Abstract:The 2015 Nobel Prize in Physiology or Medicine, shared by Professor Youyou Tu, focused worldwide attention on artemisinin, a natural product antimalarial drug inspired by traditional Chinese medicine (TCM). This is the first Nobel Prize in natural sciences presented to a Chinese scientist for her impactful research work in China in collaboration with other Chinese scientists. We are delighted to provide the background and implications of the discovery of artemisinin, along with our personal viewpoints toward the affordability of modern medicines from natural products.

Abstract:Malaria is an ancient infectious disease that threatens millions of lives globally even today. The discovery of artemisinin, inspired by traditional Chinese medicine (TCM), has brought in a paradigm shift and been recognized as the "best hope for the treatment of malaria" by World Health Organization. With its high potency and low toxicity, the wide use of artemisinin effectively treats the otherwise drug-resistant parasites and helps many countries, including China, to eventually eradicate malaria. Here, we will first review the initial discovery of artemisinin, an extraordinary journey that was in stark contrast with many drugs in western medicine. We will then discuss how artemisinin and its derivatives could be repurposed to treat cancer, inflammation, immunoregulation-related diseases, and COVID-19. Finally, we will discuss the implications of the "artemisinin story" and how that can better guide the development of TCM today. We believe that artemisinin is just a starting point and TCM will play an even bigger role in healthcare in the 21st century.

Abstract:Chinese scientist Tu You?you won the Nobel Prize in Physiology or Medicine 2015,due to her outstanding contri?bution to the discovery of antimalarial drug artemisinin. The discovery of artemisinin is a great breakthrough using modern phar?maceutical technology to excavate ancient Chinese traditional medicine resources,based on government’s coordination mecha?nisms. This article analyzes the research and development status of the current global antiparasitic products,as well as China’s needs and strengths in related areas. The authors propose that China should take the advantage of government financing and re?search capacity to strengthen the research and development of antiparasitic products,so as to contribute more to improving glob?al health equity.

Abstract:In 2015, the Chinese pharmacologist, Tu Youyou, was awarded the Nobel Prize for Physiology or Medicine for the discovery of artemisinin. Traditional Chinese medicine (TCM) was the source of inspiration for Tu's discovery and provides an opportunity for the world to know more about TCM as a source of medical knowledge and practice. In this article, the value of TCM is evaluated from an ethical perspective. The characteristics of 'jian, bian, yan, lian' are explored in the way they promote accessibility and economic efficiency for TCM. The article also examines how the increased use and prevalence of TCM reflects the scientific, cultural, and ethical values of TCM and their increasing attraction in meeting major challenges to medicine and health systems currently and in the future. The article discusses safety issues within TCM, which is a controversial area, and also comments on some shortcomings and challenges which pose difficulties for more widespread and greater uptake of TCM-derived clinical or therapeutic interventions. The article concludes that TCM is generally safe if it is used according to TCM theory and where such applications are cognizant of the strengths and weaknesses of TCM. TCM has important bioethical values which may inform potential measures for meeting challenges facing global health care systems and the article argues that it can have an increasing role in improving human health.

Abstract:In this paper, the initial publication of the chemical structure of artemisinin on(1977), as well as the disclosure of its antimalarial efficacy and R&D process on(1978) aredocumented,and both events have revealed the contribution of Tu Youyou and the Institute of Chinese Medicine of Academyof Traditional Chinese Medicine in the identification and structure determination of artemisinin. Abstract:The discovery of the artemisinin involved a number of people in an unusual social background. Most of the published articles of the early history of artemisinin discovery, including that in Cell, remain controversial because they are often based on reports relying on one side of a complicated story. The controversies have not been lessened by the award of the Lasker prize to Youyou Tu. Here we present a balanced account of the history about: 1) the effective crude extract of Qinghao (the sweet wormwood); 2) the extraction process of the effective antimalarial monomer; 3) clinical validation of the effective monomer; 4) the determination of artemisinin structure. Our article is based on original documents, conference records, diaries and interview records. These materials indicate that cooperation among different scientists and institutes played significant roles.

