Abstract:An efficient 3d metal-catalyzed reductive amination is present. Carbonyl compounds, amines and molecular hydrogen are employed as the substrate to produce the secondary or tertiary amines. The skeleton ligand EDTA collaborates with a promotor HBF4 and a hydrogen shuttle isopropanol to eventually afford efficient hydroge-nation of the imine intermediates. The mechanisms have been revealed using a combination of control experi-ments and DFT calculations. Thus, the role of each component of the catalytic system has been addressed at a molecular level. Our work may be instructive to the design of hydrogen transfer catalysts.

Abstract:Ruthenium-catalyzed sigma-bond activation-assisted meta-C-H functionalization has emerged as a useful tool to forge distal C-C bonds. However, given the limited number of mechanistic studies, a clear understanding of the origin of the site-selectivity and the complete reaction pattern is not available. Here, we present systematic computational studies on ruthenium-catalyzed C-H functionalization with primary, secondary, tertiary alkyl bromides and aryl bromides. The C-H scission and the C-C formation were carefully examined. Monocyclometalated ruthenium(II) complexes were identified as the active species, which then underwent inner-sphere single electron transfer (ISET) to activate the organic bromides. The site-selectivity results from the competition between the close-shell reductive elimination and the open-shell radical coupling. Based on this mechanistic understanding, a multilinear regression model was built to predict the site-selectivity, which was further validated by experiments.

Abstract:An efficient approach for the remote C-H alkylation of arenes, employing a variety of N-directing groups is described. This method facilitates the straightforward synthesis of valuable phenylethylamine derivatives by exclusively cleaving the benzylic C-N bond in aziridines. Furthermore, these products can easily remove the protecting groups, resulting in a variety of meta-substituted compounds, such as amines and ketones, which hold significance in synthetic chemistry.

Abstract:AbstractThe prevalence ofC‐aryl glycosides in biologically active natural products and approved drugs has long motivated the development of efficient strategies for their selective synthesis. Cross‐couplings have been frequently used, but largely relied on palladium catalyst with prefunctionalized substrates, while ruthenium‐catalyzedC‐aryl glycoside preparation has thus far proven elusive. Herein, we disclose a versatile ruthenium(II)‐catalyzedmeta‐C−H glycosylation to accessmeta‐C‐aryl glycosides from readily available glycosyl halide donors. The robustness of the ruthenium catalysis was reflected by mild reaction conditions, outstanding levels of anomeric selectivity and exclusivemeta‐site‐selectivity.

Abstract:A ruthenium‐catalyzed C(sp2)–H acyloxylation of 2‐aroyl pyridine derivatives with simple sodium carboxylate utilizing transformable directing groups is described. This protocol features broad functional group tolerance and chemo‐ and regio‐selectivity, providing the acyloxylation products in 45%‐84%yield. Furthermore, the synthetic utility of this protocol was demonstrated by the late‐stage functionalization of pharmaceutical compounds. Notably, the acyloxylation products could be further transformed into a variety of useful heterocycles under mild conditions.

Abstract:Asymmetric C−H activation has surfaced as a transformative toolbox for the efficient assembly of chiral molecules. However, despite of major advances in rhodium and palladium catalysis, ruthenium(II)‐catalyzed asymmetric C−H activation has thus far largely proven elusive. In contrast, we herein report on the unprecedented ruthenium(II)‐catalyzed highly regio‐, diastereo‐ and enantioselective C–H alkylation. The key to success was proved by the identification of C2‐symmetric chiral imidazolidine carboxylic acids (CICAs), which can be easily prepared in a one‐pot fashion. This ruthenium/CICA system enabled the efficient installation of central and axial chirality, and featured excellent branched to linear ratios with generally >20:1 dr and up to 98:2 er. Mechanistic studies by experiment and computation were carried out to unravel the catalyst mode of action.

An N-amino (i.e., hydrazine) group is used for the directed C-H functionalization with either an alkyne or an alkene, affording an indole derivative or olefination product. The synthesis features a broad substrate scope, superior atom and step economy, as well as mild reaction conditions.

Abstract:Free radical cyclization has emerged as one of the most important reaction types, which is widely used in natural product synthesis, pharmaceutical chemistry, and materials science. This report described the combination of radical cyclization and ruthenium-catalyzed meta-selective C–H functionalization for the synthesis of arylpyrrolidone derivatives. This method exhibited the highly meta-site selectivity of the primary, secondary, and tertiary alkyl radicals formed by intramolecular addition. A wide spectrum of directing groups bearing diversified N-heterocycles performed well, including biologically active molecules. Abstract:Recent years have witnessed the growing interest in the remote functionalization of alkenes for it offers a strategy to activate the challenging C-H bonds distant from the initiation point via alkene isomerization/functionalization. However, the catalytic enantioselective isomerization/functionalization with one single transition metal catalyst remains rare. Here we report a highly regio- and enantioselective cobalt-catalyzed remote C-H bond borylation of internal alkenes via sequential alkene isomerization/hydroboration. A chiral ligand featured twisted pincer, anionic, and non-rigid characters is designed and used for this transformation. This methodology, which is operationally simple using low catalyst loading without additional activator, shows excellent enantioselectivity and can be used to convert various internal alkenes with regio- and stereoisomers to valuable chiral secondary organoboronates with good functional group tolerance.

