Abstract:Achromatic performance is crucial for numerous multi -wavelength optical fiber applications, including endoscopic imaging and fiber sensing. This paper presents the design and nanoprinting of a fiber-integrated achromatic diffractive lens within the visible spectrum (450-650 nm). The 3D nanoprinting is achieved by a high-resolution direct laser writing technology, overcoming limitations in the optical performance caused by the lack of an arbitrary 3D structure writing capability and an insufficient feature resolution in the current manufacturing technology for visible light broadband achromatic diffractive lenses. A three-step optimization algorithm is proposed to effectively balance optical performance with writing difficulty. The characterization results demonstrate excellent achromatic focusing performance, paving the way towards the development of nanoprinted flat optical devices for applications such as optical fiber traps, miniature illumination systems, and integrated photonic chips.(c) 2023 Optica Publishing Group
Accurate background reduction in adaptive optical 3D-STED nanoscopy by dynamic phase switching

Abstract:Stimulated emission depletion (STED) fluorescence nanoscopy allows the three-dimensional (3D) visualization of nanoscale subcellular structures, providing unique insights into their spatial organization. However, 3D-STED imaging and quantification of dense features are obstructed by the low signal-to-background ratio (SBR), resulting from optical aberrations and out-of-focus background. Here, combining with adaptive optics, we present an easy-to-implement and flexible method to improve SBR by dynamic phase switching. By switching to a counterclockwise vortex phase mask and a top-hat one with an incorrect inner radius, the depletion pattern features a nonzero-intensity center, enabling accurate background recordings. When the recorded background is subtracted from the aberration-corrected 3D-STED image, the SBR in dense sample areas can be improved by a factor of 3–6 times. Abstract:High-performance image-forming systems often require high system complexity due to the overdetermined nature of optical aberration correction. What we present here is a novel computational imaging modality which can achieve high-performance imaging using a simple non-image-forming optical system. The presented optical system contains an aspherical non-imaging lens which is designed with the optimal transfer of light radiation between an object and a detector. All spatial frequencies of the object collected by the non-imaging lens are delivered to the detector. No image is formed on the detector, and a full-path optical diffraction calculation method is developed to recover a high-quality image of the object from multiple intensity measurements. The effectiveness and high performance of the proposed imaging modality is verified by the examples.
An Efficient Projection Defocus Algorithm Based on Multi-Scale Convolution Kernel Templates

Abstract:The focal problems of projection include out-of-focus projection images from the projector caused by incomplete mechanical focus and screen-door effects produced by projection pixilation. To eliminate these defects and enhance the imaging quality and clarity of projectors, a novel adaptive projection defocus algorithm is proposed based on multi-scale convolution kernel templates. This algorithm applies the improved Sobel-Tenengrad focus evaluation function to calculate the sharpness degree of intensity equalization and then constructs multi-scale defocus convolution kernels to remap and render the defocus projection image. The resulting projection defocus corrected images can eliminate out-of-focus effects and improve the sharpness of uncorrected images. Experiments show that the algorithm works quickly and robustly and that it not only effectively eliminates visual artifacts and can run on a self-designed smart projection system in real time but also significantly improves the resolution and clarity of the observer’s visual perception.

Abstract:Non-invasive, real-time imaging and deep focus into tissue are in high demand in biomedical research. However, the aberration that is introduced by the refractive index inhomogeneity of biological tissue hinders the way forward. A rapid focusing with sensorless aberration corrections, based on machine learning, is demonstrated in this paper. The proposed method applies the Convolutional Neural Network (CNN), which can rapidly calculate the low-order aberrations from the point spread function images with Zernike modes after training. The results show that approximately 90 percent correction accuracy can be achieved. The average mean square error of each Zernike coefficient in 200 repetitions is 0.06. Furthermore, the aberration induced by 1-mm-thick phantom samples and 300-mu m-thick mouse brain slices can be efficiently compensated through loading a compensation phase on an adaptive element placed at the back-pupil plane. The phase reconstruction requires less than 0.2 s. Therefore, this method offers great potential for in vivo real-time imaging in biological science. High-contrast and High-Stable Wavefront Shaping Multimode Fiber Imaging

