Abstract:Employing cheap Cu nanoclusters to design a novel fluorescent probe have promising opportunities in the field of optical sensors. Here, we fabricated a highly photoluminescent D-tryptophan (D-Trp)-coated Cu nanoclusters (Trp-Cu NCs) by rapid microwave-assisted method to achieve precise quantification of tetracyclines (TC). Due to protecting groups of Trp, the synthesized Trp-Cu NCs have remarkable fluorescence stability with a quantum yield reached 12.5%. A distinct fluorescence quenching with the incremental addition of TC via the internal filtration effect (IFE). Based on turn-off fluorescence within 1 min, a detection method for detecting TC was constructed with a linear range of 0.3-120 μM and a limit of detection (LOD) of 0.12 μM. Besides, the proposed fluorescent probe has been employed for the determination of practical samples such as water samples, milk and honey, and exhibited satisfactory recoveries of 96.1%-108.2%, with relative standard deviations (RSD) lower than 5.0%. This is a sensitive, rapid and easily recognizable Trp-Cu NCs based sensing platform for the determination of TC, which could offer a powerful tool for ensuring food safety.
Sensitive and selective detection of tetracycline using fluorescence-enhanced Eu(III)-functionalized silver nanoparticles with homocysteine

Abstract:Utilizing metal luminescence enhancement to design fluorescent probes is a very sensible strategy. Herein, a fluorescent probe based on europium (III)-functionalized silver nanoparticles-conjugated homocysteine (AgNPs-Hcy-Eu3+) was proposed for the selective and sensitive detection of tetracycline (TC). In this probe, Eu(III) was employed as the detection signal unit for TC, while AgNPs-Hcy was used as the ligand of fluorescence enhancement. When TC exists, it can bind to Eu3+ immobilized in AgNPs-Hcy, leading to an enhanced fluorescence signal from Eu3+ through energy transfer. Under optimal conditions, the fluorescence intensity of AgNPs-Hcy-Eu3+ increased linearly with increasing TC concentration in the range of 0.1-30 μM (R2 = 0.9964). The fluorescent probe own fluorescence enhancement, paving the way for sensitive detection with a low detection limit of 0.083 μM. It also has good selectivity for common antibiotics and anions. This work can be applied to the determination of TC in tap water and milk with recoveries of 94-98.5%. We expect AgNPs-Hcy-Eu3+ to have potential applications in environmental testing and food safety.

Abstract:Hereby, a facile-green copper nanoclusters templated by glutathione S-transferase (GST-CuNCs) has been innovatively synthesized via a simple one-pot stirring method at room temperature. Meaningfully, the as-prepared nanoclusters exhibited uniform size with satisfactory fluorescence intensity, good stability and low cytotoxicity. Significantly, the fluorescence of the obtained GST-CuNCs could be considerably enhanced by the addition of chlorotetracycline (CTC) rather than other analogues of CTC, which was ascribed to the aggregation-induced enhancement caused by the interaction between CTC and GST. The enhanced fluorescence intensity demonstrated a good linear correlation with CTC concentration in the range of 30-120 μM (R2=0.99517), and the low detection limit was 69.7 nM. Furthermore, the proposed approach showed favorable selectivity and anti-interference toward CTC among the prevalent ions and amino acids. Additionally, this nanoprobe was also profitably applied for the quantitative detection of CTC in serum samples with satisfactory outcomes, which demonstrated excellent prospects for practical applications.

Synthesis of 2MP-CuNPs Fluorescent Probes and Their Application in Tetracycline Detection

Abstract:A fluorescent probe composed of 2-mercaptopyridine–copper nanoparticles (2MP-CuNPs) was synthesized through a hydrothermal method utilizing CuCl2 and 2-mercaptopyridine (2MP). The experimental results indicate that the 2MP-CuNPs probe exhibited an excellent fluorescence emission peak at 525 nm with an excitation wavelength of 200 nm. Furthermore, this emission peak was accompanied by a substantial Stokes shift of 325 nm, which effectively minimized the overlap between the excitation and emission spectra, thereby enhancing detection sensitivity. In tetracycline (TC) detection, the dimethylamino group on TC undergoes protonation in acidic conditions, resulting in a H+ ion. Consequently, the nitrogen atom within the pyridine moiety of the 2MP-CuNPs probe forms a coordination complex with H+ via multi-toothed n-bonding interactions, leading to a significant reduction in fluorescence intensity at 525 nm. Based on this mechanism, a quantitative detection method for TC was successfully established with a linear range spanning from 0.1 to 240 μM and an impressive detection limit of 120 nM. Furthermore, during actual sample analyses involving milk and chicken feed, this analysis method based on the 2MP-CuNPs probe achieved absolute recovery rates ranging from 94% to 98%, underscoring its considerable potential for practical applications.

