Abstract:This issue of Medicinal Research Reviews is dedicated to Professor Li-He Zhang, Boya Chair Professor and academician of the Chinese Academy of Sciences, School of Pharmaceutical Sciences, Peking University, on the occasion of his 80th birthday. Professor Zhang obtained his bachelor degree in 1958 and his graduate diploma in 1967, both in medicinal chemistry, from the Department of Pharmacy, Beijing Medical College. As a graduate student, he worked on triazine chemistry and synthesis of sulfur substituted sugars under the guidance of Prof. Xu Wang. He worked in Dr. Sidney Hecht′s group as a research associate from 1981 to 1983 in the Department of Chemistry, University of Virginia (USA), where he focused on total synthesis of bleomycin A2 and its mechanism of action in DNA cleavage. He also worked on manual synthesis of oligonucleotides. Professor Zhang returned to China and became a professor of medicinal chemistry in 1985 in the School of Pharmaceutical Sciences, Beijing Medical University (now Peking University Health Science Center), a position that he has held until now. He also served as the dean of the School of Pharmaceutical Sciences (1987–1999), and founded the State Key Laboratory of Natural and Biomimetic Drugs in 1985 and served as its director (1992–2005). His research interest has centered on studies of various chemical and biological issues of nucleic acids, and development of anticancer and antiviral drugs. Specifically, he has developed a series of methods and strategies for precise modifications of antisense oligonucleotides, siRNA, and aptamers, and made significant contributions to the field of ribozyme mechanism of action, artificial oligonucleotides, and nucleosides and nucleotides as anticancer and antiviral agents. He has also developed many analogs of intracellular messenger nucleotides (cADPR and cAMP), carried out systematic studies of their chemical and biological activities, and discovered a cADPR-binding protein related to calcium release activities in cells. Professor Zhang has published extensively in the field of nucleic acids and medicinal chemistry with more than 300 research articles/reviews and five books. Prof. Zhang was elected into the Chinese Academy of Science in 1995 and chaired the Chemistry Division of National Natural Science Foundation of China between 1999 and 2006. He has a long list of other honors and positions including serving as chairman of the Federation of Asian Pharmaceutical Associations (1997–1999), vice Chairman of the Chinese Pharmaceutical Association (1997–2007), a titular member of Division of Organic and Biomolecular Chemistry, International Union of Pure and Applied Chemistry (IUPAC) (2005–2006), and a fellow of Royal Society of Chemistry (UK) (since 2006). His research awards include Japan Otani Research Award, Wu-Yang award in medicine, Colorcon award (FIP), HeliangHeli award, Nagai-Hisamitsu award (FAPA), and a National Award in Natural Sciences among others. He was granted an honorary doctorate degree by Hoshi University, Japan, in 1991, and Edgar-Snow Professorship by Missouri University in Kansas City, USA, in 1992. He has been on the editorial advisory boards of many scientific journals including Medicinal Research Reviews, Current Topics in Medicinal Chemistry, Organic & Biomolecular Chemistry, and ChemMedChem, He is associate editor of European Journal of Medicinal Chemistry, editor of Journal of Chinese Pharmaceutical Sciences, and associate editor of Journal of Chemistry of Chinese Universities. Prof. Zhang has been a thought leader in and a strong advocate for the field of medicinal chemistry and chemical biology in China. As a board member of the National Natural Science Foundation, he proposed and led two key research initiatives on chemical biology: regulation of signal pathways with small molecule probes (2008–2016) and dynamic modification and chemical intervention of biomacromolecules (2017–2025). To celebrate the 80th birthday of Professor Zhang, the International Symposium on Frontiers of Natural and Biomimetic Drugs was organized (October 2017) to honor his contributions in nucleic acid chemistry and medicinal chemistry. At the same time, we organized this special issue of Medicinal Research Reviews in his honor as a tribute to his many outstanding contributions in science, and to his devotion to mentoring many young scientists in the field of chemical biology and medicinal chemistry in China.
The Publication Process of the Article on the Chemical Structure of Artemisinin on &lt;italic&gt;chinese Science Bulletin&lt;/italic&gt;: A Tribute to the 40th Anniversary Publication of &ldquo;artemisinin: A New Type of Sesquiterpene Lactone&rdquo;