Abstract:The main challenge in multimode fiber imaging is modal scrambling caused by environmental fluctuation. How to get high contrast and high stable imaging is the main question. In this presentation, we propose some methods to increase the contrast-to-noise ratio and stability of multimode fiber imaging. Wavelength modulation is introduced to suppress the background. Exhaustive bending effect was used to improve the imaging stability. Wavelength modulation is introduced to enhance the CNR four fold in a 200 μm field-of-view imaging. We show a near diffraction limited focusing capability at imaging depths of up to 150 µm with near constant lateral resolutions of 2.1 µm. The imaging of small fluorescent beads embedded in a 3D matrix was demonstrated.

Abstract:A single multimode fiber has been applied in minimally invasive endoscopy with wavefront shaping for biological research such as brain imaging. Most of the fibers, such as step-index and graded-index multimode fibers, give rise to spatially variant blur due to limits on the numerical aperture and collection efficiency. Routines to solve this problem are based on iterative algorithms, which are often slow and computer-intense. We developed a method to synthesize datasets for driving a deep learning network to deblur and denoise the spatially variant degraded image. This approach is fast (5 ms), up to three orders of magnitude faster than the iterative way. Furthermore, our method can be applied to different types of fiber endoscopy, and two types of fiber are tested here. The performance is verified on fluorescence beads and three kinds of biological tissue sections in the experiment, demonstrating effectiveness in image enhancement.

Abstract:Spectral reconstruction method based on narrow-band measurements has been demonstrated to achieve ultrafast spectroscopic imaging with high spatial and spectral resolution, in which multiple narrow-band images are collected by using several specific filters. Although commercially available filters can be employed in such method, filters with complex transmittance that are difficult to be fabricated typically show significant improvement in spectral reconstruction accuracy. In this study, a two dimensional programmable optical filter based on digital micromirror device (DMD) is proposed, in which its transmittance spectrum can be arbitrarily and quickly switched to realize complex transmittance. Furthermore, its flexible transmittance enables directly hardware-based spectral data post-processing, which can perform data acquisition and analysis simultaneously. Those have been evaluated by the diffuse reflectance spectra from normal and occluded skin flaps, as well as Raman spectra from live, apoptosis and necrosis leukemia cells. Our simulation results show that much higher spectral reconstruction accuracies can be achieved by the optimized filters with complex transmittance. Furthermore, the classification accuracy by using the proposed method is comparable to those achieved by conventional numerical methods. Therefore, based on the proposed programmable optical filter, fast spectroscopic imaging with high spatial and spectral resolution can be achieved for observing fast changing phenomena and even real-time target identification.

Abstract:The demand for low-cost, high-performance miniaturized optical imaging systems requires creating a new imaging paradigm. In this paper, we propose an imaging paradigm that achieves diffraction-limited imaging with a non-imaging spatial information transfer lens. The spatial information transfer lens realizes a perfect match between the space-bandwidth product (SBP) of the lens and that of the image sensor so that the collected spatial information from the object can be totally recorded and fully resolved by the image sensor. A backward wave propagation model is developed to reconstruct the object by propagating the light wave modulated by the information transfer lens back from the image space to object space. The proposed imaging paradigm breaks the point-to-point imaging structure and removes the focusing-distance constraint, allowing a flexible arrangement of the object and the image sensor along the optical axis with a compact form factor of the optical system. We experimentally demonstrate the versatility and effectiveness of the proposed imaging paradigm. The proposed imaging paradigm is low-cost, simple in configuration, flexible in arrangement, and diffraction limited with great potential applications in biomedical imaging. Abstract:The contrast and resolution of images obtained with optical microscopes can be improved by deconvolution and computational fusion of multiple views of the same sample, but these methods are computationally expensive for large datasets. Here we describe theoretical and practical advances in algorithm and software design that result in image processing times that are tenfold to several thousand fold faster than with previous methods. First, we show that an 'unmatched back projector' accelerates deconvolution relative to the classic Richardson-Lucy algorithm by at least tenfold. Second, three-dimensional image-based registration with a graphics processing unit enhances processing speed 10- to 100-fold over CPU processing. Third, deep learning can provide further acceleration, particularly for deconvolution with spatially varying point spread functions. We illustrate our methods from the subcellular to millimeter spatial scale on diverse samples, including single cells, embryos and cleared tissue. Finally, we show performance enhancement on recently developed microscopes that have improved spatial resolution, including dual-view cleared-tissue light-sheet microscopes and reflective lattice light-sheet microscopes.