Abstract:Histidine‐capped copper nanoclusters were synthesized using one simple method and successfully applied in doxycycline detection with superior sensitivity and selectivity. There is a significant need to accurately measure doxycycline concentrations in view of the adverse effects of an overdose on human health. A fluorescence (FL) detection method was adopted and copper nanoclusters (CuNCs) were synthesized using chemical reduction technology. Based on FL quenching with doxycycline, the prepared CuNCs were used to explore a fluorescent nanoprobe for doxycycline detection. In an optimal sensing environment, this FL nanosensor was sensitive and selective in doxycycline sensing and displayed a linear relationship in the range 0.5–200 μM with a detection limit of 0.092 μΜ. A characterization test demonstrated that CuNCs offered active functional groups for identifying doxycycline using electrostatic interaction and hydrogen bonds. Static quenching and the inner filter effect (IFE) resulted in weakness in the FL of His@CuNCs with doxycycline with great efficiency. This suggested nanosensor was revealed to be a functional model for simple and rapid detection of doxycycline in real samples with very pleasing accuracy.

Abstract:Rampant use of tetracycline in animal feed is a threat to food security, the environment, and human health because of the risk of drug residues. Therefore, it is necessary to establish a sensitive, efficient, and reliable method for qualitative and quantitative detection of tetracycline. In this paper, we synthesized fluorescent carbon dots (FCDs) by thermal cracking of crab shell waste, and obtained a fluorescence quantum yield of 30%. Characterization of the FCDs by transmission electron microscopy, Fourier-transform infrared spectroscopy, ultraviolet visible absorption spectroscopy, and photoluminescence spectroscopy showed that they were fluorescent and evenly distributed with an average size of approximately 10 nm. We designed a sensitive probe for detecting tetracycline using the fluorescence intensity change of the FCDs. This method is sensitive, inexpensive, and environmentally friendly. The concentration of tetracycline was examined by comparing the fluorescence intensities of the FCDs before and after tetracycline addition. The limit of detection for tetracycline was 0.005 mg/L (signal-to-noise ratio = 3), which is promising for method development.

Abstract:Developing an effective method for monitoring of tetracycline (TC) concentrations is extremely crucial due to its adverse effects on both ecosystems and human health. In this study, we developed a novel glutathione-protected nickel nanoclusters (GSH-NiNCs) fluorescent (FL) sensing platform to detect TC. The GSH-Ni NCs were synthesized by a facile one-pot route with GSH as a stabilizer and ascorbic acid (AA) as reducing agent. The experimental results confirmed that the FL of GSH-NiNCs was quenched in the presence of TC based on both of the inner-filter effect (IFE) and photoinduced electron transfer (PET) mechanisms to achieve high sensitivity. Under the optimal conditions, excellent linear relationships existed between the FL intensities of GSH-NiNCs and the logarithmic TC concentrations in the range of 0.5–120.0 and 120.0–300.0 μM. Even in the absence of surface modification or any complex signal amplification techniques, the proposed FL sensing platform for TC detection showed a low limit of detection of 58 nM (3σ/k). Thus, this approach provides considerable simplicity, low-cost, timesaving and flexibility for the construction of a TC sensor. Most remarkably, we could selectively analyze TC in milk samples without the need for tedious or time-consuming pretreatment processes. This work expand the FL analysis platform for TC detection, and facilitates the design and development of NiNC-based FL sensors for the monitoring of antibiotics.

G-quadruplex Specific Thioflavin T-based Label-Free Fluorescence Aptasensor for Rapid Detection of Tetracycline.

Abstract:A label-free fluorescence aptasensor was developed for the rapid detection of tetracycline (TET) based on G-quadruplex structure of TET aptamers and G-quadruplex specific dye Thioflavin T (ThT). The fluorescence of free ThT is essentially weak in aqueous solution, whereas it selectively identifies the G-quadruplex of aptamers to form the G-quadruplex/ThT conjugates, resulting in an enormous increase of the fluorescence intensity. However, the fluorescence intensity of G-quadruplex/ThT conjugates was drastically suppressed due to the release of free ThT from G-quadruplex/ThT conjugates after the addition of TET via specific binding with TET aptamers. The key factors affecting sensitivity and selectivity including the reaction medium, binding time of ThT to TET aptamers, incubation time between TET aptamers and TET, concentration of ThT and TET aptamers were investigated in detail. The optimal conditions were as follows: ultrapure water as reaction medium, binding time of 5min, incubation time of 1 min, 9.0 mu mol/L ThT and 0.03 mu mol/L aptamers. A good linear relationship (correlation coefficient of 0.9973) was obtained between the fluorescence quenching efficiency (F-0 - F) / F0 and the logarithm of TET concentration in the range of 0.01-1.0 mu mol/L. The limit of detection was 0.001 mu mol/L (S/N = 3). The proposed assay was applied for the detection of TET in the spiked honey and milk samples with recoveries ranging from93.5% to 106.9%. The developed label-free fluorescence aptasensor showed advantages of high specificity, low cost and short time-consuming, illustrating potential application for on-site detection of TET in foodstuffs. Multicolor fluorescence assay of tetracycline: lanthanide complexed amino clay loaded with copper nanoclusters

