Abstract:Non-metal-catalyzed hydrogenation of arenes represents a sustainable and environment-friendly approach for the synthesis of cyclohexane derivatives. Herein, we report the development of a borenium-ion catalyst system which can hydrogenate a number of monocyclic arenes. Additionally, this metal-free catalytic system allows selective partial or full hydrogenation of naphthalene and anthracene by modification of the reaction conditions. Our mechanism studies suggest that the borenium-arene-mediated H2 splitting takes place before the rate-limiting step and the hydride transfer between the resulting neutral borane and protonated arene is likely the rate-limiting step in the catalytic process.

Abstract:We herein report the iridium-catalyzed enantioselective C-H borylation of aryl chlorides. A variety of prochiral biaryl compounds could be well-tolerated, affording a vast array of axially chiral biaryls with high enantioselectivities. The current method exhibits a high turnover number (TON) of 7000, which represents the highest in functional-group-directed asymmetric C-H activation. The high TON was attributed to a weak catalyst-substrate interaction that was caused by mismatched chirality between catalyst and substrate. We also demonstrated the synthetic application of the current method by C-B, ortho-C-H, and C-Cl bond functionalization, including programmed Suzuki-Miyaura coupling for the synthesis of axially chiral polyarenes.

Abstract:The regioselective derivatization of the monocarba-closo-dodecaborate anion via catalytic B-H bond activation is reported. Amide directing groups in combination with rhodium and iridium catalysts allowed for the direct functionalization of cage boron vertices. Products comprising B-C, B-N and B-Cl bonds were synthesized. As a key intermediate of the B-H activation step, an iridium complex with a direct B-Ir interaction was isolated and fully characterized by spectroscopic methods as well as X-ray crystallography.

Abstract:Exploration of new organometallic systems based on polyhedral boron clusters has the potential to solve challenging chemical problems such as the stabilization of reactive intermediates and transition-state-like species postulated for E-H (E = H, B, C, Si) bond activation reactions. We report on facile and clean B-H activation of a hydroborane by a new iridium boron cluster complex. The product of this reaction is an unprecedented and fully characterized transition metal-stabilized boron cation or borenium. Moreover, this intermediate bears an unusual intramolecular B···H interaction between the hydrogen originating from the activated hydroborane and the cyclometallated metal-bonded boron atom of the boron cluster. This B···H interaction is proposed to be an arrested insertion of hydrogen into the Bcage-metal bond and the initiation step for iridium "cage-walking" around the upper surface of the boron cluster. The "cage-walking" process is supported by the hydrogen-deuterium exchange observed at the boron cluster, and a mechanism is proposed on the basis of theoretical methods with a special focus on the role of noncovalent interactions. All new compounds were isolated and fully characterized by NMR spectroscopy and elemental analysis. Key compounds were studied by single crystal X-ray diffraction and X-ray photoelectron spectroscopy.

Abstract:Organoboron reagents are important synthetic intermediates that have a key role in the construction of natural products, pharmaceuticals and organic materials1. The discovery of simpler, milder and more efficient approaches to organoborons can open additional routes to diverse substances2-5. Here we show a general method for the directed C-H borylation of arenes and heteroarenes without the use of metal catalysts. C7- and C4-borylated indoles are produced by a mild approach that is compatible with a broad range of functional groups. The mechanism, which is established by density functional theory calculations, involves BBr3 acting as both a reagent and a catalyst. The potential utility of this strategy is highlighted by the downstream transformation of the formed boron species into natural products and drug scaffolds.

Abstract:We report a method to conduct one-pot meta cyanation of arenes by iridium-catalyzed C-H borylation and copper-mediated cyanation of the resulting arylboronate esters. This process relies on a method to conduct the cyanation of arylboronic esters, and conditions for this new transformation are reported. Conditions for the copper-mediated cyanation of arylboronic acids are also reported. By the resulting sequence of borylation and cyanation, 1,3-disubstituted and 1,2,3-trisubstituted arenes and heteroarenes containing halide, ketone, ester, amide, and protected alcohol functionalities are converted to the corresponding meta-substituted aryl nitriles. The utility of this methodology is demonstrated through the conversion of a protected 2,6-disubstituted phenol to 4-cyano-2,6-dimethylphenol, which is an intermediate in the synthesis of the pharmaceutical etravirine. The utility of the method is further demonstrated by the conversion of 3-chloro-5-methylbenzonitrile, produced through the one-pot C-H borylation and cyanation sequence, to the corresponding 3,5-disubstituted aldehydes, ketones, amides, carboxylic acids, tetrazoles, and benzylamines.