Abstract:On March 1977, an article named “Artemisinin: a new type of sesquiterpene lactone” was published on the Vol.22, No.3 of Chinese Science Bulletin . The author of the article is “Coordinating Research Group for the Structure of Artemisinin”, and the date of reception is February 20th, 1976. Based on original documents, the author of this paper studies the background and progress of the publication of this important article, in which he reveals the influence on Youyou Tu’s winning 2015 Noble Prize in Physiology or Medicine. The author of this paper points out: (ⅰ) the decision of publishing the research on the structure of Artemisinin through Chinese Science Bulletin was made by Institute of Chinese Materia Medica, Academy of Traditional Chinese Medicine; Institute of Biophysics, Chinese Academy of Sciences and Shanghai Institute of Organic Chemistry, Chinese Academy of Sciences after negotiation, and granted by Office 523, the agency managed the Artemisia Anti-Malaria Project. (ⅱ) The purpose of publication is to “win an honor for mother country and take the advantage to publish the results prior to the overseas counterpart”, because “Yugoslavia is conducting a research on Artemisia and has reported the chemical structure of one crystal”. (ⅲ) Two measures were taken then due to confidentiality requirement. The first one was the publication was in the name of Coordinating Research Group for the Structure of Artemisinin, instead of independent agencies. The second one was the article should only deal with the chemical structure of Artemisinin, and no description of its anti-malaria effect would appear in the article. But because of objections, no catering to “the need of pharmacy speculators from capitalist countries” or “the utilitarianism of unevolved intellectuals” is allowed, of the former Minister of Health, Xiangpin Liu, the article could not be published right on time. After the Great Proletarian Cultural Revolution, Institute of Chinese Materia Medica applied to issue the article again, and the Ministry of Health granted publishing the article on February 25th, 1977. Since Institute of Chinese Materia Medica is not only the leading agency in the publication of the article, but also the initiator of the structural confirmation of Artemisinin and the earliest provider of Artemisinin crystals, Youyou Tu, the former director of Artemisia Anti-Malaria Project in the institute, is considered as the major contributor to the article without controversy when the institute held her as the biggest hero of the project, unless there are direct evidences indicating the institute is wrong. In all, the author of this paper concludes that the article, which is as thin as only one page and was issued on Chinese Science Bulletin , is the first article on the Artemisinin research in China, and it holds a magnificent significance to Youyou Tu’s winning Noble Prize.

Abstract:For the first time in history, the Nobel Prize in Medicine 2015 is awarded to a researcher who is specialized in traditional Chinese medicine. The Chinese Tu Youyou received this highest award in medicine for her excellent work in malaria research. Tu Youyou and her team found out that artemisinin, the active ingredient of the medicinal herb “sweet wormwood”, represents an effective malaria therapy. In the following article, the research that led to this Nobel Prize is introduced. In addition, the insidious tropical disease malaria will be briefly explained. Results of the research on moxibustion using artemisia are also presented in this article; these studies were carried out at the TCM Research Center Graz, partly in close cooperation with the “China Academy of Chinese Medical Sciences”, the institution of the Nobel Prize winner.

Abstract:In 2015, the Chinese pharmacologist, Tu Youyou, was awarded the Nobel Prize for Physiology or Medicine for the discovery of artemisinin. Traditional Chinese medicine (TCM) was the source of inspiration for Tu's discovery and provides an opportunity for the world to know more about TCM as a source of medical knowledge and practice. In this article, the value of TCM is evaluated from an ethical perspective. The characteristics of 'jian, bian, yan, lian' are explored in the way they promote accessibility and economic efficiency for TCM. The article also examines how the increased use and prevalence of TCM reflects the scientific, cultural, and ethical values of TCM and their increasing attraction in meeting major challenges to medicine and health systems currently and in the future. The article discusses safety issues within TCM, which is a controversial area, and also comments on some shortcomings and challenges which pose difficulties for more widespread and greater uptake of TCM-derived clinical or therapeutic interventions. The article concludes that TCM is generally safe if it is used according to TCM theory and where such applications are cognizant of the strengths and weaknesses of TCM. TCM has important bioethical values which may inform potential measures for meeting challenges facing global health care systems and the article argues that it can have an increasing role in improving human health.