Abstract:A multicolor fluorescent nanoprobe has been prepared by loading bovine serum albumin–stabilized copper nanoclusters (BSA-Cu NCs) onto amino clay (AC) and grafting Eu 3+ with auxiliary ligand citric acid (Cit). Tetracycline (TC) can coordinate with Eu 3+ by β-diketone structure and transfer energy to Eu 3+ through antenna effect. When the concentration of TC is in the range 0 to 13 μM, the blue emission intensity of BSA-CuNCs is basically unchanged, and the red emission of Eu 3+ is remarkably enhanced due to the coordination with TC. The emission color gradually changes from blue to red under UV lamp ( λ = 365 nm). However, when the TC concentration is in the range 13 to 350 μΜ, the internal amino acid residues of BSA sensitize TC, and the emission color gradually changed from red to green. The nanoprobe has rich color, is simple to prepare, portable, and provide a wide detection range. The limit of detection (LOD) is 3.04 nM, which could be used for real-time visual analysis of trace TC in actual samples (lake water, milk, honey, and bovine serum albumin). In addition, a visual test paper has been designed and combined with the color scanning APP of a smartphone to complete the qualitative and semi-quantitative test of TC.
Copper doped carbon dots as the multi-functional fluorescent sensing platform for tetracyclines and pH

Abstract:The copper doped fluorescent carbon dots (Cu-CDs) with excellent biocompatibility were prepared from l-tryptophan and copper chloride as precursors through a facilely hydrothermal method. As-prepared Cu-CDs exhibited the bright blue fluorescence and excellent optical stability. In the presence of tetracyclines antibiotics (TCs), the intensely fluorescent quenching of Cu-CDs occurred from internal filtration effect (IFE), providing the foundation of TCs quantification. Meanwhile, the fluorescence intensity of Cu-CDs decreased gradually along with the increasing of pH and a sigmoidal curve was well fitted with the pKa12.49 and pKa2 6.21, respectively. The Cu-CDs as a multi-functional fluorescence sensing platform demonstrated great accuracy and selectivity for tetracyclines (TCs) and pH. On account of practical application, as-synthesized Cu-CDs not only were effectively employed for determination of TCs in antibiotic tablets and milk products, but also could trace nuclear and had been extended to sense and visualize pH fluctuations in cells due to low toxicity and biocompatibility. As a facile and effective multi-functional fluorescent sensor, the proposed Cu-CDs have a significant potential for biosensing and biomedical imaging.
A Ratiometric Fluorescent Nano-Probe for Rapid and Specific Detection of Tetracycline Residues Based on a Dye-Doped Functionalized Nanoscaled Metal–Organic Framework

Abstract:Tetracycline (TC) residues are harmful to the environment and human body, so it is necessary to develop a highly sensitive probe for rapid detection of tetracycline residues. In the present paper, a novel dye-doped porous metal–organic framework (UiO-66)-based multi-color fluorescent nano-probe was designed for sensitive ratiometric detection of tetracycline (TC). In this probe, dye-molecules doped UiO-66 was used as a fluorescent internal standard, and the externally grafted lanthanide Eu3+ complex was used as response signals. The fluorescence of the Eu3+ complex was selectively enhanced with increasing concentrations of TC, which was accompanied by a visual blue-to-red color switch. The nano-probe had a linear response between 0.1 and 6 μM with a lowest detection limit of 17.9 nM, which was much lower than the maximum residue limits set by the United States Food and Drug Administration (676 nM) and the European Union (225 nM). The applicability of this method in the analysis of actual samples was evaluated by the determination of TC in honey and milk samples, indicating satisfactory recovery and good reproducibility. In addition, a cost-effective paper-based probe for rapid and visual detection of TC was developed by fixing the nano-probe on filter papers. With the help of a smartphone camera to capture the fluorescence color, and chromaticity analysis software, the calculation and analysis of red (R) and blue (B) values can be realized, which has the potential for real-time visual detection of TC.