Abstract:Recent years have witnessed the growing interest in the remote functionalization of alkenes for it offers a strategy to activate the challenging C-H bonds distant from the initiation point via alkene isomerization/functionalization. However, the catalytic enantioselective isomerization/functionalization with one single transition metal catalyst remains rare. Here we report a highly regio- and enantioselective cobalt-catalyzed remote C-H bond borylation of internal alkenes via sequential alkene isomerization/hydroboration. A chiral ligand featured twisted pincer, anionic, and non-rigid characters is designed and used for this transformation. This methodology, which is operationally simple using low catalyst loading without additional activator, shows excellent enantioselectivity and can be used to convert various internal alkenes with regio- and stereoisomers to valuable chiral secondary organoboronates with good functional group tolerance.

Abstract:1,2,3-Trisubstituted closo-dodecaborates with B-O, B-N, and B-C bonds as well as a fused borane oxazole ring have been synthesized by rhodium-catalyzed direct cage B-H alkenylation and annulation of ureido boranes in the first reported example of regioselective B-H bond functionalization of the [B12H12](2-) cage by transition-metal catalysis. This reaction proceeded at room temperature under ambient conditions and exhibited excellent selectivity for efficient monoalkenylation with good functional-group tolerance. The urea moiety enabled B-H activation by acting as a directing group, was incorporated in the oxazole ring in situ, and also avoided multiple alkenylation. A possible mechanism is proposed on the basis of the isolation of a rhodium agostic intermediate and control experiments.

Abstract:An ideal way to synthesize multi-substituted arenes is the selective installation of any group on any position of aromatic rings with any substituent. However, reactant-controlled selective functionalization of both meta- and para-C–H bonds is essentially impossible because of their intrinsically contradictory electronic and steric demands. Here, we report the first examples of catalysts that can direct the tunable borylation of a variety of arenes with precise control at either meta or para site through the computational design of ligands. A wide range of hydrogen bond acceptors can serve as directing groups, including Weinreb amides, phosphonates, and boronates, which can be easily transformed into many different types of functional groups. This work also showcases the critical role of transition-state calculations in catalyst design for remote C–H activation.

Abstract:The metal-catalyzed C-H borylation reaction is a robust and valuable method for the installation of boronate esters into simple organic substrates. The regioselectivity observed in early examples was governed by steric control; those systems were extended to include a number of approaches that override the natural selectivity to obtain directed C-H borylation. In spite of the array of catalysts and directing groups that are now known to achieve directed reactions, no comprehensive experimental and computational study of the mechanism has been reported to date. In this study, experimental and computational results have been used to provide a detailed study of the catalytic mechanism of amine-directed C(sp(2))-H borylation. A notable result of the present study is the absence of inter- or intramolecular kinetic isotope effects at the functionalized C-H bond. The kinetic and computational data support a rate-determining reassociation of pinacolborane from the iridium catalyst to allow carbon-boron bond formation. An additional feature of note is the role of the boron source in the reaction. Computational analysis revealed the anticipated role of pinacolborane generated in the catalytic cycle. Based on this analysis, pinacolborane was examined as an additive, which overcame an induction period and provided an overall rate enhancement. Pinacolborane was found to serve as an autocatalyst in the transformation, a feature that can be utilized to improve the reactivity in directed C-H borylation reactions. The experimentally and computationally determined free energies of activation for the overall reaction are in agreement and provide valuable insights into substrate-directed C-H borylation reactions.

