Abstract:Alzheimer's disease (AD) is a neurodegenerative disease with insidious onset and progressive development. It is clinically characterized by cognitive impairment, memory impairment and behavioral change. Chinese herbal medicine and acupuncture are important components of traditional Chinese medicine (TCM), and are commonly used in clinical treatment of AD. This paper systematically summarizes the research progress of traditional Chinese medicine natural products and acupuncture treatment of AD, which combined with existing clinical and preclinical evidence, based on a comprehensive review of neuroinflammation, and discusses the efficacy and potential mechanisms of traditional Chinese medicine natural products and acupuncture treatment of AD. Resveratrol, curcumin, kaempferol and other Chinese herbal medicine components can significantly inhibit the neuroinflammation of AD in vivo and in vitro, and are candidates for the treatment of AD. Acupuncture can alleviate the memory and cognitive impairment of AD by improving neuroinflammation, synaptic plasticity, nerve cell apoptosis and reducing the production and aggregation of amyloid β protein (Aβ) in the brain. It has the characteristics of early, safe, effective and benign bidirectional adjustment. The purpose of this paper is to provide a basis for improving the clinical strategies of TCM for the treatment of AD.

Abstract:Alzheimer's disease (AD) is an age-related neurodegenerative disease that is primary characterized as a cognitive disorder. Its pathology is characterized by the formation of senile plaques in the brain from amyloid-beta (Aβ) aggregation, neuronal fibrillary tangles from hyperphosphorylated tau protein aggregation, prolonged inflammatory responses, and neuronal death. The pathogenesis and clinical manifestations of AD are complex, but aging is generally accepted as one of the most important contributing factors. In addition, there are several hypotheses, including the Aβ hypothesis based on amyloid plaques, the tau hypothesis based on neuronal fiber entanglement, the inflammation hypothesis based on long-term inflammatory responses causing brain damage, and the neuroprotection hypothesis based on synaptic dysfunction and neuronal death. Although the pathogenesis of AD has been broadly classified into four major hypotheses, there are multiple forms of interactions, which is one of the reasons for its complex pathogenesis. Numerous epidemiological studies have shown the important role of genes in AD, followed by brain damage, hyperlipidemia, diabetes, hypertension, and obesity as risk factors for the disease. Despite years of research, several mysteries in AD remain unsolved. Drugs based on various pathogenetic hypotheses are being investigated in large numbers, but the effects are unsatisfactory. In recent years, traditional Chinese medicine (TCM) has made excellent progress and is expected to provide a new possibility for AD treatment. In this review, we focus on the latest developments in studies on the risk factors-Aβ aggregates and related factors such as apolipoprotein E, synaptic loss, and fatty acids, and then present the progress in the research of TCM based on the above pathogenesis, intended to provide a research reference and treatment for AD.

Abstract:While mentioning Alzheimer's disease, it’s horrible elderly characterized by the loss of important memories and fundamental cognitive disability. Currently, machine learning has been widely applied in the early analysis and recognition of Alzheimer's disease. This paper explores the predictive methods for Alzheimer's disease, including the application of machine learning techniques in this field. The paper compares the traditional MoCa assessment method with the machine learning-based approach, highlighting the latter's advantages in diagnostic accuracy and efficiency. It provides a detailed analysis of the main components of the machine learning predictive method, introducing their functions, advantages, and disadvantages in prediction. Subsequently, through multiple research case studies, the effectiveness and advantages of machine learning methods are demonstrated, showing superiority in both accuracy and universality. Finally, the paper emphasizes the exploration of the causation of Alzheimer's disease and proposes future directions for improvement in research, the importance of interdisciplinary collaboration, and prospects for future research.

Abstract:Alzheimer's disease (AD) is a progressive age-related neurodegenerative disease characterized by memory loss and cognitive impairment. The major characteristics of AD are amyloid β plaques, apoptosis, autophagy dysfunction, neuroinflammation, oxidative stress, and mitochondrial dysfunction. These are mostly used as the significant indicators for selecting the effects of potential drugs. It is imperative to explain AD pathogenesis and realize productive treatments. Although the currently used chemical drugs for clinical applications of AD are effective in managing the symptoms, they are inadequate to achieve anticipated preventive or therapeutic outcomes. There are new strategies for treating AD. Traditional Chinese Medicine (TCM) has accumulated thousands of years of experience in treating dementia. Nowadays, numerous modern pharmacological studies have verified the efficacy of many bioactive ingredients isolated from TCM for AD treatment. In this review, representative TCM for the treatment of AD are discussed, and among these herbal medicines, the Lamiaceae family accounts for the highest proportion. It is concluded that monomers and extracts from TCM have potential therapeutic effect for AD treatment.