Abstract:In this work, A novel label-free electrochemical biosensor based on octahedral Cu 2 O @ Au nanocomposites was developed for the detection of tetracycline. The gold nanoparticles (AuNPs) on the surface of Cu 2 O nanomaterials not only improve the electrochemical performance, but also can be used as a binding site for thiol-modified tetracycline aptamers, which can specifically bind to tetracycline. Cu 2 O @ Au nanocomposites provide a synergistic effect of electrochemical signal amplification and tetracycline recognition strategy. Under the optimal conditions, the proposed biosensor exhibited different electrochemical reactions for different concentrations of tetracycline, with a linear range of 1 nM-1000 μM and a detection limit of 0.16 nM. This work provides a new idea for rapid and sensitive detection of tetracycline by using a simple and economical tetracycline aptamer combined with Cu 2 O @ Au biosensor platform.

Abstract:Tetracycline (TET) is a broad-spectrum antibiotic commonly used in the treatment of animals. TET residues in food inevitably threaten human health. High-performance analytical techniques for TET detection are required in food quality assessment. The objective of this study was to establish a label-free fluorescent biosensor for TET detection using specific aptamer-templated silver nanoclusters (AgNCs). An aptamer with a high specific binding ability to TET was used to synthesize a novel DNA-templated AgNCs (DNA-AgNCs). When TET is present, the aptamer's conformation switched from an antiparallel G-quadruplex to a hairpin structure, altering the connection between AgNCs and the aptamer. Following the transformation of AgNCs into large sized silver nanoparticles (AgNPs), a fluorescence decrease was detected. When used to detect TET in milk, the proposed biosensor displayed high sensitivity and selectivity, with a limit of detection of 11.46 ng/mL, a linear range of 20 ng/mL−10 g/mL, and good recoveries of 97.7–114.6% under optimized conditions. These results demonstrate that the proposed biosensor was successfully used to determine TET quantitatively in food samples, suggesting that our method provides an efficient and novel reference for detecting antibiotics in food while expanding the application of DNA-AgNCs in related fields.

Abstract:Gold nanoclusters (AuNCs) have shown great potential in mimicking the redox properties of nanozymes. However, limited fundamental knowledge and a lack of an effective means have hindered the enhancement of their catalysis efficiency. In this study, we report a simple protocol for the synthesis of D-trytophan methyl ester protected AuNCs and exhibit their peroxidase-like activity in a chromogenic reaction coupling with 3,3',5,5'-tetramethylbenzidine-H2O2. Interestingly, the addition of tetracycline resulted in a remarkable acceleration of the redox reaction rate with AuNCs as the nanozymes due to the increased levels of reactive oxygen species. The UV-vis adsorption intensity of oxidized 3,3′,5,5′-tetramethylbenzidine presented a good linear relationship with the concentration of tetracycline in the range of 1.5 to 30.0 µM and the limit of detection of the method was 0.2 µM. The proposed assay was further applied in the analysis of serum tetracycline. This study enriches our basic understanding of the enhancement of the catalytic efficiency of AuNCs-based nanozymes.

Abstract:Concentration-dependent photoluminescence carbon dots (CDs) have been successfully synthesized through the one-step hydrothermal treatment of o-phthalic acid and ethylenediamine. The CDs possessed higher fluorescence quantum yield, up to 39.22%, exhibiting distinguished optical property, water solubility, and stability. The CDs that emit strong blue-green fluorescence can visually identify and determine tetracycline (TC), oxytetracycline (OTC), and chlortetracycline (CTC). TC quenched the fluorescence of CDs at 500 nm owing to the inner filter effect; OTC behaved similarly, but the emission wavelength of CDs was red-shifted to 515 nm. Inversely, once CTC was introduced to CDs solution, the fluorescence increased and the emission peak was blue-shifted to 450 nm. Bandgap transition and electrostatic interaction were proposed to be the mechanisms for the detection of OTC and CTC by CDs. Wide linear relationships were established for TC, OTC, and CTC with the limits of detection to be 50 nM, 36 nM, and 373 nM, respectively. Furthermore, the nanoscale probe constructed by this system has been applied to detect tetracyclines (TCs) in complex samples with satisfying recoveries (93.2–114%) and was designed as a portable test strip sensor for visually on-site TCs of honey sample screening. Accordingly, the preparation process of the nano fluorescent probe is simple and environmentally friendly, and the probe has a specific recognition ability for tetracyclines. The synthesized CDs in this work provide a new orientation for fast, effective, and visual real-time detection of tetracycline in actual samples.