Abstract:Herein, we report a boron-centered arene extrusion reaction to afford putative cyclic(alkyl)(amino) carbene (CAAC)-ligated chloroborylene and bromoborylene. The borylene precursors, chloro-boranorbornadiene (ClB(C 6 Me 6 ), 2 Cl ) and bromo-boranorbornadiene (BrB(C 6 Me 6 ), 2 Br ) were synthesized through the reaction of the corresponding 1-halo-2,3,4,5-tetramethylborole dimer (XBC 4 Me 4 ) 2 (X = Cl, 1 Cl ; X = Br, 1 Br ) with 2-butyne. Treatment of 2 Cl with CAACs resulted in the release of di-coordinate chloro-borylene (CAAC)BCl from hexamethylbenzene (C 6 Me 6 ) at room temperature. In contrast, the reaction of 2 Br with CAAC led to the formation of a boronium species [(CAAC)BC 6 Me 6 ] + Br – ( 7 ) at room temperature. Heating 7 in toluene promoted the release of di-coordinate bromo-borylene (CAAC)BBr as a transient species. Surprisingly, heating 7 in dichloromethane resulted in the C–H activation of hexamethylbenzene. The conversion of a CAAC-stabilized bromo-borepin to a borylene, a boron-centered retro Büchner reaction, was also investigated.

Abstract:Nickel-catalyzed C-H bond functionalization reactions provide an impressive alternative to those with noble metal catalysts due to their unique reactivity and low cost. However, the regioselective C(sp2)-H borylation reaction of arenes accomplished by nickel catalyst remains limited. We herein disclose a silyl-directed ortho C(sp2)-H borylation of substituted arenes with a Ni(cod)2/PMe3/KHMDS catalyst system. Using readily available starting materials, this protocol provides easy access to ortho-borylated benzylic hydrosilanes bearing flexible substitution patterns. These products can serve as versatile building blocks for the synthesis of sila or sila/borine heterocycles under mild conditions. Control experiments and DFT calculations suggest that a catalytic amount of base prompts the formation of Ni(II)-Bpin-ate complex, likely related to the C(sp2)-H bond activation. This borylation reaction might follow an unusual Ni(II)/Ni(IV) catalytic cycle.

Abstract:<p>Site‐selective borylation of simple arenes was realized in one pot via an electrophilic thianthrenation/Pd‐catalyzed borylation sequence. The key to achieve this operatically simple process is the use of Pd catalysis which could tolerate the solvent and acidic conditions used in the thianthrenation step. Abstract:Due to boron's metalloid properties, aromatic boron reagents are prevalent synthetic intermediates. The direct borylation of aryl C-H bonds for producing aromatic boron compounds offers an appealing, one-step solution. Despite significant advances in this field, achieving regioselective aryl C-H bond borylation using simple and readily available starting materials still remains a challenge. In this work, we attempted to enhance the reactivity of the electron-donor-acceptor (EDA) complex by selecting different bases to replace the organic base (NEt3) used in our previous research. To our delight, when using NH4HCO3 as the base, we have achieved a mild visible-light-mediated aromatic C-H bond borylation reaction with exceptional regioselectivity (rr > 40:1 to single isomers). Compared with our previous borylation methodologies, this protocol provides a more efficient and broader scope for aryl C-H bond borylation through the use of N-Bromosuccinimide. The protocol's good functional-group tolerance and excellent regioselectivity enable the functionalization of a variety of biologically relevant compounds and novel cascade transformations. Mechanistic experiments and theoretical calculations conducted in this study have indicated that, for certain arenes, the aryl C-H bond borylation might proceed through a new reaction mechanism, which involves the formation of a novel transient EDA complex.

Abstract:Organoborons have emerged as versatile building blocks in organic synthesis to achieve molecular diversity and as carboxylic acid bioisosteres with broad applicability in drug discovery. Traditionally, these compounds are prepared by the substitution of Grignard/lithium reagents with electrophilic boron species and Brown hydroboration. Recent developments have provided new routes for the efficient preparation of organoborons by applying reactions using chemical feedstocks with leaving groups. As compared to the previous methods that used organic halides (I, Br, and Cl), the direct borylation of less reactive C-Het and C-C bonds has become highly important to get efficiency and functional-group compatibility. This Review aims to provide a comprehensive overview of this topic, including (1) C-F bond borylation, (2) C-O bond borylation, (3) C-S bond borylation, (4) C-N bond borylation, and (5) C-C bond borylation. Considerable attention is given to the strategies and mechanisms involved. We expect that this Review will inspire chemists to discover more efficient transformations to expand this field.