Among the most prominent transition metals for C–H activations, ruthenium offers several salient features, including cost-effective, robust, and mild C–H activations with unique reactivities for remote functionalization. Indeed, ruthenium(II) catalysis by carboxylate assistance offers a highly functional group tolerant and predictable toolbox for directed as well as nondirected C–H functionalizations, with a major impact to improve the sustainability in industrial settings. These assets have allowed the use of nontoxic, nonflammable, and cost-effective H2O as a reaction medium for sustainable ruthenium(II)-catalyzed C–H activations. In this perspective, we summarize the potential of water for sustainable ruthenium(II)-catalyzed C–H activations up to February 2022.

Abstract:A ruthenium-catalyzed electrochemical dehydrogenative C(sp2)−H acyloxylation of aniline derivatives with carboxylic acids has been developed. Electric current is used to recycle the active ruthenium-based catalyst and promote H2 evolution. This method significantly expands the scope and enhances the selectivity of metal-electron catalyzed mono- and di-acyloxylations, which remain challenged. Our findings allowed mono-selectivity in broadly effective late-stage diversification of structurally complex drugs and natural product molecules, tolerating drugs, natural products and amino acids. The method and its underlying strategy are expected to have widespread uses in functional aniline synthesis and drug discovery process.

Abstract:Ruthenium-catalyzed arylation of ortho C-H bonds directed by a bidentate 8-aminoquinoline moiety not only is important to construct new biaryl derivates but also merges important research areas. In this study, the density functional theory (DFT) method M11-L was employed to predict the mechanism of this C-H bond arylation reaction. The computational results indicate that the initial step for this reaction is catalyst loading by electrophilic deprotonation to generate a substrate-coordinated Ru(II) intermediate, which is the key compound in the complete catalytic cycle. The catalytic cycle includes electrophilic deprotonation by carbonate, oxidative addition of bromobenzene, reductive elimination to form a new aryl aryl bond, proton transfer to release the product, and ligand exchange to regenerate the initial Ru(II) intermediate. Theoretical calculations suggest that the oxidative addition of bromobenzene is the rate determining step of the whole catalytic cycle, and the apparent activation free energy is 32.7 kcal/mol. The ligand effect was considered in DFT calculations, and the calculated results agree well with experimental observations.

Abstract:C(sp2)–H functionalization offers an efficient strategy for the synthesis of various elaborated N-containing heteroarenes. Along these lines, oxazino pyridines that can be readily prepared from pyridines, have been introduced as powerful substrates in radical- and ionic-mediated meta-C–H functionalization. However, the regioselective meta-C–H arylation of pyridines remains a great challenge. Herein, a copper-catalyzed meta-selective C–H arylation of pyridines and isoquinolines through bench-stable dearomatized intermediates is reported. Electrophilic aryl-Cu(III) species, generated from readily accessible aryl I(III) reagents, enable the efficient meta-arylation of a broad range of pyridines and isoquinolines. The method also allows the meta-selective alkenylation of these heteroarenes using the corresponding alkenyl I(III)-reagents. Late-stage arylation of drug-derived pyridines and larger-scale experiments demonstrate the potential of this synthetic methodology.

Abstract:The past decades have witnessed rapid development in organic synthesis via catalysis, particularly the reactions through C–H bond functionalization. Transition metals such as Pd, Rh and Ru constitute a crucial catalyst in these C–H bond functionalization reactions. This process is highly attractive not only because it saves reaction time and reduces waste,but also, more importantly, it allows the reaction to be performed in a highly region specific manner. Indeed, several organic compounds could be readily accessed via C–H bond functionalization with transition metals. In the recent past, tremendous progress has been made on C–H bond functionalization via ruthenium catalysis, including less expensive but more stable ruthenium(II) catalysts. The ruthenium-catalysed C–H bond functionalization, viz. arylation, alkenylation, annulation, oxygenation, and halogenation involving C–C, C–O, C–N, and C–X bond forming reactions, has been described and presented in numerous reviews. This review discusses the recent development of C–H bond functionalization with various ruthenium-based catalysts. The first section of the review presents arylation reactions covering arylation directed by N–Heteroaryl groups, oxidative arylation, dehydrative arylation and arylation involving decarboxylative and sp3-C–H bond functionalization. Subsequently, the ruthenium-catalysed alkenylation, alkylation, allylation including oxidative alkenylation and meta-selective C–H bond alkylation has been presented. Finally, the oxidative annulation of various arenes with alkynes involving C–H/O–H or C–H/N–H bond cleavage reactions has been discussed.