Abstract:A number of design options for image slicers for NGST and Gemini are being investigated. These image slicers are all of the focussing type and both refractive and reflective solutions are being explored. One such device, an image slicer that focuses 10 slices on a spectrograph slit is now in operation at the McMath Solar telescope. It consists of three lenslet arrays, and additionally acts as a focal reducer and provides correction for astigmatism of the telescope. A combined refractive and reflective slicer designed for use on NGST delivers near-diffraction limited images for up to 40 slices.

Abstract:A fundamental challenge in fluorescence microscopy is the defocused background caused by scattering light, optical aberration, or limited axial resolution. Severe defocus backgrounds will submerge the in-focus information and cause artifacts in the following processing. Here, we leverage a priori knowledge about dark channels of biological structures and dual frequency separation to develop a single-frame defocus removal algorithm. It stably improves the signal-to-background ratio and structural similarity index measure of images by approximately 10-fold, and recovers in-focus signal with 85% accuracy, even when the defocus background is 50 times larger than in-focus information. Our Dark-based optical sectioning approach (Dark sectioning) is fully compatible with various microscopy techniques, such as wide-filed microscopy, polarized microscopy, laser-scanning / spinning-disk confocal microscopy, stimulated emission depletion microscopy, lightsheet microscopy, and light-field microscopy. It also complements reconstruction or processing algorithms such as deconvolution, structure illumination microscopy, and super-resolution optical fluctuation imaging.

Abstract:Multi-order diffraction lenses can focus different wavelengths at the same focal point by utilizing various diffraction orders. However, due to material dispersion, the focal position shifts when the incident wavelength moves away from the primary wavelength, resulting in limited spectral bandwidth. This paper presents a computationally optimized multi-order diffractive lens design and a related image reconstruction algorithm, enabling broadband high-resolution imaging with a single element. We introduce a new type of lens, the Spectral Multi-Order Diffractive Lens (SMODLs), which redistributes the energy across different diffractive orders to achieve relatively high focusing performance within specified wavelength bands. Subsequently. The relatively sharp spectral images generated by SMODL within these specified bands serve as priors for reconstructing images across the full spectrum. A prototype lens with a 40 mm aperture and a 320 mm focal length was designed and fabricated. The prior images' wavebands were designated as 525 nm to 575 nm and 675 nm to 725 nm, with peak Strehl ratios of 0.47 and 0.35, respectively. Finally, grayscale images within the 500∼800 nm wavelength range were reconstructed. Experimental results confirm the effectiveness of our approach, achieving high-resolution imaging across a 300 nm bandwidth with a single diffractive element.

Abstract:We propose a three-dimensional nonlinear diffusion method to implement the similar autofocusing function of multiple micro-objects and simultaneously remove the defocused images, which can distinguish the locations of certain sized scattering particles that are overlapping along z-axis. It is applied to all of the reconstruction slices that are generated from the captured hologram after each back propagation. For certain small sized particles, the maxima of maximum gradient magnitude of each reconstruction slice appears at the ground truth z position after applying the proposed scheme when the reconstruction range along z-axis is sufficiently long and the reconstruction depth spacing is sufficiently fine. Therefore, the reconstructed image at ground truth z position is remained, while the defocused images are diffused out. The results demonstrated that the proposed scheme can diffuse out the defocused images which are 20 um away from the ground truth z position in spite of that several scattering particles with different diameters are completely overlapping along z-axis with a distance of 800 um when the hologram pixel pitch is 2 um. It also demonstrated that the sparsity distribution of the ground truth z slice cannot be affected by the sparsity distribution of corresponding defocused images when the diameter of the particle is not more than 35um and the reconstruction depth spacing is not less than 20 um.

