Abstract:In this contribution, we demonstrate a wide-field hyperspectral mid-infrared (MIR) microscope based on multidimensional single-pixel imaging (SPI). The microscope employs a high brightness MIR supercontinuum source for broadband (1.55 [Formula: see text]-4.5 [Formula: see text]) sample illumination. Hyperspectral imaging capability is achieved by a single micro-opto-electro-mechanical digital micromirror device (DMD), which provides both spatial and spectral differentiation. For that purpose the operational spectral bandwidth of the DMD was significantly extended into the MIR spectral region. In the presented design, the DMD fulfills two essential tasks. On the one hand, as standard for the SPI approach, the DMD sequentially masks captured scenes enabling diffraction-limited imaging in the tens of millisecond time-regime. On the other hand, the diffraction at the micromirrors leads to dispersion of the projected field and thus allows for wavelength selection without the application of additional dispersive optical elements, such as gratings or prisms. In the experimental part, first of all, the imaging and spectral capabilities of the hyperspectral microscope are characterized. The spatial and spectral resolution is assessed by means of test targets and linear variable filters, respectively. At a wavelength of 4.15 [Formula: see text] a spatial resolution of 4.92 [Formula: see text] is achieved with a native spectral resolution better than 118.1 nm. Further, a post-processing method for drastic enhancement of the spectral resolution is proposed and discussed. The performance of the MIR hyperspectral microsopce is demonstrated for label-free chemical imaging and examination of polymer compounds and red blood cells. The acquisition and reconstruction of Hadamard sampled 64 [Formula: see text] 64 images is achieved in 450 ms and 162 ms, respectively. Thus, combined with an unprecedented intrinsic flexibiliy gained by a tunable field of view and adjustable spatial resolution, the demonstrated design drastically improves the sample throughput in MIR chemical and biomedical imaging.

Abstract:We introduce a wide field hyperspectral microscope using the Fourier-transform approach. The interferometer is based on the Translating-Wedge-Based Identical Pulses eNcoding System (TWINS) [Opt. Lett. 37, 3027 (2012)], a common-path birefringent interferometer which combines compactness, intrinsic interferometric delay precision, long-term stability and insensitivity to vibrations. We describe three different implementations of our system: two prototypes designed to test different optical schemes and an add-on for a commercial microscope. We show high-quality spectral microscopy of the fluorescence from stained cells and powders of inorganic pigments, demonstrating that the device is suited to biology and materials science. We demonstrate the acquisition of a 1Mpixel hyperspectral image in 75 seconds in the spectral range from 400 to 1100 nm. We also introduce an acquisition method which synthesizes a tunable spectral filter, providing band-passed images by the measurement of only two maps.

Abstract:Spectral measurements in the infrared (IR) optical range provide unique fingerprints of materials which are useful for material analysis, environmental sensing, and health diagnostics. Current IR spectroscopy techniques require the use of optical equipment suited for operation in the IR range, which faces challenges of inferior performance and high cost. Here we develop a spectroscopy technique, which allows spectral measurements in the IR range using visible spectral range components. The technique is based on nonlinear interference of infrared and visible photons, produced via Spontaneous Parametric Down Conversion (SPDC). The intensity interference pattern for a visible photon depends on the phase of an IR photon, which travels through the media. This allows determining properties of the media in the IR range from the measurements of visible photons. The technique can substitute and/or complement conventional IR spectroscopy techniques, as it uses well-developed optical components for the visible range.

Abstract:Optical photothermal microscopy is a powerful, emerging method that overcomes the diffraction limit in infrared hyperspectral imaging by utilizing a visible probe laser beam to detect local temperature-induced modulation at the visible diffraction limit. However, the spectral range of this technique has been limited by the tuning range of infrared sources, which is generally restricted to the fingerprint window with commercially available quantum cascade lasers. In this work, ultra-broadband synchrotron infrared radiation was used for infrared photothermal imaging and spectroscopy, spanning the entire mid-infrared range. Both optical- and fluorescence-detected photothermal modalities were performed, demonstrating improved spectral range when compared to optical photothermal microscopy using commercial sources and improved spatial resolution when compared to synchrotron micro-spectroscopy measurements. Following these initial validation studies, synchrotron Fourier-transform optical photothermal infrared spectroscopy (FT-OPTIR) in combination with synchrotron micro-spectroscopy measurements were used to differentiate cells in mouse brain tissue sections.

Abstract:Infrared nanospectroscopy enables novel possibilities for chemical and structural analysis of nanocomposites, biomaterials or optoelectronic devices. Here we introduce hyperspectral infrared nanoimaging based on Fourier transform infrared nanospectroscopy with a tunable bandwidth-limited laser continuum. We describe the technical implementations and present hyperspectral infrared near-field images of about 5,000 pixel, each one covering the spectral range from 1,000 to 1,900 cm−1. To verify the technique and to demonstrate its application potential, we imaged a three-component polymer blend and a melanin granule in a human hair cross-section, and demonstrate that multivariate data analysis can be applied for extracting spatially resolved chemical information. Particularly, we demonstrate that distribution and chemical interaction between the polymer components can be mapped with a spatial resolution of about 30 nm. We foresee wide application potential of hyperspectral infrared nanoimaging for valuable chemical materials characterization and quality control in various fields ranging from materials sciences to biomedicine.

