear response between 0.1 and 6 μM with a lowest detection limit of 17.9 nM, which was much lower than the maximum residue limits set by the United States Food and Drug Administration (676 nM) and the European Union (225 nM). The applicability of this method in the analysis of actual samples was evaluated by the determination of TC in honey and milk samples, indicating satisfactory recovery and good reproducibility. In addition, a cost-effective paper-based probe for rapid and visual detection of TC was developed by fixing the nano-probe on filter papers. With the help of a smartphone camera to capture the fluorescence color, and chromaticity analysis software, the calculation and analysis of red (R) and blue (B) values can be realized, which has the potential for real-time visual detection of TC.