Abstract:Background: Recent development in neuroimaging and genetic testing technologies have made it possible to measure pathological features associated with Alzheimer's disease (AD) in vivo. Mining potential molecular markers of AD from high-dimensional, multi-modal neuroimaging and omics data will provide a new basis for early diagnosis and intervention in AD. In order to discover the real pathogenic mutation and even understand the pathogenic mechanism of AD, lots of machine learning methods have been designed and successfully applied to the analysis and processing of large-scale AD biomedical data. Objective: To introduce and summarize the applications and challenges of machine learning methods in Alzheimer's disease multi-source data analysis. Methods: The literature selected in the review is obtained from Google Scholar, PubMed, and Web of Science. The keywords of literature retrieval include Alzheimer's disease, bioinformatics, image genetics, genome-wide association research, molecular interaction network, multi-omics data integration, and so on. Conclusion: This study comprehensively introduces machine learning-based processing techniques for AD neuroimaging data and then shows the progress of computational analysis methods in omics data, such as the genome, proteome, and so on. Subsequently, machine learning methods for AD imaging analysis are also summarized. Finally, we elaborate on the current emerging technology of multi-modal neuroimaging, multi-omics data joint analysis, and present some outstanding issues and future research directions.

[Treatment of Alzheimer Disease: an Evidence-Based Review].

Abstract:OBJECTIVE:The purpose of this research was mainly two-fold: first, to get an understanding of current researches conducted on Alzheimer disease in China; second, to systematically evaluate and compare Alzheimer's treatment delivered by traditional Chinese medicine (TCM) and Western medicine.METHODS:Two steps were employed in this research. They were data collection and cleaning, followed by systemic review and qualitative analysis. The data were selected from the following two databases: CNKI (http://www.cnki.net) and Wanfang Data (http://www.wanfangdata.com.cn). Inclusion criteria were: (1) Chinese literature; (2) Published between year 1994 and year 2004; (3) Using TCM as treatment and Western medicine as control; (4) Similar research purposes and methodology; (5) Subjects were diagnosed as Alzheimer disease. Descriptive analysis, homogeneity test, meta analysis, sensitivity analysis and subgroup analysis were performed in the second step.RESULTS:Supposing all qualified studies were of high quality, we got the following conclusion: the advantage of TCM was losing because of the newly-developed acetylcholinesterase inhibitors came in market. Moreover, the studies conducted after year 2002 were more homogeneous in comparison with those conducted in early years. Those studies using mini-mental status examination (MMSE) as outcome measurement were also more homogeneous than non-MMSE measurement groups. Combined odds ratio in comparative studies was 1.5 fold higher than that in experimental studies. Regarding to different outcome measurement, those studies using TCM assessment profile were 2.58(4.79/1.86) fold higher than those using MMSE as outcome measurement.CONCLUSIONS:After systemic literature search, we found that only 40 out of 2,403 studies met our inclusion criteria. Moreover, those qualified studies were of low quality. Therefore, the external validity of this study would be compromised. The solution to this would be to improve study quality by strengthening study design and carefully select more homogeneous subjects in terms of syndrome differentiation, by so doing, the results of meta analysis will be more convincing and easily recognized by international society.

Abstract:Alzheimer's disease (AD) stands as a formidable neurodegenerative ailment that poses a substantial threat to the elderly population, with no known curative or disease-slowing drugs in existence. Among the vital and time-consuming stages in the drug discovery process, disease modeling and target identification hold particular significance. Disease modeling allows for a deeper comprehension of disease progression mechanisms and potential therapeutic avenues. On the other hand, target identification serves as the foundational step in drug development, exerting a profound influence on all subsequent phases and ultimately determining the success rate of drug development endeavors. Machine learning (ML) techniques have ushered in transformative breakthroughs in the realm of target discovery. Leveraging the strengths of large dataset analysis, multifaceted data processing, and the exploration of intricate biological mechanisms, ML has become instrumental in the quest for effective AD treatments. In this comprehensive review, we offer an account of how ML methodologies are being deployed in the pursuit of drug discovery for AD. Furthermore, we provide an overview of the utilization of ML in uncovering potential intervention strategies and prospective therapeutic targets for AD. Finally, we discuss the principal challenges and limitations currently faced by these approaches. We also explore the avenues for future research that hold promise in addressing these challenges.