Abstract:Hyperspectral imaging captures both spectral and spatial information from a sample. The near infrared (NIR, > 800 nm) is advantageous for biomedical imaging as it falls into the tissue transparency window but also contains vibrational overtone and combination modes useful for molecular fingerprinting. Here, we demonstrate hyperspectral NIR imaging using a spectral phasor transformation (HyperNIR). This method employs a liquid crystal variable retarder (LCVR) for tunable, wavelength-dependent sine-, cosine and no filtering that transforms optical signals into phasor space. Spectral information is thus obtained with just three images. The LCVR can be adjusted to cover a spectral range from 900 nm to 1600 nm in windows tunable from 50 nm to 700 nm. This approach enables distinguishing NIR fluorophores with emission peaks less than 5 nm apart. Furthermore, we demonstrate label-free hyperspectral NIR reflectance imaging to identify plastic polymers and to monitor in vivo plant health. The approach uses the full camera resolution and reaches hyperspectral frame rates of 0.2 per second, limited only by the switching rate of the LCVR. HyperNIR facilitates straightforward hyperspectral imaging with standard NIR cameras for applications in biomedical imaging and environmental monitoring.

Abstract:Hyperspectral imaging (HSI) acquires a three-dimensional dataset called hypercube, with two spatial dimensions and one spectral dimension. HSI is an emerging imaging modality for various medical applications, especially in disease diagnosis of early and biomedical research based on their unique spectral signatures. A visible-near-infrared HSI for microspectroscopy is designed, the measured spectrum is from 450 nm to 900 nm, which is sampled by 256 spectral channels. The spatial resolution is 1.25μm.The modulation transfer function (MTF) value of full spectrum and full field of view is close to the diffraction limit. At last, this microspecroscopy have been fabricated and preliminary tests have been implemented. The results indicated this visible-near-infrared hyperspectral microscopy optical systems have excellent optical performances. This hyperspectral microscopy will be well developed and used in the life sciences fields.

Abstract:We demonstrate a new scheme for infrared spectroscopy with visible light sources and detectors. The technique relies on the nonlinear interference of correlated photons, produced via spontaneous parametric down conversion in a nonlinear crystal. Visible and infrared photons are split into two paths and the infrared photons interact with the sample under study. The photons are reflected back to the crystal, resembling a conventional Michelson interferometer. Interference of the visible photons is observed and it is dependent on the phases of all three interacting photons: pump, visible and infrared. The transmission coefficient and the refractive index of the sample in the infrared range can be inferred from the interference pattern of visible photons. The method does not require the use of potentially expensive and inefficient infrared detectors and sources, it can be applied to a broad variety of samples, and it does not require a priori knowledge of sample properties in the visible range.

Abstract:Spectrally-resolved imaging provides a spectrum for each pixel of an image that, in the mid-infrared, can enable its chemical composition to be mapped by exploiting the correlation between spectroscopic features and specific molecular groups. The compatibility of Fourier-transform interferometry with full-field imaging makes it the spectroscopic method of choice, but Nyquist-limited fringe sampling restricts the increments of the interferometer arm length to no more than a few microns, making the acquisition time-consuming. Here, we demonstrate a compressive hyperspectral imaging strategy that combines non-uniform sampling and a smoothness-promoting prior to acquire data at 15% of the Nyquist rate, providing a significant acquisition-rate improvement over state-of-the-art techniques. By illuminating test objects with a sequence of suitably designed light spectra, we demonstrate compressive hyperspectral imaging across the 700-1400 cm-1 region in transmission mode. A post-processing analysis of the resulting hyperspectral images shows the potential of the method for efficient non-destructive classification of different materials on painted cultural heritage.

Abstract:Hyperspectral imaging aims at providing information on both the spatial and the spectral distribution of light, with high resolution. However, state-of-the-art protocols are characterized by an intrinsic trade-off imposing to sacrifice either resolution or image acquisition speed. We address this limitation by exploiting light intensity correlations, which are shown to enable overcoming the typical downsides of traditional hyperspectral imaging techniques, both scanning and snapshot. The proposed approach also opens possibilities that are not otherwise achievable, such as sharper imaging and natural filtering of broadband spectral components that would otherwise hide the spectrum of interest. The enabled combination of high spatial and spectral resolution, high speed, and insensitivity to undesired spectral features shall lead to a paradigm change in hyperspectral imaging devices and open-up new application scenarios.