Abstract:Half of the 2015 Nobel Prize in Physiology or Medicine was awarded to Chinese scientist Youyou Tu in recognition of her pioneering work on the antimalarial artemisinin, extracted from Artemisia annua, a traditional Chinese herbal remedy used to treat fever. This is clearly a great encouragement for scientists who engage traditional medicine research. As Youyou Tu stated in her Nobel Prize awarding lecture, artemisinin is a gift from traditional Chinese medicine (TCM) to the world, this definitely will generate great interest in not only TCM but also other traditional medicine systems from other parts of the world, including Europe, Africa, India, Americas, etc. Though the Nobel Prize selection committee did not confess that the prize is to award traditional Chinese medicine, rather a new drug discovery inspired from TCM, this is indeed the exciting and encouraging event for Chinese TCM scientists. In my opinion, it is meaningless to dispute if artemisinin is still belonging to TCMor westernmedicine. Any contribution or new discovery derived from TCM to benefit for the human health should be recognized and respected. TCM needs to be developed and modernized. Any standpoint that developed or modernized TCM does not belong to TCM may drastically hinder the TCM modernization and integration with modern medicine process, or even deepen the gap between TCM and modern medicine. Evidence-based medicine is the future of TCM by means of modern biomedicine advances and novel approaches. In the past 20 years, TCM has undergone rapid development period evidenced by a number of achievements including the geometric growth of TCM research papers, booming TCM industry, TCM new drug research and development, etc. However, TCM is still facing grand challenges in the future development. Shortage of modern scientific evidence for safety, effectiveness, quality and mode of action should be emphasized and dealt with solutions. TCM experienced-based feature should be turned into evidence-based and science-based one. Following several aspects are suggested for the future modern TCM research: a) TCM-based new drug discovery. TCM is still a non-fully explored treasure house for new drug discovery and development. According to TCM resource survey, there are over 12000 TCM species, among which majority are herbs. Apart from artemisinin and its derivatives, a number of single chemical entity drugs have been developed and successfully marketed, such as arsenic trioxide, ginsenoside Rg3, bifendate, β-elemene, indirubin, etc. It is anticipated that more new drugs could be discovered from this vast TCM resource with modern drug discovery technology and methodology. b) Research on the holistic TCM quality control. The current quality control approach is more focused on a single marker or a few marker determinations, which follows the western drug quality control model and hence, could not really monitor the quality of TCM products. Systemic and comprehensive quality control method should be developed for the TCM crude drugs, prepared slices and finished products, especially for those multiple-ingredient herbal products. c) Research on TCM processing. All TCM crude drugs should be processed in different extent before they can be used either for decoction or for herbal production. TCM processing method is mainly based on the traditional knowledge or experience. Mechanism of processing either for the purpose of enhancing efficacy or eliminating toxicity should be thoroughly investigated and clarified. d) TCM efficacy and safety research. During the development of TCM, greater attention needs to be paid to the integrated evaluation of the effectiveness and safety based on TCM theory and modern scientific research. New evaluation methodology fit for TCM complex feature should be developed with international collaborations. Globally recognized evaluation criteria should be constructed. Other TCM research focuses include pharmacokinetics, geo-authenticity, active principles and giant data library. It is firmly believed that TCM will contribute more than ever to the human health along with the rapid advances of biomedicine science and eventually developed into an evidence-based medicine system.

Abstract:Inhibitor of nuclear factor kappa-B kinase subunit beta (IKKβ) is a kinase principally known as a positive regulator of the ubiquitous transcription factor family Nuclear Factor-kappa B (NF-κB). In addition, IKKβ also phosphorylates a number of other proteins that regulate many cellular processes, from cell cycle to metabolism and differentiation. As a consequence, IKKβ affects cell physiology in a variety of ways and may promote or hamper tumoral transformation depending on hitherto unknown circumstances. In this article, we give an overview of the NF-κB-dependent and -independent functions of IKKβ. We also summarize the current knowledge about the relationship of IKKβ with cellular transformation and cancer, obtained mainly through the study of animal models with cell type-specific modifications in IKKβ expression or activity. Finally, we describe the most relevant data about IKKβ implication in cancer obtained from the analysis of the human tumoral samples gathered in The Cancer Genome Atlas (TCGA) and the Catalogue of Somatic Mutations in Cancer (COSMIC).
The Publication Process of the Article on the Chemical Structure of Artemisinin on &lt;italic&gt;chinese Science Bulletin&lt;/italic&gt;: A Tribute to the 40th Anniversary Publication of &ldquo;artemisinin: A New Type of Sesquiterpene Lactone&rdquo;

Abstract:On March 1977, an article named “Artemisinin: a new type of sesquiterpene lactone” was published on the Vol.22, No.3 of Chinese Science Bulletin . The author of the article is “Coordinating Research Group for the Structure of Artemisinin”, and the date of reception is February 20th, 1976. Based on original documents, the author of this paper studies the background and progress of the publication of this important article, in which he reveals the influence on Youyou Tu’s winning 2015 Noble Prize in Physiology or Medicine. The author of this paper points out: (ⅰ) the decision of publishing the research on the structure of Artemisinin through Chinese Science Bulletin was made by Institute of Chinese Materia Medica, Academy of Traditional Chinese Medicine; Institute of Biophysics, Chinese Academy of Sciences and Shanghai Institute of Organic Chemistry, Chinese Academy of Sciences after negotiation, and granted by Office 523, the agency managed the Artemisia Anti-Malaria Project. (ⅱ) The purpose of publication is to “win an honor for mother country and take the advantage to publish the results prior to the overseas counterpart”, because “Yugoslavia is conducting a research on Artemisia and has reported the chemical structure of one crystal”. (ⅲ) Two measures were taken then due to confidentiality requirement. The first one was the publication was in the name of Coordinating Research Group for the Structure of Artemisinin, instead of independent agencies. The second one was the article should only deal with the chemical structure of Artemisinin, and no description of its anti-malaria effect would appear in the article. But because of objections, no catering to “the need of pharmacy speculators from capitalist countries” or “the utilitarianism of unevolved intellectuals” is allowed, of the former Minister of Health, Xiangpin Liu, the article could not be published right on time. After the Great Proletarian Cultural Revolution, Institute of Chinese Materia Medica applied to issue the article again, and the Ministry of Health granted publishing the article on February 25th, 1977. Since Institute of Chinese Materia Medica is not only the leading agency in the publication of the article, but also the initiator of the structural confirmation of Artemisinin and the earliest provider of Artemisinin crystals, Youyou Tu, the former director of Artemisia Anti-Malaria Project in the institute, is considered as the major contributor to the article without controversy when the institute held her as the biggest hero of the project, unless there are direct evidences indicating the institute is wrong. In all, the author of this paper concludes that the article, which is as thin as only one page and was issued on Chinese Science Bulletin , is the first article on the Artemisinin research in China, and it holds a magnificent significance to Youyou Tu’s winning Noble Prize.