Abstract:Alzheimer's disease (AD) is a neurodegenerative disorder that causes memory degradation and cognitive function impairment in elderly people. The irreversible and devastating cognitive decline brings large burdens on patients and society. So far, there is no effective treatment that can cure AD, but the process of early-stage AD can slow down. Early and accurate detection is critical for treatment. In recent years, deep-learning-based approaches have achieved great success in Alzheimer's disease diagnosis. The main objective of this paper is to review some popular conventional machine learning methods used for the classification and prediction of AD using Magnetic Resonance Imaging (MRI). The methods reviewed in this paper include support vector machine (SVM), random forest (RF), convolutional neural network (CNN), autoencoder, deep learning, and transformer. This paper also reviews pervasively used feature extractors and different types of input forms of convolutional neural network. At last, this review discusses challenges such as class imbalance and data leakage. It also discusses the trade-offs and suggestions about pre-processing techniques, deep learning, conventional machine learning methods, new techniques, and input type selection.

Abstract:Alzheimer's Disease (AD) is an irreversible neurodegenerative disease common in the elderly. The application of artificial intelligence technology to the early diagnosis of AD can not only improve the accuracy of prediction compared with traditional methods, but also save the complicated manual feature extraction of traditional methods and speed up the diagnosis. This paper reviews various applications of artificial intelligence algorithms in AD diagnosis, including machine learning, convolutional neural network, graph convolutional neural network, cyclic neural network and other mainstream deep learning technologies. The advantages and disadvantages of each approach are discussed, and finally, we discuss limitations and future prospects.

Abstract:Several (inter)national longitudinal dementia observational datasets encompassing demographic information, neuroimaging, biomarkers, neuropsychological evaluations, and muti-omics data, have ushered in a new era of potential for integrating machine learning (ML) into dementia research and clinical practice. ML, with its proficiency in handling multi-modal and high-dimensional data, has emerged as an innovative technique to facilitate early diagnosis, differential diagnosis, and to predict onset and progression of mild cognitive impairment and dementia. In this review, we evaluate current and potential applications of ML, including its history in dementia research, how it compares to traditional statistics, the types of datasets it uses and the general workflow. Moreover, we identify the technical barriers and challenges of ML implementations in clinical practice. Overall, this review provides a comprehensive understanding of ML with non-technical explanations for broader accessibility to biomedical scientists and clinicians.

Abstract:Alzheimer’s disease (AD), a growing global health concern, has been posing a significant threat to the health of the aging population. The factors contributing to the occurrence and development of AD are extremely complex, including multiple neural networks and multiple targets, which join together to formulate enormous challenges in AD treatment. Traditional Chinese medicine (TCM) possesses the characteristics to regulate multiple targets at the same time, which is consistent with the pathogenesis of AD, moreover, clinical results in TCM treating AD reveal promising effects. In this paper, we first collected anti-Alzheimer’s prescriptions and their therapeutic effects from commonly used literature databases and expanded the data to form the anti-Alzheimer’s TCM dataset. Next, we combined machine learning models to train and analyze the dataset, which was used to predict the effectiveness of new TCM prescriptions. For the first time, we proposed to use the artificial intelligence method to train the properties of nature, flavor, and channel tropism in TCM prescriptions. The accuracy of the prediction model for the effectiveness of anti-Alzheimer’s can reach up to 85%. The experimental results demonstrated that our method can precisely predict the effectiveness of prescriptions against Alzheimer’s disease, and have great value in providing guidance for the development of new anti-Alzheimer’s drugs. Finally, we built a distributed model training architecture based on federated learning to train and predict the effectiveness of TCM prescriptions under the premise of ensuring data security.

Abstract:Alzheimer's disease (AD) is a multifactorial complex disease. The pathogenesis of AD is very complicated, and involves the β-amyloid (Aβ) cascade, tau hyperphosphorylation, neuroinflammation, oxidative stress, mitochondrial dysfunction, reduced levels of neurotrophic factors, and damage and loss of synapses as well as cholinergic neurons. The multi-target characteristics of traditional Chinese medicine (TCM) may be advantageous over single-target drugs in the treatment of complex diseases. These drugs have therefore attracted more attention in the research and development of AD therapies. This review describes advances made in experimental studies of TCM for AD treatment. It discusses research, from our group and other laboratories, on TCM compound drugs (Shenwu capsule) and approximately 10 Chinese medicinal herb extracts (tetrahydroxystilbene glucoside, epimedium flavonoid, icariin, cornel iridoid glycoside, ginsenoside, puerarin, clausenamide, huperzine A, and timosaponins).