Abstract:An approach is described for spectrally parallel hyperspectral mid-infrared imaging with spatial resolution dictated by fluorescence imaging. Quantum cascade laser (QCL)-based dual-comb mid-infrared spectroscopy enables acquisition of infrared spectra at high speed (<1 millisecond) through generation of optical beat patterns and radio-frequency detection. The high-speed nature of the spectral acquisition is shown to support spectral mapping in microscopy measurements. Direct detection of the transmitted infrared beam yields high signal-to-noise spectral information, but the long infrared wavelength imposes low diffraction-limited spatial resolution. Use of fluorescence detected photothermal infrared (F-PTIR) imaging provides high spatial resolution tied directly to the integrated IR absorption. Computational imaging using a multi-agent consensus equilibrium (MACE) approach combines the high spatial resolution of F-PTIR and the high spectral information of dual-comb infrared transmission in a single optimized equilibrium hyperspectral data cube

Abstract:Mid-infrared photothermal microscopy is a new chemical imaging technology in which a visible beam senses the photothermal effect induced by a pulsed infrared laser. This technology provides infrared spectroscopic information at submicrometer spatial resolution and enables infrared spectroscopy and imaging of living cells and organisms. Yet, current mid-infrared photothermal imaging sensitivity suffers from a weak dependence of scattering on the temperature, and the image quality is vulnerable to the speckles caused by scattering. Here, we present a novel version of mid-infrared photothermal microscopy in which thermosensitive fluorescent probes are harnessed to sense the mid-infrared photothermal effect. The fluorescence intensity can be modulated at the level of 1% per Kelvin, which is 100 times larger than the modulation of scattering intensity. In addition, fluorescence emission is free of interference, thus much improving the image quality. Moreover, fluorophores can target specific organelles or biomolecules, thus augmenting the specificity of photothermal imaging. Spectral fidelity is confirmed through fingerprinting a single bacterium. Abstract:Mid-infrared microscopy is an important tool for biological analyses, allowing a direct probe of molecular bonds in their low energy landscape. In addition to the label-free extraction of spectroscopic information, the application of broadband sources can provide a third dimension of chemical specificity. However, to enable widespread deployment, mid-infrared microscopy platforms need to be compact and robust while offering high speed, broad bandwidth and high signal-to-noise ratio (SNR). In this study, we experimentally showcase the integration of a broadband, high-repetition-rate dual-comb spectrometer (DCS) in the mid-infrared range with a scanning microscope. We employ a set of 1-GHz mid-infrared frequency combs, demonstrating their capability for high-speed and broadband hyperspectral imaging of polymers and ovarian tissue. The system covers 1000 $\mathrm{cm^{-1}}$ at $\mathrm{\nu_c=2941 \; cm^{-1}}$ with 12.86 kHz spectra acquisition rate and 5 $\mathrm{\mu m}$ spatial resolution. Taken together, our experiments and analysis elucidate the trade-off between bandwidth and speed in DCS as it relates to microscopy. This provides a roadmap for the future advancement and application of high-repetition-rate DCS hyperspectral imaging.

Abstract:Infrared (IR) spectroscopy depicts molecular structure and dynamics based on vibrational absorption of chemical bonds. Spatially resolved IR spectroscopy, i.e. IR imaging, further enabled label-free in situ chemical imaging for dynamics in complex systems. However, IR imaging suffers from low spatial resolution at a few micrometers due to diffraction limit, thus having difficulty in applications such as sub-cellular imaging. Recently, by visible light probing of the photothermal effect of vibrational absorption, mid-infrared photothermal imaging (MIP) overcomes the limitations of conventional IR microscopy and has achieved sub-micron resolution. In this work, we built an optimized MIP system to boost the spatial resolution and sensitivity, and demonstrated MIP imaging of nanometer-sized polymeric microspheres and living cells with a high spatial resolution of 200 nm.

Abstract:Optical micro-spectroscopy is an invaluable tool for studying and characterizing samples ranging from classical semiconductors to low-dimensional materials and heterostructures. To date, most implementations are based on point-scanning techniques, which are flexible and reliable, but slow. Here, we describe a setup for highly parallel acquisition of hyperspectral reflection and photoluminescence microscope images using a push-broom technique. Spatial as well as spectral distortions are characterized and their digital corrections are presented. We demonstrate close-to diffraction-limited spatial imaging performance and a spectral resolution limited by the spectrograph. The capabilities of the setup are demonstrated by recording a hyperspectral photoluminescence map of a CVD-grown MoSe$_2$-WSe$_2$ lateral heterostructure, from which we extract the luminescence energies, intensities and peak widths across the interface.

Hyperspectral fluorescence imaging using a high-speed silicon photomultiplier array

Abstract:High-speed multiplex imaging of fluorescent probes is limited by a combination of spectral resolution, sensitivity, high cost and low light throughput of detectors, and filters. In this work, we present a hyperspectral detection system based on a silicon photomultiplier array that enables high-speed, high-light throughput hyperspectral imaging at low cost. We demonstrate 16 spectral channel imaging at 50 MP/s (800M spectra per second) with a conventional two photon microscope combined with a generalized spectral unmixing model that enables extraction of spectrally overlapping fluorophores. We show that the high spectral resolution combined with high throughput enables the multiplexing of multiple contrast agents over large areas and the detection of subtle spectral shifts associated with molecular binding. Silicon photomultiplier arrays may be a promising method to extend multiplex fluorescence imaging in a variety of scenarios.