Abstract:We combine the 3D hybrid-Weickert nonlinear diffusion regularizer with digital holography, which can distinguish the locations of certain small-sized particles that are overlapped along the z-axis, to realize autofocusing of multiple micro-objects and simultaneously remove the defocused images. The proposed method is applied to all the reconstruction slices generated from the hologram after each back-propagation. After applying the proposed scheme for certain small-sized particles, the reconstructed image at the ground truth z position is retained, the defocused images are diffused out when the reconstruction range along the z-axis is sufficiently long and the reconstruction depth spacing is sufficiently fine. The results demonstrated that the proposed scheme can diffuse out the defocused images that are 20 mu m from the ground truth z position, despite several scattering particles of different diameters being completely overlapped along the z-axis with a distance of 800 mu m when the hologram pixel pitch is 2 mu m. In addition, the results revealed that the sparsity distribution of the ground truth z slice is not affected by the sparsity distribution of the corresponding defocused images when the diameter of the particle is less than 35 mu m and the reconstruction depth spacing is more than 20 mu m.
A New Deep Learning Method for Image Deblurring in Optical Microscopic Systems

Abstract:Deconvolution is the most commonly used image processing method to remove the blur caused by the point-spread-function (PSF) in optical imaging systems. While this method has been successful in deblurring, it suffers from several disadvantages including being slow, since it takes many iterations, suboptimal, in cases where experimental operator chosen to represent PSF is not optimal. In this paper, we are proposing a deep-learning-based deblurring method applicable to optical microscopic imaging systems. We tested the proposed method in database data, simulated data, and experimental data (include 2D optical microscopic data and 3D photoacoustic microscopic data), all of which showed much improved deblurred results compared to deconvolution. To quantify the improved performance, we compared our results against several deconvolution methods. Our results are better than conventional techniques and do not require multiple iterations or pre-determined experimental operator. Our method has the advantages of simple operation, short time to compute, good deblur results and wide application in all types of optical microscopic imaging systems. The deep learning approach opens up a new path for deblurring and can be applied in various biomedical imaging fields.

Efficient Image Deblurring Networks based on Diffusion Models

Abstract:This article presents a sliding window model for defocus deblurring, named Swintormer, which achieves the best performance to date with remarkably low memory usage. This method utilizes a diffusion model to generate latent prior features, aiding in the restoration of more detailed images. Additionally, by adapting the sliding window strategy, it incorporates specialized Transformer blocks to enhance inference efficiency. The adoption of this new approach has led to a substantial reduction in Multiply-Accumulate Operations (MACs) per iteration, drastically cutting down memory requirements. In comparison to the currently leading GRL method, our Swintormer model significantly reduces the computational load that must depend on memory capacity, from 140.35 GMACs to 8.02 GMACs, while improving the Peak Signal-to-Noise Ratio (PSNR) for defocus deblurring from 27.04 dB to 27.07 dB. This innovative technique enables the processing of higher resolution images on memory-limited devices, vastly broadening potential application scenarios. Abstract:The increasing throughput of experiments in biomaterials research makes automatic techniques more and more necessary. Among all the characterization methods, microscopy makes fundamental contributions to biomaterials science where precisely focused images are the basis of related research. Although automatic focusing has been widely applied in all kinds of microscopes, defocused images can still be acquired now and then due to factors including background noises of materials and mechanical errors. Herein, we present a deep-learning-based method for the automatic sorting and reconstruction of defocused cell images. First, the defocusing problem is illustrated on a high-throughput cell microarray. Then, a comprehensive dataset of phase-contrast images captured from varied conditions containing multiple cell types, magnifications, and substrate materials is prepared to establish and test our method. We obtain high accuracy of over 0.993 on the dataset using a simple network architecture that requires less than half of the training time compared with the classical ResNetV2 architecture. Moreover, the subcellular-level reconstruction of heavily defocused cell images is achieved with another architecture. The applicability of the established workflow in practice is finally demonstrated on the high-throughput cell microarray. The intelligent workflow does not require a priori knowledge of focusing algorithms, possessing widespread application value in cell experiments concerning high-throughput or time-lapse imaging.