Abstract:Alzheimer's disease (AD) is one of the major neurological disorders causing death in the elderly worldwide. As a neurodegenerative disease that is difficult to prevent and cure, the pathogenesis of AD is complex and there is no effective cure. A variety of natural products derived from plants have been reported to have promising anti‐AD activities, including flavonoids, terpenes, phenolic acids and alkaloids, which can effectively relieve the symptoms of AD in a variety of ways. This paper mainly reviews the pharmacological activity and mechanisms of natural products against AD. Although the clinical efficacy of these plants still needs to be determined by further high‐quality studies, it may also provide a basis for future researchers to study anti‐AD in depth.

Abstract:The incidence of Alzheimer's disease (AD) is increasing as the life expectancy goes up. AD is characterized by progressive memory loss, associated with agnosia, dyscalculia, apraxia, and deficits in executive functioning. Although many hypotheses have been raised, the pathogenic mechanism of AD is still uncertain. Recently more researchers have been putting their eyes on herbal medicine in searching for the active candidates in AD therapy. Traditional Chinese medicine (TCM) has shown a potential effect on Alzheimer's disease with various mechanisms of action, such as modulation of nervous transmitter metabolism, alleviation of neuronal apoptosis, influence on protein and nucleic acid metabolism etc. The study of utilizing TCM and their active component to prevent and treat AD has got a rapid progress. This article reviewed the recent advances in the studies of AD treatment by using TCM.

Abstract:Alzheimer's disease (AD), a neurodegenerative disease, is closely related to the genetic and environmental factors, about 70% of which are caused by the genetic factors, but its pathogenesis is still unclear. Along with the advent of high-throughput gene sequencing, the processing of genetic data using machine learning (ML) has become a hot spot. In this paper, the applications of ML in AD are mainly reviewed, including the diagnosis and prognosis of AD based on genetic data, the analysis of genetic variation of AD, the analysis of gene expression profile of AD, and the further development of ML for AD. Firstly, during the diagnosis and prognosis of AD, the genetic data combining with other modalities, such as imaging data, clinical data and histological data, would be greatly improved the accuracy of ML methods. It is valuable for the early diagnosis of AD, and effectively delays the progression of AD. Secondly, the application of ML in the analysis of genetic variation of AD, single nucleotide polymorphisms (SNPs) of new genes were dug out, and the pathogenic mechanism of AD was further explored. Thirdly, the analysis of gene expression profile of AD mainly focuses on the discovery of the pathways of genes which could provide the possibility of gene targets for AD therapy. In the future, the multi-level model of ML might be developed for high-quality, diverse and large data, and provide scientific strategies for exploring the pathogenesis of AD.

Abstract:This study aims to investigate the network pharmacology of Chinese medicinal formulae for treatment of Alzheimer’s disease.Machine learning algorithms were applied to construct classifiers in predicting the active molecules against 25 key targets toward Alzheimer’s disease(AD).By extensive data profiling, we compiled 13 classical traditional Chinese medicine(TCM) formulas with clinical efficacy for AD. There were 7 Chinese herbs with a frequency of 5 or higher in our study. Based on the predicted results, we built constituent-target, and further construct target-target interaction network by STRING(Search Tool for the Retrieval of Interacting Genes/Proteins) and target-disease network by DAVID(Database for Annotation,Visualization and Integrated Discovery) and gene disease database to study the synergistic mechanism of the herbal constituents in the Chinese traditional patent medicine. By prediction of blood-brain penetration and validation by TCMsp (traditional Chinese medicine systems pharmacology) and Drugbank, we found 7 typical multi-target constituents which have diverse structure. The mechanism uncovered by this study may offer a deep insight into the action mechanism of TCMs for AD. The predicted inhibitors for the AD-related targets may provide a good source of new lead constituents against AD.

Abstract:Traditional Chinese medicine (TCM) is practiced in the Chinese health care system for more than 2,000 years. In recent years, herbal medicines, which are used to treat Alzheimer's disease (AD) in China based on TCM or modern pharmacological theories have attracted considerable attention. In this paper, we discuss etiology and pathogenesis of AD, TCM therapy, and herbal extracts for the treatment of AD. There is evidence to suggest that TCM therapy may offer certain complementary cognitive benefits for the treatment of AD. Chinese herb may have advantages with multiple target regulation compared with the single-target antagonist in view of TCM.