Abstract:In this paper, the initial publication of the chemical structure of artemisinin on(1977), as well as the disclosure of its antimalarial efficacy and R&D process on(1978) aredocumented,and both events have revealed the contribution of Tu Youyou and the Institute of Chinese Medicine of Academyof Traditional Chinese Medicine in the identification and structure determination of artemisinin. The author argues that thedisclosure of the particular details in artemisinin’s antimalarial research actually did not bring any disadvantages to theinvention patent application of artemisinin;instead,it has played a certain role to make Tu Youyou,a 2015 Nobel Prizewinner in Physiology or Medicine.

Abstract:Dr. Chen-Lu Tsou (1923-2006) obtained his doctorate from Cambridge in 1951, and became one of the founders of Chinese biochemistry. He spent most of his life on protein structure and function, making a number of original contributions. To commemorate his one hundred years of birth, two of his research achievements are elaborated in detail here, one of which is relation between the functional groups of proteins and their biological activity, and the other is on kinetics of irreversible modification of enzyme activity. This commemorative article intends to share with audiences how much can be achieved by the older generations of Chinese scientists with great minds but few resources, as epitomized in the two great pieces of work by Dr. TSOU.

Abstract:The story of discovery of artemisinin highlights the diversity of scientific values across time and space. Resituating artemisinin research within a broader temporal framework allows us to understand how Chinese drugs like qinghao came to articulate a space for scientific experimentation and innovation through its embodiment of alternating clusters of meanings associated with tu and yang within scientific discourse. Tu science, which was associated with terms like native, Chinese, local, rustic, mass, and crude, articulated a radical vision of science in the service of socialist revolutionary ideals. Yang science, which signified foreign, Western, elite, and professional, tended to bear the hallmarks of professionalism, transnational networks in education and training, and an emphasis on basic or foundational research. With respect to medical research, the case of artemisinin highlights how the constitution of socialist science as an interplay of tu and yang engendered different scientific values and parameters for scientific endeavor. Modern medical research in Maoist China could harness the productive energies of mass participation to technical expertise in its investigations of Chinese drugs, and under the banner of tu science, it became possible and scientifically legitimate to research Chinese drugs in ways that had previously provoked resistance and controversy.

Tu Youyou and the Discovery of High Antimalarial Potency of Sweet Wormwood

Abstract:In 1995, a note entitled Returning the Due Rights of the Inventers of Artemisinin was jointly drafted and signed by Zhang Jianfang and other eight participants of the national initiative on antimalarial research, appealing for a clarification from the government on the intellectual patent on artemisinin. The note argued that the high antimalarial potency of sweet wormwood(and its abstract artemisinin) was not Tu Youyou’s own discovery, pointing out that 1) sweet wormwood as an antimalarial therapy hadbeen recorded in ancient Chinese medical texts and 2) the antimalarial potency ofsweet wormwoodhadbeen widely applied in folk medical practices, and 3) sweet wormwoodwas already listed among the object drugs in the 523 Project of 1967. The first two points are well grounded but the third is not accurate. Mainly based on original documents and diaries, this article investigates the historical stages of the research of traditional Chinese medicine on antimalarial drugs from 1969 to 1972. Though it might be disputable to claim that Tu was the one who firstintroduced sweet wormwood into the 523 Project, it is totally grounded that she was the first to discover the neutral extract of sweet wormwood with diethyl ether as the substantive active antimalarial component and to apply it to clinical trials.

Abstract:Professor Fosong Wang was born in Guangdong, China in 1933 and received his undergraduate education at Wuhan University between 1951 and 1955. Following graduation, he went to the Former Soviet Union to continue his graduate studies whereupon, in 1960, he received his Ph.D. in chemistry from the Institute of High Molecular Compounds of the Soviet Union Academy of Science. After he returned to his motherland, Professor Wang joined the Chinese Academy of Sciences where he remains to this day. From 1985 to 1988, he served as Director of the Changchun Institute of Applied Chemistry, and from 1988 to 1994, Professor Wang served as Vice President of the Chinese Academy of Sciences.