Abstract:In 1995, a note entitled Returning the Due Rights of the Inventers of Artemisinin was jointly drafted and signed by Zhang Jianfang and other eight participants of the national initiative on antimalarial research, appealing for a clarification from the government on the intellectual patent on artemisinin. The note argued that the high antimalarial potency of sweet wormwood(and its abstract artemisinin) was not Tu Youyou’s own discovery, pointing out that 1) sweet wormwood as an antimalarial therapy hadbeen recorded in ancient Chinese medical texts and 2) the antimalarial potency ofsweet wormwoodhadbeen widely applied in folk medical practices, and 3) sweet wormwoodwas already listed among the object drugs in the 523 Project of 1967. The first two points are well grounded but the third is not accurate. Mainly based on original documents and diaries, this article investigates the historical stages of the research of traditional Chinese medicine on antimalarial drugs from 1969 to 1972. Though it might be disputable to claim that Tu was the one who firstintroduced sweet wormwood into the 523 Project, it is totally grounded that she was the first to discover the neutral extract of sweet wormwood with diethyl ether as the substantive active antimalarial component and to apply it to clinical trials.

Abstract:Artemisinin and its derivatives belong to a family of drugs approved for the treatment of malaria with known clinical safety and efficacy. In addition to its anti-malarial effect, artemisinin displays anti-viral, anti-inflammatory, and anti-cancer effects in vivo and in vitro. Recently, much attention has been paid to the therapeutic role of artemisinin in liver diseases. Several studies suggest that artemisinin and its derivatives can protect the liver through different mechanisms, such as those pertaining to inflammation, proliferation, invasion, metastasis, and induction of apoptosis and autophagy. In this review, we provide a comprehensive discussion of the underlying molecular mechanisms and signaling pathways of artemisinin and its derivatives in treating liver diseases. Further pharmacological research will aid in determining whether artemisinin and its derivatives may serve as promising medicines for the treatment of liver diseases in the future.

Abstract:<p>Malaria has been a global epidemic health threat since ancient times. It still claims roughly half a million lives every year in this century. Artemisinin and its derivatives, are frontline antimalarial drugs known for their efficacy and low toxicity. After decades of wide use, artemisinins remain our bulwark against malaria. Here, we review decades of efforts that aim to understand the mechanism of action (MOA) of artemisinins, which help explain the specificity and potency of this anti-malarial drug. We summarize the methods and approaches employed to unravel the MOA of artemisinin over the last three decades, showing how the development of advanced techniques can help provide mechanistic insights and resolve some long-standing questions in the field of artemisinin research. We also provide examples to illustrate how to better repurpose artemisinins for anti-cancer therapies by leveraging on MOA. These examples point out a practical direction to engineer artemisinin for broader applications beyond malaria.</p>

Abstract:Though traditional medicines have been developed through practice for thousands of years, limited research has discussed the research and development (R&D) pattern of traditional medicines. China's discovery of artemisinin accumulated valuable experiences to explore traditional medicine under low-resource settings. With limited R&D resources, China mobilized all domestic research units at different levels and departments to develop artemisinin collaboratively. The discovery of artemisinin not only based on valuable experiential wisdom of traditional medicine, but also relied on excellent synergy among all units. In this study, we reviewed the story of how artemisinin was discovered in China, summarized key factors for new drug development from traditional medicines under limited resources, and raised suggestions to utilize traditional medicines in low- and middle-income countries. This case suggested that the vitality of traditional medicine could be extended by promoting new drug development based on modern methods and collaboration.

Abstract:Malaria is the one of major diseases which threatens people's life in Africa. Out of humanitarianism, Chinese scientists has contributed to research of Artemisinin and ACTS more than 30 years, China provides long-term antimalaria assistance to Africa and gain great achievements. In Moheli, the island of Comoros, the antimalaria group used a new strategy which universal medication and proactive intervention. They established not only an effective antimalaria system and reporting system but also a local antimalaria team. Furthermore, they enhanced publicity and put mass protection and treatment into effect. Finally, they achieved significant result. In order to apply those successful experience to other countries in Africa, this paper summed up those experience and inspirations.