Abstract:Abstract Metal–organic frameworks (MOFs) with high stability and porosity have gained great attention in bioanalysis due to their potential in improving sensitivity and robustness of assays. Herein, to improve both the stability and the emission intensity of Cu nanoclusters (CuNCs), in situ entrapment strategy of CuNCs into zeolitic imidazolate framework-8 (ZIF-8) is described. Blue emissive and stable CuNCs was prepared, for the first time, using thiamine hydrochloride as capping agents, and showed strong and stable emission at 440 nm when excited at 375 nm with fluorescence quantum yields 12%. Encapsulation of CuNC into ZIF-8 showed dramatic enhancement of the fluorescence intensity up to 53% fluorescence quantum yield. Furthermore, the CuNCs@ZIF-8 possesses better stability (more than three months) due to protective and confinement effect of MOFs. Upon the addition of tetracycline to CuNCs@ZIF-8 solution, the blue emission intensity was significantly decreased. The fluorescence ratio (Fo/F) against the concentration of tetracycline exhibited a satisfactory linear relationship from 1.0 to 10.0 μM with a detection limit (LOD) of 0.30 μM. Abstract:Tetracycline (TC) and copper ion (Cu2+), as important additives in animal feed, play a crucial role in disease prevention and growth regulation. However, the abuse leads to concentration accumulation, which seriously threatens human health and the ecological environment. There is an urgent need to develop a detection method to achieve fast and synchronous detection of these pollutants without cross-interference. Here, a carbon dots-doped lanthanide-based fluorescent nanosensor (CDs@Tb-MOFs@SiO2-NH2-Eu) was synthesized, which can detect TC in the 380 nm channel by "antenna effect" and internal filtering effects (IFE), and identify Cu2+ in the 320 nm channel. The sensor was highly sensitive to TC within 0-4 μM with a detection limit as low as 3.64 nM, and Cu2+ could be detected within 0-40 μM with a detection limit of 38 nM. A portable dual-channel visual fluorescence sensor was obtained by loading the probes onto test paper and cotton swabs in food samples, which indicates the practicability of this sensing strategy.

Abstract:A ratiometric fluorescence probe is proposed for sensitive and visual detection of tetracyclinee (TC) based on cascade fluorescence signal amplification induced by bovine serum albumin–stabilized copper nanoclusters (BSA-CuNCs) and yttrium ions (Y 3+ ). TC can combine with Y 3+ to form the complex (TC-Y 3+ ) to enhance the fluorescence of TC at 515 nm. Then, positively charged TC-Y 3+ and negatively charged BSA-CuNCs was bonded together by electrostatic interactions to achieve the fluorescence resonance energy transfer (FRET) process. With the increase of TC concentration, the fluorescence intensity of TC-Y 3+ at 515 nm (F 515 ) gradually increased; meanwhile, the fluorescence intensity of BSA-CuNCs at 405 nm (F 405 ) decreased gradually. The ratio of F 515 and F 405 was used for the quantitative determination of TC. The linear range of the constructed fluorescent probe is 1.0 to 60.0 μM, and the limit of detection is 0.22 μM. The method was successfully applied to the determination of TC in spiked milk with recoveries ranging from 94.3 to 112%. Furthermore, the color of this platform can be observed from dark violet to bright green under the UV lamp. Since the response time of the reaction is less than 10 s, an intelligent sensing platform based on the use of the smartphone as image acquisition equipment was also established to realize rapid on-site and portable detection of TC through the colorimetric recognition application.

Abstract:In this study, a water-soluble fluorescent probe based on copper nanoparticles (CuNPs) was synthesized using a hydrothermal method with bovine serum albumin (BSA) (BSA@CuNPs) as a protectant, and it was utilized to efficiently detect the concentration of tetracycline (TC). Upon the addition of TC to the BSA@CuNPs probe, its fluorescence intensity is obviously diminished due to the keto-enol functions C=O group in TC connects with the hydroxyl group on the BSA@CuNPs probe to form a non-fluorescent complex, which can be successfully utilized for detecting TC concentration. Linear range and detection limit for TC detection are 0.1–130 μM and 90 nM. Furthermore, the BSA@CuNPs probe was applied to detect TC concentrations in real meat samples, with recoveries ranging from 95.40% to 102.13%, and the relative standard deviation ( RSD ) was below 2.73%. Therefore, BSA@CuNPs probe exhibits high sensitivity and selectivity for TC, which will be used for the study of TC analysis in meat.
